Martinek Author-Workplace-Name: Czech Republic Title: DNA Markers for High Molecular weight Glutenin Subunits 5+10 Used in Wheat and Triticale Breeding Abstract: Prediction of flour breadmaking quality was verified using DNA markers in seven genotypes of winter wheat (T. aestivum L., 2n = 6x = 42, AABBDD) of different quality classes, four genotypes of triticale (× Triticosecale Wittmack, 2n = 6x = 42, AABBRR), and selected progenies of triticale Presto with the T1R.1D5+10-2 translocation. DNA isolated from fresh leaves (the stage of the first true leaf) was used to detect the Glu D1 5+10 allele based on the SPLAT protocol according to D'Ovidio and Anderson (1994). The presence of the Glu D1 5+10 allele was verified using a product of 450 bp size. It was detected in the wheat genotypes Athlet, Brea, Bruneta, Iris, Lívia, Mona, Sida, and in all analysed progenies derived from the Presto triticale sample with the T1R.1D5+10-2 translocation. Effects are discussed of other loci on the final breadmaking quality in the wheat varieties Athlet, Lívia, and Mona with Glu D1 5+10 and a lower grain breadmaking quality. Debre Author-Workplace-Name: Gene Bank of the Slovak Republic, Research Institute of Plant Production, Piešťany, Slovak Republic Title: Diversity of Common Bean Landraces Collected in the Western and Eastern Carpatien Abstract: The study of diversity in common bean was based on morphological and agronomical characteristics, differentiation of collected accessions by morphological and molecular markers, detection of genetic variation, and duplicates detection in bean landraces. The analysed 82 accessions of common bean (Phaseolus vulgaris L.) were collected in the Western andEastern Carpatien as landrace mixtures. Their seeds were segregated and pooled according to their characteristics; they were further multiplicated, and introduced into the collection. An extensive variation in plant and seed traits was discovered in thirty-three morphological and agronomical characteristics. Nevertheless, some of the accessions were identical in these characteristics. Cluster analysis grouped genotypes into two main branches, reflecting the growth type, seed size parameters, and thousand-seed weight. Molecular differentiation studies were performed by multilocus polymorphism detection in microsatellite and minisatellite DNA regions. Cluster analysis based on molecular data also grouped genotypes but no linkage to morphological traits was revealed. Bean accessions with very similar or identical morphological characters were clearly distinguished by DNA banding patterns. The presence of duplicates was excluded. Abstract: In the case of poor germination of seed samples and minimal number of seedlings obtained, in vitro methods can be used to revitalise and recover the gene resource. The highest germination of meadow vetchling (Lathyrus pratensis L.) seeds was achieved after scarification with H2SO4 and cultivation in MS medium. The seedlings were used as a material for micropropagation. Regeneration passed through nodal segments cultivated on basal MS medium solidified with a combination of agar and phytagel. This culture medium was also suitable for the plant maintenance. An addition of cytokinin to the induction medium did not support multiplication and growth. In the basal MS medium rooted 72.5% (gene resource 62) or 42.5% (gene resource 28) of shoots. The rooting of gene resource 28 was increased to 63% by the addition of indolylbutyric acid to the culture medium. The regenerated plants were successfully transferred to the soil. This protocol can be used to rescue gene resources of this species. Šíp Author-Workplace-Name: Varieties Registered in the Czech Republic Title: Detection of Norin 10 dwarfing genes in winter wheat varieties registered in the Czech Republic Abstract: Insensitivity to the applied gibberellic acid, indicating the presence of Norin 10 dwarfing genes, was detected in 20 of 57 (35.1%) winter wheat varieties registered in the Czech Republic in 2002. Hybrid analyses in F2 generation showed a prevalent occurrence of Rht2 gene (in 14 varieties). Only 6 varieties were found to carry Rht1 gene. Among the recently developed Rht2 varieties, Clever possesses also a high bread-making quality which was not observed with the older materials. Rht2 varieties were found, on average, less winter-hardy, later in heading and more resistant to lodging than Rht1 varieties.
