Sauerwein Author-Workplace-Name: Institute of Animal Science, Physiology and Hygiene Group, University of Bonn, Bonn, Germany Title: Qualitative detection of haptoglobin mRNA in bovine and human blood leukocytes and bovine milk somatic cells Abstract: Local expression of haptoglobin (Hp) mRNA in tissue homogenates from different sites of the bovine mammary gland has recently been established. Besides alveolar and stromal tissue, blood cells are one potential cellular location of Hp expression. The aim of this study was to investigate whether bovine leukocytes might contribute to the Hp mRNA transcripts found in the mammary gland after their migration into this organ. For this purpose RT-PCR was carried out with total RNA extracted from leukocytes in blood and from somatic cells in milk of three dairy cows. In comparison, human leukocytes of four donors were examined. Both bovine blood and milk somatic cells showed distinct signals for Hp mRNA in all samples in contrast to human leukocytes where Hp mRNA was detected in only two out of four donors after reamplification. In conclusion, the presence of Hp mRNA in blood derived leukocytes indicates that these cells contribute to the Hp mRNA transcripts found in the bovine mammary gland. It remains to be clarified whether other cells within the mammary gland are also capable of Hp mRNA synthesis. The results were obtained by the determination of the iodine content in leg muscles and by the relationship between those concentrations and the iodine intake by the human population. The iodine content was assessed by the Sandell-Kolthoff method in 108 samples of leg muscles (m. gracilis) of market pigs from 18 herds in 10 districts of the Czech Republic collected during the period April 2004 to August 2004. Average iodine content in leg muscles of market pigs was 25.6 ± 15.54 μg I/kg fresh matter, median 20.2 and coefficient of variation 60.6%. Significantly higher (P < 0.05 to P < 0.001) iodine levels were recorded in the leg muscles was of herds LI2, ZU, BR, FU, BU, ST and some others. Iodine level variations in the samples from respective farms were expressed in the levels range of 8.5 to 66.2 μg I/kg. The detected variations might have been caused by different iodine saturation of the pigs from different herds, their physiological requirement, manifestation of physiological ability of respective animals to utilize the iodine source, potential effect of goitrogens and environmental conditions. It is necessary to consider the iodine content in pork in the balance of the iodine supply in the shopping basket of consumers. Provided that the average annual consumption of pork is 40.9 kg with iodine content of 25.6 μg/kg(8.5 to 66.2 μg/kg), the average annual iodine intake is 1047 μg, which represents 1.4 to 2.4% of the required intake per person per year. Using the AGP test used for serotyping, serotype A was found to be the predominant serotype, only one strain was serotyped as serotype B. By RAPD assay, the tested ORT strains were found to have different RAPD profiles. In addition, the RAPD assay showed almost similar DNA profiles among the tested strains of the serotypes A, B, D and E. The strain of serotype C did give a different RAPD profile. Within strains of the same serotype (A), different profiles were found but the strain of serotype (B) had an identical profile as strains of serotype A. This study suggests that more genotypes of ORT strains are present within the same serotype and thus that no relationship exists between the RAPD pattern of ORT and their serotype. Although cytotoxic and genotoxic activity of ZEA was detected, the majority of its toxic influence is related to the ability of binding to estrogen receptors and disrupting the endocrine regulation of the reproductory functions in females. It was previously found that ZEA inhibits proliferation of cells in porcine ovaries, as detected with immunostaining for proliferating cells nuclear antigen (PCNA). The number of PCNA-positive cells was inversely proportional to the dose of ZEA. We decided to answer the question of whether ZEA induces apoptosis in porcine ovaries. Experimental gilts (before first estrus) were fed ZEA in a dosage of 20 (group II) or 40 (group III) µg/kg of body weight/day for 63 days. Control animals (group I) were fed a placebo. After that period animals were sacrificed, ovaries were extirpated, fixed in paraformaldehyde solution, cut into sections with a cryostat and studied for apoptosis with TUNEL kit, and for the presence of apoptosis-promoting protein Bax with immunohistochemistry. It was found that apoptosis was detected with TUNEL only in medium-sized antral ovarian follicles in animals of groups I and II. No apoptosis signal was found in the ovaries of animals in group III. No differences in the distribution and intensity of staining for Bax were found between animals of all investigated groups. The results indicate that ZEA do not induce apoptosis in porcine ovaries, and the inhibition of proliferation must be associated with other mechanisms. In this study, totally 272 fishes were investigated for the presence of L. intestinalis plerocercoids. The prevalence value was 40.01% of the whole fish sample and the mean intensity was 2.48. The lower IP values and mean intensity levels may provide some evidence of the strategy of Ligula in order to complete its life cycle. None of the parasitization parameters showed significant differences between the sexes. Phenotypic methods are time-consuming and sometimes lead to intermediate results, therefore replacement by more specific and rapid methods are needed. Out of a total of 911 campylobacter strains tested, 800 human isolates were received from the clinical bacteriology laboratories from 5 regions and 111 foodstuff isolates (raw chicken and pork meat from retail market) originated from the routine examination in our laboratory. Based on the PCR method 85.1% of these strains were identified as C. jejuni, 12.5% as C. coli and 2.3% as mixed cultures of C. jejuni and C. coli. When species determination of campylobacters was based on conventional methods (hippurate hydrolysis test), 28.5% of the isolates were not identified correctly. The mixed cultures of campylobacters have not been detected without further subculturing of strains, which takes several days and enormously extends the identification process. The aim of this study was to investigate how common N. caninum infection is among dairy herds in the Czech Republic. Bulk milk samples were collected from 495 dairy herds and analysed for the presence of specific antibodies by a commercial N. caninum iscom ELISA. Five out of 495 dairy herds (1.01%) had percentage positive values ≥20 and were considered positive. In the positive herds, blood samples were collected from cows and pregnant heifers and analysed by the ELISA test. The within-herd seroprevalence ranged from 2.5 to 50%. The bulk milk ELISA could be a useful and inexpensive method for rapid screening of Neospora caninum infection in dairy herds in large areas. This condition is usually followed by uterine stump pyometra, which can be described as the infection of uterine body tissue remaining after the operation. However, both conditions may also be encountered separately. In this study, the clinical findings, therapies and long term postoperative conditions of five ovarioectomized bitches with uterine stump pyometra that were brought to the clinic with symptoms like lethargy, loss of appetite, purulent or sero-sanguinous vaginal discharge, polydipsia, polyuria within a four year period, were presented.
