Organic fertilization induces changes in soil nitrogen mineralization and enzyme activitiesOriginal PaperMaría Rosa Yagüe, Carmen Lobo, Pilar GarcíaPlant Soil Environ., 2023, 69(1):38-43 | DOI: 10.17221/274/2022-PSE In this study, we addressed the reuse of two organic waste products as fertilisers. To this end, soil fertilised with the spent mushroom substrate (SMS) or with an anaerobic digestate (DIG) was subjected to an incubation assay, and the results were compared with those from soil treated with a mineral fertiliser (MIN) and an unfertilised soil (CO). The soil was sampled after fertilisation and after 90 days of aerobic incubation. Nitrogen (N) mineralisation (NH4+ and NO3–) and oxidable carbon (OC) were determined. The impact of the treatments on the soil was evaluated by measuring the enzymatic activity of arylsulfatase (ARYL), ß-galactosidase (GAL), and urease (URE). The highest OC content was observed in the SMS treatment. After 90 days of incubation, the SMS treatment showed a lower mineral N content than the CO treatment. This finding was associated with N immobilisation. However, mineral N significantly increased ARYL activity in the DIG and MIN treatments, and URE activity was always higher at both sampling times in the SMS treatment. Initially, GAL activity was notably high in the DIG treatment but decreased after incubation, reaching similar values to those registered in the CO treatment. Organic fertilisation treatments induced different effects on soil N mineralisation, showing changes in the activity of the enzymes analysed. |

Eng., 2022, 68(4):169-179 | DOI: 10.17221/83/2020-RAE A combined roaster and oil expeller was developed with locally available and accessible materials and the efficiency of the machine was evaluated. The obtained result shows that the efficiency of the machine is a function of the roasting temperature, the roasting duration, the moisture contents of the processed seeds and the machine feeding rate. The expeller is movable, simple in design for local fabrication, is easy to operate, requires few repairs and little maintenance and is cost effective. It is powered using a gear reduction electric motor of 5.5 Hp, the expeller has an average oil yield of 25.77% and an extraction efficiency of 70.26% and is designed to work for 8 hours per day of operation. The shaft diameter was designed to be 30 mm, while the roaster heater capacity was 2.8 kW. The designed machine is good for castor oil expression for both small- and medium-scale processing among rural and urban communities. |

This study evaluated the rice yield and components, the CH4 and N2O emissions and the global warming potential between the triple rice (R-R-R) and sesame-rice rotation (S-R-R) systems in Can Tho city, Vietnam. The experiments were conducted in 3 cropping seasons: Spring-Summer 2016, Summer-Autumn 2016, and Winter-Spring 2016-2017. The results showed that there was no significant difference in yield components and grain yield between triple rice and rotation systems. The application of sesame rotation in rice-based could reduce the CH4 and N2O emission by 30.5% and 18.7%, respectively. Global warming potential in the S-R-R rotation was 9860 kg CO2e/ha, significantly lower than the R-R-R rotation (12410 kg CO2e/ha) by 20.6%. These results show that the S-R-R rotation has the potential to mitigate GHG emissions, especially CH4, which contributes to a large amount of emissions in rice cultivation. |
Prediction of the soil organic carbon in the LUCAS soil database based on spectral clusteringOriginal PaperBaoyang Liu, Baofeng Guo, Renxiong Zhuo, Fan Dai, Haoyu ChiSoil & Water Res., 2023, 18(1):43-54 | DOI: 10.17221/97/2022-SWR The estimation of the level of the soil organic carbon (SOC) content plays an important role in assessing the soil health state. Visible and Near Infrared Diffuse Reflectance Spectroscopy (Vis-NIR DRS) is a fast and cheap tool for measuring the SOC. However, when this technology is applied on a larger area, the soil prediction accuracy decreases due to the heterogeneity of the samples. In this paper, we first investigate the global model performance in the LUCAS EU-wide topsoil database. Then, different clustering strategies were tested, including the k-means clustering based on the principal component analysis (PCA) and hierarchical clustering, combined with the partial least squares regression (PLSR) models, and a clustering based on a local PLSR approach. The best validation results were obtained for the local PLSR approach with R2 = 0.75, root mean squared error of prediction (RMSEP) = 13.38 g/kg and ratio of performance to interquartile range (RPIQ) = 2.846, but the algorithm running time was 30.05 s. Similar results were obtained for the k-means clustering method with R2 = 0.75, RMSEP = 14.61 g/kg and RPIQ = 2.844, at only 4.52 s. This study demonstrates that the PLSR approach based on k-means clustering is able to achieve similar prediction accuracy as the local PLSR approach, while significantly improving the algorithm speed. This provides the theoretical basis for adapting the spectral soil model to the needs of real-time SOC quantification. |

In the term of developing of components for electronic devices, it is important to provide the good physical properties of the used dielectric. The main parameters are optional and stable value of the relative permittivity and the minimum dielectric losses of the material. The paper is focused on testing of loss factor and relative permittivity of following materials: polylactic acid (PLA, in two dye modifications), polyethylene terephthalateglycol (PET- G) and acrylonitrile butadiene styrene (ABS) in the frequency range of 1-100 MHz. It was proven, that the values of permittivity of the tested materials were 2.9-4.2 and loss factor 0.8-4%. Concerning relative permittivity, the tendency to mild linear drop of relation was observed by increasing frequency, especially expressed in PLA materials. In loss factor, PLA materials displayed increasing values with increasing frequency, whereas the declining curve was observed in PET-G. Absolute value of ABS loss factor varied between 0.9-1.5%. The reasonable influence of added dyes was found out. |

Continuous versus intermittent enteral nutrition in hospitalised dogs and cats using a new scoring system: A prospective clinical studyOriginal PaperA Rado-Blozonova, L Rauserova-Lexmaulova, L Cerna, M Pavlik, M Rado, M FusekVet Med - Czech, 2023, 68(6):246-252 | DOI: 10.17221/26/2023-VETMED Nutrition is an important part of the critical care. The goals of this prospective clinical study were to create a scoring system for the assessment of patient nutritional status and to compare continuous and intermittent bolus feeding of enteral nutrition (EN). A total of 38 patients (21 dogs and 17 cats) were divided into Group C (continuous feeding; 23 patients) and Group I (intermittent feeding; 15 patients). The following variables were recorded for each patient in both groups: gastrointestinal (GI) complications, changes in body weight (BW), body condition score (BCS), muscle condition score (MCS), veterinary nutritional status score (VetNSS), length of hospitalisation and outcome. The normality of the data was assessed using the Shapiro-Wilk test. Fisher’s exact test, Mann-Whitney, Wilcoxon signed-rank tests, and the t-test were used in statistical analyses. Significant changes were found in VetNSS and BW between the 1st and 5th day in Group C. The VetNSS seems to be useful for monitoring the nutritional status of hospitalised patients. Anorectic dogs and cats can be successfully supported with either continuous or intermittent feeding methods with a similar risk of GI complications and outcomes. |
Presence of zoonotic black-pigmented periodontal pathogens in the oral microbiota of pet and stray catsOriginal PaperV Ozavci, HTY Dolgun, S KirkanVet Med - Czech, 2023, 68(2):62-68 | DOI: 10.17221/59/2022-VETMED Black-pigmented bacteria are one of the neglected species to cause periodontal disease in cats, and they are also zoonotic agents that pose an infection risk to humans. In this study, we aimed to determine the presence of Porphyromonas gingivalis, Porphyromonas gulae and Prevotella nigrescens in the oral microbiota of pet and stray cats. Dental swab samples were taken from 25 pet cats and 25 stray cats with symptoms of periodontal disease and then investigated by multiplex polymerase chain reaction using 16S rRNA species-specific primers. As a result of the multiplex PCR analysis, P. gingivalis 3/25 (12%), P. nigrescens 1/25 (4%), P. gingivalis + P. gulae 7/25 (28%), P. gingivalis + P. nigrescens 1/25 (4%), P. gulae + P. nigrescens 1/25 (4%), and P. gingivalis + P. gulae + P. nigrescens 2/25 (8%) were molecularly typed in the pet cats. In addition, 1/25 (4%) of P. gulae and 21/25 (84%) of P. gingivalis + P. gulae were typed in the stray cats. In 10/25 (40%) pet and 3/25 (12%) stray cat samples, no bacteria were detected by molecular typing. In summary, the results provide strong evidence that black-pigmented zoonotic pathogens are associated with cat periodontal disease. |
Verification of accuracy of qPCR method for intravital diagnostics of Macrorhabdus ornithogaster in avian droppingsOriginal PaperL Vrbasova, D Molinkova, P Linhart, Z KnotekVet Med - Czech, 2023, 68(2):69-74 | DOI: 10.17221/85/2022-VETMED The objective of this study was to verify the accuracy of the quantitative PCR (qPCR) method for in vivo detection of Macrorhabdus ornithogaster. A total of 100 faecal and gastric mucosa samples from avian cadavers were investigated, using cytological and qPCR techniques (budgerigars, Fischer’s lovebirds, red-crowned parakeets, scarlet-chested parrots, eastern rosellas, domestic canaries, zebra finches, white Java sparrow). Using qPCR, the probability of detecting positive samples of droppings was significantly higher than in the faecal smear microscopy (P < 0.01). Cytology detected the presence of Macrorhabdus ornithogaster in 34 faecal samples, whereas qPCR detected 54 positive samples. In all 46 qPCR negative faecal samples, gastric smear qPCR was performed and also yielded negative results. Gastric smear qPCR was also performed in 20 cadavers where faecal qPCR has detected the presence of Macrorhabdus ornithogaster and in all samples confirmed the positive result. This verifies the accuracy of faecal sample qPCR for intravital diagnostics. Overall, the faecal qPCR technique appears to be extremely reliable, as it made it possible to detect all infected individuals, including those with negative stool or gastric cytology. |

Echocardiographic and echo-Doppler measurements were performed on twenty healthy adult Heterodon nasicus (10.10). The values of the ventricular length, aortic diameter, pulmonary trunk diameter, the mean thickness of the interventricular septum, and thicknesses of the wall of the cavum pulmonale (Cav. P) and cavum arteriosum (Cav. A), were measured. The aortic flow and pulmonary trunk flow were recorded. Two dead specimens (1.1) were dissected. The male’s pulmonary trunk diameter was bigger compared to the female’s in both the long and short axis. The reproductive ecology of Heterodon nasicus has yet to be fully elucidated upon, however, male territorialism and dispersal from the hibernacula, and multiple male courtships toward a single female were described, hence, the more active reproductive activity of the male and the consequent sexual selection toward a higher aerobic performance can be hypothesised. A moderate interventricular right to left shunt was noticed in the Cav. V of all the specimens, which is considered normal and should not confuse the clinician. Congenital defects, cardiomyopathies, valvulopathies, and pericardial diseases are known to occur in ophidians and other reptiles. Reliable data and profound knowledge of the anatomy and physiology of the ophidian heart are fundamental for the in vivo diagnosis of cardiac diseases in snakes. |

Screening and quantification of pesticide residues in ciders by liquid chromatography-high resolution mass spectrometryOriginal PaperVeronika Zu¹»áková, Martin Du¹ek, Vladimíra Jandovská, Jana Ol¹ovskáCzech J. Food Sci., 2023, 41(1):29-35 | DOI: 10.17221/158/2022-CJFS The present study aimed to apply a multi-residue method for the screening and quantification of pesticide residues in ciders, low alcoholic beverages made by fermentation of apple juice. Twenty bottled craft cider samples purchased from the Czech market were analysed for pesticide residues. The residues of pesticides were extracted from samples using the QuEChERS (Quick, Easy, Cheap, Effective, Rugged, and Safe) procedure in combination with additional solid-phase extraction (SPE) sample clean-up to achieve the lowest detection limit possible. In this study, targeted screening of pesticide residues in the samples was performed with the Q-Orbitrap mass spectrometry instrument. We identified 18 pesticides in cider samples analysed by screening method using an accurate-mass database of about 500 pesticide compounds, including their retention times, empirical formulas, and characteristic fragments. Additionally, liquid chromatography with high-resolution tandem mass spectrometer (LC-HRMS/MS) was used for re-analysis of positive findings of pesticides in samples and allowed to quantify compounds of interest at 0.2 µg L–1 concentration level. The monitoring scheme was applied to the set of craft ciders, and the results revealed the presence of pesticide residues in most of the samples at trace levels ranging from 0.5 to 5 µg L–1 and rarely at a level higher than 10 µg L–1. |

Identification and functional characterization of a new terpene synthase gene from Chrysanthemum indicum var. aromaticumOriginal PaperYu Liu, Wenyue Ji, Xiang Wang, Bin Xia, Miao He, Yunwei ZhouHort. (Prague), 2023, 50(1):61-71 | DOI: 10.17221/61/2022-HORTSCI Terpene synthase (TPS) is a key enzyme in the synthesis of terpenoids in plants and plays an important role in the regulation of the synthesis of aromatic substances in plants. In this study, we treated Chrysanthemum indicum var. aromaticum as the experimental material to clone the CiTPS gene. The open reading frame of CiTPS is 1 818 bp, encoding a putative protein of 605 amino acids. There is a monoterpene synthase conserved domain and a highly conserved sequence, DDXXD, (N,D)D(L,I,V)X(S,T)XXXE and RRX8W, respectively, which belong to the monoterpene synthase family. The multiple sequence alignment and phylogenetic analysis showed that the CiTPS gene belongs to the TPS-b subfamily. The pBI121-TPS-GFP recombinant plasmid and the pBI121-GFP empty vector plasmid were introduced into Agrobacterium by electroporation and transferred into wild-type tobacco by the Agrobacterium-mediated method. Three transgenic CiTPS lines were screened, and the secretions of wild-type tobacco and transgenic tobacco T1 leaves in the vigorous growth period were analysed. The volatiles of the transgenic lines were similar to those of wild-type tobacco, but their monoterpenes increased significantly, and the sesquiterpenoids and diterpenoids decreased or remained unchanged, indicating that the CiTPS gene is related to the synthesis of the monoterpenes. |
Change in β-glucosidase activity in root zone of ferns under toxic elements soil contaminationOriginal PaperMilan Novák, Veronika Zemanová, Milan Pavlík, Simona Procházková, Daniela PavlíkováPlant Soil Environ., 2023, 69(3):124-130 | DOI: 10.17221/448/2022-PSE The influence of toxic elements, such as arsenic (As), cadmium (Cd), lead (Pb), and zinc (Zn), in the root zone of As-hyperaccumulator Pteris cretica 'Albo-lineata' and non-As-hyperaccumulator P. straminea, on the enzymatic activity of β-glucosidase, dissolved organic carbon (C) in soil, toxic element accumulation in fern roots, and root biomass were evaluated in a pot experiment. Ferns were cultivated in soils from the locality of Suchdol (control) and Litavka (high contamination) for six months. For all toxic elements, an increasing trend in their contents in the roots was observed with soil contamination for both ferns. Differences between ferns were observed in As and Zn accumulation. Pteris cretica had a significantly higher As accumulation than P. straminea. Zinc accumulation in the roots showed an opposite trend. A significant difference between ferns was confirmed in the dissolved organic C content. Our results showed a significantly higher content of dissolved organic C in the P. straminea root zone than in P. cretica. The significant effect of toxic elements in the soil on β-glucosidase activity was observed. Toxic elements inhibited β-glucosidase activity in the root zone of P. cretica, and an increase in P. straminea was determined in the Litavka soil. The results suggest a higher sensitivity of P. straminea to toxic element contamination in soil, leading to increased β-glucosidase activity and increased dissolved organic C content. |

In this experimental study, the efficacy of both cold atmospheric plasma/nitric oxide (NO) and NPH insulin ointment, recently known to have beneficial effects on wound healing, was investigated in diabetic wound healing. Twenty-four (24) diabetic rats were divided into four groups DC, DI, DNO and DINO (diabetic control, diabetic insulin, diabetic nitric oxide, diabetic insulin + nitric oxide groups). No treatment was applied to the DC group, NPH insulin was applied to the DI group, CAP/NO was applied to the DNO group, and CAP/NO + NPH insulin was applied to the DINO group once daily for 14 days. The wound area reduction and the wound contraction rate were calculated on the basis of the tissue sections taken, and histopathological and genetic analyses were carried out. Compared to the control group, exogenous NO gas was found to be a potent antibacterial agent in the diabetic wound healing, causing a reduction in the wound area (P = 0.034), an increased contraction rate (P = 0.021), epithelialisation (P = 0.02), collagen organisation (P = 0.006) and a reduction in the number of inflammatory cells (P = 0.002). A significant increase in the expression of IL-8 mRNA was observed (P = 0.026). It was concluded that NPH insulin alone contributes to wound healing, but it is not necessary to use it together with exogenous NO gas. |

Seven crops were tested: lettuce (Lactuca sativa L.) cv. Novosadska majska maslena, oil pumpkin (Cucurbita maxima Duch) cv. Olivija, oilseed rape (Brassica napus L.) cv. NS Ras, pepper (Capsicum annuum L.) cv. Kurtovska kapija, soybean (Glycine max (L.) Merr) cv. ZP Laura, sunflower (Helianthus annuus L.) cv. NS Kruna, and tomato (Solanum lycopersicum L.) cv. Dunavski Rubin. Herbicide dicamba in the range of 0.14 to 1 155.6 g a.i. (active ingredient)/ha inhibited biomass, height, leaf area, and visual injury of all crops, while glyphosate doses from 0.48 to 3 840 g a.i./ha also reduced the growth of all tested species. A rate of 116 g a.i./ha mesotrione was needed to reach 80% visual injury in oilseed rape, while the same effects on lettuce only required 1.8 g a.i./ha of mesotrione. Tomato and oil pumpkin were also sensitive to low mesotrione doses, where only 1.3 g and 0.5 g a.i./ha of mesotrione was needed for 80% of biomass reduction, respectively. Lettuce was the most sensitive crop of all tested species; biomass was reduced by 80% by dicamba, glyphosate, mesotrione, and nicosulfuron at the low rates of 33 g a.i./ha, 19 g a.i./ha, 1.25 g a.i./ha, and 2.7 g a.i./ha, respectively. Among all herbicides, visible injuries were detected in dicamba at the lowest rates. Soybean was the most tolerant of glyphosate, mesotrione, and nicosulfuron. Based on the available literature and obtained results, herbicide off-target movement must be mitigated to maximise herbicide efficacy and decrease the negative influence on susceptible plants and the environment. |

This task is more difficult to perform in high-mountainous landscapes, at an altitude of more than 4 000 m a.s.l. Moreover, these high-mountain ecosystems are the most vulnerable to various kinds of anthropogenic impacts, and therefore the relevance of bioremediation is obvious. The research was conducted in the high-altitude ecosystems of the Kyrgyz Republic at the Kumtor mine. In this study was carried out on the bioremediation of oil contaminated soil using biostimulation, bioaugmentation and biostimulation + bioaugmentation remediation techniques for 90 days in the climatic conditions of high mountain region. The biostimulation treatment showed the highest total petroleum hydrocarbons (TPH) biodegradation percentage 62.78% compared to the bioaugmentation 50.63% and biostimulation + bioaugmentation 49.11%. Thus, the method of biostimulation proved to be the most effective method for bioremediation of soils contaminated with petroleum products. The application of this method could be one of the successful methods of recycling contaminated soils. This study demonstrated the possibility of restoring TPH-polluted soils using biological methods of soil treatment in climatic cold conditions of high mountains. |

Food Sci., 2023, 41(2):118-126 | DOI: 10.17221/54/2022-CJFS his study optimised the conditions for ultrasound-assisted extraction (UAE) of defatted wheat germ protein (WGP) and evaluated its effect on the functional properties. Single-factor and orthogonal experiment designs were combined to optimise the UAE extraction condition. The extraction of WGP reached the highest level, at 88.66%, with a solid : liquid ratio of 1 : 25 g·mL–1, pH value of 9.0, ultrasonic time of 10 min, and ultrasonic power at 400 W. Under these conditions, albumin, globulin, prolamin, and glutenin accounted for 32.26, 28.52, 5.42, and 22.40% of total protein, respectively. In addition, this study compared the functional properties of WGP extracted by UAE with the results based on a commercially available soy protein (SP) isolate (SPI). The UAE of WGP had better oil absorption, foaming, and emulsifying properties. Therefore, UAE is a promising technique for food protein extraction because it can change the protein efficiencies and properties of the extract. |
Network-based business model in the agri-food sector: A case study of Green FingersCase StudySagit Barel-ShakedAgric. This research study examines a network-based business model of a new venture offering services to avocado farmers and its potential impact on customer performance. A theoretical framework of a network-based business model in the agri-food sector was developed and implemented in the production of avocados. The Israeli agri-food firm, Green Fingers, was selected as the case study to analyse the value drivers of the network-based business model. The proposed business model can serve as a benchmark in evaluating other business models and developing practical strategies in the agri-food sector. The model is flexible, adaptable in meeting the evolving needs of customers and farmers, and applicable to other sectors. The model's building blocks emphasise the importance of networking and diffusing knowledge among key stakeholders, adapting to changing environments, maintaining environmental sustainability, collaborating with policymakers, and supporting farmers. The extended business model canvas may enhance the currently limited research on network-based business models in the agri-food sector from theoretical and empirical perspectives. |
Effect of glyphosate on the foraging activity of European honey bees (Apis mellifera L.)ReviewAneta Bok¹ová, Jan Kazda, Jan Barto¹ka, Martin KamlerPlant Soil Environ., 2023, 69(5):195-201 | DOI: 10.17221/86/2023-PSE Glyphosate is a widely used agrochemical. Nevertheless, only a few studies have investigated its effect on bees, specifically its influence on their foraging activity. This article provides a summary of the prominent research results on this issue, published in journals in the field of experimental biology. The effect of commonly used concentrations of glyphosate on honey bee navigation has been evaluated in several studies, as well as concentrations that are reportedly sublethal. Exposure to this herbicide increases the flight time back to the hive and affects the flight trajectories of these bees. These results imply that glyphosate at certain concentrations reduces their sensitivity to nectar rewards in associative memories. The contact of bees with non-lethal concentrations of glyphosate results in sublethal effects that affect foraging. In the future, the behaviour of glyphosate and its effect on bees in their natural environment need to be explored. |
Effects of Pseudomonas chlororaphis strain AFS009 and Beauveria bassiana strain GHA against Plumeria rust in HawaiiShort CommunicationPhilip Waisen, Zhiqiang Cheng, Richard CrileyPlant Protect. Sci., 2023, 59(2):202-207 | DOI: 10.17221/86/2022-PPS This study examined the effects of beneficial rhizobacterium Pseudomonas chlororaphis strain AFS009 and entomopathogenic fungus Beauveria bassiana strain GHA against plumeria rust Coleosporium plumeriae. Two preemptive or three curative laboratory experiments and a curative field experiment were conducted to examine the effects of these commercially available biocontrol products. Treatments included the application of B. bassiana at 1.23 g/L and P. chlororaphis at 3 or 9 g/L. Systemic fungicide azoxystrobin applied at 0.12 g/L and water were included as positive and negative controls, respectively. While its effect was insignificant in the field trial, B. bassiana reduced the rust pustule development in one of two preemptive and two of three curative laboratory trials. In contrast, P. chlororaphis applied at 9 g/L suppressed the number of rust pustules in both laboratory and field experiments, demonstrating its potential biological activity against plumeria rust. In the field trial, the effect of P. chlororaphis was observed at 14 days post-treatment, suggesting that an application interval of 14 days on infected plants can take the rust under control. |
Detection of Coxiella burnetii and characterisation by multiple-locus variable-number tandem repeat analysis in bovine bulk tank milk samplesOriginal PaperB Yanmaz, EK OzgenVet Med - Czech, 2023, 68(5):185-190 | DOI: 10.17221/87/2022-VETMED Coxiella burnetii is the aetiological agent of Q fever, which is highly prevalent in Turkiye, but information on the genetic profiles of the bacterium is limited. This study aimed to investigate the presence of C. burnetii in bovine bulk tank milk (BTM) samples by polymerase chain reaction (PCR) and to investigate the genotypes by means of multiple-locus variable-number tandem repeat analysis (MLVA). A total of 25 markets that sold raw cow’s milk were analysed by conventional PCR analysis. An MLVA analysis was performed at six loci, namely MS23, MS24, MS27, MS28, MS33, and MS34, to determine the genotypic variations of C. burnetii found in the positive DNA samples. The DNA of C. burnetii was detected in 16% of the BTM samples. The C. burnetii strains identified in the bovine milk samples collected in this study were found to belong to the same genotypic group as those detected in the bovine milk samples gathered in Greece. As a result, both the presence and genotyping studies of C. burnetii on the BTM samples in Turkiye will contribute to the determination of the geographical distribution of the agent. |
