Phytoextraction of cadmium, copper, zinc and mercury by selected plants

During the years 2006 and 2007 the phytoextraction ability of maize (Zea mays), willow-tree (Salix smithiana) and poplar (Populus nigra × P. maximowiczii) to accumulate cadmium, copper, mercury and zinc was investigated. Small scale field experiment was carried out on soil contaminated with chemicals from the waste incineration plant in Hradec Kralové (Czech Republic). Screening of this allotment showed very different contamination of all observed risk elements on places where the material intended to be burnt without safety of leakage into soil. Grown plants showed the different accumulation of observed elements in plant tissues as well as the influence of total content of the risk elements in soil. The highest Cd (1.5-1.73 mg/kg) and Zn (242-268 mg/kg) concentrations were found in willow-tree biomass mainly in the leaves. Cu and Hg were mostly accumulated by maize roots (14.6-15.8 mg Cu/kg and 1.3-7.4 mg Hg/kg) and lower amount was found out in willow-tree leaves again. In reference to total production of each plant the maximum Cd and Zn uptake by aboveground biomass was found in poplars (201 mg Cd/m2 and 38 200 mg Zn/m2) and maize, which showed high Zn uptake. The biggest amount of copper (2563 mg Cu/m2) was accumulated by aboveground maize biomass on the collection point with the highest Cu concentration in soil and by poplar (2394 mg Cu/m2) on the other collection point. The highest Hg uptake differs in reference to total Hg content in soil; willow-tree has the highest uptake on the place with lower Hg content in soil (44.6 mg Hg/m2) and maize has the highest uptake on the place with higher Hg content in soil (92 mg Hg/m2).

Effects of putrescine and low temperature on the apoplastic antioxidant enzymes in the leaves of two wheat cultivars

The effects of putrescine (a polyamine), low temperature and their combinations on the activities of apoplastic antioxidant enzymes were studied in the leaves of two wheat cultivars, winter (Dogu-88) and spring (Gerek-79). Fifteen-day-old wheat seedlings were treated with putrescine solutions (0.1, 1 and 10mM) prior to cold treatment (5/3°C). The activities of apoplastic catalase, peroxidase and superoxide dismutase were determined in the leaves both under normal and cold conditions at 1, 3 and 5 days. The results indicate that cold treatment significantly increased the activities of apoplastic catalase, peroxidase and superoxide dismutase in winter wheat while not generally affecting spring wheat. Under control conditions, the putrescine treatments were more effective in increasing the enzyme activities in winter wheat than in spring wheat. However, under cold conditions, the putrescine treatments surprisingly induced enzyme activities in spring wheat while generally reducing those in winter wheat leaves. The results show that putrescine may act as an agent inducing primary changes in the apoplastic antioxidant system of wheat leaves during reactive oxygen species-mediated damage caused by low temperature stress.

Interannual dynamics of available potassium in a long-term fertilization experiment

Dynamics of the plant-available potassium (K) has been studied in polyfactorial long-term fertilization experiments since 1980. The fertilization scheme includes 10 combinations of K muriate and farmyard manure application rates (annually 0-230 kg K/ha). At medium treatment (annually 153 kg K/ha), the K balance within an 8-year crop rotation reflected crop specific K application rates with positive annual balances in years of growing silage maize and sugar beet (high K input), and negative in two years of growing alfalfa. Available K clearly corresponded to the dynamics of the K balance, with statistically significant fluctuations from 88 to 149 mg K/kg within one crop rotation cycle. Periodic fluctuations of available K induced by crop rotation were observed also in non-fertilized treatments. The variability of available K contents was influenced primarily by crop plants and experimental unexplained factors; interannual weather fluctuations and field differences were of low significance. In the paper, the importance of interannual K dynamics for the construction of correct long-term time trends is shown and discussed.

The effects of treatment with polyamines on dry matter and some metabolites in salinity - stressed chamomile and sweet majoram seedlings

The seeds of two medicinal plants, namely Chamomilla recutita (Babong) and Origanum majorana (Bardaqoush) were subjected to germination in different NaCl concentrations, polyamines (putrescine, spermidine, spermine) and to combination of both. The results revealed that the growth alterations induced by NaCl were alleviated by various levels of polyamines. The organic solutes of both plant seedlings exhibited somewhat variable responses to various salinity levels or polyamines treatments and in combination of both treatments. Putrescine in Ch. recutita seedlings was more effective in alleviating the stress effects of salinization than spermidine and spermine, while in O. majorana seedlings spermidine was more effective. Generally, the degree of stimulation differed according to the type, concentration of the additive used and the type of the plant tested.

Impacts of root sulfate deprivation on growth and elements concentration of globe amaranth (Gomphrena globosa L.) under hydroponic condition

Sulfur (S) regarded as the fourth key element is mainly taken by the plant roots. However, some plants can also absorb atmospheric sulfides, which may be of great importance for ameliorating the environment and for farming as a green organic S fertilizer used to balance insufficient soil S content for intensive cultivation in China; H2S and mainly SO2 are emitted to air as a result of the rapid industrialized and economic development. Globe amaranth (Gomphrena globosa L.) might be one of the plants that can use atmospheric sulfides for its growth. Therefore the effects of sulfate deprivation from root on its growth, S status and other elements concentration under hydroponic culture were explored firstly. Based on measurements of plant growth, biomass, nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), S, iron (Fe), manganese (Mn), copper (Cu), zinc (Zn), boron (B), and molybdenum (Mo) concentration, the results showed that S concentration in flower, shoot and root of plant without root sulfate supplied was increased with plant growth and development, symptoms of S deficiency disappeared and other elements concentration in plant tended to be nearly the same as the root sulfate-supplied plants. The interesting results might imply that globe amaranth may be able to live on the atmospheric sulfides as sulfur source.

Morpho-anatomical characterization of root in recurrent selection cycles for flood tolerance of maize (Zea mays L.)

Changes in root anatomical structures at successive cycles of selection (cycle 1 to cycle 18, alternating) were observed in the study of maize (Zea mays L. cv. Saracura-BRS 4154) capable to survive and produce in temporarily flooded soils; this cultivars was developed by the Maize and Sorghum National Research Center through stratified phenotypic recurrent selection for cultivation wetland soils. Field trial was carried out and flooding of the soil was initiated at the six-leaf stage; the soil was flooded with water (20-cm deep) three times per week. Root sample was collected, fixed, and selected for observation in photon microscope. A gradual increase in the number of aerenchyma, the proportion of vascular cylinder, smaller metaxylem, and phloem and epidermis width, and a decrease in exodermis and cortex were observed in successive selection cycles. Such phenotypic changes impart the flood tolerance ability to this maize cultivar.

The effect of zeolite, organic and inorganic fertilizers on soil chemical properties, growth and biomass yield of apple trees

The objective of this study was to evaluate the influence of organic (cattle manure) and inorganic fertilizers [composite NPK (15:15:15)] and natural zeolites on soil properties, vegetative growth and yield of apple plants grown on vertisol under Cacak conditions (Western Serbia). The results showed that the combined fertilization induced a decrease in acidity, an increase in humus content, a partial increase in total nitrogen (NTOT), and, primarily, a rise in available phosphorus (P) and potassium (K) levels. A highly significant (P < 0.01) interactive effect of the apple cultivars and nutrients on one-year-old shoot length and trunk cross-sectional area (TCSA) and a significant effect (P < 0.05) on yield per tree and yield per hectare were determined.

Microbial biomass dynamics after addition of EDTA into heavy metal contaminated soils

An incubation experiment with addition of EDTA and alfalfa into soils contaminated with heavy metal over 200 years was carried out in order to evaluate the EDTA effects on microbial properties. Alfalfa was added to soils together with EDTA to examine its abilities to improve microbial activities affected by EDTA. The obtained results showed that the addition of EDTA led to a significant decrease of microbial biomass C during the first 24 days of incubation. At the end of the experiment the microbial biomass C significantly increased quite close to the original level. The EDTA amendment caused, probably due to the toxic effects, a significant increase in respiratory activities and of the metabolic quotient qCO2. An addition of alfalfa significantly improved the microbial biomass C contents in arable soils treated together with EDTA. Both, respiratory activities and qCO2 significantly increased after the soil treatment with EDTA together with alfalfa. EDTA alone decreased the microbial biomass, alfalfa alone as organic substrate was mineralised and utilised by soil microorganisms for their metabolism.

Leaf growth under temperature and light control

Dynamics of crop growth and photosynthesis are two main processes that are of major importance for adaptation of plants to their environment. Two experiments were carried out during 2005 and 2006 with sun and shaded barley plants. The results showed that leaf area increased with leaf position on the main stem up to leaf position 5 for sun and leaf position 6 for shaded plants, and then declined towards flag leaves. Air temperature affected leaf appearance, mainly at the beginning of the growing season. A positive correlation between leaf expansion duration (LED) and accumulated temperature sum was measured for both variants, LED linearly increased with temperature sum. Leaf expansion rate (LER) showed a similar dependence on accumulated air temperatures in both light variants. A strong relation was found between soil temperature and LER for the first four leaves of the main stem of shaded plants in 2006. In shaded environment the higher LER was associated with lower accumulated irradiance sum when light restriction supported an increase of leaf elongation and final leaf length.

Effect of abrasive particle size on abrasive wear of hardfacing alloys

Hardfacing is one of the most useful and economical ways to improve the performance of components submitted to severe wear conditions. This study has been made for the comparison of microstructure and abrasion resistance of hardfacing alloys reinforced with chromium carbides or complex carbides. The hardfacing alloys were deposited onto ČNS EN S235JR low carbon steel plates by the gas metal arc welding (GMAW) method. Different commercial hardfacing electrodes were applied to investigate the effect of abrasive particle size on abrasive wear resistance. The abrasion tests were made using the two-body abrasion test according to ČSN 01 5084 standard, abrasive cloths were of grits 80, 120, 240, and 400. Microstructure characterisation and surface analysis were made using optical and scanning electron microscopy. The results show the different influence of abrasive particles size on the wear rate for different structures of Fe-Cr-C system. The structures without primary carbides are of high abrasive wear rate, which increases nonlinearly with the increasing abrasive particle size. On the contrary, the structures containing primary carbides are of low abrasive rates and theses rates increase linearly with the increasing abrasive particle size.

Utilization of microbial inoculation and compost for revitalization of soilsShort Communication

Improving the quality of reclaimed soils requires an active population of microorganisms which can promote plant growth. Increasing the activity of microorganisms can be done by adding nutrients, making agrotechnical soil improvements and by the inoculation of beneficial microorganisms. We investigated the role of fertilizer treatments on plant growth and nitrogen fixation in a pot experiment conducted under green house conditions. Influence of the fertilizer type on numbers of bacteria was also investigated. The seeds were inoculated with the mixture of Azotobacter spp. and Rhizobium spp. The pot experiment was set up with the substrate from the mine spoil (North Bohemia coal basin, the Czech Republic) using Medicago sativa as test plants. The following treatments were used: compost 0, 20, 40, 120, 400, 800 t/ha and mineral fertilizer - ammonium sulphate. The doses of ammonium sulphate were calculated to be equivalent (in nitrogen content) to those doses of compost. Control variants without bacteria inoculation and fertilizers were also included. Inoculation with the mixture of AzotobacterRhizobium spp. significantly increased plant growth and nitrogenase activity. The nitrogenase activity was inhibited by mineral fertilizers in all doses used. The results of the study have proved that compost application stimulated the growth of Azotobacter spp. and Rhizobium spp.

Soluble and insoluble pollutants in fog and rime water samples

Fog and rime water samples were collected at the meteorological observatory Milešovka in February and June 2006. In the samples, the soluble and insoluble pollutant concentrations were evaluated separately and the differences between the fog and rime water samples were studied. The comparison of the fog and rime water samples indicates that the mean soluble component concentrations in the air appear to be higher during the rime events than during the fog events at Milešovka. We recorded a larger mean particle size of the insoluble compounds in the fog water samples than in those of rime water. Some elements contained in the insoluble particles like Ca, Cl, C, Cu, Ag, were present largely in fog whole others, like Fe, Al, Si, Ti, prevailed in rime. In addition to the overall evaluation, the backward air trajectories were determined for each fog/rime event and the concentrations are presented as depending on the direction of the air particle transfer.

Mycobacteria isolated from the environment of pig farms in the Czech Republic during the years 1996 to 2002

Sources of mycobacterial infections in 50 pig herds in the Czech Republicwere investigated during the years 1996 to 2002. A total of 2 412 samples from the external environment (feeds, bedding materials, drinking water, biofilms on drinkers, scrapings from the walls, floors and pen barriers, dust, spider webs, peat, kaolin, faeces, organs of rodents, and birds, etc.) were examined. After staining by the Ziehl-Neelsen technique, acid-fast rods were detected in 95 (3.9%) samples by direct microscopic examination and mycobacteria were cultured from 575 (23.8%) samples. From Mycobacterium avium complex (MAC), M. avium subsp. hominissuis (genotype IS901-, IS1245+) of serotypes 4, 6, 8, and 9 (272; 47.0% isolates), M. a. avium (genotype IS901+, IS1245+) of serotype 2 (13; 2.2% isolates) and M. intracellulare (genotype IS901-, IS1245-) of different serotypes (2; 0.3% isolates) were detected most frequently. Other isolates from among 14 other mycobacterial species ranked as follows: 64 M. gordonae, 47 M. fortuitum, 17 M. chelonae, 14 M. flavescens, 11 M. terrae, seven M. phlei, seven M. scrofulaceum, three M. diernhoferi, three M triviale, three M. smegmatis, two M. xenopi, one M. szulgai, one M. gastri, and one M. ulcerans. The remaining 111 isolates of unidentified species did not contain specific sequences IS901 and IS1245 characteristic for the pathogenic members of MAC (M. a. avium and M. a. hominis­suis). Peat, drinking water, biofilms on drinkers, bedding materials, feeds, free living birds, kaolin and charcoal were identified as potential sources of mycobacterial infections for pigs. Peat given to piglets as a feed supplement was the most important source of mycobacteria (65.1% positive of 327 examined samples); 81.2% of them were positive for M. a. hominissuis of serotypes 4, 6, 8, and 9. By contrast, mycobacteria of other species (M. gordonae, M. fortuitum, M. chelonae, M. flavescens, etc.) were the main isolates obtained from drinking water and biofilms on drinkers for pigs. By culture examination, the detection rate was higher in the biofilm samples (36.4%) than in the samples of drinking water (29.6%). The third group of sites with detected high levels (26.4%) of mycobacterial contamination were various types of beddings of woody material. M. a. hominissuis of serotypes 6, 8, and 9 were the most frequent isolates from sawdust; M. a. avium serotype 2 was sporadically detected. Mycobacterial findings in other samples from the external environment (wall and floor scrapings, dust, soil from the runs, and invertebrates) gave an indication of the pressure of infection in the herds. High contamination levels in faecal samples (15.6%) and in scrapings (18.4%) from respective parts of pens and stables indicated exposure of pigs to mycobacteria. Diagnostic testing of different stages of avian tuberculosis in naturally infected hens (Gallus domesticus) by the tuberculin skin and rapid agglutination tests, faecal and egg examinations

Avian tuberculosis (ATBC) is a significant cause of morbidity and mortality in birds in zoos and breeding establishments. The primary sources of Mycobacterium avium subsp. avium (MAA) of serotypes 1, 2 and 3 are infected animals (esp. birds), who shed MAA in different ways and thus contaminate the environment. The first aim of this work was to compare the efficiency of the diagnostic methods that are routinely used for the diagnosis of ATBC (skin test, serology and culture of faeces and eggs) in naturally infected hens with different levels of infection. The second aim was to determine the excretion rate of MAA in faeces and eggs. The tuberculin skin test gave a positive result in nine (42.9%) infected hens of which four (57.1%) and one (14.3%) were heavily and slightly infected hens, respectively. A positive serological response to MAA-b antigen (water bird isolate of serotype 1) was observed in five (23.8%) and to MAA-p antigen (pig isolate serotype 2) in seven (33.3%) hens. No correlation between serological and skin-test data was found. The results show that both techniques, serological and skin-test data are inadequate for the diagnosis of ATBC. In consecutively euthanized hens, with heavy infection and tuberculous lesions, serological positivity was significant (P < 0.05) in comparison with slightly infected hens lacking tuberculous lesions. Faecal culture detected MAA in 50 (29.8%) of 168 samples collected for eight days before euthanasia. MAA excretion in faeces was intermittent, but significantly (P < 0.01) higher in heavily infected hens. No mycobacteria were detected in any of the 43 examined eggs, which implies that the shedding of MAAand/or transmission of ATBC through eggs may not be frequent events.

Detection methods for Mycobacterium avium subsp paratuberculosis in milk and milk products: a review

Mycobacterium avium subsp. paratuberculosis (MAP) is the etiologic agent of paratuberculosis, a disease with considerable economic impact, principally on dairy cattle herds. Animals with paratuberculosis shed viable MAP especially in their milk, faeces and semen. MAP may have a role in the development of Crohn's disease in humans via the consumption of contaminated milk and milk products. The current methods of milk pasteurization are not sufficient to kill all MAP cells present in milk and MAP has been cultured from raw or pasteurized milk and isolated from cheese. The purpose of the present study was to review the different methods used for detection of MAP in milk and milk products. We analyze the current methods for direct or non direct identification of MAP and culture and molecular biology methods that can be applied to milk and milk products.

Spirocercosis and its complications in stray dogs in Shiraz, southern Iran

Spirocerca lupi is accepted in Veterinary Medicine as the cause of true malignancies in dogs. Aortic lesions and esophageal nodular granulomas are pathological lesions of spirocercosis that are considered to be pathognomic for this disease. Post mortem and histopathological studies of 105 stray dogs both male and female of different age groups from various residential areas were performed and their esophagus were examined for any lesions and helminthic parasites. Twenty dogs (19.04%) were infected with S. lupi. The infection rate was significantly higher in dogs greater than one year old (23.5%) and in western areas (36.4%) with similar prevalence rate in males and females. At histopathologic level, two isolate but closely related fibrosarcoma masses were situated at the distal portion of the thoracic esophagus of one dog and in the site of junction of esophagus and stomach of another one. Metastasis were observed in the lung and mediastinal lymph nodes of one of the dogs. Despite presence of large population of stray dogs little information is available about S. lupi in this country, so this study was undertaken to obtain an overall picture of this infection in stray dogs of Shiraz area, southern Iran.

Detection of nongroup A rotaviruses in faecal samples of pigs in the Czech Republic

Besides group A rotaviruses, group B and C rotaviruses have been detected as the cause of diarrheal diseases in pigs. Of a set of 329 faecal samples from pigs, 16 samples were selected in which rotavirus was detected by electron microscopy and at the same time group A rotavirus was excluded by ELISA method. Rotaviruses were assayed using specific primers for detection of group B and C rotaviruses, and RT-PCR and semi-nested PCR methods. In one sample, no rotavirus of group B or C was detected; in the remaining 15 samples rotavirus group C was detected, in two samples together with group B rotavirus. Sequencing of the obtained PCR products and comparison with corresponding gene sequences revealed 80% nucleotide sequence identity between group B rotaviruses and available sequences of porcine isolates. A nucleotide sequence identity of 92% was obtained in group C rotaviruses as compared with the Cowden strain.

Evaluation of the activity of intraarticular hyaluronic acid in the repair of experimentally induced osteochondral defects of the stifle joint in dogs

The present study examined the results of using hyaluronic acid with autogenetic cancellous grafts in the treatment of experimentally induced osteochondral defects in the stifle joints of dogs. In this study, 10 mature dogs of different breeds, weights and of both sexes were used. General anesthesia and usual operation procedures were followed. A 10 mm deep defect was created on the femoral sulcus of the trochlea with a drill tip of 8 mm in diameter. The defects in the right and left legs were filled with autogenic cancellous grafts taken from the metaphysial region of the tibia. The left legs constituted the experimental group while the right legs served as control group. In the experimental group, 2 mg/kg intraarticular hyaluronic acid was twice administered into the stifle joint, i.e., immediately subsequent to the operation and 1 month afterwards. Parenteral antibiotics were prescribed postoperatively for ten days. Five animals were sacrificed at the third and sixth month after surgery. Macroscopic and microscopic findings obtained from each case were evaluated. On macroscopical examination, trochlear defects were determined to be incompletely filled at the third month in both control and experimental groups. On histopathologic examination, a loose fibrovascular formation in the area where the graft was applied was observed to be present in both control and experimental groups. However, in the experimental group this formation was more superficial, ossification activity was greater and trabeculous bone formation had been initiated. Macroscopical examination carried out in the sixth month determined that in the control group the defect surface did not fill up to the trochlear sulcus level. In the histopathologic examination, in control groups it was found that fibrocartilageous structures were developing in the fibrovascular space even though ossification was incomplete. The macroscopic examination showed that in the experimental group, the defect surface reached the trochlear sulcus level of defects in this month. The histopatologic examination revealed that fibrous tissue comprised a thin layer, under which ossification processes were complete and bone trabeculates fully formed. It was concluded that the usage of autogenic cancellous graft along with hyaluronic acid may be useful in the repair of large osteochondral defects.

Detection of toxigenic Clostridium difficile in pig feces by PCR

: Clostridium difficile is considered an important uncontrolled cause of neonatal diarrhea. Also, the presence of bacteria in the feces of the animal could represent a zoonosic risk for the contamination of meat products. Therefore, it is necessary to have procedures available for the early detection of C. difficile in animals. The current study describes a new semi-automated procedure for the recovery of C. difficile DNA from pig feces and subsequent amplification by polymerase chain reaction (PCR) of three different sequences: the triose phosphate isomerase gene tpi, specific for this bacterial species, and the tcdA and tcdB genes, which code for the A and B toxins of C. difficile, respectively. Twenty-two fecal samples microbiologically positive for C. difficile were used. The tpi and tcdA genes were amplified in all of them. The internal fragment of tcdB was detected from 21 of these extracts; the negative sample gave a positive result when a different primer pair was used. None of the 10 DNA extracts obtained from culture-negative samples gave a positive result. The method presented in this article eliminates the interference caused by the possible presence of PCR inhibitors. To the authors' knowledge, this is the first description of a PCR procedure for detection of C. difficile DNA from domestic animal feces.

Serum deprivation affects secretory activity of cultured porcine ovarian follicles and granulosa cells and their response to hormones

The aim of the present study is to understand the hormonal mechanisms of the effect of malnutrition on ovarian follicle functions. For this purpose, we examined the effect of malnutrition/serum deprivation, addition of metabolic hormones and gonadotropin (IGF-I, leptin and FSH) and their combination on the release of progesterone (P4), testosterone (T), estradiol (E2) and insulin-like growth factor I (IGF-I) by cultured whole ovarian follicles and on P4 and IGF-I output by cultured granulosa cells isolated from porcine ovaries. It was observed that in ovarian follicles cultured with nutrients/serum addition of IGF-I reduced release of P4, but not of T or E2. Exogenous leptin reduced output of E2, but not of P4 or T, and increased IGF-I output. No significant effect of FSH on release of steroid hormones by isolated follicles was found. Serum deprivation did not affect release of P4, but reduced output of T and E2, and promoted IGF-I release by cultured ovarian follicles. Addition of hormones failed to prevent the effect of malnutrition on the secretory activity of cultured ovarian follicles. In cultured granulose cells, all the tested hormones promoted release of both P4 and IGF-I. Food restriction/serum deprivation reduced both P4 and IGF-I output. Additions of either IGF-I, leptin and FSH prevented the inhibitory action of malnutrition on both P4 and IGF-I release. The present observations (1) confirm the involvement of the hormones IGF-I, leptin and FSH in the control of secretory activity of ovarian cells, (2) demonstrate, that both isolated ovarian granulosa cells and whole follicles cultured in the absence of serum nutrients could be an adequate in-vitro model for studying the effect of malnutrition on ovarian secretory functions, and (3) suggest, that malnutrition could affect ovarian functions through changes in the release of ovarian hormones.

Analysis of economic risks of hop growing

The hitherto development of agricultural production shows that one of the characteristic attributes of the present period is great economic instability. Monitoring of the development of prices of inputs and outputs as well as climatic conditions reveals that similar problems are not limited to the Central European countries. Prosperity and competitiveness of the production is a function of mutual relations of costs, prices and yields. For the sound managerial decision-making, it is necessary to continually analyse and evaluate the rate of risk - soundness of the planned results (Rataj, Kavka 1999; Rataj 2001). For that reason, this contribution concerns the analysis of economic risks of the hop growing that takes into account statistical data in the time horizon of the last 15 years for the "Žatec poloranný červeňák" of traditional planting (further only ŽPČT) and 7 years for ŽPČ virus free (farther only ŽPČV) and the hybrid sorts (farther only HYBR).

Globalization in trade, the continual expansion of the significant European and world-wide foreign trade networks into the countries of Middle andEastern Europe and the rise in the number of shopping and entertainment centres cause changes in consumer behavior in the Slovak, as well as in theCzechRepublic. Results of the marketing researches conducted by the Slovak Agricultural University employees in Nitra and the Mendel University of Agriculture and Forestry employees in Brno in the Polus and Aupark shopping and entertainment centres of Bratislava and the Olympia centre of Brno Modřice in 2006 gave us some more detailed information about the social-demographic features and consumer orientation in Slovakia and the Czech Republic. Shopping in shopping - entertainment centres in both countries is becoming a part of the present lifestyle, spending free time and having fun.

Effect of dietary esterified glucomannan on performance, serum biochemistry and haematology in broilers exposed to aflatoxin

The amelioration of aflatoxicosis in broiler chickens was examined by feeding two concentrations of yeast component (esterified glucomannan; EG). EG, incorporated into the diet at 0.5 and 1 g/kg, was evaluated for its ability to reduce the detrimental effects of 2 mg total aflatoxin (AF; 82.72% AFB1, 5.50% AFB2, 10.20% AFG1 and 1.58% AFG2) in diet on growing broiler chicks from 1 to 21 d of age. A total of 240 male broiler chicks (Ross-308) were divided into 6 treatment groups [control, AF, EG (0.5 g/kg), AF plus EG (0.5 g/kg), EG (1 g/kg), and AF plus EG (1 g/kg)]. Compared to the control, AF treatment significantly decreased body weight gain from week 2 onwards. AF treatment also caused significant decreases in serum total protein, albumin, total cholesterol, triglyceride, glucose, inorganic phosphorus, creatinine levels and alanine-aminotransferase (ALAT) activity but increased the aspartate-aminotransferase (ASAT) activity. Red blood cell, haematocrit, haemoglobin, thrombocyte, and lymphocyte counts and tibial crude ash levels were significantly reduced by AF treatment, while significant increases were seen in heterophil counts. The addition of EG (1 g/kg) to an AF-containing diet significantly improved the adverse effects of AF on haematological parameters, total protein, albumin values and ASAT activity. EG (1 g/kg) also partially improved body weight gains (59%) and the other biochemical parameters influenced by AF treatment. The addition of EG (both 0.5 and 1 g/kg) to the AF-free diet did not cause any considerable changes in the investigated values. These results clearly indicated that EG (1 g/kg) addition effectively diminished the adverse effects of AF on the investigated values. Also, the higher dietary concentration of EG (1 g/kg) was found more effective than the lower concentration (0.5 g/kg) against the adverse effects of AF on the variables investigated in this study.

Total phenolics and phenolic acids content in low (Chrysopogon gryllus) and mediocre quality (Festuca vallesiaca) forage grasses of Deliblato Sands meadow-pasture communities in Serbia

Chrysopogon gryllus and Festuca vallesiaca are components of a number of meadow-pasture communities in Serbia. We performed the analyses of phenolics that influence quality and digestibility of grasses to a great extent. Total phenolics were measured spectrophotometrically and phenolic acids by HPLC analysis. The aboveground parts of C. gryllus contained 10.6 mg/g whereas F. vallesiaca of 21.6 mg/g total phenolics. Bound phenolics dominated over free ones in both species. The content of both free and bound p-coumaric, ferulic, p-hydroxybenzoic and syringic acid was higher in C. gryllus (6.34 mg/g) than in F. vallesiaca (3.96 mg/g). Derivatives of cinnamic acid prevailed in both species compared to the benzoic acid derivatives. Low quality of C. gryllus is connected with its high tissue phenolic acids and mediocre quality of F. vallesiaca with the high content of total phenolics that act unfavourably on digestibility of such grasses.

Influence of lucerne and meadow hay quality on the digestibility of nutrients in the roe deer

It was confirmed in metabolic experiments with 4 individuals of roe deer (2 males and 2 females with the live weight of 20.8 and 17.9 kg, respectively) that due to the animals' selective choice of a part of feeds there was no significant difference in the intake of lucerne hay of different quality and high-quality meadow hay (515.86 ± 66.97 and 597.82 ± 94.87 g, respectively). However, different quality of lucerne hay influenced the animals' intake of crude protein and crude fibre. When the animals were fed meadow hay, they took in significantly less crude protein and fibre and significantly more nitrogen-free extract, fat and ash. Digestibility of crude protein was observed to be significantly decreased in hay of lower quality (by 10.1%). Digestibility coefficients for crude protein, crude fibre, organic matter and fat were significantly lower in grass hay compared to lucerne hay. Based on the coefficients of nutrient digestibility it can be stated that meadow hay, even if of excellent quality, is not a suitable supplement for the roe deer.

Influence of lecithin emulsifier on the utilisation of nutrients and growth of piglets after weaning

The influence of supplementation of a lecithin emulsifier (0.1%) to the feed mixture for piglets after weaning was studied. 16 piglets in the control group (C - without lecithin) and 16 piglets in an experimental (E) group were monitored in three replications in a 28-day experimental period. We observed higher digestibility of monitored nutrients in the experimental group (crude fat: C = 66.28 ± 12.11, E = 69.75 ± 9.85%, crude protein: C = 78.72 ± 5.47, E = 82.17 ± 2.23%, crude fibre: C = 56.70 ± 12.85, E = 59.10 ± 12.04%, nitrogen free extract: C = 86.67 ± 3.32, E = 87.75 ± 3.00%, ash: C = 63.63 ± 11.59, E = 65.73 ± 10.35%). Statistically insignificantly higher daily average gain (C = 374.1 ± 107.0, E = 399.1 ± 104.7 g) of experimental groups and a lower feed conversion ratio (C = 2.285 ± 0.416, E = 1.768 ± 0.048 kg/kg) were found out. It was also confirmed in an additional field experiment (C = 279, E = 312 piglets from birth to 71 days of age) where average daily gain weight was 270 g in control and 290 g in experimental group. The influence of lecithin on the occurrence of haemolytic strains of Escherichia coli was not proved. This experiment confirmed a positive effect of the lecithin emulsifier supplement on the efficiency of piglets.

Nutritional value of amaranth (genus Amaranthus L.) grain in diets for broiler chickens

The following characteristics were determined in raw and popped amaranth grain: crude protein (158.1 and 168.5 g/kg), ether extract (71.5 and 69.4 g/kg), neutral detergent fibre (NDF) (99.2 and 111.8 g/kg), cellulose (86.6 and 60.0 g/kg) and essential amino acids (Cys 4.2 and 4.1, Thr 6.0 and 6.5, Ala 8.8 and 9.2, Val 6.8 and 7.4, Ile 5.2 and 5.6, Lys 9.2 and 8.8, Arg 12.8 and 14.2 g/kg). In vitro protein digestibility was 68.1 and 50.6% in raw and popped amaranth grain, respectively. In balance experiments with broiler chickens ROSS 308 the following coefficients of apparent digestibility (%) were determined for control and experimental diets containing 0, 10% raw and 10% popped amaranth: crude protein 85.4, 86.5 and 83.0, ether extract 88.3, 88.2 and 86.1, NDF 21.2, 27.6 and 15.9, cellulose 25.0, 38.4 and 36.3, nitrogen free extractives 76.1, 82.6 and 81.1, organic matter 77.3, 81.8 and 80.6, gross energy 77.5, 80.6 and 78.2.

The effect of sex and rearing system on carcass composition and cut yields of broiler chickens

The objective of this study is an analysis of meat quality in broilers of standard fast-growing hybrid Hybro G, reared in two different non-industrial systems (extensive rearing in a poultry house or "extensive indoor" system and rearing in a poultry house using a free range or "free-range" system). Quality parameters, i.e. proportions of basic carcass parts (breasts, drumsticks and thighs), abdominal fat, and proportions of basic tissues (muscles, bones and skin) in more valuable carcass parts, were investigated. The aim of the investigation was to analyse the effect of the two non-industrial rearing systems and broiler sex on the above-mentioned traits of broiler meat quality. The used rearing systems (free-range and extensive indoor ones) did not have a statistically significant effect on the proportions of major basic carcass parts and of abdominal fat in broiler chickens (P > 0.05). Heavier carcasses at slaughter (on the 56th day of fattening) were recorded in the male broilers compared to the female ones, the differences being statistically significant (P < 0.01). The drumstick proportion in the male broiler carcasses was statistically significantly higher (P < 0.05) than the proportion in the female broilers. The interaction between the investigated effects (of sex and rearing system) did not exert a statistically significant effect on the proportions of major basic carcass parts and abdominal fat in broiler chickens (P > 0.05). The rearing system (free-range and extensive indoor) and the interaction between the investigated effects (of sex and rearing system) did not affect statistically significantly the differences expressed in the meat, skin and bone proportions in breasts, drumsticks and thighs of the broilers (P > 0.05). The sex effect on the skin proportions in breasts, drumsticks and thighs was not statistically significant (P > 0.05). Average muscle proportions in drumsticks and thighs of the female broilers were statistically highly significantly (P < 0.01) and significantly (P < 0.05) higher compared to those in the male broilers. Average bone proportions in the breasts of the male broilers were statistically significantly higher (P < 0.05) than those in the female ones, and statistically highly significantly higher (P < 0.01) in drumsticks and thighs.

Breeding value evaluation in Polish fur animals: Estimates of (co)variances due to direct and litter effects for fur coat and reproduction traits

5 540 records of the arctic fox fur coat and reproductive traits collected in 1983-1999 were studied. The analyzed traits were: body size (BS), colour type (CT), colour purity (CP), coat density (CD), hair length (HL), general appearance (GA), total score (TS), skin length (SL), litter size at birth (LSB), litter size at weaning (LSW), number of dead pups (NPD), pup weight at weaning (PW), and pregnancy length (PL). (Co)variance components were estimated using a derivative-free algorithm of REML and a multi-trait animal model. Random effects were direct additive, common litter environment and residual. The genetic parameters for the fur coat traits (discrete characters) were estimated twice: using the original data set, and the data set in which the distribution of fur coat scores was normalised using a probit link function. Direct heritability estimates obtained from the original data set ranged from 0.108 for SL to 0.276 for HL, and were somewhat lower than those estimated using the transformed data set (they ranged from 0.109 for GA to 0.315 for CT). Reproductive traits were lowly heritable with direct heritabilities ranging from 0.060 for PW to 0.174 for LSB. Estimates of the portion of litter variation calculated from the original and transformed data set were comparable ranging from 0.045 for GA to 0.156 for CP, and from 0.059 for GA to 0.185 for TS, respectively. Genetic correlations between fur coat traits ranged from high favourable (0.948 between SL and BS, original data) to strong negative ones between CP and GA (-0.405, transformed data). High positive genetic correlations were found between LSB and LSW (0.954), and between LSB and NPD (0.783), whereas PL was negatively correlated with all other reproductive traits.
