China Author-Name: LI Xing Author-Name: Daqun LIU Title: Molecular identification of wheat leaf rust resistance genes in sixty Chinese wheat cultivars Abstract: Common wheat (Triticum aestivum L.) is the major crop cultivated in Xinjiang and Anhui provinces of China. The climate in these two provinces is favourable for wheat leaf rust (Puccinia triticina) (Pt) infection. Here, we demonstrate a detailed investigation on the leaf rust resistance of 60 major wheat cultivars cultivated in these two regions. A mixture of high virulent Pt races (THTT, THTS, THTQ and PHPS) were used to phenotype all the collected wheat cultivars at an adult plant stage. Phenotypic disease severity (FDS) and the area under the disease progress curve (AUDPC) for each of these wheat cultivars were calculated. Among all the tested wheat cultivars, three cultivars (Xindong20, Xindong 29 and 99AR142-1) with the lowest FDS and AUDPC may carry major resistance genes. Twenty-seven cultivars (45% of the total tested ones) showed a relatively lower resistance with an average of 12.52% FDS and 126.3 AUDPC. Minor resistance or slow rusting genes may be present in this group of cultivars. Molecular markers for leaf rust resistance genes Lr1, Lr9, Lr19, Lr24, Lr26 and Lr34 were further used for the genotypic screening. Lr1, Lr19, Lr26 and Lr34 were detected in 19 (31.7%), 1 (1.7%), 12 (20%) and 6 (10%) wheat cultivars, respectively. Neither Lr9 nor Lr24 could be detected in any of the tested cultivars. These results will greatly improve wheat molecular breeding for leaf rust resistance in these areas. Abstract: Although haploids were successfully produced via irradiated pollen technique and anther culture in Cucurbita maxima and Cucurbita moschata, the haploidization efficiency is still low due to genotype dependence. Thus, as an alternative technique, the efficacy of the ovule culture was investigated. Ovules were extracted at different flowering time and then cultured on a solid MS medium supplemented with 2,4-dichlorophenoxyacetic acid (2,4-D), benzylaminopurine (BAP), thidiazuron (TDZ), and naphthaleneacetic acid (NAA) to induce callogenesis and plant regeneration. The gynogenic response was influenced by the combination of plant growth regulators, genotype and culture time. The medium containing of 4.0 mg/l BAP + 0.05 mg/l NAA + 0.1 mg/l TDZ provided the highest response at anthesis time. Plantlets were rooted and elongated on a solid MS medium supplemented with 0.01 mg/l indole-3-acetic acid (IAA) + 1.0 mg/l BAP. The ploidy observations of 122 plants revealed that 70 plants were haploid, 46 plants were diploid and the others were mixoploid. In total, 48 693 unigenes with an average length of 703.34 bp were obtained by de novo assembly. 3392 potential EST-SSRs (expressed sequence tag-simple sequence repeat) were identified as potential molecular markers from unigenes with lengths exceeding 1 kb. A total of 80 pairs of PCR primers were randomly selected to validate the assembly quality and develop EST-SSR markers from genomic DNA. Of these primer pairs, 14 primer pairs successfully amplified DNA fragments and detected significant amounts of polymorphism within the lacquer tree population in Langao, Shaanxi province, China. There were high genetic diversities (number of alleles per locus (A) = 2.93, polymorphic information content (PIC) = 0.53, observed heterozygosity (Ho) = 0.62 and expected heterozygosity (He) = 0.85) in the lacquer tree natural population. The four loci were significantly deviated from Hardy-Weinberg equilibrium. These results suggested high homozygosity in the population and low or deficiency in heterozygosity (inbreeding coefficient (Fis) = 0.27). These polymorphic EST-SSR markers will provide the base for further studies of genetic structure and breeding in T. vernicifluum. China Author-Name: Yang HAN Title: Lignin biosynthesis regulated by the antisense 4CL gene in alfalfa Abstract: The Antisense 4CL gene was transfected into alfalfa through Agrobacterium-mediated transfer. The test results indicated that the antisense 4CL gene was successfully integrated into the genome DNA of alfalfa and was stably transmitted to the offspring. Compared to the wild-type plants, the lignin content of T0 and T1 generation plants was reduced by 45.77% and 31.97%, respectively; there were no significant differences in height and weight of T0 and T1 plants, compared to the wild-type plants. However, the transgenic plant differed from the wild-type plant by softer stems and leaves, larger leaves, fewer flowers and a fewer seeds. The T0 line was susceptible to disease infection, but significantly improved in the second year. The results suggest that the 4CL gene from Amorpha fruticosa can be used to regulate lignin biosynthesis in transgenic forage crops. PCR-based markers have been developed for numerous traits in different plant species including several genes conferring resistance to viruses in potato. In the present work, rapid and reliable approaches were developed for the simultaneous detection of Ryadg and Ry-fsto, Ns, and PLRV.1 genes conferring resistance to Potato virus Y, Potato virus S and Potato leafroll virus, respectively, on the basis of previously published and newly modified markers. The sequence characterized amplified region (SCAR) markers for Ryadg, Ns and PLRV1 and the newly modified cleaved amplified polymorphic sequences (CAPS) marker for Ry-fsto were amplified in one PCR reaction which could simply characterize Ryadg and PLRV.1 resistance. Additional digestion of amplicons with EcoRV and MfeI for genotyping the Ry-fsto and Ns resistance genes, respectively, was needed. The effectiveness of genotyping in triplex and tetraplex PCRs was tested on 35 potato varieties used for potato seed production and breeding programs. For those reasons, apricot production and consumption are lower than in other fruit species. This study was conducted for the purpose of breeding new late ripening apricot varieties. To this aim, a total of 3718 hybrid genotypes of apricots were obtained from 42 crosses. Phenological, pomological and yield analyses were performed and variations of fruit development period, fruit size, polarimetric dry matter (Brix), total acidity, yield and other traits were determined. Two years of results for important traits are given for 12 promising genotypes. Fruit development period varied between 148 and 167 days, fruit weight 25.2 and 41.2 g, Brix 16.3 and 22.6% and total acidity 0.56 and 1.25%.
