Champion Author-Workplace-Name: South Frontier Federal University, Realeza, Brazil Title: Heart rate variability in Mangalarga Marchador horses after physical exercise Abstract: The aim of this study was to evaluate the influence of physical exercise (marcha gait) on heart rate variability indices in Mangalarga Marchador horses. Twenty-five animals, fourteen females and eleven males, aged 6.4 ± 2.7 years and with a mean weight of 428.3 ± 24.7 kg, were used. Electrocardiograms were recorded at two different time points: rest and immediately after exercise (40 min of aerobic exercise, marcha gait). Time domain analyses of heart rate variability were made using vasovagal tonus index (VVTI) and standard deviation of normal intervals (SDNN). Variables were analysed for normality using the Shapiro-Wilk test and comparisons were made between T0 and T1 using the paired t-test (VVTI) or Mann-Whitney (SDNN), considering P < 0.05. The Pearson coefficient was calculated to evaluate the correlation between heart rate and VVTI and the Spearman coefficient was calculated for heart rate and SDNN. There was an increase in heart rate after exercise (P < 0.0001), from 45.7 ± 12.7 to 77.3 ± 13.5 beats/min, and a reduced VVTI (P < 0.0001), from 8.66 ± 1.45 to 4.65 ± 1.25 and a reduced SDNN (P < 0.0001), from 80.09 to 11.67 ms. No correlation was observed between heart rate and VVTI or SDNN. It was possible to conclude that the physical activity carried out by Mangalarga Marchador horses influenced the autonomic response and consequently heart rate variability indexes, suggesting the possible use of these indices for the evaluation of Mangalarga Marchador performance. Ejaculates were collected using artificial vaginas from five Merino rams, microscopically evaluated and pooled at 37 °C. The pooled semen samples were diluted in a Tris-based extender, including cysteine (2 mM and 4 mM), trehalose (10 mM and 25 mM) and no antioxidant (control). Diluted semen samples were kept in tubes and cooled from 37 to 5 °C in a cold cabinet, and maintained at 5 °C. Cooled samples were evaluated for sperm motility, viability, mitochondrial activity and acrosome integrity at 0, 24, 48, 72 and 96 h. Extender supplemented with trehalose (10 and 25 mM) and cysteine (2 and 4 mM) led to higher motility in comparison to the control at 24, 48, 72 and 96 h of liquid storage (P < 0.05). Trehalose at the doses of 10 mM, 25 mM and 2 mM cysteine led to higher viability between 24-48-72 h and at 96 h of liquid storage (P < 0.05). Further, 4 mM of cysteine improved sperm viability rates at 24 and 48 h of storage compared to the control group (P < 0.05), and resulted in improved acrosome integrity rates compared to the control group at 72 and 96 h of storage (P < 0.05). Extender supplemented with 10 and 25 mM trehalose at 24 and 72 h and 4 mM cysteine at 24 and 96 h of storage led to higher sperm mitochondrial activity rates when compared to the control group (P < 0.05). In conclusion, the findings of this study show that trehalose and cysteine provided significant protection to ram sperm parameters during liquid storage. Proskura Author-Workplace-Name: West Pomeranian University of Technology, Szczecin, Poland Title: Expression profile of the SOX9 gene in the testes of sexually immature and mature male goats (Capra hircus), and its potential influence on postnatal testis development Abstract: The aim of this study was to compare the expression levels of the SOX9 (SRY-box 9) gene in the testes of 18 White improved male goats (Capra hircus) divided into three age groups (one, 10 and 15 months of age with seven, eight and three individuals per group, respectively). Abnormalities in testis development were observed in three individuals from the group of 10-month-olds. Additionally, differences in SOX9 expression that may affect the process of testis maturation and testicular spermatogenic activity were investigated among individuals. The expression of the SOX9 gene in testicular tissues was analysed using qPCR. Maximal SOX9 expression was observed in 10-month-old males with normal testes, while expression was significantly reduced in the same age group of males with abnormal testes. Abnormalities in testis development were associated with low parameters of semen quality. The lowest expression levels of SOX9 were observed in 15-month-old goats. The results of the present study indicate that SOX9 expression changes significantly during the development of male goats. The effect of orally administered Budesonide (Entocort) on the HPA axis was analysed in 21 dogs with inflammatory bowel disease. Biochemical analyses were carried out to evaluate the activity levels of alanine aminotransferase, asparagine aminotransferase, alkaline phosphatase, cortisol and endogenous adrenocorticotropic hormone. Urine samples were collected from each patient before the study and after 30 days of the experiment to determine the composition and the physical and chemical properties of urine sediments. Considerably lower serum concentrations of cortisol and endogenous adrenocorticotropic hormone were observed after 30 days of treatment. A significant increase in alkaline phosphatase levels was noted on Day 30. In the studied dogs, the drop in HPA axis activity was correlated with side effects associated with the administered glucocorticosteroid (polyuria, polydipsia). In conclusion, we have shown that oral administration of Budesonide to dogs diagnosed with inflammatory bowel disease significantly suppressed the activity of the HPA axis. The aim of this study was to determine the presence of, and risk factors for T. blagburni among a cohort of cats living in different conditions in the Czech Republic. Samples were collected from 170 cats living in different environments. The InPouch™ TF-Feline medium method was used for diagnosis of feline trichomonosis. A single case (0.6%) with motile trichomonads identified as Pentatrichomonas hominis was found in a cat from a multi-cat household. Our study suggests that trichomonads and in particular, T. blagburni, infection may be much less common in the Czech Republic than in neighbouring countries, despite the inclusion of cats that were likely to be from higher-risk groups. A review of studies of the association of trichomonads and feline diarrhoea carried out in different countries revealed variation in the frequency of trichomonads detected. Different combinations of PCR or culture methods for screening or confirmation have been utilised, with or without species differentiation; however, this could not solely account for the variation in the occurrence between countries. From those studies where differentiation was performed, we calculated from the combined studies that T. blagburni occurred in six cats without diarrhoea (1.1%) and 47 cases with diarrhoea (5%). This finding supports an association with diarrhoea as well as the occurrence of asymptomatic cases. We note that in many studies, including our own, the occurrence of T. blagburni may well be underestimated and suggest that future studies use a combination of PCR screening of both faeces and faecal cultures, with differentiation of trichomonad species. Forty-eight pregnant Sprague-Dawley rats were used as material. Five groups were subjected to treatments while one group served as a control. The numbers of animals in each group were equal (n = 8). Treatment groups were administered carprofen (10 mg/kg, single or double dose, s.c.), flunixin meglumine (5 mg/kg, single or double dose, i.m.) and meloxicam (2 mg/kg, a single dose, s.c.) on the third day after mating. The control group received saline. The rats were sacrificed on Day 7 of gestation. Luteal spots and implantation sites were recorded. Pre-implantation loss was calculated by subtracting the number of luteal spots from the number of implantation sites. Compared with the control, the administration of flunixin meglumine (double dose), carprofen (double dose) and meloxicam highly significantly decreased the implantation rate (P < 0.001). Single dose administration of flunixin meglumine and carprofen led to significant decreases (P < 0.01). In conclusion, this study indicates that carprofen, flunixin meglumine and meloxicam treatment cause a decline in implantation rate in rats. Thoracotomy was performed in the fourth inter-costal space under general, isoflurane-maintained, anaesthesia. Cutting of the arterial ligament failed to free the oesophagus. Dissection of further vascular structures revealed the left and right aortic arches to be of similar diameter. A vascular clamp was put on the left arch for 10 min, while the patient's cardiovascular parameters were monitored. The lack of significant cardio-respiratory symptoms indicated the necessity of ligating and cutting the left aortic arch. Removal of additional tissue from the region of the cut on the left arch completely released the oesophagus which was continuously monitored by video-oesophagoscopy. The chest was closed according to a standard thoraco-surgical procedure. The cutting of the left aortic arch and the release of the surrounding tissue restored the normal position of the oesophagus. After surgery, the animal was returned to its owners in a very good condition. Control examination showed complete amelioration of clinical symptoms and subsequent normal development of the dog. Intra-operative clamping of the left aortic arch with simultaneous monitoring by oesophagoscopy ensured a positive outcome of the surgery of the intra-operatively diagnosed double aortic arch. On initial evaluation, severe hyperglycaemia (blood glucose concentration of 34.9 mmol/l), ketonuria, systemic inflammation (fever, panting, left-shift neutrophilia, and a high C-reactive protein level of 980.97 nmol/l, abnormal pancreatic lipase immunoreactivity, and periodontitis were found. With consideration of possible insulin resistance, blood glucose (BG) levels were monitored using a portable glucose meter (q 1-3 h) and a continuous glucose monitoring system (CGMS) for 72 h (three consecutive trials); intensive insulin therapy was initiated using regular insulin (2.2 IU/kg intravenously). The insulin doses needed, based on the nadir, peak, and duration of insulin action from a traditional intermittent glucose curve were higher than those based on the CGMS results. Meanwhile, transient hyperglycaemic and hypoglycaemic periods, occurring between the intermittent measurements, were easily identified with the CGMS. Therefore, insulin resistance and the Somogyi phenomenon are less likely to occur with use of the CGMS than with intermittent BG measurements. By comparing data from a CGMS to those from an intermittent portable BG measurement system, this case report emphasises the importance and usefulness of a CGMS. The granulomatous colitis diagnosis was made on the basis of PAS-positive histiocytes in the lamina propria of the colonic mucosa in biopsy samples taken at colonoscopy. Remission of granulomatous colitis has been reported using fluoroquinolones leading to the idea that invasive Escherichia coli strains in the colonic mucosa are involved. Oral enrofloxacin (Baytril 150 mg, Bayer, Spain) at 10 mg/kg per day for eight weeks was prescribed to both dogs in this study. A first course of therapy resolved the problem in dog No. 1, which, however, was followed by relapse three months later without enrofloxacin response. No clinical remission was seen in dog No. 2 and 4.4 mg/kg marbofloxacin (Marbocyl P 20 mg, Vetoquinol, Spain) per day for 10 weeks was administered but without any response. From both dogs, biopsy samples from the colonic mucosa were taken during colonoscopy. Samples were homogenised for microbial culture in different agar media to identify invasive microbes. Escherichia coli were largely isolated and antibiotic sensitivity testing (MIC of E. coli to selected antimicrobials, CLSI 2013) was carried out. In both cases, E. coli was resistant to fluoroquinolones. In dog No. 1 E. coli was susceptible to amoxicillin-clavulanate, cefazolin, amikacin and gentamicin whereas in dog No. 2 it was susceptible to doxycycline and amoxicillin-clavulanate. Clinical remission was achieved in dog No. 1 with amoxicillin-clavulanate (Synulox 250 mg, Pfizer, Spain) therapy for eight weeks. No response was found in dog No. 2 with any of the antimicrobials alone or combined with metronidazole. The cat presented with incontinence, pollakiuria, vomiting and weight loss. An abdominal mass was palpated and ultrasonography revealed severe segmental thickening of the jejunal wall with loss of layering. Complete resection of the jejunal mass was performed. The resected mass was composed of branching trabeculae of dense collagen, intermixed with bundles of large fibroblasts and inflammatory cells, which is consistent with FGESF. One year later, a new mass reoccurred in another intestinal segment. Between both FGESF occurrences, mycoplasmas were cultured from urine and fine needle aspiration of the urinary bladder wall showed numerous eosinophils only; therefore, a presumptive diagnosis of eosinophilic cystitis was made.
