Food Sci., 2019, 37(5):366-373 | DOI: 10.17221/57/2018-CJFS The effect of taxifolin (TXF) with starter cultures (SC), such as Leuconostoc carnosum, or a mixture of strains Pediococcus pentosaceus and Staphylococcus xylosus, on the TXF stability was evaluated. UPLC analysis demonstrated that after 181 days of storage total TXF content was the highest in samples with TXF and L. carnosum (60%), compared to the 1st day of storage. The sausages with TXF and the mixture of P. pentosaceus and S. xylosus (56%) followed next. The samples treated only with TXF retained 40% of TXF, compared to the 1st day of storage. TXF had no significant effect on the growth of lactic acid bacteria. The accumulation of biogenic amines (BA), including histamine and putrescine, was more effectively reduced in sausages inoculated with the TXF plus P. pentosaceus and S. xylosus mixture. Using this mixture, the rate of lipolysis and processes of lipid oxidation were effectively slowed down. Fatty acid (FA) composition was stable in all cases. |

Evaluation of genetic variability within actual hop (Humulus lupulus L.) cultivars by an enlarged set of molecular markersShort CommunicationJosef PATZAK, Alena HENYCHOVÁCzech J. Genet. Plant Breed., 2018, 54(2):86-91 | DOI: 10.17221/175/2016-CJGPB Traditional hop (Humulus lupulus L.) cultivars have been used in the brewing industry for a long time. Globally, about ten new breeding lines were released to the market in each decade from ~1970 to 1999. Since 2006, the rate of release of new cultivars has increased tenfold. It is, therefore, important to identify their genotype and origin. Molecular genetic methods based on DNA are the most appropriate technology for this purpose. Recently, we developed an efficient marker system for the authenticity control of hop genotypes based on expressed sequence tag-simple sequence repeats (EST-SSR). In the present study, we enlarged the previously established EST-SSR set with 27 new polymorphic markers and evaluated molecular genetic variability within 135 traditional and new world hop cultivars. Two sets of 10 markers effectively differentiated all used cultivars, with the exception of cultivars derived from the same original genotype such as Saaz, Spalt, Tettnang and Nadwislawsky. Results of molecular genetic variability analyses corresponded with the genealogical and geographical origin of the key cultivars. |

New generation sequencing allowed single nucleotide polymorphism (SNP) chip technologies for genotyping, description of functional apple genes, characterization of the evolutionary results in apple genome fragment transition, as well as phylogenetic reconstruction of the genus Malus, being the confirmed progeny of M. sieviersii. Based on these technologies, newly developed putative markers may give the most important biological data such as age, geographical origins, tissue type determination, and external visible characters. The new generation genotyping platforms, representing very high efficiency, are now successfully applied for random apple genome-wide association (GWA) studies as well pedigree-based analysis and marker-assisted selection (MAS). |

Characterization of one new non-S-RNase of Armeniaca cathayanaShort CommunicationMengpei Liu, Pei Hou, Xiaoyuan Wang, Yu Dong, Wei ZongCzech J. Genet. Plant Breed., 2019, 55(1):39-41 | DOI: 10.17221/15/2018-CJGPB Armeniaca cathayana, a new species described in 2010, belongs to gametophytic self-incompatibility (GSI) system which is under S-allele control. One new non-S-ribonuclease (non-S-RNase) was found in A. cathayana through comparing its nucleotide and amino acid sequences with sequences of the S-allele in Genbank. The BLAST analysis showed that the one new non-S-RNase S68-RNase (GenBank Accession No. MH155952) had the highest 96% nucleotide sequence homology with Prunus webbii non-S-RNase PW1 (EU809938.1). Alignment of deduced amino acid sequences of A. cathayana S68-RNase shared 83% similarity with P. webbii PW1. The new non-S-RNase determined in this study will provide new information to GSI of Rosaceae. |
Relationship between abnormal spermatozoa and seminal plasma free amino acids in boarsJ. Čeřovský, S. Frydrychová, A. Lustyková, M. RozkotCzech J. Anim. Sci., 2007, 52(2):44-49 | DOI: 10.17221/2346-CJAS The objective of this study was to provide some data on concentrations of seminal plasma free amino acids (FAAs) in order to compare these values with different total contents of morphologically abnormal spermatozoa (MAS) in the semen of 37 experimental adult boars. The boars were kept in the same housing, feeding and breeding conditions. Immediately after collection, semen gel free volume, sperm concentration and total MAS were determined microscopically. The boars were divided into two groups (A - n = 24, B - n = 13) according to the significantly different total MAS content (x: A = 9.46 ± 4.95, B = 46.00 ± 14.54%, P < 0.01). Deproteinized seminal plasma samples (supernatant) were analysed for concentrations of 13 FAAs: Tau, Asp, Thr, Ser, Glu, Gly, Ala, Val, Met, Ile, Leu, Tyr and Lys by liquid chromatography (AAA 339 M analyser). A highly significant correlation was determined between total MAS and total FAA content in all experimental boars (n = 37, r = -0.60, P < 0.01). Glutamic acid was a predominant FAA in seminal plasma in both groups of boars (x: A = 28.49 vs. B = 17.60μM/100 ml) but its concentration was significantly higher in group A (P < 0.01). The proportion (%) of glutamic acid concentration in the total content of FAAs in both groups of boars was nearly equal (A = 38.1 vs. B = 38.9%, P > 0.05) as well as the proportion index of the individual FAAs calculated from glutamic acid (= 100%). The differences in the particular FAAs recorded between group A and group B were statistically significant for 11 out of the 13 FAAs observed (P < 0.05 and P < 0.01) in favour of group A as well as in the total calculated content of FAAs per boar (x: 74.70 vs. 45.23μM/100 ml, P < 0.01). A significantly negative relationship between the MAS content and the concentration of seminal plasma FAAs (r = -0.60, P < 0.01) is the main result of this study with regard to the markers for potential boar semen fertility estimation. |
RE-NUM-OR: Python-based renumbering and reordering software for pedigree filesOriginal PaperKemal YazganCzech J. Anim. Sci., 2018, 63(2):70-77 | DOI: 10.17221/64/2017-CJAS RE-NUM-OR is a new, flexible, and user-friendly renumbering and reordering data arrangement software for unprocessed pedigree files used in genetic evaluation systems for personal computers. RE-NUM-OR was written using Python (Ver. 2.7.13) 64 byte programming language and compiled with PyInstaller-3.2.1 software which is a set of utilities for freezing Python scripts into executable. RE-NUM-OR not only runs on 32 or 64 byte MS Windows but also runs on 64 byte GNU/Linux. The program has new, practical, and simple interface and the user does not need to create a parameter file for running the program and .txt, .xlsx or .et extension files can be used as input files directly. Output files (.txt, .xlsx, .et, .dat) can be obtained. For animal, sire and dam IDs, it can read all standard characters (ASCII codes from 32 to 126) in input files. The program supports both dot (.) and comma (,) for numerical data. Pedigree lines for parents do not need to be arranged prior to their progeny. If pedigree lines for parents follow their progeny, the program can detect this and it can reorder the animals as pedigree lines placing parents prior to their progeny. Another feature of this software is that it has a pop-up window with error notification. Also, it supports repeated observations. Comparison of different semen extenders and cryoprotectant agents to enhance cryopreservation of rabbit spermatozoaOriginal PaperPaula Domingo, Maite Olaciregui, Noelia González, Ignacio De Blas, Lydia GilCzech J. Sci., 2019, 64(2):59-66 | DOI: 10.17221/53/2018-CJAS The purpose of this research was to find a suitable protocol to enhance frozen rabbit sperm preservation analysing the role that seminal plasma (SP) plays and the effect of different cryoprotectant agents on sperm quality 0 and 2 h after thawing. Sperm samples were pooled and divided in eight fractions. Four of them were diluted with BotuCrio® (extender A), INRA 96® plus 6% glycerol (extender B), 6% N, N-dimethylformamide (extender C) and 6% N-methyl-2-pyrrolidone (extender D), respectively. The other four fractions were centrifuged and the supernatant was discarded in order to eliminate SP. Each sample was then resuspended with extender A, B, C and D. Samples were cooled progressively, loaded into 0.5 ml freezing straws and frozen with liquid nitrogen vapour. Thawing was performed by placing the straws into a bain-marie at 37°C for 21 s. Straws were dried and sperm samples placed into Eppendorf tubes to be analyzed by ISAS software, vitality test, HOS test and acrosome integrity test. The best motility and velocity parameters were obtained by extender A (P < 0.050) even when the motility parameter was compared with previous studies using other diluents. Additionally, sperm quality decreased over incubation time (P < 0.050) and no differences were found in samples processed with or without SP. This research revealed that BotuCrio® could be used for rabbit sperm cryopreservation and moreover the improvement of the cryopreservation process of rabbit sperm due to the demonstration that SP removing is not required. |

However, under cold and/or moderate ambient temperature, these genes might possess different behaviour. The main objective of this study was to evaluate laying performance, immune response, and antioxidant status of native laying hens segregating for naked neck (Na) and frizzle (F) genes under low ambient temperature. Five genotypes were studied: homozygous naked neck (NaNaff), heterozygous naked neck (Nanaff), homozygous frizzle (nanaFF), heterozygous frizzle (nanaFf), and normally feathered (nanaff). The hens were raised under temperature range 22.2-16.7°C. No adverse effect due to ambient temperature was detected in laying performance for naked neck genotypes. Significant decrease in egg weight was recorded in nanaFF genotype compared to the other genotypes leading to significant decrease in egg mass. Significant improvement in shell thickness was associated with Na and F genes. NaNaff genotype had a significantly higher eggshell strength compared to nanaff. Furthermore, Na and F genes improved cellular mediated immune responsiveness, whereas this improvement did not extend to humoral immunity. Birds carrying F gene in homozygous state had a higher total antioxidant activity compared to the remaining genotypes. It could be concluded that the presence of Na and F genes in laying hens raised under low ambient temperature significantly increased shell thickness and, in turn, improved shell strength. Moreover, they greatly enhanced cellular immunity, particularly in heterozygous naked neck status. |
Effect of the composition of starter diet fed in the rearing phase on the performance and certain physiological parameters of Holstein calvesOriginal PaperSzandra Tóth, Melinda Kovács, Brigitta Bóta, Judit Szabó-Fodor, Gábor Bakos, Hedvig FébelCzech J. Sci., 2019, 64(9):367-376 | DOI: 10.17221/34/2019-CJAS The objective of this study was to compare the effects of calf starters of different starch and fibre content on the growth, blood and rumen parameters of Holstein female calves during the pre-weaning period. A total of 60 calves were divided into two groups. Group A/B was fed according to a two-phase calf nutrition system: the calves received calf starter "A" (28.5% starch content, 16% neutral detergent fibre (NDF), 9.2% acid detergent fibre (ADF)) in days 7-45 and then calf starter "B" (14.2% starch content, 32.4% NDF, 14.3% ADF) in days 46-70. Calves of group B received calf starter "B" throughout the experimental period. The calves were weaned between 52 and 61 days of life. The data on body weight, weight gain, starter feed intake, blood metabolites and rumen fermentation parameters were collected individually. In the first phase of experiment, the starter feed consumption of A/B group was higher than that of B group (P < 0.05), but it did not manifest in differences in body weight at weaning time. At some of the sampling times, the different composition of starter diets significantly (P < 0.05) influenced the plasma urea, albumin, glucose and triglyceride concentrations. The consumption of diet with different starch and NDF content had no significant effect either on the composition of the rumen microbiota or on the ruminal concentration of volatile fatty acids (VFA) and ammonia. In some cases significant correlations (-0.83 and -0.93; 0.82 and 0.90) were found between the composition of rumen microbiota and ruminal VFA concentration. Feeding a diet of higher starch content (28.5 vs 14.2%) in the first 45 days of life did not affect rumen fermentation, rumen microbiota and the most important blood metabolites, apart from a few exceptions. The production parameters of calves, including total feed intake and body weight gain, were similar in the experiment. |

Sci., 2019, 64(12):491-503 | DOI: 10.17221/226/2019-CJAS This study aimed to evaluate the impact of selection on the genome structure of beef cattle through identification of selection signatures reflecting the breeding standard of each breed and to discover potential functional genetic variants to improve performance traits. Genotyping data of six beef breeds (Aberdeen Angus, Hereford, Limousin, Charolais, Piedmontese and Romagnola) were used to perform genome-wide scans for selection signatures. The approaches applied were based on an assumption that selection leads to linkage disequilibrium or to a decrease of genetic variability in genomic regions containing genotypes connected with favourable phenotypes. Thus, the selection signatures were analysed based on Wright's FST index, distribution of runs of homozygosity segments in the beef genome and determination of linkage disequilibrium variability between breeds. The number and length of detected selection signals were different depending on the breeds and methodological approaches. As expected due to the breeding goals of analysed breeds, common signals were located on autosomes 2, 6, 7, 13 and 20 close to the genes associated with coat colour (KIT, KDR), muscle development (GDF9, GHRH, GHR), double muscling (MSTN), meat tenderness (CAST) and intramuscular fat content (SCD). But, across the genomes of analysed breeds, unique selection signals were found as well. The subsequent analysis of those single nucleotide polymorphism markers can be beneficial for the genetic progress of studied breeds in future. |
First characterisation of minor and neglected Vitis vinifera L. cultivars from Mount EtnaOriginal PaperFilippo Ferlito, Elisabetta Nicolosi, Stefano La Malfa, Antonio Cicala, Alessandra GentileHort. (Prague), 2018, 45(1):37-46 | DOI: 10.17221/198/2016-HORTSCI Eight minor and neglected cultivars of Vitis vinifera L. were characterised according to their ampelographic and agronomic traits and discriminated by molecular analysis with SSR markers. These results are the first complete ampelographic description of these minor and neglected cultivars from the Mount Etna region. The results of this study reveal high morphological diversity of ancient grapevines growing in this region. SSR markers enabled us to discriminate the cultivars and revealed the genetic divergence between them and several autochthonous cultivars. Our efforts could contribute to a better knowledge of grape biodiversity based on morphological and molecular data and could be useful for the development of a reliable germplasm conservation strategy. |
Cold hardiness of peach flowers at different phenological stagesOriginal PaperLászló Szalay, Imre Gergő Gyökös, Zsuzsanna BékefiHort. Sci. (Prague), 2018, 45(3):119-124 | DOI: 10.17221/146/2016-HORTSCI At the colder peach production regions it is important to know the cold hardiness of peach cultivars at different phenological stages of flowering. In our experiment, artificial freezing tests were conducted in a climate chamber in five selected years between the period of 2007 and 2016 to determine the freeze tolerance of generative organs of three peach cultivars ('Venus', 'Redhaven', 'Piroska') at different phenological stages of bloom. Based on the results of the laboratory freezing tests LT50 values were calculated. Our results showed that LT50 values of examined peach cultivars in swelled bud stage averaged over five years were between -6.8 and -11.2°C according to cultivar, and as phenological phases progressed, cold hardiness of generative organs decreased. At the end of bloom LT50 values varied between -1.7 and -4.1°C. Cultivar 'Piroska' had the highest freeze tolerance and cultivar 'Venus' showed the lowest in each year studied. This study shows that trees with delayed development are more prone to cold damage to flowers. |
Effect of planting time and supplemental irradiation on growth and flowering of Lachenalia 'Romaud'Original PaperAnna KapczyńskaHort. Sci. (Prague), 2019, 46(2):72-80 | DOI: 10.17221/203/2017-HORTSCI Growth and flowering of lachenalia 'Romaud' was studied with reference to its commercial potential as pot plant and the need to obtain flowering plants at a specific time. The experiment was carried out in a heated glasshouse. Lachenalia bulbs were planted in November, December, January and February. The plants were exposed to two lighting regimes, natural lighting and natural lighting with supplemental irradiation (HPS lamps). The later the planting date was, the faster the bulbs flowered, and they produced thicker inflorescence stems with greater number of florets. Depending on the bulb planting date and light conditions, the plants flowered from February to May. The leaves obtained from the bulbs planted in November and December were longer than those produced by the bulbs planted in January and February. Compared with control, supplemental irradiation accelerated flowering by 10-13 days and positively affected plant features by promoting the growth of thicker inflorescence stems with more abundant and longer florets. The leaves of irradiated bulbs were shorter (apart from the bulbs planted in February) and were characterised by a higher content of chlorophyll a, chlorophyll a + b and carotenoids as compared with control. Plants grown under HPS light also had the higher dry weight of bulbs, leaves and stems. |

(Prague), 2019, 46(3):138-145 | DOI: 10.17221/218/2017-HORTSCI Research in southern Italy assessed the effects of biodegradable mulch on fruit yield and quality of two greenhouse tomato cultivars, 'Coronel F1' and 'Kero F1'. Three mulching types (two MaterBi biodegradable black films, MB N2/12 amnd MB N8; black polyethylene film, low-density polyethylene (LDPE)) and not mulched control were compared. 'Coronel F1' showed higher values of fruit yield, total crop biomass and leaf area index (LAI). MB N8 and LDPE films led to the highest fruit yield and growth indexes, whereas not mulched control to the lowest. Fruit dry residue and soluble solids were highest under MB N2/12 and MB N8, titratable acidity was highest under MB N8. Fruits grown under MB N8 and LDPE mulches attained the highest levels of colour components "L" and "b" respectively, and MB N8 the highest fruit firmness. MB N2/12 and MB N8 showed the highest levels of antioxidants and antioxidant activity. Biodegradable polymers improved root growth conditions and fruit quality, showing suitable features for sustainable vegetable production. |

Freshly harvested asparagus is available only during a short part of the year and the demand varies. The produce is perishable and only proper handling can ensure its quality after a reasonable shelf life. In this study, we investigated a new method to reduce one of the most important reasons for quality decline: fresh matter losses. The potential benefits or disadvantages of moist asparagus packaging for unpeeled white asparagus (cv. 'Gijnlim') was tested. We used polythene sleeves or bags, lined with cellulose fleece to hold added water for moisturizing. 500 g bunches of spears were filled in the bags. Spears were partly covered by the sleeves or totally covered by the bags. All spears were stored for 3 to 11 days at the temperature of 2°C, 8°C and a combination of 2°C/22°C.The last temperatures were to simulate retail conditions. In wet bags spear fresh weight increased, losses in bags with dry fleece were low. Maintaining of fresh weight in wet bags coincided significantly with higher bending firmness, and lower shear force, which points to less fibrousness. Wet storage also reduced the incidence of dried cut surfaces but promoted the incidence of lengthwise fractures and, in combination with higher temperature, grey-brown discolouration and colour saturation. Wet packaging may be beneficial primarily under low temperature and low air humidity storage conditions. |

(Prague), 2019, 46(4):171-179 | DOI: 10.17221/12/2019-HORTSCI The promising branching agent cyclanilide [1-(2,4-dichlorophenylaminocarbonyl)-cyclopropane carboxylic acid] is not permitted in the EU and the low vigour rootstocks preferred by growers make the branching of young sweet cherry (Prunus avium) trees difficult in the temperate zone production regions. To increase the number and length of new branches ("feathers") on such trees, chemical (6-benzyladenine [BA], gibberellic acid [GA4/7]), mechanical (leaf pinching, summer notching) and various combined treatments were tested from 2015-2018. The numbers of feathers divided into three different lengths (1-10 cm, 10-30 cm and > 30 cm) were assessed as the main indicator of quality, supplemented with other morphological characteristics. Summer notching induced an insufficient number of feathers and led to some negative effects on the trees. The number of feathers from BA or BA with GA4/7 treatment also was insufficient. The effect of pinching alone was variable. The number of all feathers generally was highest when pinching was combined with BA, regardless of the year or cultivar. |
Snow damage to birch stands in Northern MoraviaA. Martiník, O. MauerJ. For. Sci., 2012, 58(4):181-192 | DOI: 10.17221/48/2011-JFS The condition and snow damage to stands dominated by birch were studied in the area of interest in Northern Moravia. Based on basal area, the share of birch ranged from 68 to 88%. The stands came to existence on fertile sites through natural regeneration and exhibited growth at a level of the best site classes for the Czech Republic. The most severe snow damage to the stands was recorded at the upper stand height of 8-15 m. The extent of damage in the analyzed stands amounted to 67-95% of all trees. The unambiguously predominant type of damage was bending while breakages were recorded only in up to 4% of trees. Slenderness ratios of intact trees differed according to the age (height) of stands. The highest values of about 180 were observed in the youngest, ca 5-years-old and about 5 m high stand. Slenderness ratio values of intact trees in a 17-m high stand ranged about 100. The analyzed stands did not show any differences in the slenderness ratio values between intact and damaged trees. Lower values of the ratio for solitary trees (ca 130) in a 5-years-old stand were accompanied by markedly longer crowns (80% of stem) as compared with trees growing in the stand (60% of stem). Differences in the root system architecture were revealed between intact and damaged trees within a stand of about 9 m in height. |

The effect of the Sequoiadendron giganteum (Lindl.) Buchholz cone crystals on germinationOriginal PaperHana PrknováJ. For. The inhibitory effect of this extracted material on Norway spruce, Scots pine and European larch germination was newly examined. Sown seed representative samples without and with added cone crystals were compared after their incubation in the same appropriate conditions. All these cases have brought compelling evidence that cone crystals totally inhibit germination. However, the chemically inhibited seeds being rinsed afterwards germinated very well. This switch effect caused by the cone crystals of the three above-mentioned conifers proved to be a nonspecific tool. Seeds of the giant sequoia, naturally affected by the cone crystals, were sown as rinsed and non-rinsed samples. Both the samples, grown on a wet peat substrate in appropriate conditions for 16 weeks, exhibited an equal, yet very low viability of 1.3% at the same time. This low number, fixed in seeds of the tree being far from the ecological optimum, does not allow any disputation with other authors. |

Sci., 2018, 54(3):129-137 | DOI: 10.17221/154/2016-PPS The Bacillus amyloliquefaciens Y1 strain was evaluated for its effects on soil properties, pepper seedling growth, rhizosphere bacterial flora and soil enzyme activities. Y1 solubilised insoluble phosphate, produced chitinase, and released siderophores in plate detection assay. In order to evaluate the plant growth promotion potential in vivo, strain Y1 was grown in media containing chitin powder and complex fertiliser. The pot experiment was conducted by treating pepper seedlings with C1/1 (Y1 culture, 50 ml), C2/3 (Y1 culture, 33 ml), C1/2 (Y1 culture, 25 ml), F1/1 (complex fertiliser, 50 ml), F1/2 (complex fertiliser, 25 ml), and W (water) at 10, 20, 30, 40, and 50 days after transplantation (DAT). Plants receiving Y1 had 52% (C1/2) and 68% (C1/1) more root and shoot biomass than W, and 14% (C1/1) and 18% (C2/3) more compared to F1/1 at 80 DAT. Total numbers of flowers per plant at 80 DAT were found significantly higher with the application of Y1 having 34 (C1/1), 35 (C2/3), and 22 (C1/2) compared to 4 (W), 12 (F1/1) and 10 (F1/2). In addition, chlorophyll content in pepper leaves was found to improve with the application of Y1. Furthermore, Y1 has significantly improved nutritional assimilation of total NPK, population of total culturable bacteria and chitinase producing bacteria and activities of chitinase and dehydrogenase in soil. At 60 and 80 DAT, the number of B. amyloliquefaciens at C1/1, C2/3, and C1/2 ranged from 2.3 × 104 to 4.6 × 104 CFU/g of soil. Our results concluded that B. amyloliquefaciens Y1 has positive effects on soil properties and can be suggested as a bio-fertiliser to minimise fertiliser application in modern agriculture. |

WDV-Wheat and WDV-Barley isolates of WDV were transmitted to wheat and barley, respectively, using the leafhoppers under four temperature regimes of constant 20, 25, 30, and 35°C. Infection rate, symptom development and virus content of the virus-inoculated plants were determined and the data was statistically analysed. The results showed that the temperature of 25°C was associated with the highest infection rate caused by the viruses. Moreover, P. alienus nymphs were found to be more efficient vectors of WDV than adults, highlighting the importance of nymphs in the epidemiology of wheat dwarf disease. WDV-infected plants incubated at 35°C showed less symptoms than those kept at 20, 25, and 30°C. ELISA results showed that these plants had comparatively low virus content. However, there was no significant difference between the infection rate, symptom development and virus content in plants infected by WDV-Wheat or WDV-Barley. |

Damage was found in an apple orchard under organic farming, in Central Moravia (Czech Republic), at 700 m a.s.l. There were two cultivated apple cultivars Red Spring and Melodie/Angold. Damage occurred in winter with the snow cover lasting from December to February. In total 1 012 trees and 95.7% of trees were damaged. The cv. Red Spring was damaged more than cv. Melodie/Angold. Almost 90% of the dead trees were killed by common voles. While hares damaged both cultivars equally, voles damaged the cv. Red Spring to a significantly greater extent (P = 0.04). The study confirms the need of further research on the development of methodologies for orchard protection from damage caused by small mammals. |

Using RDP4 analysis the natural recombinant isolate PSB204CZ bearing two breakpoints in nucleotide positions 4053 and 6080 was identified. The isolate was composed of fragment 2028 nt in length partially covering CI and 6K2 regions of the minor parent (PSB262CZ) incorporated into the major parent (PSB194CZ). The results suggest that the observed recombination in CI-6K2 region is responsible for severity of developed symptoms. This observation detected for the first time natural recombination within PSbMV isolates of an important pathogen of leguminous plants. |

We investigated the occurrence of bark necrotic disease in beech thicket and health status of the trees. Our objective was to examine how bark necrotic wounds persist and the regenerative potential of young beech thicket. The research experiment was carried out at two isolated localities in the Western Carpathians in the period 2015-2017. A terminal shoot (stem) and a lateral shoot (branch) of each of the 30 sampled beech trees were examined. Young beech stands were affected by the necrotic disease in 87-94% cases (stems 69-83%, branches 33-56%). Shoot diameter and age had a significantly negative linear correlation (P < 0.05) with the development of all analysed disease characteristics on the beech bark. The encouraging finding is a positive regenerative capacity of young beech trees. Over the same period more annual shoots were healed in comparison with those that were newly infected (10.8% on stems and 0.6% on branches). |
The effect of sites and years on the technological quality of winter wheat grainJ. KučerováPlant Soil Environ., 2005, 51(3):101-109 | DOI: 10.17221/3562-PSE The results of three-year trials (1999 to 2001) conducted with six winter wheat varieties in which was investigated the effect of sites, years and varieties on the final grain yield and technological quality showed statistically significant differences (LSD, 95%) among sites in grain yield and these parameters of quality: test volume mass, wet gluten content, sedimentation test, protein content, falling number and flour water absorption capacity. As to the quality, the Žatec site showed itself as generally most suitable, and the Jaroměřice site as the least suitable one. Climate conditions showed a statistically significant effect (LSD, 95%) on grain yield and all quality parameters, beside the percentage of complete grains and swelling capacity of gluten and falling number, which were insignificant. The most favourable weather conditions, a lot of precipitation and high temperature in the course of ripening from three years were proved in the year 2000. Among the varieties were statistically significant differences (LSD, 95%) in grain yield and these parameters of quality: test volume mass, thousand grain weight, ash content, percentage of complete grains, sedimentation test, protein content, reologic properties of dough and baking tests. From the six varieties the best profits were shown from variety Sulamit (loaf volume 595 ml/100 g of flour), the worst was Semper (loaf volume 543 ml/100 g of flour) and Vlasta (loaf volume 532 ml/100 g of flour). The best grain yield was in 2001 (average of sites 8.84 t/ha) and the variety Semper had the highest grain yield of 9.17 t/ha, that is higher at 13.41% compared with Sulamit (7.94 t/ha). |

Our study reported the consumption of CH4 by soil in a lowland broadleaf mixed temperate forest in the Czech Republic (Central Europe). The motivation of our study was to determine the importance of CH4 fluxes in context of carbon dioxide (CO2) fluxes of a broadleaf mixed forest. CH4 and CO2 emissions from the soil were measured during the 2016 vegetation season on a long transect applying the chamber technique. The average daily consumption of atmospheric CH4 by the forest soil ranged from 0.83 to 1.15 mg CH4-C/m2/day. This consumption of CH4 during summer and autumn periods was not significantly affected by soil temperature and soil moisture. However, during spring period the consumption of CH4 was positively significantly affected by soil temperature and moisture. Estimated amount of carbon (CH4-C) consumed by the forest soil makes up a very small part of carbon (CO2-C) participated in the ecosystem carbon cycle. |
Use of biologically active substances in hopsShort CommunicationPavel PROCHÁZKA, Přemysl ŠTRANC, Kateřina PAZDERŮ, Jan VOSTŘEL, Jan ŘEHOŘPlant Soil Environ., 2018, 64(12):626-632 | DOI: 10.17221/655/2018-PSE In four-year experiments, hop was treated with 7 biologically active substances in two terms during vegetation: Lignohumate max (a mixture of humic acids and fulvic acids), Lexin (a mixture of humic acids and fulvic acids enriched with auxins), Lexenzym (a mixture of humic acids and fulvic acids enriched with auxins, phytohormones and enzymes precursors), Ascophyllum nodosum seaweed extract, synthetic auxin, humic acids and fulvic acids alone. The chlorophyll content was monitored after the application both in the vine leaves and in the branch leaves. After harvesting of the hops from the individual treatments, the yield of dry hops was determined and the cones were analysed for the content of alpha bitter acids. The results show that the most effective hop treatment was the application of Lexin and Lexenzym. The Lexenzym treatment provided a yield of dry hops of 1.86 t/ha, i.e. 0.47 t/ha higher compared with untreated control. The Lexin treatment provided yield higher by 0.41 t/ha of dry hops compared with the untreated control, while the harvested cones contained the most alpha-bitter acids (4.57%). |
