Effect of chokeberry juice consumption on antioxidant capacity, lipids profile and endothelial function in healthy people: a pilot studyFood Analysis, Food Quality and Nutrition

Many studies show that the consumption of juices rich in polyphenols may increase serum antioxidant capacity, improve blood lipid profile, and endothelial function. The relation between the consumption of chokeberry juice and changes in the antioxidant capacity, blood lipid profile, and endothelial function as important indicators in the assessment of cardiovascular risk were determined. The study was conducted on a group of 11 healthy volunteers, who consumed chokeberry juice for three weeks. The research included determination of serum antioxidant capacity, blood lipid profile analysis, and measurements of endothelial function. The 3-week consumption of chokeberry juice significantly increased serum antioxidant capacity, and the best results were observed just after one week of the experiment. However, there was no significant change in the blood lipid profile, except for the persons with a higher level of triglycerides, in whom the consumption of chokeberry juice reduced these compounds to normal values. The endothelial function was normal in all patients and did not significantly change during the study.

The important nutritional characteristics of the Rubiota spelt variety, grown in the Czech Republic, were reported. We evaluated the contents of total dietary fibre, protein, wet gluten, total starch, and starch fractions, i.e. rapidly digested starch, slowly digested starch, and resistant starch in pearled grain of spelt and in pearling fines. We measured these properties depending on the degree of kernel abrasion. Small differences were found between the sequential pearling cycles in the pearled spelt but significant differences were observed in the fines. In this study we also compared two methods for determining total and resistant starch.

Antioxidant activity, S-alk(en)yl-l-cysteine sulfoxide and polyphenol content in onion (Allium cepa L.) cultivars are associated with their genetic backgroundFood Chemistry and Safety

Six onion cultivars Bingo, Dormo, Elenka, Elbrus, Spirit, and Sturon grown in the Czech Republic for commercial purposes were analysed to investigate the content of health-promoting compounds. The results showed that at harvest time, cysteine sulfoxide content varied from 32.38 to 44.16 g/kg of dry weight, polyphenol content was between 2.66 and 3.37 g/kg of dry weight, and antioxidant activity ranged from 0.75 to 0.83 g/kg. Cv. Bingo had the highest level of the analysed compounds. The cultivars were concurrently analysed by DNA (microsatellite) markers. Dendrograms based on the chemical composition and DNA analysis were almost identical. This finding confirms the dependence of the secondary metabolite content on onion genotype.

Fermented buttermilk-based beverage: impact on young volunteers' health parametersFood Analysis, Food Quality and Nutrition

The technology of a functional fermented buttermilk-based beverage was developed and the impact of beverage consumption on the health parameters of young volunteers was studied. Three functional beverages were made on the basis of buttermilk, which differed in the composition. The beverage containing the buttermilk-skimmed milk-milk protein concentrate (0.3%) as a product with the preferred quality characteristics (synaeresis, viscosity, sensory properties, and acceptability) was used for further studies regarding the impact on human health. The medical nutrition experiments did not show any statistically significant effect on the biochemical blood parameters of 25 young volunteers after a 21-day period of the consumption of fermented buttermilk beverage with milk protein concentrate, although some of them (e.g. total, low- and high-density cholesterol, triacylglycerol concentrations, etc.) slightly decreased. The anthropometric and body composition, arterial blood pressure, and pulse evaluation indicators of recipients did not change significantly.

Influence of chestnut flour addition on quality characteristics of pasta made on extruder and minipressFood Technology and Economy, Engineering and Physical Properties

Durum semolina was replaced with 10, 15, and 20% of chestnut flour. Pasta was produced on a single screw extruder with the temperature profile of 80/90/90°C and on a laboratory minipress. Pasta samples were dried at room temperature and physical and sensory properties were determined. Generally, the chestnut flour addition to durum wheat pasta decreased optimum cooking time, hardness, cohesiveness, and chewiness, but increased cooking losses and pasta adhesiveness. Samples made on an extruder showed shorter optimum cooking times in relation to samples made on a minipress. Absorbed water was decreased with chestnut flour addition. Pasta made on an extruder showed a higher sensory score in comparison with pasta made on a minipress. The addition of chestnut flour influenced the colour of the samples. Samples made on an extruder showed darker colour in dried and cooked pasta samples in relation to minipress samples. In dried pasta samples, the extruder gave yellower samples in relation to the minipress, while in cooked pasta samples it was reversed. Overall, the extruder gave higher-quality pasta compared to the minipress.

The effects of red wine maturation on the contents of selected parameters of bioactive compounds in wine were determined. Samples of Alibernet, Cabernet Sauvignon, and Torysa wines were studied by spectrophotometric analysis after 3, 7, 11, 19, and 28 months of aging and the selected parameter content was statistically evaluated. Statistically highly significant changes in total polyphenol content, total anthocyanin content, antioxidant activity, and wine colour density were found depending on the aging according to the used statistical analyses. The obtained results can be used for the optimisation of the wine aging process and they allow producers to time the optimal date of wine release onto the market, depending on the desired content parameters.

Heavy metals in the common carp (Cyprinus carpio L.) from three reservoirs in the Czech RepublicFood Analysis, Food Quality and Nutrition

Toxic metals (cadmium, lead, and mercury) in the tissues of common carp fished from three reservoirs (Pilská, Domaninský, and Matějovský) in the Bohemian-Moravian Highlands were measured in the period from April 2013 to September 2014 and the risk of consuming the fish from these localities was evaluated. During this project 25 fish specimens from each locality were analysed. The concentration of metals in muscle tissue and liver was determined by atomic absorption spectrometry. A variation in the content of heavy metals in fish samples and fish tissues was demonstrated and discussed. Higher concentrations of cadmium in the fish tissue were found in 50 samples of carp liver. Because the fish liver is not commonly consumed, common carp from the Moravian-Moravian Highlands does not pose any health risk for consumers. Concentrations of the other monitored toxic metals in fish tissue were low and were complying with the maximum residue limit in all cases.

Morphological characterization and turf performance of Paula hard fescue and Casero colonial bentgrass selections under low maintenance conditionsOriginal Paper

Planting grasses that require low maintenance is a good option for reducing the management input required. The objective of this study was to obtain information about turfgrass characteristics based on spaced-plant characterization and turf performance in turf plots of two selections (Paula hard fescue and Casero colonial bentgrass) recently released for public distribution by the Plant Production Area of the University of Oviedo. The plants were grown under conditions of no irrigation, no fertilizer, no pesticide application and minimal mowing over a 3-year period (2012-2014). The following morphological measurements were done in a spaced-plant nursery: heading date, plant height, inflorescence length and flag leaf length and width. The turf plots were evaluated for several traits including overall turfgrass quality (turf score), leaf texture and genetic colour. In both the spaced-plant nursery and turf trials, hard fescue genotypes displayed better morphological characteristics (narrow leaves, reduced plant height) and performed better (overall high turfgrass quality, fine leaf texture, dark green colour) than bentgrass genotypes under conditions of low maintenance. For areas in which climate, soil characteristics and management conditions are similar to those encountered in this trial, the selection of Paula hard fescue is recommended for use in low-maintenance turf settings because of the good turfgrass performance and morphological characteristics of this selection.

Association of the ADRB3, FABP3, LIPE, and LPL gene polymorphisms with pig intramuscular fat content and fatty acid compositionOriginal Paper

The aim of the present study was to investigate the associations of single nucleotide polymorphisms (SNPs) in candidate genes with fatness traits in the Longissimus dorsi muscle of pigs. The polymorphisms of genes were investigated, which included beta-3-adrenergic receptor gene (ADRB3), heart fatty acid-binding protein gene (FABP3), and hormone-sensitive lipase gene (LIPE) as well as lipoprotein lipase gene (LPL). The intramuscular fat (IMF) content and fatty acid composition contents in Longissimus dorsi muscle samples were measured. Results showed that ADRB3, LIPE, and LPL SNPs were associated with IMF content (P < 0.05). ADRB3 AG heterozygotes exhibited higher IMF content. LIPE A allele was associated with greater IMF content. LPL CT heterozygotes exhibited the lowest IMF content. ADRB3 c.1192G>A had highly significant association with the total monounsaturated fatty acid (MUFA) (P < 0.01) and the total polyunsaturated fatty acid (PUFA) (P < 0.01). LIPE c.442G>A was significantly associated with the contents of C12:0 and C14:0 (P < 0.05). LPL c.624C>T was significantly associated with the percentage of C16:1 (P < 0.05) and the percentage of total saturated fatty acid (SFA) (P < 0.05). The pigs with ADRB3 G allele had more MUFA, and the pigs with LPL T allele had less SFA, implying that the ADRB3 G and LPL T in pigs may be beneficial to human health. In conclusion, the results suggest that these genetic markers are important sources of the variations for the pork selection to obtain favourable meat with higher IMF levels and appropriate fatty acid composition.

Composition of cultivable enteric bacteria from the intestine of Antarctic fish (family Nototheniidae)Original Paper

Selenium (Se), copper (Cu), and zinc (Zn) play important antioxidant role during the transition period of dairy cattle. However, there is limited information about their blood fluctuations during the entire transition period, especially in heifers. Furthermore, it is questionable whether the use of clinoptilolite, a natural zeolite, affects the availability of these trace elements during this period. The objective of the present study was to monitor the blood concentrations of Se, Cu, and Zn during the transition period of dairy heifers and to investigate whether the dietary inclusion of clinoptilolite has any effect on them. Forty clinically healthy Holstein heifers were used in the experiment. They were randomly allocated in two equal groups (n = 20) formed according to their body condition score. The control group was fed only the basal ration whereas the daily feed of treatment group was supplemented with 200 g clinoptilolite. The experiment started 28 days before the expected day of calving and lasted until day 21 after parturition. Blood samples were taken at the onset of the experiment and then at weekly intervals until parturition, at the day of calving, and on days 7, 14, and 21. All samples were analyzed for blood Se and plasma Cu and Zn concentrations. The results indicate that the levels of Se, Cu, and Zn in blood change significantly (P < 0.05) throughout the transition period in dairy heifers and increase significantly (P < 0.05) immediately after calving. Furthermore, the dietary administration of clinoptilolite does not significantly affect their blood concentration (P > 0.05). Blood levels of Se, Cu, and Zn, although undergoing significant changes throughout the transition period in dairy heifers, remain practically stable until parturition and increase significantly immediately after calving. Clinoptilolite does not impair the dietary availability of the trace elements evaluated when added in heifers' rations during this period.

Haematological and biochemical response of burbot (Lota lota L.) exposed to four different anaestheticsOriginal Paper

The aim of this study was to compare the effect of four anaesthetics on haematological and biochemical blood profiles in burbot Lota lota L. Blood profiles of burbot were evaluated 10 min and 24 h after anaesthesia with tricaine methanesulfonate (MS 222) (100 mg/l), clove oil (33 mg/l), 2-phenoxyethanol (0.3 ml/l), Propiscin (1 ml/l) and compared to non-anaesthetized control. The tested anaesthetics had no effect on haematological profile of burbot. The exposure to clove oil, 2-phenoxyethanol, and Propiscin significantly (P < 0.01) influenced the level of ammonia and glucose. The level of lactate was significantly (P < 0.01) increased following anaesthesia with 2-phenoxyethanol and Propiscin. The levels of total protein, aspartate aminotransferase, and calcium were higher (P < 0.01) with clove oil, 2-phenoxyethanol, and Propiscin compared to control. The use of MS 222 showed the lowest effect on haematological and biochemical blood profile and is recommended as a suitable anaesthetic for burbot.

Effects of thermal manipulation during late incubation period on post-hatch thermotolerance in ostrichOriginal Paper

The effects of thermal manipulation (TM) during late ostrich embryonic development on hatchability, body weight, biochemical and hormonal changes, and the ability of hatching chicks to cope with thermal challenge in days 6-8 of age were examined. At 35 days of incubation, two hundred fertile eggs were weighed and divided into two equal groups with five replicates. The first group was exposed to 36.5°C and 25% relative humidity (RH) (control), while the second group was exposed to 38.5°C and 45% RH for 3 h daily in days 35-37 of incubation (thermal manipulation). At days 6-8 of age, the hatched chicks from each treatment were randomly divided into two groups: control group (exposed to 32 ± 1°C), and thermal challenge group (exposed to 40 ± 1°C for 3 h daily). Hatchability rate was significantly lower with high incubation temperature as compared to the normal incubation temperatures. Embryonic TM and thermal challenge in days 6-8 of age reduced significantly total protein, albumin, and triiodothyronin concentrations and elevated uric acid, creatinine, triglycerides, and glucose concentrations as compared with the control. The level of corticosterone was significantly higher in the thermal challenge group as compared to the control. In conclusion, exposing the ostrich embryos to TM (38.5°C) during late embryonic development induced physiological changes that may represent epigenetic adaptation to TM. The same mechanisms are employed for increasing the ability to improve thermotolerance post-hatch.

Equilibration and freezing interactions affecting bull sperm characteristics after thawingOriginal Paper

The objective was to determine effects of equilibration length and freezing curve type as well as their interactions on motility and live spermatozoa proportion in bull sperm after thawing. The ejaculates of 6 sires were repeatedly collected. Fresh semen was diluted with one extender and divided into 3 groups equilibrated for 30, 120, and 240 min. Subsequently, half straws of each group were frozen using standard 3-phase or 2-phase freezing curve differing in the rate of temperature decrease. The spermatozoa motility (M) was evaluated immediately after thawing and at 30, 60, 90, and 120 min of thermodynamic test (TDT). Live spermatozoa proportion was evaluated after thawing and at the end of TDT. Average of spermatozoa motility (AM), decrease of spermatozoa motility (MD), average proportion of live spermatozoa (ALS), and decrease of live spermatozoa proportion (DLS) through the TDT were calculated. Significant inter-sire differences in AM (0.45-17.0%; P < 0.05-0.01), MD (0.76-12.57%; P < 0.05-0.01), and ALS (0.99-23.8%; P < 0.01) were detected. The longest equilibration ensured the highest M during TDT and AM (+2.72 and +4.58%; P < 0.05-0.01), however higher MD (+4.06%; P < 0.01) compared to standard length as well. Straws freezed using 2-phase curve achieved higher M through TDT, AM (+7.3%; P < 0.01) as well as ALS (+11.77%; P < 0.01). The 2-phase curve presented higher M compared to the 3-phase freezing curve within all equilibration lengths. Significant differences in AM, MD, and ALS (0.45-6.78%, 0.62-5.35%, and 20.79-21.11%; P < 0.05-0.01) between equilibration length vs freezing curve interactions were determined. Results document the importance of equilibration length, freezing curve, and their interaction effect on live spermatozoa proportion and sperm motility after thawing as well as necessity of individual conditions for bulls semen processing and insemination doses production.

Plum cultivars Zlatka and Pozna Plava (Prunus domestica L.) bred at the Fruit Research Institute in ČačakOriginal Paper

This study was carried out to compare the most relevant biological characteristics and field resistance to economically important diseases of the new (Zlatka and Pozna Plava) and standard plum cultivars (Čačanska Lepotica and Čačanska Rodna) in the Region of Čačak. The experimental area climate belongs to the temperate type. The mean annual air temperature during the experimental period was 12.65°C, and humidity was 80.4%. The average annual rainfall was 571.2 mm. The ripening time of the cv. Zlatka occurred between August 3-23, while the cv. Pozna Plava ripened between August 26-September 25. Compared to cv. Čačanska Lepotica, cv. Zlatka had a significantly lower mean fruit weight, reducing sugars and total acids content, but a significantly higher proportion of flesh, yield and sucrose content. Compared to cv. Čačanska Rodna as the standard cultivar, cv. Pozna Plava had a significantly higher mean individual fruit weight and total acids content, but also a significantly lower orchard yield and sweetness measured as fruit soluble solids, total sugars, reducing sugars and sucrose content. As regards the field resistance to economically important diseases such as red leaf spot, rust, fruit root, pockets plum disease and sharka disease, both new cultivars demonstrated better results than the reference cultivars.

Headed Chinese cabbage growth and yield influenced by different manure types in organic farming systemOriginal Paper

Chinese cabbage is a less known, cultivated and consumed vegetable species in Transylvania, although previous studies have shown that it can be grown successfully in this area. The main purpose of the present research was to underline the effectiveness of various treatments on Chinese cabbage growth, yield and quality of the final product in organic farming system. The treatments included: different fertilisation types, place of the culture and several hybrids. Results showed that all the studied factors had a high influence on cabbage heads characteristics such as length, diameter, and weight, on the number of leaves and also on yield. The best yields were registered when horse manure was used (between 76.50 and 99.30 t/ha), followed by cattle manure (between 76.80 and 93.75 t/ha). A higher yield of better quality cabbage was obtained in a polyethylene tunnel (89.27 t/ha) compared to the open field culture (77.50 t/ha). Hybrid Super provided the highest yield (92.70 t/ha), followed by hybrid Vitimo (86.09 t/ha).

Effects of rootstock genotypes on compatibility, biomass, and the yield of WelschrieslingOriginal Paper

The aim of this work is to determine the compatibility, the scion biomass, and the yield of the grapevine variety Welschriesling grafted onto 12 grapevine rootstocks. As an index of compatibility, the callus development and graft success were determined. Dry weight of canes was measured at the end of the growing seasons (2011-2014), while root dry weight only in the first year in the nursery. The grape yield was measured in the first production year. Welschriesling showed good compatibility with all examined rootstocks. More than 85% of grafts had a complete callus development (8BČ rootstock 100%). The average of graft success in the nursery was 67%, but the average of 5BB, G251, and G103 was above 80%. The G103 rootstock had the highest root dry weight after one season. The dry weight of canes in vineyards was above the average with 5BB, SO4, Binova, Börner, and M V rootstocks. All Georgikon rootstocks had a lower cane dry weight per vine than the others. The highest yields were recorded on SO4, G251, and Börner rootstocks.

An economic aspect of conversion of Scots pine (Pinus sylvestris L.) stands in the Polish Lowland

Pine stands at fertile sites need conversion into broadleaved or mixed forests. Yet, according to the age of stand, conversion may have different economic results. Therefore, in order to reduce costs of the process, the analysis on the basis of the potential income from wood sale has been done. The aim of the analysis has been to find an optimal date of conversion beginning. The date of conversion beginning has been fixed on the basis of the relation of the value of wood increment from the stands which need conversion to the value of average wood increment from stands conformable to the sites. By this means, three age intervals have been appointed.

Conditions conducive to an epidemic of Gremmeniella abietina, European race, in red pine plantationsOriginal Paper

In North America, Gremmeniella abietina, European race (GaEU), was reported in 1975. Our objective was to follow the spread of GaEU on red pines growing on flat land and on slope. Annual height infection varied significantly on flat land, ranging from 60 to 110 cm in 1991 to 0 to 50 cm in 1992. On the slope, pines in the bottom were killed by the disease, but survived on the top. Favorable conditions follow a horizontal line about 10 m over the lower elevation and are probably related to fog or mist. The horizontal disease spread over a 3-year period was only 20 m and this is mainly explained by the absence of ascospores in North America.

Why was browsing by red deer more frequent but represented less consumed mass in young maple than in ash trees?!Original Paper

European ash (Fraxinus excelsior) and sycamore maple (Acer pseudoplatanus) are considered amongst tree species as some of the most attractive forage for red deer (Cervus elaphus). The aim of this paper is to estimate branch and foliage mass browsing by red deer in young Sycamore maple and European ash in Central Slovakia and to analyze interspecific differences in browsing frequency and quantity. Altogether 665 ash trees and 701 maple trees were measured for height and stem diameter at the ground level. Red deer browsing, defined as leader shoot or lateral branch cropping, was recorded for all trees. A combination of the diameter of browsed branches and branch regression models allowed for the estimation of browsed (potentially consumed) mass by red deer. Results show that browsing occurred more frequently on maple (5 cases per tree) compared to ash (3 cases per tree); however, more total branch biomass was consumed in ash (10.7 g per tree) compared to maple (2.6 g per tree). This is because browsed branches were larger in ash than in maple. We assume that the difference in the size of browsed branches between ash and maple is relative to species-specific branch morphological and structural features.

Objective and subjective assessment of selected factors of the work environment of forest harvesters and forwardersOriginal Paper

This article was aimed at assessment of selected factors of the work environment, which influenced the operators of harvesters and forwarders. The selection of the work environment factors was based on a thorough literature survey. From the survey exposure to whole-body vibrations, noise (equivalent and peak), microclimatic conditions and mental load were selected for detailed observations. The measurements were conducted according to international standards and assessed according to European or national legislation. Subjective assessment of the effects of the work environment was conducted through a structured interview. The exposure to whole-body vibrations was 0.5 m.s-2. The equivalent noise exposure was 78 dB, peak noise exposure was 130 dB. The average temperature inside the machine cabs was 17.9°C and average airflow velocity was 0.06 m.s-2. Mental load was high according to the Meister questionnaire, three operators considered the load inadequate, two considered it adequate. Subjective assessment showed that operators considered other factors to be light or no load

Impact of climatic conditions on the reproductive phenological phases of European hazel (Corylus avellana L.) in SlovakiaOriginal Paper

The work presents the results of phenological observations of flowering and fruit ripeness of the European hazel (Corylus avellana L.) in Slovakia in two time periods: 1964-1983, and 1994-2013. The phenological stations at elevations from 100 to 875 m a.s.l. were grouped to 7 elevation zones, each 100 m wide. In the first studied period, flowering started on 19 March on average, while in the second period it started 5 days earlier. The average duration of flowering equal to 7-9 days did not depend on the elevation, but on the air temperature and weather patterns in the different elevation zones. During the first period, the average elevation gradient of flowering was 5.6 days per 100 elevation meters, while in the second period it was reduced to 3.5 days in the entire elevation profile. In the elevation zones between 300 and 500 m, a high correlation (P < 0.01) between the onset of flowering and elevation was reduced to a moderate level of correlation due to changing environmental conditions. In 1994-2013, the ripeness of hazel nuts started on average on 30 August in Slovakia, showed low variability and moderate correlation with elevation. The phenological elevation gradient was 2.9 days per 100 m of elevation.

Relation between selected indicators of forest stand diversity and quality of timber production in managed Central European forestsOriginal Paper

The present study examines the relationship between the quality of timber production and the species and structural diversity of forest stands. The data used came from a regional forest inventory of the University Forest Enterprise "Kostelec nad Černými lesy", Czech Republic. The inventory was performed from 2009 to 2011 on 1,188 sample plots that represented 86 strata defined by the combination of three variables: site (5 categories), age (12 categories) and canopy cover (5 categories). On each sample plot, we quantified 171 partial biodiversity indicators that represented species or structural diversity. The quality of timber production was specified by four indicators quantified using local assortment tables. In total, we analysed 58,824 univariate linear regressions describing the relationships between diversity indicators and timber quality in individual strata. The results revealed that their relationship changes with stand age. The proportion of the best-quality assortments increases with the increasing species richness in all age categories.

Effect of permanent waterlogging on the growth of poplar clones MAX 4, MAX 5 (J-104, J-105) (Populus maximowiczii A. Henry × P. nigra Linnaeus) and evaluation of wood moisture content in different stem parts - Short CommunicationShort Communication

The study deals with differences in the growth of 2-year mix of poplar clones MAX 4, MAX 5 in the first rotation period at a site with the average moisture regime and at a neighbouring site affected by waterlogging. Average diameter at breast height (DBH) at the site without the observable water influence was 19.6 mm (average height 380 cm). Average DBH at the water-affected site was 8.1 mm (height 220 cm). The difference was statistically significant, which corroborates the hypothesis of the adverse influence of waterlogging on the growth of poplar clones. Furthermore, moisture content was ascertained in wood samples. A comparison was made of samples from the butt, middle part and top part of the stem. Differences between the respective stem parts were not statistically significant. Total average wood moisture content at the time of harvest (January 2012) was 59.7%.

To enhance the induction of shoots, the excised nodes were cultured on Murashige and Skoog medium containing different cytokinins and various concentrations. The best conditions for shoot induction and growth were with 6-benzyladenine at 1.0 mg.l-1. For axillary shoot proliferation, the excellent result was obtained using 2.0 mg.l-1 meta-topoline. Well-developed shoots (more than 2 cm in length) were successfully rooted ex vitro at 82% by treatment with commercial rooting powder (2% indole-3-butyric acid; Rhizopon®). Rooted plantlets were acclimatized in the greenhouse and the survival rate of transplantation reached 80%.
