Yield, nitrogen use efficiency and balance response to thirty-five years of fertilization in paddy rice-upland wheat cropping systemOriginal Paper

Optimal soil nitrogen management is vital to crop production and environment protection. Little knowledge is available on crop yield, nitrogen uptake, use efficiency and balance in paddy rice-upland wheat cropping system of China. A thirty-five-year long-term field experiment was designed with nine treatments, including an unfertilized treatment (control), nitrogen (N), phosphorus (P), potassium (K) fertilizer, manure (M), and manure combined with mineral fertilizer treatments. Crop yield, N uptake, use efficiency, and N surplus or deficit amount were determined. The results indicated that rice, wheat yield and N uptake amount in the manure combined with mineral fertilizer treatments were higher than that in the manure alone or mineral fertilizer alone treatments. N use efficiency was the highest in the treatment with manure alone. Soil N input indicated a surplus in the mineral fertilizer in combination with manure treatment, but soil N input indicated a deficit in the control, NPK and M treatments. Considering crop yields, N use efficiency and N balance, recommended N application amount is almost 220 kg N/ha/year in the paddy rice-upland wheat cropping system. Taking into account labour and fertilizer sources, half mineral N and half organic N applications were recommended.

Soil phosphorus sorption properties in different fertilization systemsOriginal Paper

The study aimed at the evaluation of the accumulation and vertical distribution of different forms of phosphorus (P) in reference to phosphorus sorption properties subject to mineral (NPK), mineral-organic (NPK + M), and organic (M) fertilisation. It was carried out in a long-term experimental field in Skierniewice (Central Poland) conducted since 1923 under rye monoculture. Total P content in the M and NPK soil profile was similar and lower than in the NPK + M soil. The content of organic P in Ap and Eet horizons of both manured soils was similar and higher than in the NPK soil. The Langmuir P sorption maximum (Smax) in the studied soils ranged from 39.7 to 90 mg P/kg, while the Freundlich P sorption coefficient aF ranged from 6.9 to 41.9 mg P/kg. Higher variability of parameters related to the binding energy from the Lanqmuir (k) and Freundlich (aF) equations was determined between soil horizons than between the fertilisation systems. Nonetheless, in M and NPK + M soils, sorption parameters aF and Smax and binding energy (k, bF) were considerably lower than in the NPK soil. The content of water extracted P in manured soils was higher than in the NPK soil.

Effect of biochar on nitrogen use efficiency, grain yield and amino acid content of wheat cultivated on saline soilOriginal Paper

Biochar can potentially increase crop production in saline soils. However, the appropriate amount of biochar that should be applied to benefit from resource preservation and increase both grain yield (GY) and quality is not clear. A pot experiment was conducted to evaluate the effects of biochar applied at various rates (i.e., 0, 5, 10, 20, 30, 40 and 50 t/ha) on the nitrogen use efficiency (NUE), GY and amino acid (AA) contents of wheat plants in saline soils. The results showed that the application of 5-20 t/ha biochar increased wheat NUE by 5.2-37.9% and thus increased wheat GY by 2.9-19.4%. However, excessive biochar applications (more than 30 t/ha) had negative effects on both the NUE and GY of wheat. Biochar had little influence on leaf soil and plant analyzer development (SPAD) values, the harvest index or yield components. The AAs were significantly affected by biochar, depending on the application rate. Among the application rates, 5-30 t/ha biochar resulted in relatively higher (by 5.2-19.1%) total AA contents. Similar trends were observed for each of the 17 essential AAs. In conclusion, the positive effects of biochar occurred when it was applied at appropriate rates, but the effects were negative when biochar was overused.

Comparison of growth of annual crops used for salinity bioremediation in the semi-arid irrigation areaOriginal Paper

The decline of soil organic carbon (SOC) has aggravated salinity-related problems in semi-arid irrigation areas of the Awash river basin, Ethiopia. This study aimed at evaluating the performance of potential remediation crops on saline soil and their effectiveness for remediating soil salinity and improving pH, SOC, bulk density (BD) and hydraulic conductivity (HyCo). Rhodes grass (RHG), alfalfa (ALF), sudangrass (SUG) and blue panicgrass (Retz) (BPG) were grown in saline (3-13.9 dS/m) field plots. The crop biomass was incorporated into the soil immediately before flowering. The results show that at high soil salinity levels, BPG and SUG grew well, with the harvesting frequency of BPG being much higher than for SUG. Conversely, the growth of ALF and RHG was strongly inhibited by high soil salinity. Significant (P < 0.05) reduction of soil salinity levels (-3.2 dS/m) and related ionic concentrations, an increase of SOC (0.8% to 1.6%) and improvement of BD and HyCo were observed in BPG plots. The fast-growing nature of BPG in the hot climate of the experimental area resulted in harvests every three weeks and promoted the incorporation of high amounts of biomass to the soil and efficient soil salinity remediation. At moderately saline conditions, ALF also showed a great potential for salinity reclamation (-1.8 dS/m) and SOC accumulation. The cultivation of fast-growing annual crops proved an efficient and low-cost strategy for soil salinity mitigation and the reclamation of salinity-associated soil degradation in irrigation agriculture in Ethiopia.

Experimental study on soil erosion under different soil composition using rainfall simulatorOriginal Paper

Soil erosion is one of the major environmental problems in open-cut mines in tropical regions. It causes negative impacts including the removal of nutrient-rich topsoil, destroys aquatic habitat, dam and pond siltation, clogs river by deposition of sediment, and causes water pollution in the rehabilitation process. Soil texture is an important factor to affect soil erosion. In this study, artificial rainfall experiment in the laboratory scale was conducted to clarify the mechanism of soil erosion under the different soil composition and to discuss the methods for minimizing soil erosion. The obtained results showed that the soil seal generated due to the presence of fine particle under high rainfall intensity is the main contributor to accelerate the soil erosion. Additionally, the surface coverage by the cover crops is the most effective measure to reduce soil erosion because both the coarse and fine contents runoff can be minimized while arranging of the slope angle is effective for reducing the runoff of coarse contents and the soil compaction is effective to reduce that of fine contents. Soil erosion can be minimized by selecting prevention method considering the type of soil because the prevention effect on soil erosion is different depending on the type of soil.

Evaluation of fertigation technique for phosphorus application of maize in the semi-arid region of Northeast ChinaOriginal Paper

To determine the best phosphorus (P) fertilizer management strategy in chernozem soil in the semi-arid region of Northeast China, a field experiment under film mulched drip irrigation was conducted for two consecutive years. Five P application methods were tested, including no P fertilizer applied (P0); P fertilizer one-time basal application (P1); one fertigation one time (P2); fertigation twice (P3) and fertigation four times (P4). The shoot dry matter weight, phosphorus accumulation, yield and POlsen in soil were measured regularly during the maize growing season. The results demonstrated that P fertilizer application methods imposed significant effects on dry shoot matter, the apparent P loss, P fertilizer use efficiency and yield of maize (P < 0.05). The yield, P agrinomic efficiency and P recovery efficiency of P4 treatment were significantly higher than P1 treatment by 4.2, 39.7, and 66.4% across two year. In addition, P4 treatment significantly enhanced the shoot dry matter weight after V12 stage, post-silking P uptake and reduced the apparent P loss. In conclusion, P fertigation and a rational frequency (e.g., fertigation four times) enable to keep the maximum grain yield, the shoot dry matter, and recovery efficiency of P fertilizer (66.4%) though changing agronomic methods for efficient acquisition of P toward a sustainable and productive agricultural fertigation system.

Is maize suitable for substitution planting in arsenic-contaminated farmlands?Original Paper

The efficacy of using maize (Zea mays L.) as a suitable substitute for other crops with high arsenic (As) accumulation in As-contaminated farmlands remains debated. Here, the As uptake capacity and the stability of accumulated As of different maize cultivars were studied using pot and field experiments, outdoor investigations and literature data analysis. When the total and available soil As levels were 238.8 and 8.1 mg/kg, respectively, grain As ranged from 0.03 to 0.07 mg/kg, significantly lower than the acceptable As limit (0.5 mg/kg) for maize in China. The results of field investigations and literature data analysis also supported this observation. Maize is a crop with low grain As, thus, making it suitable for substitution planting in As-contaminated farmlands. Further, grain As concentration varied among different maize cultivars. The planting of normal and waxy maize is prioritized over the sweet maize as the first one has lower available bio-concentration factor (aBCF) of 0.007 for grain and higher accumulated As stability among its cultivars (CV < 10%) than those for sweet maize (aBCF = 0.01 and CV = 35.5%). Arsenic compartmentalization in the roots and low As upward migration into the grain were responsible for the low grain As of maize.

Assessment of plants for phytoremediation of hydrocarbon-contaminated soils in the Sudd Wetland of South SudanOriginal Paper

Hydrocarbon contaminants have become a global concern due to their long-term adverse effects on soil ecosystems and human health. Successful implementation of phytoremediation to clean up hydrocarbon contaminants requires the identification of the most effective remediation plant species. Twelve native plant species of the Sudd Wetland in South Sudan were evaluated for their potential application as phytoremediators. The treatments included six total petroleum hydrocarbon (TPH) concentrations of 0, 25, 50, 75, 100 and 125 g/kg soil. The twelve native plant species tested were: Sorghum arundinaceum Desv., Oryza longistaminata A. Chev. & Roehrich, Hyparrhenia rufa Nees, Abelmoschus ficulneus L., Gossypium barbadense L., Nicotiana tabacum L., Sorghum bicolour L. Moench, Eleusine coracana Gaertn., Capsicum frutescens L., Zea mays L., Tithonia diversifolia Hemsl. and Medicago sativa L. Significant differences in phytoremediation rates were observed amongst the treatments with exception of the 125 g/kg soil concentration of hydrocarbon that was lethal to all the plant species. Over 50% TPH reduction in the 75 g/kg soil concentration was observed in contaminated soil phytoremediation in H. rufa, G. barbadense, O. longistaminata, T. diversifolia and S. arundinaceum, making them potential phytoremediators of hydrocarbon-contaminated soil in the Sudd-Wetland of South-Sudan.

Weed suppressive ability of cover crops under water-limited conditionsOriginal Paper

The water demand for cover crops (CC) should be considered to achieve competitive crop stands for weed control also under unfavorable conditions. This study aims to estimate the weed suppressive ability of winter CC, as Sinapis alba L., Phacelia tanacetifolia Benth., Vicia sativa L. and Avena strigosa Schreb., under a water-limited regime. The water deficit tolerance of different CC was determined in a greenhouse experiment by measuring the maximum quantum efficiency of photosystem II. Moreover, soil moisture, CC, and weed establishment were measured in field experiments in Southwest-Germany during two contrasting growing seasons in 2016 and 2017. A. strigosa showed a higher water deficit tolerance than S. alba in the greenhouse. In the field, A. strigosa showed the highest weed cover reduction (98%) in the field, along with an increasing effect on the soil moisture compared to the untreated control. S. alba performed most sensitive to water deficit in the greenhouse but reached the significantly highest weed control efficacy (94%) during the dry field season in 2016. Even though the selected CC showed differing sensitivities to water deficit in the greenhouse, their weed suppression ability was independent of the water supply under field conditions.

Effect of drought stress on oil content and fatty acids composition of some safflower genotypesOriginal Paper

To assess the response of promising safflower genotypes to late-season drought stress in delayed planting conditions, an experiment was conducted in two years (2016-2017 and 2017-2018) in Iran. The irrigation regime was specified in two levels, including normal irrigation and irrigation cut off in the seed filling stage in main plots, and five safflower genotypes, including Soffe, Goldasht, Golmehr, Padideh, and Parnian were categorized in subplots. Applied drought stress significantly reduced the seed yield and yield components of all genotypes, which accompanied with a substantial decrease in oil content and oil yield of all genotypes. However, the highest seed and oil yield in drought stress conditions obtained in the Parnian genotype by value 2338 and 561 kg/ha, respectively. Moreover, a significant drought-induced increase in palmitic, stearic, and oleic acids, as well as a decrease in linoleic acid content, was observed in all genotypes. Parnian genotype with high unsaturated fatty acids content (90.9%) and the minimum amount of saturated fatty acids (8.7%) might be a promising genotype to starting a formal crop improvement program to achieve more drought-tolerant safflower genotype.

Effect of sowing density on grain yield, protein and oil content and plant morphology of soybean (Glycine max L. Merrill)Original Paper

To find out exactly how sowing density and sowing pattern affect soybean grain yield, quality and its components in non-traditional soybean growing regions, such as Germany, two field trials have been conducted at two locations in Bavaria in 2016 and 2017. The experiments were carried out with four sowing densities (30, 50, 70, 90 seeds/m2) and four cultivars from different maturity groups (Viola 000, Lissabon 000, ES Mentor 00, Orion 00) as randomized complete block design with four replicates. Almost all evaluated traits varied significantly depending on year × location. There was no interaction between the main factors (cultivar × sowing density). The results revealed that grain yield and height of the first pod increased with increase of sowing density. The number of pods per plant and branching decreased with increasing sowing density. At higher sowing densities at flowering leaf area index was significantly higher than at lower sowing densities. The cv. ES Mentor (00) with 70 seeds/m2 has proved to be a suitable cultivar in terms of yield and quality in southern Germany (> 3.6 t/ha grain yield and 40% protein).

Blood chemistry reference intervals of captive Asiatic black bears (Ursus thibetanus)Original Paper

Data on blood chemistry values can make fundamental contributions to our understanding of physiological changes. However, there is a lack of information regarding blood chemistry in Asiatic black bears (Ursus thibetanus). Thus, the objects of this study were to determine reference ranges for 29 blood chemistry variables, and to evaluate differences between age groups and between seasons. Blood samples (n = 138) were collected from 44 (20 males, 24 females; age range, 1-15 years) clinically healthy, captive Asiatic black bears (Ursus thibetanus) in the Republic of Korea. Young and adult bears showed significantly higher levels of creatinine and total cholesterol, and lower levels of blood urea nitrogen, blood urea nitrogen/creatinine ratio, lactate dehydrogenase and creatine kinase MB during hibernation compared to during non-hibernation. Adults also showed significantly higher levels of triglyceride, but lower levels of inorganic phosphorus, aspartate transaminase, alkaline phosphatase, high density lipoprotein cholesterol and creatine phosphokinase during hibernation than during non-hibernation. During hibernation, the urea nitrogen/creatinine ratio and levels of alkaline phosphatase, lactate dehydrogenase, and creatine phosphokinase in young bears were significantly higher than in adults, whereas creatinine levels were lower than in adults. During non-hibernation, the urea nitrogen/creatinine ratio and levels of calcium, alkaline phosphatase, lactate dehydrogenase, creatine phosphokinase and creatine kinase MB in young bears were significantly higher, whereas creatinine, total protein, albumin, gamma-glutamyl transferase and haemoglobin levels were lower than in adults. The results of this study provide reference values that will aid in understanding the physiology of Asiatic black bears and in assessing the health of these animals in captive environments.

Long-term evaluation of autologous lamellar corneal grafts for the treatment of deep corneal ulcer in four dogs: a case reportCase Report

Autologous lamellar corneal grafts were performed on four dogs with two deep corneal ulcers and two corneal perforations to achieve better visual and aesthetic outcomes. The donor corneal graft was harvested from the relatively normal corneal region of the affected eye and used to cover the corneal defect. At the two-year follow-up examination, ultrasound biomicroscopy was performed to confirm the thickness and endothelial continuity of the transplanted grafts and donor site of the cornea. The evaluations revealed that the dogs had healed without incident and that their vision had been maintained. Furthermore, the owners were satisfied with the aesthetic outcomes in all cases. Our findings show that autologous lamellar corneal grafts are useful and effective in treating canine corneal ulcers and perforations.

Carotid body tumour in a dog: computed tomography and histopathology findings and evaluation of surgical managementCase Report

A 2.5-kg, eight-year-old, neutered male Yorkshire Terrier was presented for evaluation of a cervical mass that had first been noticed a year earlier. A firm spherical mass located caudal to the left mandible was found on physical examination. Ultrasonography revealed a well-defined, round-shaped mass located medial to the left mandibular salivary gland that was approximately 2.6 cm in height, 2.3 cm in width and 3 cm in length. Volume-rendered images obtained by computed tomography (CT) showed that the left external and internal carotid arteries and internal jugular vein were encased in the mass. A definitive diagnosis of carotid body tumour was made based on histopathology. The tumour and the carotid arteries and internal jugular vein encased in the tumour were resected using CT-based surgical planning. The regional neural structures were preserved by careful blunt dissection. The dog had a mild hacking cough after swallowing and hoarseness that disappeared spontaneously five days and two months, respectively, after surgery. There was no evidence of recurrence or distant metastasis at 18 months postoperatively. This case report describes successful surgical management of a carotid body tumour with ligation and transection of the carotid arteries in a dog. To the authors' knowledge, this is the first report of successful surgical treatment of an advanced carotid body tumour based on CT findings.

The efficacy of fenbendazole against tapeworm infections in the common carp (Cyprinus carpio L.) in Veľké Blahovo pondsOriginal Paper

A survey of the pond system in Veľké Blahovo, Dunajská Streda district, Slovak Republic, confirmed the occurrence of tapeworms in carp with a prevalence of 63.5% and mean intensity of 1.7 ± 2.72. The species Khawia sinensis and Atractolytocestus huronensis were identified. We attempted to reduce the occurrence of tapeworms using a medicated feed mixture (granules) with 1% fenbendazole as active substance under controlled conditions. Faeces collected from carp (K2) were examined and the presence of tapeworm eggs was confirmed by the flotation method. Positive carp were divided into three groups; each group, labelled F1 to F3, included 15 individuals. The carp were placed in concrete pools (storage ponds) nourished by fish pond water at the peak of the vegetation period from August 7-17, 2015. The medicated feed mixture was administered to the first two groups (F1 and F2), while the third group was fed with a non-medicated feed mixture and served as the control (F3). A single dose was administered to the F1 group; tapeworm prevalence 24 hours after administration in the F1 group was 26.66% with a mean intensity of 1.3 ± 0.57. The F2 group was fed three times in total with 24-hour intervals between feeding. Tapeworm prevalence 24 hours after the last administration in the F2 group was 13.33% with a mean intensity of 2.0 ± 0.77. The prevalence in the control group was 66.66% with a mean intensity of 2.2 ± 1.62. Khawia sinensis tapeworms responded well to fenbendazole, while A. huronensis tapeworms responded poorly and, despite repeated administration of the active substance, this species remained attached to the intestinal mucosa. When compared with the control group, the reduction in the number of tapeworms was statistically significant (P ≤ 0.05) in the F2 group; however, it was not statistically significant in the F1 group.

Gas-filled urachal abscess with a pinging sound in a heifer calfCase Report

A 6-month-old crossbred of a Holstein and Japanese Black heifer calf weighing 95 kg presented with a history of intermittent abdominal distension and failure to thrive. The physical examination identified a pinging sound over the dorsal left flank. The abdominal radiography showed a huge gas-filled mass. The intravenous urography revealed no communication between the mass and the urinary bladder. Although the visual examination and palpation of the umbilicus did not reveal visible abnormalities, an umbilical disease was suspected because the animal exhibited poor growth, depression, and a hunched back posture. When the eschar adhering to the centre of the umbilicus was removed, the presence of a fistulous tract was revealed. The umbilical ultrasound examination revealed an intra-abdominal abscess and the fistulography demonstrated that the abscess communicated with the umbilicus. The abscess, compressing into the rumen, was observed by computed tomography. From these images, it was diagnosed as an umbilical cord remnant abscess and a definitive diagnosis of a urachal abscess was obtained by open abdominal surgery and the subsequent removal of the mass. The calf was discharged from the university hospital on day 14 after the operation. This case shows that a urachal abscess should be considered when a pinging sound is present, even if the animal exhibits no swelling or pain of the umbilicus.

Surgical treatment of an oesophageal achalasia in a small breed dogCase Report

A 6-year-old, male Yorkshire Terrier dog presented with persistent regurgitation and severe weight loss. Based on the clinical signs, physical and other various diagnostic examinations, including fluoroscopy, were performed. Myasthenia gravis was ruled out through a serum acetylcholine receptor antibody titre measurement and a negative response to neostigmine bromide (0.02 mg/kg) within 4 hours of injection. The dog was diagnosed with idiopathic oesophageal achalasia. As a treatment, a modified Heller's oesophagomyotomy was performed, and the dog recovered well with no signs of recurrence for 18 months.

The molecular and morphometric identification of Dictyocaulus capreolus in clinically affected roe deer (Capreolus capreolus L.)Original Paper

The poor state of health and increased mortality rate of young roe deer, as reported by South Moravian hunters, caused by the increasing numbers of adult nematodes in the lungs of roe deer prompted us to identify the parasites using a combination of morphological measurements and a phylogenetic SSU rRNA analysis. The study was conducted in a 294 ha game reserve in South Moravia, Czech Republic. Molecular and morphometric techniques were used to identify adult nematodes collected from the respiratory tracts of nine 4-5 months old roe deer in poor health (low body weight of 3-4 kg, poor haircoat quality, and, in some cases, symptoms of diarrhoea). The morphological identification was based on a combination of adult worm characteristics corresponding to Dictyocaulus capreolus. A small subunit rRNA (SSU) partial sequence analysis showed the highest identity scores (99%) corresponding to the sequences of D. capreolus from a roe deer (GenBank: AY168859) from Sweden and the outcomes of the phylogenetic analyses resulted in a tree with a high branch support for two groups, with our sequences forming a well-supported clade with D. capreolus and Dictyocaulus sp. ex Capreolus capreolus (FJ589016) and Dictyocaulus sp. ex Rupicapra rupicapra (FJ589019) sequences from Spain. The examined roe deer have shown symptoms of diarrhoea, anorexia, and respiratory tract inflammation indicating that there might be a connection to the clinical importance of the Dictyocaulus infection.

The occurrence of mastitis and its effect on the milk malondialdehyde concentrations and blood enzymatic antioxidants in dairy cowsOriginal Paper

Early identification of mastitis is a serious challenge for dairy farmers and veterinarians in ensuring the health of an animal and the hygienic quality of the produced milk. The purpose of this study was to detect the occurrence and aetiology of mastitis in a dairy herd of 153 milked cows localised in a farm in west Slovakia. During the complex investigation, 606 quarter milk samples were examined (6 quarters were discarded) and classified based on the clinical status, the presence of abnormal udder secretions, the result of the California mastitis test (CMT), the somatic cell count (SCC) and the bacteriological identification of the pathogens causing the intramammary infection (IMI). The study was augmented by the detection of malondialdehyde (MDA) in the milk and the measurements of the blood enzymatic activities of glutathione peroxidase (GPx) and superoxide dismutase (SOD) as potential biomarkers for the udder health screening. A positive CMT score was recorded in 19.5% (118) of the examined quarters and 12.5% (76) of the quarters were infected with bacterial pathogens causing latent mastitis (LM; 1.3%), subclinical mastitis (SM; 8.3%), and clinical mastitis (CM; 2.9%). The most commonly isolated bacteria from the infected quarters were coagulase-negative staphylococci (55.2%), Staphylococcus aureus (11.8%) and streptococci (10.5%). The concentration of MDA and SCC were significantly higher from both the SM and CM cases than in the milk samples from the healthy cows, while the blood activities of SOD and GPx were lower in the cows with CM compared to the healthy cows. The higher MDA concentrations in the SM and CM milk observed in this study showed the presence of an oxidative stress in the infected milk, accompanied by a decrease in the antioxidative enzymatic activity in the blood of the cows. Therefore, the measurement of the milk MDA concentration and the activity of the blood SOD and GPx may prove insightful for the better screening of the udder health in the early diagnosis of mastitis.

Evidence for wheat, rye, and barley presence in gluten free foods by PCR method - comparison with ELISA method

A method of the evidence for the presence of wheat, rye, and barley in gluten free foods, based on the polymerase chain reaction (PCR), was validated. DNA was isolated from foods by chaotropic solid phase extraction. The PCR method applied was focused on the intron of the chloroplast gene trnL and utilised primers WBR11 and WBR13. Electrophoresed wheat and rye DNAs were characterised by a 201 bp fragment, barley DNA by a 196 bp fragment. The validated PCR method was applied to the selection of 18 gluten free foods, previously found by ELISA method to contain 1 mg or more of gliadin per 100 g food. The presence of wheat was confirmed by PCR method in all foods analysed. The comparison with the results obtained by ELISA method reliably verified the detection limit of PCR method, i.e., 0.02% wheat.

ELISA for free S-equol in human urine

The popularity of phytoestrogen food supplements of plant origin steadily increases. They are used to solve the problems related to the climacterium. Their efficiency depends on woman's capability to metabolise the active components - isoflavonoids. Women able to convert daidzein to S-equol are supposed to get a greater benefit from these supplements. To determine S-equol urine levels, the competitive ELISA was developed. Carboxymethylequol coupled to BSA via 4'-O-position was used as an immunogen. The assay conditions, including the concentrations of the coating antigen and antisera, were optimised. The detection limit was 0.1 ng/ml (5 pg/well) and 50% intercept was 1 ng/ml (50 pg/well). The intraassay coefficients of variation (CV) varied from 4.7% to 9.9%, for the interassay CV the corresponding values of 2.6-11.6% were obtained. The recovery of the standard added to urine ranged between 89% and 104%. All examined samples were obtained from women in climacterium. The volunteers had been taking phytoestrogen food supplements for 3 months. The urine samples were collected before and after the therapy. The levels of free urinary S-equol after the therapy ranged between 0 ng/ml and 42.4 ng/ml. The equol production was observed in 57% of the examined patients. The laboratory results were confronted with patients' subjective evaluations.

Influence of different essential oils on refrigerated fish patties produced from bonito fish (Sarda sarda Bloch, 1793)Food Analysis, Food Quality and Nutrition

The effects of different concentrations of thyme (group T), clove (group Cl), and rosemary (group R) extracts on the microbiological, chemical, and sensory attributes of fish patty made from the bonito fish (Sarda sarda Bloch, 1793) were investigated. The numbers of total mesophilic aerobic bacteria, coliform bacteria, Staphylococcus-Micrococcus spp. and yeasts and moulds in group R were lower than those of the other groups during storage (P < 0.05). During the storage period, total volatile basic nitrogen (TVB-N), thiobarbituric acid index (TBA-i), peroxide values, and free fatty acid content gradually increased in all groups, and significant differences were found between the groups (P < 0.05). The results of sensory evaluation showed that the shelf life of fish patties was 4 days for the control group, and 8, 10, and 14 days for the groups treated with thyme, clove and rosemary essential oil, respectively (P < 0.05). In conclusion, the addition of essential oils showed a positive effect on the product shelf-life; and in particular, rosemary essential oil produced a remarkable effect.

Analysis of chemical and sensory parameters in different kinds of escolar (Lepidocybium flavobrunneum) productsFood Analysis, Food Quality and Nutrition

The chemical composition (nutritional and freshness parameters) and sensory characteristics were evaluated of escolar (Lepidocybium flavobrunneum) defrosted raw fillets and steaks that were marinated differently (pepper, onion, chilly, herbs, knusper, and hot) and packaged in a manner suitable for microwave heating (Darfresh® Simple Steps®). The study was carried out by using 8 skinless fillets of Lepidocybium flavobrunneum. The selected 18 parameters, i.e. dry matter/moisture, crude protein, net protein, collagen, net muscle protein, lipid, ash, saccharide, energy value, pH, water activity, salt content, total volatile basic nitrogen, trimethylamine, free fatty acids, peroxide value, and thiobarbituric acid assay were evaluated. The results indicate that the sample of Chilly marinated steaks was evaluated as the best one (90.29 ± 9.82), however, the samples of Herbs (89.6 ± 13.89) and Knusper (88.64 ± 15.59) marinated steaks also received more than 85 points for the overall impression. Control sample received the smallest number of points (71.7 ± 26.54) for the overall impression.

The effect of addition of ripened cheese slurry into retentate on the proteolysis of Iranian ultrafiltered cheese was investigated during 60-day ripening. No significant differences were found in gross composition between experimental samples, however, pH values were significantly higher (P > 0.05) in slurry-containing cheese. Urea-polyacrylamide gel of the pH 4.6 insoluble fractions showed that αs1-casein was hydrolysed more rapidly than β-casein. There was no significant difference in the hydrolysis rate of αs1- and β-casein between the two cheeses. The levels of water soluble nitrogen were significantly higher (P > 0.05) in slurry-containing samples from day 15. The incorporation of ripened slurry into retentate influenced the peptide profiles obtained by RP-HPLC of the pH 4.6-soluble fractions of the cheeses. The ratio of hydrophilic to hydrophobic peptides was higher in slurry-containing cheese. Total and individual free amino acids were significantly higher (P > 0.05) in cheeses containing slurry. It was concluded that the incorporation of ripened cheese slurry into retentate has a beneficial effect on proteolysis acceleration and development of aroma and flavour in Iranian ultrafiltered white cheese.

Phosvitin phosphopeptide preparation using immobilised trypsin and enhancing calcium absorption in growing ratsFood Analysis, Food Quality and Nutrition

We demonstrate a new, efficient method for the preparation of phosvitin phosphopeptides using immobilised-trypsin enzymolysis technology. Immobilised trypsin was prepared using a covalent binding method, and then was added to degrade egg yolk phosvitin for the production of phosphopeptides. In our results, the prepared immobilised trypsin demonstrated a higher hydrolysing activity toward phosvitin than free trypsin, and the hydrolysing activity was retained well even after trypsin was repeatedly used up to five times. Interestingly, the phosvitin phosphopeptides prepared with immobilised trypsin demonstrated a lower N/P ratio and a higher calcium-binding efficiency than those prepared with free trypsin. Furthermore, phosphopeptides significantly increased the rate of calcium absorption and serum calcium content in vivo. Based on these results, we conclude that trypsin immobilised onto chitosan has a greater phosvitin hydrolysing activity than free trypsin, and the prepared phosphopeptides can be used as a new calcium supplement to significantly increase calcium absorption in growing rats.

This experiment was carried out to compare morphological egg quality parameters, as well as lipid and protein profiles, of brown eggs laid by chickens reared under different production systems: cage, free-range, and family type. A total of 270 brown eggs were obtained from commercial poultry companies raising Lohmann Brown laying hens in a cage system and free-range unit as well as families possessing hens in their yards. The egg lipid and protein contents, as well as lipid and protein profile, varied among the production systems. However, eggs from the free-range system had similar characteristics to those from the cage system. Quality of eggs from the family type system was quite variable. In conclusion, egg quality can be affected by the production system.

Preparation of conjugated linoleic acid enriched derivatives by conventional and biphasic isomerisationFood Chemistry and Safety

The preparation of conjugated linoleic acid (CLA)-enriched free fatty acids by industrial processes compared with our biphasic isomerisation experiments in a special designed reactor enabling the preparation of CLA esters was evaluated. Our experiments further revealed the main disadvantage of semi-synthetic alkali isomerisation to be the formation of conjugated E,E-octadecadienoic acid isomers (2.92-3.44%) and the bioavailability of free fatty acid products. Urea fractionation technology improved the quality of the reaction mixture, but at the same time the yield of rumenic acid was decreased on purification. Therefore, we decided to apply complexes of noble metals in order to isomerise linoleic acid ester derivatives. The known Wilkinson's hydrogenation catalyst, RhCl (PPh3)3, was found to be the most effective. We investigated the preparation of bioavailable CLA-enriched triacylglycerols. Special attention was paid to recycling of Wilkinson's catalyst.
