Comparison of indirect optical methods for estimating effective leaf area index in young Norway spruce standsOriginal PaperJakub Èerný, Jaroslav Èepl, Zdenìk Vacek, Jan Cukor, Stanislav VacekJ. For. Sci., 2026, 72(6):285-296 | DOI: 10.17221/42/2026-JFS Forest canopy structure is crucial for regulating light interception, carbon exchange, and water balance, making accurate estimation of leaf area index (LAI) essential for forest ecology and management. This study compared three indirect optical methods (LAI-2200 PCA, LaiPen LP 110, and digital hemispherical photography – DHP) for estimating effective leaf area index (LAIe) in young Norway spruce [Picea abies (L.) Karst.] stands. It evaluated the influence of the field of view (FOV) and seasonal timing on inter-method agreement. The research was conducted in young, even-aged Norway spruce stands at the Køivina experimental site in the Czech Republic, using three research plots with varying stand densities. Results showed that LAIe estimates were influenced by the selected FOV, with mean values generally decreasing as FOV increased, particularly for the LAI-2200 PCA. Narrow FOV configurations produced greater local variability among sampling positions, whereas broader FOVs resulted in more homogeneous spatial canopy patterns. The strongest agreement among methods was observed between LaiPen LP 110 and LAI-2200 PCA, with correlation coefficients ranging from 0.71 to 0.84 across temporally matched measurements. In contrast, DHP exhibited weaker and more variable relationships with LaiPen LP 110. Differences among research plots were most pronounced between the dense control stand and the heavily thinned stand, indicating that canopy density and structural heterogeneity substantially affected LAIe estimation. The study demonstrates that indirect optical methods are sensitive to both canopy structure and measurement configuration; therefore, careful instrument selection, FOV standardisation, and synchronised measurements are essential for obtaining reliable and comparable LAIe estimates. |
Production and quality assessment of fig wine: A comparative study on Turkish fig varietiesOriginal PaperZiya Binat, Ayºegül Kırca-ToklucuCzech J. Food Sci., 2026, 44(3):192-203 | DOI: 10.17221/201/2025-CJFS This study aimed to produce technologically viable fig wines from two main Turkish fig varieties (Sarilop and Bursa Siyahi) and to evaluate their quality and acceptability. Fig musts were prepared at two soluble solids concentrations (17 and 24 °Bx) using fresh and sun-dried Sarilop (yellow-coloured) and fresh Bursa Siyahi (dark purple-coloured) figs. Total phenolic content of fig wines ranged from 223.21 to 267.98 mg gallic acid equivalents (GAE)·L–1, while total antioxidant capacity varied from 3.73 to 4.42 µmol Trolox·(100 mL)–1 (ABTS) and 35.09 to 69.30 µmol Trolox·(100 mL)–1 (DPPH). Wines produced from fresh Bursa Siyahi figs exhibited the highest antioxidant activity. Both fig variety and fruit form (fresh or dried) significantly affected phenolic composition. Epicatechin and chlorogenic acid were predominant in wines produced from fresh Bursa Siyahi figs, whereas rutin was dominant in wines made from fresh Sarilop figs. Wines produced from musts with 24 °Bx showed significantly higher descriptive sensory scores than those produced from musts with 17 °Bx (P < 0.05). No significant difference in overall impression was observed between wines made from fresh and dried Sarilop figs. |
Current trends and future perspectives in dairy by-product valorisationReviewAslıhan TüğenCzech J. Food Sci., 2026, 44(3):167-180 | DOI: 10.17221/7/2026-CJFS The increase in milk production and demand for dairy products worldwide has led to the emergence of a wide variety of products in the sector, whilst a high volume of solid (sludge) and liquid waste is also generated. Dairy industry wastes, due to their high organic load (biological oxygen demand, chemical oxygen demand), high moisture content and variable composition, can cause significant environmental and economic problems when disposed of directly. The aim of this research is to determine current methods and possible future trends for the valorisation of dairy industry by-products (whey, casein and lactose-containing wastes, wastewater and sewage sludge). Literature from 2010–2025 was systematically screened in databases such as Web of Science, ResearchGate, ScienceDirect, PubMed and Google Scholar in line with the PRISMA guide, and a total of 34 documents were examined. According to the results, various technologies aimed at conversion into valuable products (valorisation) and integration into the circular economy have come to the fore. Thermochemical methods such as hydrothermal carbonisation (HTC), hydrothermal liquefaction (HTL), pyrolysis and gasification increase energy and chemical substance recovery from wastes with high moisture content. Biological methods (anaerobic and aerobic digestion, composting, microbial fermentation) yield value-added products such as biogas, compost, bioplastic and single cell protein (SCP). The necessity of integrated systems combining both thermochemical and biological approaches is also established in the literature. |
Characterisation of Colletotrichum species associated with anthracnose disease in red chilli pepper (Capsicum annuum L.) in the South of Vietnam and the effectiveness of the consortium Bacillus amyloliquefaciens and Pseudomonas fluorescens Original PaperVo Thi Ngoc Ha, Huynh Thuong Vuong, Tran Bao Thang, Huynh Thanh HungPlant Protect. Sci., 2026, 62(3):239-251 | DOI: 10.17221/41/2025-PPS Anthracnose is one of the most destructive diseases that limits pepper production and quality worldwide. In this study, the causal agent of anthracnose in red chilli pepper in Southern Vietnam was collected and identified based on morphological characteristics and multilocus sequence regions (ITS, β-tubulin, GPDH, ACT). The antifungal activity of Bacillus amyloliquefaciens and Pseudomonas fluorescens was evaluated in vitro and in vivo under greenhouse conditions. The results revealed that the morphological analysis categorised the Colletotrichum isolates into three species: C. acutatum, C. gloeosporioides, and C. scovillei. Sequence analysis of the four genes confirmed that C. scovillei was the causal agent of anthracnose in chilli pepper in Southern Vietnam. B. amyloliquefaciens and P. fluorescens bacteria demonstrated antifungal activity against C. scovillei in vitro, with mycelial growth inhibition rates ranging from 20.79% to 78.69%. The consortium of B. amyloliquefaciens CC-LD2.4, P. fluorescens CC-FN1.1, and P. fluorescens O-BT1.2 achieved 84.4% control efficacy at 7 days after inoculation (DAI), which declined to 68.5% at 14 DAI and 41.7% at 21 DAI, at the flowering stage, and achieved 100% control efficacy at the fruiting stage. B. amyloliquefaciens CC-LD2.4 showed very high chitinase, protease, and cellulase activities (halo diameter of 26.7 mm, 22.7 mm, 21.5 mm), whereas P. fluorescens CC-FN1.1 was very high in protease and cellulase (14.3 mm, 12.4 mm) but weak in chitinase (5.1 mm), and P. fluorescens O-BT1.2 exhibited overall lower activities (3.4–9.9 mm). There is still considerable room to optimise bacterial consortia to develop bio-fungicides that meet the requirements for an alternative or advanced solution for controlling anthracnose in red chilli peppers in sustainable agriculture. |
Modern advanced food safety and smart traceability systemsReviewAslıhan Tüğen, Louiza Belkacemi[Ahead of Print]CAAS Agricultural Journals, X:X | DOI: 10.17221/130/2025-CJFS This systematic review article aimed to examine the effects of digitalisation in the field of food safety and traceability on applications and the challenges encountered in the context of the food industry. The decentralised structure of blockchain technology prevents forgery in records. It has been determined that tools such as IoT sensors and Radio Frequency Identification (RFID) facilitate quality control and logistic processes, ensure consumer trust, savings and cost reduction in logistics and supply. High investment and energy costs make it difficult to use these technologies. The incompatibility of regulatory policies and standards in different countries is a factor that makes difficult to create a global food traceability ecosystem. Despite all these challenges, blockchain and IoT technologies are expected to contribute more to food safety and traceability applications in the future. |

The application of choice experiments in a study on consumer preference for agri-food products: A literature reviewReviewArif Yustian Maulana Noor, Hery Toiba, Budi Setiawan, Abdul Wahib Muhaimin, Adhitya Marendra KiloesAgric. Econ. - Czech, 2022, 68(5):189-197 | DOI: 10.17221/429/2021-AGRICECON A choice experiment (CE) is a stated preference method to elicit the respondent's preference. The CE can predict consumer valuation for a hypothetical product's attributes. Many scholars have discussed the CE's design, analysis, reliability, and validity. Still, no scientific papers reviewed its application in agri-food studies in a broad spectrum, particularly in investigating food product categories and their attributes. Additionally, this review emphasises the technical aspects of CEs, such as the sample size, software, data analysis, and research implications. The result discovered that most CE studies are relevant to developed countries. Most reviewed studies observed wine and meat as objects in the CE studies, in the theme of health, food safety, origin, and sustainability. The future research trend was related to health benefits and natural ingredients. Thus, this review provides recommendations for future studies to explore consumer preference using CE in agri-food research. |
Nutritional evaluation of the full-day dietOriginal PaperBarbora Pohoøelá, Andrea Poláchová, Markéta Rù¾ièková, Marek Dole¾al, Jana Pulkrabová, Jan PánekCzech J. Food Sci., 2022, 40(2):118-129 | DOI: 10.17221/273/2021-CJFS Nutrition plays an important role in human life. So far, there have been discussions focusing on the nutritional value of individual foods, separate dishes, or daily meals. However, they have not taken into account the composition of the diet in the longer term. The aim of this work was to evaluate a full-day diet from a currently renowned company that is producing box diets on the Czech market against a full-day diet of twenty randomly selected students. The results showed that the box diet met the required amount of protein, fat, vitamin E, cholesterol, and also the optimal intake of n-6 and n-3 fatty acids. The amount of saturated fatty acids (SFA) was only slightly increased. Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) content averaged 38 mg day-1, which did not meet the current requirements. The students' full-day diet was variable, the individual differences were large. Protein intake could be assessed as sufficient in most cases. The total fat intake ranged from 21 g day-1 to 126 g day-1. Seven out of twenty samples would satisfy the recommendation for SFA. The content of the essential acids EPA and DHA was below the detection limit with the exception of one sample (containing herring fillets). |

Forestry violations as a global issue of legal regulation in the field of forest control and supervision: the Krasnodar territory and the Republic of Bashkortostan case studyOriginal PaperRadmir Iksanov, Olga Khalikova, Igor Vladimirov, Ravil Gizzatullin, Regina Baiturina, Vitaliy Kovshov, Aigul Selezneva, Sofya KhasanovaJ. For. Sci., 2021, 67(6):272-284 | DOI: 10.17221/99/2020-JFS Control and supervision activities in the field of forest management are a type of activity of state authorities to detect, prevent forest violations. The purpose of this study is to analyse the dynamics and nature of violations in the forest sector on the example and in comparison of such regions of Russia as the Krasnodar territory and the Republic of Bashkortostan. The research methodology is selected taking into account the characteristics of the object under study. With the assistance of employees of the environmental inspectorate, search operations were organized in the areas where the greatest amount of forest damage occurs in the studied regions. The authors concluded that one of the reasons for the existing problems is the lack of interest of the state in the development of this sector of economy. The paper focuses on specific types of forest violations and the problems of their prevention. The authors believe that illegal logging belongs to the most common and socially dangerous forest violations. The article concludes that when adopting new regulations for control and supervisory activities in the field of forest management, the legislator must take into account requirements of administrative reform, regulatory guillotine, and risk-based approach. |
Spruce forest litter structure, distribution, and water retention along hiking trails in the Ukrainian CarpathiansOriginal PaperYurii Ivanenko, Ganna Lobchenko, Volodymyr Maliuha, Vasyl YukhnovskyiJ. For. Sci., 2022, 68(7):241-252 | DOI: 10.17221/12/2022-JFS Almost 24% of the Ukrainian Carpathian ecosystems are assigned to the conservation fund due to their ecological values and attraction to numerous tourists. The forest litter in mountain forest stands plays an important role in terms of its ability to mitigate the impact of tourist activities, and erosion processes along with its contribution to the soil mineralization. Water interception, infiltration, and retention ability of forest litter have an impact on hydrological processes of forest ecosystems. At the same time, the accumulation and spatial distribution of litter can be affected not only by environmental conditions, but also by tourism. In this study, 13 Norway spruce (Picea abies L.) stands distinguished by average distance to the trail were chosen to investigate whether there are any differences in litter structure, water retention, and infiltration abilities as well as litter accumulation in areas along popular hiking trails in the Ukrainian Carpathians. Results showed that the litter thickness has increased with altitude and slope steepness. Moreover, results of one-way ANOVA demonstrate a significant difference (P < 0.05) in litter stock between two groups of sample plots: established directly to adjoin hiking trails and at a distance. Therefore, the forest litter stock nearby hiking trails may indicate that trampling caused by tourists has a negative impact on litter accumulation. However, no statistical difference was found (P > 0.05) in the accumulation of litter along three trails and altitudes across all surveyed forest stands. Results of the immersion test showed that the litter infiltration rate has a significant negative correlation with the maximum mass of absorbed water (r = -0.62, P < 0.05), litter stock (r = -0.69, P < 0.01), and retained precipitation (r = -0.62, P < 0.05). Despite our assumption, the infiltration rate was lower for sites distanced from the hiking trail in comparison with adjoining ones. According to our measurements, the water holding capacity of the litter varies from 42.3 t.ha-1 to 187.3 t.ha-1 regardless of the stand composition. Further, the precipitation amount retained by litter varies between 4.2 mm and 18.7 mm. Insignificant differences in litter fractional structure and accumulation were observed in pure spruce stands and mixed spruce-beech with an admixture of beech up to 20%. Further observational and modelling studies are necessary to clarify the role of the beech share in stand composition in relation to fractional structure and water interception and retention ability. |
Radial growth, present status and future prospects of west Himalayan fir (Abies pindrow Royle) growing in the moist temperate forest of Himalayan mountains of PakistanOriginal PaperZahid Rauf, Adam Khan, Samina Siddiqui, Sidra Saleem, Tahir Iqbal, Safdar Ali Shah, Nowsherwan Zarif, Wahiba IqbalJ. For. Sci., 2022, 68(9):344-356 | DOI: 10.17221/3/2022-JFS Forests play a significant role for maintaining the biodiversity. In order to manage sustainable forests, tree species history, distribution, and their future prospects are vital. Using standardized quantitative approaches, the age, radial growth, and size class distribution of Abies pindrow (Himalayan fir) were determined from three different altitudinal sites (i.e. high, middle, and lower). The results indicate that Himalayan fir growing in the high-altitude site (Ayubia, 2 917 m a.s.l.) of moist temperate forests of the Himalayan mountains showed lower radial growth (0.13 cm) than in the middle (Bara Gali, 2 617 m a.s.l.; radial growth = 0.13 cm) and lower (Kuldana, 2 455 m a.s.l.; radial growth = 0.22 cm) altitude sites. Correlation analysis demonstrated that age showed a significant positive correlation (P < 0.001) with diameter at breast height. The tree-ring width chronology (totally 80 core samples) of Himalayan fir was developed from moist temperate forests of Himalayan mountains of Pakistan. At Ayubia site it possesses a long time-span (1703-2020 C.E.), followed by Bara Gali (1862-2020 C.E.) and Kuldana (1864-2020 C.E.). Further, the tree-ring width (TRW) chronology of Ayubia showed a significant positive correlation (P < 0.05) with May and June temperature, and a significant negative correlation (P < 0.05) with June and October precipitation, indicating that summer temperatures are the key factor for the radial growth of Himalayan fir. For the Kuldana site, the response of TRW chronology to temperature and precipitation was the same, however, it was significant only for June temperature at Bara Gali. The size class distribution of the high-altitude region (Ayubia) showed a higher number of individuals than the lower altitude region, indicating the lowest disturbance conditions. The absence of individuals in the early size classes and the gap in middle and mature size classes indicate a lower regeneration potential and anthropogenic impact. The pointer year analysis indicated that the Bara Gali forest is more sensitive to abnormal climate events than the other sites. Based on the present study, we suggest that proper attention and conservation strategy should be provided to Himalayan fir growing in the moist temperate forests of Pakistan. |
Effect of endophytic bacterium, Stenotrophomonas maltophilia JVB5 on sunflowersOriginal PaperBartholomew Saanu Adeleke, Ayansina Segun Ayangbenro, Olubukola Oluranti BabalolaPlant Protect. Sci., 2022, 58(3):185-198 | DOI: 10.17221/171/2021-PPS Identifiable endophytic bacteria with plant growth-promoting traits promise to ensure sustainable agriculture. However, information on the versatility and exploration of sunflower-associated bacteria as bioinoculants is less studied. Here, we present the whole-genome sequence and annotation of Stenotrophomonas maltophilia JVB5 isolated from the sunflower root endosphere from the North West province, South Africa. The whole-genome analysis revealed a genome size of 4 771 305 bp, a sequence read count of 8 764 890, a 66% guanine-cytosine content, 57 tRNAs, 268 contigs, and 4 160 protein-coding genes with functions in various metabolic pathways. Pathways involved in the indole acetic acid production were found in the S. maltophilia JVB5 genome. The whole-genome annotation predicted notable genes involved in bacterial colonisation, antibiosis, and plant growth promotion. The predicted genes are involved in the sulfur metabolism, and the oxidative stress may enhance the plant growth promotion and boost plant the resistance to stress. Upon inoculation, S. maltophilia JVB5 efficiently colonised the sunflower root under greenhouse conditions with a significant improvement on the wet plant weight of 437.20 g compared to the uninoculated control with a 331.04 g wet weight. The genomic analysis revealing specific functional genes in the bacteria genome suggests their bioprospecting in agriculture. Hence, understanding the mechanisms employed by S. maltophilia JVB5 based on the predicted multifunctional genes will help harness their bioresource in sustainable plant health. |
Measurement of post-dispersal invertebrate seed predation of some relevant weed species in maize fields in Hungary: An ecosystem service provided in crop fields contributing to weed managementOriginal PaperMohammed Gaafer Abdelgfar Osman, Mark Szalai, Mihaly Zalai, Zita Dorner, Jozsef KissPlant Protect. Sci., 2022, 58(4):351-359 | DOI: 10.17221/159/2021-PPS Invertebrate seed predation is a potential ecosystem service that substantially reduces weed seeds in crop fields, decreasing the seedling emergence and, thus, limiting the weed competition next season. It may, thus, be considered as a natural component of the long-term weed management toolbox. This study aimed to measure the post-dispersal invertebrate seed predation levels of the following relevant weed species in Hungarian maize fields: Ambrosia artemisiifolia, Datura stramonium, Chenopodium album, and Echinochloa crus-galli, and to compare the predation levels among them. We hypothesised that invertebrate seed predators will predate weed seeds, but the predation levels may vary with the weed species. Two sampling rounds were performed, in November 2019 and October 2020, in Gödöllõ, Hungary. A total of 100 seed cards/round were placed on the soil surface inside a maize field prior to harvest, 10 m from the field's edge, along 25 transects, with four cards/transect. A distance of 10 m was set between the transects and 1 m between the cards. Twenty seeds of each weed species were glued onto sandpaper (25 × 10 cm, P-60), and a wire mesh was used to exclude vertebrate predators. The seed removal was calculated every 24 h, for seven days in 2019 and for five days in 2020, and then the seed predation was measured using the number of removed seeds on each card. The results showed high seed predation levels on all the seed cards, with an overall average of 85.9 ± 13.7%. Besides, there was a decrease in the % of remaining seeds on the cards starting from the first day after exposure due to seed predation in both years. The optimum exposure period for measuring the seed predation was found to be three to four days, though the number of predated seeds on days 3 and 4 significantly differed between years (P < 0.001), with higher predation rates in 2020 than in 2019. However, no differences were detected in the predation rates among the weed species (P = 0.962, 0.079). These findings indicate the potential contribution of seed predation by invertebrates to weed management in Hungarian maize fields. |
Nitrogen addition turns a temperate peatland from a near-zero source into a strong sink of nitrous oxideOriginal PaperBoli Yi, Fan Lu, Zhao-Jun BuPlant Soil Environ., 2022, 68(1):49-58 | DOI: 10.17221/411/2021-PSE Peatlands, as important global nitrogen (N) pools, are potential sources of nitrous oxide (N2O) emissions. We measured N2O flux dynamics in Hani peatland in a growing season with simulating warming and N addition for 12 years in the Changbai Mountains, Northeastern China, by using static chamber-gas chromatography. We hypothesised that warming and N addition would accelerate N2O emissions from the peatland. In a growing season, the peatland under natural conditions showed near-zero N2O fluxes and warming increased N2O emissions but N addition greatly increased N2O absorption compared with control. There was no interaction between warming and N addition on N2O fluxes. Pearson correlation analysis showed that water table depth was one of the main environmental factors affecting N2O fluxes and a positive relationship between them was observed. Our study suggests that the N2O source function in natural temperate peatlands maybe not be so significant as we expected before; warming can increase N2O emissions, but a high dose of N input may turn temperate peatlands to be strong sinks of N2O, and global change including warming and nitrogen deposition can alter N2O fluxes via its indirect effect on hydrology and vegetation in peatlands. |
Responses of nitrogen accumulation and translocation in five cytoplasmic hybrid rice cultivarsOriginal PaperNoor Muhammad, Yating Zheng, Farhan Nabi, Guotao Yang, Sumbal Sajid, Abdul Hakeem, Xuechun Wang, Youlin Peng, Zartasha Khan, Yungao HuPlant Soil Environ., 2022, 68(2):73-81 | DOI: 10.17221/354/2021-PSE To study the difference among cytoplasm at the different nitrogen conditions, a research experiment was conducted using five different cytoplasmic male sterile (CMS) hybrid rice with nitrogen levels at N0, N1, N2, and N3; the nitrogen application rates were 0, 90, 180, and 270 kg/ha, from 2018 to 2019. Results showed that among tested cultivars of CMS hybrid rice, JW (J803A × Chenghui727) showed the highest yield in both years for the low nitrogen and high nitrogen treatments. The dry matter accumulation and translation of JW type in nutritive organs were higher than that of others during the low nitrogen level (N1). We concluded that the nutrient translocation within plants organs and dry biomass accumulation were highly dependent on CMS type and nitrogen application. This research indicates that selecting a rice cultivar with greater efficiency of nitrogen is favourable for raising the number of grains per panicle, grain yield, and nitrogen use efficiency. JW cytoplasm displayed great efficiency in low nitrogen, which is a potential cytoplasmic resource. |
Biochar-based fertiliser improved the yield, quality and fertiliser utilisation of open field tomato in karst mountainous areaOriginal PaperMeng Zhang, Yanling Liu, Quanquan Wei, Lingling Liu, Jiulan GouPlant Soil Environ., 2022, 68(4):163-172 | DOI: 10.17221/471/2021-PSE Biochar-based fertiliser (BF) is beneficial to improve yield and quality, but the effect of BF on open field tomato remains unclear, especially in karst mountainous areas. The objective of this study was to identify the application effect and optimum application rate of BF. A field experiment was carried out in Southwestern China from 2019 to 2020 to study the effects of different application amounts of BF on the yield, quality, nutrients accumulation and fertiliser utilisation of open field tomatoes. The results showed that compared with the traditional fertilisation practice, BF can significantly increase the yield of open field tomato by 5-9% (2019) and 12-23% (2020), and significantly reduce nitrate content and increase vitamin C content of fruits. Meanwhile, nutrient accumulations, agronomic efficiency, and recovery efficiency of BF treatments were all significantly improved. In conclusion, the BF rate of 2 326 kg/ha improves yield and fertiliser utilisation in open-field tomatoes and could be recommended for tomato production in karst mountainous areas. |
Clinical and radiographic evaluation of minimally invasive plate osteosynthesis (MIPO) in dogs with tibial fracturesOriginal PaperC Istim, M AricanVet Med - Czech, 2022, 67(6):316-322 | DOI: 10.17221/42/2021-VETMED The aim of this study was a clinical and radiographic evaluation of the minimally invasive plate osteosynthesis (MIPO) method for treatment in dogs with tibial fractures. Ten dogs of different breeds, ages, and sex with tibial fractures were used as the subjects. A medial approach to the tibia was used in all the cases. The tibial tuberosity was palpated proximally and a short incision was made along the midline. After reduction of the fracture, proximal and distal insertion incisions were made to approach the tibia. The mean times for the MIPO operations were 21 minutes. The patients were able to stand and walk postoperatively on the first day and recovery was rapid for all of the dogs. The fracture healing was completed on day 45 based on the radiographic evaluations of these cases. Dogs with tibia fractures treated by MIPO healed rapidly without any complications (90%). Correct fracture selection is important for the use of MIPO to be successful. Further studies with a larger group of dogs should be considered to compare the efficacy of MIPO and other fixation techniques and for the long-term evaluation of the repaired tibial fractures. |
Immune responses and protective efficacy of a trivalent combination DNA vaccine based on oprL, oprF and flgE genes of Pseudomonas aeruginosaOriginal PaperQ Gong, Y Li, W Zhai, M NiuVet Med - Czech, 2022, 67(12):611-619 | DOI: 10.17221/86/2021-VETMED Pseudomonas aeruginosa is an infectious pathogenic bacteria infecting many different species of animals. Currently, it lacks a commercial vaccine. In this study, three monovalent DNA vaccines (poprL, poprF, and pflgE), three bivalent combination DNA vaccines (poprL+poprF, poprL+pflgE, poprF+pflgE), and a trivalent DNA vaccine (poprL+poprF+pflgE) were constructed. Consequently, we immunised chickens with these DNA vaccines and used inactivated vaccines as the positive controls. Then, the immune efficacy was evaluated through serum antibody detection, a lymphocyte proliferation assay, and cytokine concentration determination. Lastly, we assessed the protection rate through a challenge experiment. Following vaccination, the serum antibody levels induced using these DNA vaccines were different due to the different coating antigens. In the trivalent combination DNA vaccine group, we established that the lymphocyte proliferation (SI values), IFN-γ, IL-2, and IL-4 levels were significantly higher than those of the other six DNA vaccine groups and the inactivated vaccine group. However, the protection provided was slightly lower than that of the inactivated vaccine and higher than those of other DNA vaccines. The protection rate of poprL, poprF, pflgE, poprL+poprF, poprL+pflgE, poprF+pflgE, poprL+poprF+pflgE, and the inactivated vaccine were 50, 45, 60, 75, 80, 80, 90, and 95%, respectively. The results of this study indicated the trivalent DNA vaccine based on oprL, oprF and flgE genes represents a promising approach for the prevention of Pseudomonas aeruginosa infections. |
Molecular evaluation of selected mango genotypes grown in Egypt using SCoT and ISSR markersOriginal PaperIbrahim El-Shenawy Ghounim, Mohammed Raafat Mustafa, Ibrahim HmmamHort. Sci. (Prague), 2022, 49(4):213-224 | DOI: 10.17221/112/2021-HORTSCI The mango (Mangifera indica L.) is a common tropical and subtropical fruit and the second main fruit crop in Egypt. In this work, start codon targeted (SCoT) and inter simple sequence repeat (ISSR) markers were employed to study the genetic relationship among twenty-three mango genotypes grown under the El-Giza district conditions. These genotypes consist of four newly introduced cultivars, six new progenies and thirteen closely related mango cultivars which were included as cultivar references for the best definition. Based on the overall SCoT and ISSR data, a total of 52 bands were generated (18 for SCoT and 34 for ISSR) with a total polymorphic percentage equal to 53.84% (44.44% for SCoT and 58.82% for ISSR). Based on the whole set of SCoT and ISSR marker data, the genetic similarity coefficients ranged from 80 to 97% with a mean equal to 88.5% and the first two principal axes explained about 31% of the total genetic variation among the mango accessions. The SCoT and ISSR data revealed that the four newly introduced mango genotypes, namely 'Aya', 'Kasturi', 'Maya' and 'Omer', are novel genetic resources based on their genetic profiles, which can be used for future breeding programmes in Egypt. |

Sci., 2023, 59(1):19-30 | DOI: 10.17221/90/2022-PPS Sisal purple leafroll disease (SPLD) is a destructive disease affecting sisal in China, however, its etiology remains unknown. This disease is characterized by purple margins and rolling and yellowing of the leaves. Recently, phytoplasma was found to be present in SPLD-affected sisal according to PCR assay. To further verify the presence of phytoplasma, determine its association with SPLD and identify the possible pathogenic phytoplasma infecting sisal, 80 symptomatic and 65 asymptomatic sisal samples were collected from the main sisal farms in three Chinese provinces, including Hainan, Guangdong and Guangxi provinces, from 2018 to 2021. PCR assays and transmission electron microscopy further revealed the presence of phytoplasma. Nested PCR using the universal primer R16mF2/R16mR1 followed by R16F2n/R16R2 was confirmed as the most effective molecular method for the detection of phytoplasma in sisal. A comparison of the detection rate of phytoplasma in symptomatic (83%) versus asymptomatic (17%) sisal populations showed some association between phytoplasma and SPLD. Sequencing analyses, phylogenetic analyses, and virtual restriction fragment length polymorphism analyses confirmed the presence of phytoplasma in sisal belonging to 16SrI-B. This is the first report of 16SrI-B phytoplasma infecting sisal in China. |

Effect of elicitors and biostimulants on the content of bioactive compounds in raspberry fruitsOriginal PaperNayelli Azucena Sigala Aguilar, Jose Antonio Gonzales Fuentes, Luis Alonso Valdez Aguilar, Mercedes Guadalupe López Pérez, Julia Medrano Macias Medrano Macias, Adalberto Benavides-Mendoza Benavides Mendoza, Susana González MoralesHort. Sci. (Prague), 2023, 50(2):101-111 | DOI: 10.17221/98/2021-HORTSCI Raspberry has acquired great interest in human health due to its content of bioactive compounds that provide protection against diseases caused by non-communicable diseases. Bioactive compounds are mainly represented by secondary metabolites such as phenols, anthocyanins, and flavonoids. Biostimulants and elicitors are substances or microorganisms that provide protection and defence to the physiological processes of plants. The present study evaluated the effect of two elicitors (hydrogen peroxide, salicylic acid) and three biostimulants (humic and fulvic acids, glutamic acid, seaweed extracts) on the content of bioactive compounds in raspberry fruits, agronomic and fruit yield parameters in plants. Hydrogen peroxide increased the content of bioactive compounds such as flavonoids, anthocyanins, omega 3 and oleic acid. Salicylic acid increased the content of flavonoids, anthocyanins, and citric acid in raspberry fruits; the number of fruit loaders and fruits per plant was also increased. Humic and fulvic acids, glutamic acid, and glutamic acid combined with seaweed extracts increased the content of flavonoids and anthocyanins, without affecting growth parameters and fruit yield. Glutamic acid and seaweed extracts were the only treatments that increased the content of palmitic acid, while seaweed extracts increased °Brix content in fruits. |
Phenotypic proximity and remoteness of seedless vine varieties depending on their ampelographic characteristicsOriginal PaperVenelin Roychev, Neli KeranovaHort. Sci. (Prague), 2023, 50(1):72-89 | DOI: 10.17221/59/2021-HORTSCI The degree of the phenotypic similarity and differences between 61 seedless vine varieties was studied by grouping them according to economically important phenological, agrobiological and technological indicators. A multidimensional statistical approach is applied – a cluster analysis, which provides possibilities of establishing relatively homogeneous groups (clusters), based on a set of certain indicators. As the cluster analysis does not perform any tests for the statistical significance of the results, it is combined with a factor analysis, which provides information on the reasons leading to the distribution of the individual varieties in different clusters and the merging of others into one cluster. The applied factor analysis transformed all the studied indicators into seven main components, explaining 76% of the total variance. The transformation of these traits to the corresponding number of factors and the indicators in them, which explain that the varieties belong to the separate clusters, was established. The limits of the phenotypic similarity and differences between the studied seedless vine varieties and the conditional parameters of the polymorphism with their economically significant ampelographic characteristics were determined. The presented grouping can be used for taxonomic purposes, as well as in the selection to create new seedless vine varieties. By applying targeted agro-technical measures, the desired change in the indicators of the first and second factors in the individual groups of traits can be actively influenced. |

Colonization of ‘Sampion’ apple tree roots and rhizosphere by mycorrhizal fungi following the application of AquaGel or organic compost enriched with beneficial microorganismsOriginal PaperEdyta Derkowska, Beata Sumorok, Lidia Sas-Paszt, Krzysztof Górnik, S³awomir G³uszek, Anna Lisek, Augustyn Mika, Mateusz Fr±cHort. Sci. (Prague), 2023, 50(4):253-261 | DOI: 10.17221/1/2023-HORTSCI Apple trees of the cultivar ‘Sampion’ were planted at the Experimental Orchard of National Institute of Horticultural Research in D±browice. In a three-year experiment (2019–2021) the following fertilization combinations were applied: AquaGel (commercial hydrogel), AquaGel combined with a consortium of beneficial bacteria, AquaGel combined with a consortium of beneficial filamentous fungi, organic compost alone, compost combined with a consortium of beneficial bacteria, and compost combined with a consortium of beneficial filamentous fungi. The presence of arbuscular mycorrhizal fungi was assessed both in the rhizosphere soil and in the roots of apple trees. The results of the study indicate the most advantageous effect of compost combined with bacteria or fungi on the degree of root colonization by arbuscular mycorrhizal fungi and the formation of their spores in the soil. |
Thin layer drying characteristics of alligator pepper, ginger and turmericOriginal PaperAdebayo Olasunkanmi Salau*, Babatunde Sunday Ogunsina, Adewale Waliu Adebayo, Adekunle Felix OkunadeRes. Agr. Eng., 2023, 69(1):1-8 | DOI: 10.17221/71/2021-RAE The delightful pungency, aromatic compounds and phytochemicals in some spices make them indispensable in local food systems and native medicine, hence, driving a robust market in many parts of the world. The understanding of their drying characteristics is very important for processing and adding value, and a thin layer drying study is a proven method for achieving this. In this study, changes in the moisture content, moisture ratio, drying rate and effective diffusivity of alligator peppers, ginger and turmeric were investigated at three drying temperatures 50, 60, and 70 °C following standard procedures. Five models were proposed to simulate the drying process. Non-linear regression was used to establish the coefficient of determination (R2), sum of square error (SSE) and root mean square error (RMSE) for each model to determine the model of the best fit. The Page model gave the best fit for ginger while the logarithmic model was best fitted for alligator peppers and turmeric. The effective diffusivity ranged from 1.79–3.08 × 10–9, 8.44–9.74 × 10–9, and 4.06–6.49 × 10–9 m2·s–1 for alligator peppers, ginger and turmeric, respectively. The activation energy ranged from 16.5–22 kJ·mol–1·K–1 for the three spices. These findings promise improvement in the drying, processing and handling of spices, thereby boosting the obtainable income from the value chain. |
