Three scenarios for tree species composition and stand age in new and permanent forest areas: A case study of LatviaOriginal PaperZigmārs Rendenieks, Līga LiepaJ. For. Sci., 2023, 69(10):438-450 | DOI: 10.17221/25/2023-JFS Land abandonment and the emergence of new forest areas create novel dynamics for forest ecosystems and landscapes. Modelling is often used to forecast tree species composition, age group distribution and spatial patterns in the future. The aim of this study was to develop three scenarios for changes in tree species composition, stand age distribution and spatial patterns of new forest areas and permanent forests using Latvia as a case study. We selected 19 study areas of the size 10 km × 10 km to sample the variety of forest cover patterns, tree species, and stand age. Using GIS tools, we developed three scenarios: baseline, commercial and conservation. Results showed that the conservation scenario resulted in the most even-aged group distribution. Scenarios predicted the increase of Picea abies area (reaching 29.3% in permanent forests and even 45.7% in new forests) and the reduction of Pinus sylvestris in most cases. Changes in the median patch area were the best indicator for evaluation of different scenarios with the largest patches of new forest areas for the conservation scenario (1.92 ± 1.23 ha). The existing structural and compositional integrity of sampled forest landscapes was best retained under the baseline and conservation scenarios, while the commercial scenario indicated more fragmented forest landscapes in the future. |

Sci., 2023, 59(3):233-244 | DOI: 10.17221/103/2022-PPS Tan spot, caused by Pyrenophora tritici-repentis, is a severe worldwide wheat foliar disease and has become common in Tunisia. Two hundred wheat field surveys were conducted in northern Tunisia during the 2017–2019 seasons to determine disease intensity and the correlation of the tan spot with agricultural practices and environmental factors. The disease infested 67% of the visited regions. The highest prevalence (87.5%), incidence (> 30%), and severity (> 10%) were observed in sub-humid regions, contrary to the middle semi-arid regions. Logistic regression was used to analyse the correlation of incidence and severity with various factors as independent variables. In a multiple variable model, the bioclimatic stages, variety, previous crop, humidity, and rainfall were significantly associated with tan spot incidence and severity. High incidence and severity were significantly correlated to the sub-humid regions, varieties Maâli, Razzak, Karim, and Carioca, rainfall, high level of humidity, and monoculture. The correlation between the tan spot and several environmental factors must be considered to ensure successful and sustainable disease management strategies. |
Culture of Steinernema glaseri on three solid media and their virulence against Galleria mellonella larvaeOriginal PaperCarlos I. Sci., 2023, 59(3):278-283 | DOI: 10.17221/1/2023-PPS Steinernema glaseri is a potential biocontrol agent against white grubs of the Phyllophaga spp. complex; however, its suitability for in vitro multiplication has been scarcely investigated. In this study, the effects of the surface culture of NJ-43 strain with its symbiotic bacteria Xenorhabdus poinarii on egg-yolk agar (P2), chicken-liver agar and nutrient meat-peptone (MP) agar on infective juvenile (IJ) productivity and their virulence against Galleria mellonella larvae were investigated. The bacteria on the surface of the agar were first incubated in darkness at 30 °C for 54–102 h, and then 100 surface-sterilised IJs were added. After two harvests, the accumulated productivity was higher on chicken-liver agar (536 × 103 IJs/m2 day) and P2 agar (534 × 103 IJs/m2 day) than on nutrient MP agar (58 × 103 IJs/m2 day). The mean virulence of the in vitro produced IJs was 46–60% and showed no statistically significant difference among the three culture media. In conclusion, the maximum multiplication factor of S. glaseri NJ-43 on solid media was 385, and its original virulence was retained. |

Gly-Leu instead of Gly promoted the proliferation and protein synthesis of chicken intestinal epithelial cellsOriginal PaperJing Liang, Baolong Du, Minyan Wan, Yapeng Zhao, Defu TangCzech J. Anim. Sci., 2023, 68(9):391-398 | DOI: 10.17221/213/2022-CJAS Amino acids have positive regulatory effects on the function of intestinal epithelial cells (IEC), but in the field of animal nutrition, research on the regulatory effects of amino acids on IEC is still in the initial stages. This study aims to explore the effects of Gly, Gly-Gly, and Gly-Leu on IEC proliferation and their possible mechanisms. Chicken small intestinal epithelial cells were separated using the tissue block method, and other miscellaneous cells were removed for digestion and passage culture. The IEC were cultured in the medium containing 20 nmol/l Gly, Gly-Gly and Gly-Leu for 24 h, and the expression of enterokinase and cytokeratin in cells, the growth curve and activity of IEC, cell cycle, differentially expressed genes, mRNA expression, and protein expression levels of p-mTOR and p-S6K1 in IEC were detected. Enterokinase and cytokeratin were expressed specifically in IEC. The results of growth curve and MTT revealed that the cell viability of IEC was significantly increased after treatment with Gly, Gly-Gly and Gly-Leu. The cell cycle results showed that compared with the control group, Gly, Gly-Gly and Gly-Leu intervention could increase the proportion of IEC in G1 phase, and the proportion in S phase of IEC was decreased. Transcriptome sequencing showed that compared with the control group, there were 54, 28 and 30 differential genes in Gly group, Gly-Gly group and Gly-Leu group, respectively. These genes were mainly enriched in nitric oxide synthesis and protein kinase B signalling, PI3K signal and cellular amino acid biosynthesis and transport signal pathways. RT-PCR results showed that the mRNA expression levels of PCYT2, SPP1, EMC6, GRIA2 and PKD2 were consistent with the sequencing results. Western blot results showed that compared with the control group, the protein expression of p-mTOR and p-S6K1 in Gly group, Gly-Gly group and Gly-Leu group was significantly increased. Gly-Leu can promote the protein synthesis in IEC by activating protein synthesis of mTOR signalling pathway in chicken IEC. |
Retrospective study using biosensor data of a milking Holstein cow with jejunal haemorrhage syndromeCase ReportS Ha, S Kang, M Jung, E Jeon, S Hwang, J Lee, J Kim, YC Bae, J Park, UH KimVet Med - Czech, 2023, 68(9):375-383 | DOI: 10.17221/73/2023-VETMED Jejunal haemorrhage syndrome (JHS) is a sporadic and fatal enterotoxaemic disease in dairy cows associated with acute development and poor prognosis despite treatment. A 5-year-old Holstein cow with no reported pregnancy, three calving numbers, and 303 days in milk presented with hypothermia, discomfort, and inappetence. Anaemia, dehydration, faeces with blood clots, and absence of rumen and bowel movements were observed. We identified the presence of neutrophilia, hyperglycaemia, hypoproteinaemia, azotaemia, hyperlactatemia, hypocalcaemia, hypermagnesemia, hypokalaemia, and hypochloraemia through blood analyses. Necropsy and histopathologic examination revealed a dilated bluish-purple jejunum, blood clots within the jejunum, neutrophil infiltration into the submucosa of the jejunum, and vascular necrosis. Retrospective examination revealed extraordinary patterns of rumination time, activity, rumen mobility, and rumen temperature using biosensors and decreased milk yield. The abnormalities in the affected cow were detected before recognition by farm workers. To the best of our knowledge, this is the first report to examine data from biosensors in a cow with JHS. Our findings suggest that using biometric data may help understand the development of JHS. |
Effects of different habitats on Achnatherum inebrians (Hance) Keng ex Tzvelev (drunken horse grass) soil seed banks and aboveground vegetationOriginal PaperSaimilakezi Taiwaikuli, Guili Jin, Shazhou An, Yiqing Dong, Peng WeiPlant Soil Environ., 2023, 69(9):421-428 | DOI: 10.17221/191/2023-PSE The soil seed bank, as a potential source of ground vegetation renewal, plays an important role in the natural recovery and succession of vegetation as well as in the construction of ecosystems. To clarify the characteristics of the soil seed bank of Achnatherum inebrians and its relationship with the aboveground vegetation, the soil seed bank density, species composition and aboveground vegetation of three different grassland types, namely, desert, steppe and meadow, were investigated by means of field survey sampling and indoor germination experiments. The results showed that the seed bank densities of the three habitats were ranked as desert (1 422.22 seeds/m2), steppe (2 077.78 seeds/m2) and meadow (3 722.22 seeds/m2). The numbers of species were 16, 11 and 17, respectively. With respect to the vertical allocation, the soil seed banks in each habitat were shallow, and the seeds were mainly concentrated in the soil surface layer (0–5 cm). The species richness of the soil seed banks in the three habitats was higher than that of the aboveground vegetation, but there were some differences in richness, evenness and dominance. The species richness and diversity of soil seed banks and aboveground vegetation in meadow habitats were higher than those in desert and steppe habitats, indicating that the soil seed banks and aboveground vegetation in meadow habitats had higher stability. There was a significant positive correlation between the density of temporary soil seed banks and the density of aboveground plant communities in grassland habitats. The results may provide some reference for the prevention and control of Achnatherum inebrians in the three habitats. |

Plant Breed., 2024, 60(1):1-11 | DOI: 10.17221/113/2022-CJGPB The present study was to determine the genotypic and environmental variability and stability in seed yield, oil content, oil yield, oleic and linoleic acid of 10 safflower lines derived from a cross of Dinçer 5-18-1 × Montola 2000 together with six cultivars under six environments at five locations. The effects of genotypes, environments and genotype × environment interactions were highly significant (P < 0.01) for seed yield and oil content. Averaged across all environments, the seed yield was lowest in the cultivar Olas (2 352 kg/ha), and highest in the line Bay-Er 5 (2 869 kg/ha). According to mean (xi) and regression coefficient (bi) values, the Bay-Er 16 was better adapted to unfavourable environmental conditions, whereas the Bay-Er 1, Bay-Er 5 and Bay-Er 14 were better adapted to favourable environmental conditions. The highest oil content across environments, over 35%, was recorded in the line Bay-Er 15 and the cultivars Olas and Linas. The best adaptability to the environments was observed in the cultivar Olas. The oleic acid content of genotypes increased and the linoleic acid contents decreased from the north to the south latitudes. The oil content of genotypes grown in Southeastern Anatolia was higher than in the other regions. Within the regions, seed yield and oil content was higher after autumn sowing than after spring sowing. |
Gallbladder atrophy associated with pancreatitis: Clinical and advanced imaging diagnosis in a dogCase ReportD Han, DI JungVet Med - Czech, 2023, 68(11):435-442 | DOI: 10.17221/76/2023-VETMED Gallbladder atrophy (GBA) is characterised by a reduction in the size and volume of the gallbladder. In human medicine, it is well-established that GBA frequently occurs together with pathologies affecting the gallbladder and pancreas. However, to the best of our knowledge, there is currently a dearth of reported cases of GBA in dogs within the veterinary field. In this study, we present a case report of GBA in a 7-year-old Yorkshire Terrier. The diagnosis of GBA was confirmed using abdominal ultrasonography and advanced imaging techniques, including computed tomography, which were performed over a 4-year period. The patient initially presented with predominantly gastrointestinal symptoms, which were subsequently diagnosed and treated as pancreatitis. Concurrently, a gallbladder nodule and an anomalous structure suspected to be cholelithiasis were identified. However, during the 4-year follow-up, the gallbladder structure regressed, leaving only the presence of the gallbladder nodule. Notably, cholecystectomy was not performed, and apart from pancreatitis-related symptoms, the patient did not show any gallbladder-related problems throughout the spontaneous atrophic process. Based on these findings, we propose that the observed GBA was likely induced by cholecystitis associated with pancreatitis. This case underscores the significance of considering GBA as a potential diagnosis in canine patients presenting with pancreatitis and gastrointestinal symptoms. Furthermore, it highlights the value of comprehensive diagnostic imaging in accurately determining the underlying cause of these symptoms. |

Evaluation of five rootstocks on the growth, gas exchanges and chlorophyll fluorescence of Juglans regia L. cv. ‘Xiangling’Original PaperBing Hua Liu, Liang Cheng, Ji Xia Zhang, Lian Jia Yui, Hai Lin Ma, Fangchun LiuHort. (Prague), 2023, 50(3):208-218 | DOI: 10.17221/152/2022-HORTSCI Our study comparative analyzed the morphological and physiological performance of Juglans regia L. cv. ‘Xiangling’ (JRX) grafted onto five rootstock species originated from China. A wide range of coefficient of variation (CV), 1.155–39.848%, was detected for fourteen indexes among the studied seedlings. CV of plant height, total dry biomass, total leaf area, root/shoot ratio, chlorophyll index, and stomatal conductance was higher than 15.00%, suggesting higher variation. Principal component analysis, hierarchical cluster analysis and subordinate function analysis were used to screen engrafted seedlings for their quality. All three methods produced the same result that seedling quality of ‘Xiangling’ engrafted with the five rootstock species from high to low were – Juglans regia L. cv. ‘Jizhaomian’ (JRJ), JRX, Juglans regia L. cv. ‘Lvxiang’ (JRL), Juglans mandshurica, Maxim., and Juglans hopeiensis Hu.. The evaluation results were highly consistent with the actual growth performance of the engrafted seedlings, it verified the reliability of our results and the evaluation methods. The results demonstrated that JRJ, JRX, and JRL are potential rootstock candidates in the studied walnut planting regions. It would provide important implication for the selection of proper rootstock species for a certain particular walnut variety. |

Most biostimulators are capable of reacting against the effect of biotic and antibiotic stresses as they increase the crop production of the cultures and represent an opportunity of growing quality and healthy food. The article affects the results of the influence of RadiCifo L24 and Euroradix over the growth and development of grafted cuttings in vine rootings. The study was carried out through the dessert seedlees grapevine variety ‘Zornitsa’ grafted over the Berlandieri × Riparia (SO4) pad. Before rooting, paraffin is applied to 2/3 of the stratified grafted cutting’s length and is then immersed in a 2% solution of RadiCifo L24 (for 20 minutes) and Euroradix (for 5 minutes), after that the cuttings are rooted to 15-17 cm in depth (to the non-paraffined part). According to the biometrics rating (number of developed shoots, number of roots), the variant with the growth stimulator RadiCifo L24 seems to be a more effective method for the production of first-class vines of the ‘Zornitsa’ variety, thus it can be recommended for the production of vine planting material. |

Development and evaluation of an electromagnetic device to improve the physiological properties of some crop seedsOriginal PaperAhmed Shawky El-SayedRes. Agr. Eng., 2023, 69(4):167-178 | DOI: 10.17221/108/2022-RAE This research aims to develop an eco-friendly technique for treating seeds before sowing and improving their physiological features. The developed technique depends on utilizing synchronized electro-magnetization and microwave radiation. An electromagnetic device was evaluated to provide multiple treatment ranges. The treatments are regulated electronically according to the seeds' physiological properties and storage duration. The device was designed to accommodate small and medium seeds for a variety of strategic crops, including wheat, barley, etc. Three different treated wheat varieties were tested and compared to the control. Also, eight different levels of synchronized electro-magnetization and microwave radiation were tested. The treated wheat seeds' vegetative properties, such as germination percentage, germination rate index, germination speed coefficient, and vigour indexes, were highly significant compared to the control. The treated wheat seeds' physiological properties were highly significant. The device productivity ranged from 0.023 to 0.059 Mg·h–1 with minimum energy consumption rates of 0.396 to 0.018 kWh·Mg–1, while the operating costs decreased to 11.53–44.13 USD·Mg–1. |

(Prague), 2023, 50(4):262-274 | DOI: 10.17221/127/2022-HORTSCI Effect of pre-harvest calcium chloride (CaCl2) treatment on fruit quality attributes of three apple cultivars (‘Golden Delicious Reinders®’, ‘Granny Smith’ and ‘Red Chief’) at harvest and after two and four months of cold storage during two consecutive years was studied. Trees were sprayed four times starting from the beginning of June at 20-day intervals. After harvest, fruits were stored in a regular atmosphere (RA) for further analysis. Measurements included fruit weight, weight loss, firmness, total soluble solids (TSS), titrable acidity (TA), total sugars (TS), TS/TA, total phenolic content (TPC) and antioxidant activity (AA). CaCl2 had a positive impact on fruit firmness of all cultivars in both years. Higher values of TSS and TS of ‘Golden Delicious Reinders®’ were determined in CaCl2 treatment in both years. TSS, TS and TS/TA were increased by prolonging storage in ‘Red Chief’ in CaCl2 treatment and control, in both years. TSS and TS proved similarly stable in ‘Granny Smith’ during storage in CaCl2 treatment and control in the second year of examination, but a decrease of TA and TS/TA has been detected in both years of storage. The AA of cultivars ‘Golden Delicious Reinders®’ and ‘Red Chief’ was stronger affected by the storage compared to CaCl2 treatment. Through a comprehensive analysis of apple cultivars it can be concluded that pre-harvest application of CaCl2 in cultivars ‘Golden Delicious Reinders®’ and ‘Red Chief’ can be used to improve fruit quality during storage. |

Sci., 2023, 69(11):463-469 | DOI: 10.17221/73/2023-JFS This paper is focused on the description of the black mulberry (Morus nigra), its ecology and the possibilities of its in vitro propagation for quick and efficient obtaining of a large quantity of clones in a relatively short time for subsequent planting or sale. Due to ongoing climate change, it is considerable to use mulberry trees in horticulture, agroforestry and forestry under the conditions of Central Europe. The use of the mulberry is conditioned by the availability of planting stock. A proven and successful method of mulberry propagation is in vitro cultivation. Based on literature review, the recommended composition of planting media and other procedures for in vitro cultivation of mulberries are presented. The aim of the article is to inform foresters about the possibilities of using the black mulberry tree in our conditions and, using its example, to point out the possibility of using non-standard species of trees both as part of adaptation measures to the expected climate change and as one of the options for increasing the biodiversity of the landscape. |

Anim. Sci., 2023, 68(11):443-450 | DOI: 10.17221/107/2023-CJAS Lipid-rich bovine serum albumin (LR-BSA) has been reported to increase the survival rate of blastocysts after freezing and thawing. However studies on the early development of in vitro fertilised (IVF) embryos are still insufficient in cattle. This study investigated the blastocyst productivity of in vitro culture (IVC) medium (synthetic oviductal fluid medium with amino acids, mSOFaa) supplemented with 5% fetal bovine serum (FBS, Control) or LR-BSA (1, 2.5, 5 mg/ml dosages, respectively) and the qualitative characteristics of produced blastocysts. Cleavage rates on day 2 were similar between all groups. Significant differences were obtained in the blastocyst rate in the 2.5LR-BSA group compared with the control (23.5% vs 35.7%, P < 0.05). Hatching rates were significantly higher in all treatment groups than in control (P < 0.05). The cell number in blastocysts did not differ among groups, but the apoptotic index in the 5.0LR-BSA group was higher than the control (P < 0.05). The expression of embryo quality-related markers was affected. Octamer-binding transcription factor 4 (Oct4), placenta associated 8 (Plac8), and sex-determining region Y-box 2 (Sox2) genes expression in the 2.5LR-BSA group were significantly up-regulated compared to the control (P < 0.05). Cytoplasmic lipid contents and the lipid droplet formation-related gene, sterol regulatory element binding transcription factor 1 (Srebf1), were significantly increased in the 5.0LR-BSA group (P < 0.05). Therefore, the LR-BSA supplementation (2.5 mg/ml) to mSOFaa medium could contribute to the early development of bovine IVF embryos and the qualitative improvement of the resulting blastocysts. |

This study was conducted to evaluate the effect of exogenous ascorbic acid (AsA) on photosynthesis, chlorophyll fluorescence, and xanthophyll cycle in alfalfa leaves subject to under drought and heat stress. The results suggest that drought and heat stress caused decreases in the net photosynthetic rate (Pn) in alfalfa leaves, but stomatal conductance (gs), transpiration rate (Tr), and intercellular CO2 concentration (ci) were increased. The application of AsA could alleviate these changes to some extent. Besides, the decreased photosystem II (PSII) maximum photochemical efficiency (Fv/Fm) and violaxanthin (V) contents and significantly increased non-photochemical quenching (NPQ) levels. The increased NPQ corresponds to the de-epoxidation state (DPS) of xanthophyll pigments. In the AsA-pretreated alfalfa plants, the Fv/Fm and the NPQ were elevated, indicating that AsA could alleviate the adverse effects on photosynthesis induced by this stress. The violaxanthin de-epoxidase (VDE) enzyme activity was inhibited by drought and heat stress, and AsA significantly increased VDE enzymatic activity on the 2nd and 8th days. In summary, photoinhibition of PSII occurred in alfalfa leaves under drought and heat stress, resulting in decreased photosynthetic activity. Exogenous AsA can enhance the photosynthetic capacity of the plant, and enhance the drought and heat resistance of alfalfa. |

Sci., 2023, 68(10):399-406 | DOI: 10.17221/194/2022-CJAS This research paper addresses the hypothesis that the peptidoglycan recognition protein 1 (PGLYRP1) gene polymorphism (Tyr76His; dbSNP ID: ss104796364) has an influence on some performance traits and causes of culling in Polish Holstein-Friesian red-white cows. The study involved 134 cows kept on a farm in the south-west of Poland. PGLYRP1 genotypes TT, CT, and CC were detected. It was shown that compared with cows with genotypes CT and TT, the individuals with genotype CC were characterised by higher lifetime yields and higher amounts of lactation milk, fat, and protein. A beneficial effect of genotype CC, compared with genotype TT, was also noted in the case of the lifespan and, consequently, the length of the productive life and the average number of lactations. Diseases of the musculoskeletal system (genotypes CC and TT) and disorders of the reproductive system (genotype CT) were the most common causes of culling. An essential practical observation was the potentially higher susceptibility of cows with genotype CC to mastitis, which resulted in the necessity to cull over one-fifth of the animals in this group. Simultaneously, no cows in this group were culled due to low performance or metabolic, gastrointestinal, and respiratory diseases. Therefore, the PGLYRP1 gene seems to be a promising potential herd health marker; however, to consider it the main gene, it is necessary to extend the investigations to include more individuals and other breeds of dairy cattle. |

Several chemical parameters and their relationships were analysed in earthworm casts of two urban parks, namely the pH, total organic carbon (TOC), total nitrogen (Nt), plant available phosphorus (P), plant available potassium (K), and calcium water soluble (Ca). It was statistically significantly found that the TOC, Nt, P and K are reciprocally determined in the earthworm casts: 74.4% of the pH variability is co-determined by the N, P, K, TOC, and Ca contents; 95.9% of the Nt variability is co-determined by the pH, P, K, TOC, and Ca contents; 95.4% of the P variability is co-determined by the pH, N, K, TOC, and Ca contents; 94.5% of the K variability is co-determined by the pH, N, P, TOC, and Ca contents; 86.6% of the TOC variability is co-determined by the pH, N, P, K, and Ca contents. This study revealed the complexity of the chemical relationships inside earthworm casts, their reciprocal dependencies, and highlighted the complexity of the earthworms’ contribution to biogeochemical cycles in urban areas. Our findings propose earthworms as indicators of the integrative conservation management of urban ecosystems. |
Implications of surfactant application on soil hydrology, macronutrients, and organic carbon fractions: An integrative field studyOriginal PaperCansu Almaz, Recep Serdar Kara, Markéta Miháliková*, Svatopluk MatulaSoil & Water Res., 2023, 18(4):269-280 | DOI: 10.17221/88/2023-SWR This study investigates the effects of repeated applications of the non-ionic soil surfactant H2Flo (ICL-SF Inc., Israel) on the soil water content, hydraulic conductivity, nutrient distribution, and organic carbon fractions (OCFs) in non-hydrophobic loamy sand soils under subsurface drip irrigation. Our results indicate that H2Flo treatment reduces both saturated and unsaturated hydraulic conductivity while promoting the uniform irrigation distribution, consistent with previous findings on surfactants’ effects on sandy soils. An increase in soil pH levels, organic carbon content, and extractable magnesium, calcium, and potassium was observed in treated soils, with elevated levels of potassium permanganate oxidizable organic carbon (POXC) implying accelerated decomposition rates. Notably, a positive linear relationship was found between POXC and the increased NO3–-N content of treated soils, suggesting induced conditions of nitrification. However, the carbon fractions water-soluble organic carbon (Cws) and hot water-soluble organic carbon (Chws) remained quantitatively unchanged, even though they exhibited a positive linear relationship with the soil’s hydraulic conductivity. The study highlights the crucial role of monitoring changes in OCFs and nutrient dynamics after surfactant application to optimize soil organic matter utilization and chemical fertilizer management. |
