Pařízek Author-Workplace-Name: Agricultural Research Institute Kroměříž, Ltd., Kroměříž, Czech Republic Title: Resistance of Spring Barley Varieties to Powdery Mildew in the Czech Republicin 1971-2000 Abstract: Resistance to powdery mildew of 127 spring barley varieties was evaluated in 702 official variety trials, using scores from 1 to 9. Trials with sufficient disease severity were only analysed. Varieties possessing the resistance genes Mla7 (Elgina), Ml(Kr) (BR-1519), Mla13 (Koral) and mlo (Forum) were among the most resistant ones. The varieties Diamant, HE-3527, HE-3631, II/61-FUDII and Zefir showed high susceptibility. Fifteen varieties, carrying the genes Mla1, Mla3, Mla6, Mla9, Mla13, Ml(Kr) and Ml(Sc), were in the first years of testing highly resistant, but became susceptible later. The score of the most resistant variety ranged each year from 8.05 to 9.00, only in 1987-1991 it was lower. Since the pathogen population rapidly adapted to most resistance genes in the mid eighties, no resistant variety was found in 1987-1989. From 1986 to 1995 on average only 6.6% of the tested varieties were resistant, but there was no variety with resistance score above 8.50. High resistance was typical for varieties possessing an effective major resistance gene. Since the resistance of such varieties was not durable, we recommend breeding of varieties with at least two fully effective resistance genes, using molecular markers. Milotová Author-Workplace-Name: Mendel University of Agriculture and Forestry in Brno, Brno, Czech Republic Title: Use of Prolamin Polymorphism to Describe Genetic Variation in a Collection of Barley Genetic Resources Abstract: The polymorphism of prolamin storage proteins was studied in seed samples of 20 historical cultivars of spring barley (Hordeum vulgare L.) of Czech and Slovak origin, using polyacrylamide gel electrophoresis (PAGE). Only two samples were uniform. Most heterogeneity of prolamin patterns was observed in the oldest accessions. By means of a prolamin identity index it was possible to distinguish sister lines from admixtures within the seed samples. The obtained spectra will be used as additional descriptors for the spring barley core collection of the Collection of Genetic Resources of the Agricultural Research Institute Kroměříž, Ltd. The highest frequency of the HMW-GS 0, 7 + 9 and 5 + 10 was found among the winter wheat cultivars. The highest GQ score of 10 was found in the cultivars SK-30 and FD-92017/1. A GQ score of 9 was found in the cultivars Astella, Ilona, Velta and MV-06-95. The HMW-GS 0 and 7 + 8, associated with a GQ of 4, were found in T. durum. The electrophoretic spectra of the spelt wheat cultivars were not homogeneous and represented 4 different lines. Van De Wiel Author-Workplace-Name: Department of Botany, Faculty of Science, Palacký University, Olomouc-Holice, Czech Republic Title: Relationships among Morphological Characters, Isozymes Polymorphism and DNA Variability - the Impact on Lactuca Germplasm Taxonomy Abstract: Fifty one accessions of nineteen Lactuca species, the hybrid L. serriola × L. sativa and the related species Mycelis muralis were evaluated for morphological variability, esterase (EST) polymorphism, Amplified Fragment Length Polymorphism (AFLP) and relative DNA content. Sixteen Lactuca accessions were classified taxonomically on the basis of morphology, isozyme analysis and AFLP. Twenty-eight bands (isoforms) of EST were recorded allowing 82% of accessions to be distinguished. The relative DNA content, measured using flow-cytometry (DAPI staining), ranged from 2.02 pg in L. capensis to 17.96 pg in L. canadensis. The results from AFLP analysis and the relative DNA content measurement corresponded well with recent taxonomic classification of the genus Lactuca.
