The effect of rainfall and extensive use of grasslands on water regime

Water regimes of extensively used grasslands (one cut per year, two cuts per year, no cut, mulching) were determined and compared by drainage lysimeters in 1998-2000. Although the botanical composition and yields of experimental swards were different, there was no statistically significant difference in their water regime (only the soil moisture content of no-cut variant was significantly higher than in other variants). A determinant factor for the water regime of grasslands (GR) is the sum of rainfall over the growing season while the GR water regime is influenced by land use immediately after the cut. Water runoff from the soil profile 0.0-0.60 m (water supply to the groundwater level) was found to be negligible in the growing season, a substantial groundwater recharge occurs in an off-season period and/or at the beginning of growing season. Mulching was not proved to reduce evaporation. The best type of management providing for the economical water regime appears to be a one-cut variant. Relationships between botanical composition and GR water regime are also described.

The influence of special natural amendments based on zeolite tuff and different lime materials on some soil chemical properties

This paper deals with the changes in soil active acidity, mobile aluminium, base saturation, iron and manganese under the influence of quicklime (QL), mixture of soft lithothamnian limestone (SLL) and dolomite (D), and special natural amendments (SNA) based on zeolite tuff. Investigations were carried out on pseudogley of mesoelevations, dystric. The four-year trial was set up according to the Latin rectangle method with 18 trial treatments in four replications. While SNA based on zeolite tuff had little effect on changes of the studied parameters, traditional lime materials (LM), owing also to the fact that they were applied at several times higher rates, had a very positive effect. Soil acidity, iron and manganese were reduced under their influence, mobile aluminium, particularly under their higher rate, was fully blocked or reduced within tolerable limits, and base saturation was raised to a satisfactory level. Effects of SNA depended on the ratio of zeolite tuff and the lime component in them. It could be presupposed that their main efficiency happened in the domain of ion exchange with a positive impact on soil fertility.

The use of spelt wheat (Triticum spelta L.) for baking applications

Five cultivars of spelt wheat (Rouquin, Bauländer Spelz, Schwabenkorn, Franckenkorn and Holstenkorn) have been evaluated for baking quality by means of direct and indirect indicators. Three-year values of the indirect indicators have been statistically processed by the analysis of variance. Based on the results obtained we can say that all evaluated indicators except the extensibility of gluten were significantly influenced mainly by the year of growing. The cultivar itself was a less important source of variance, although some values of important indicators differed significantly according to cultivars. Based on indirect indicators such as the content of wet gluten, its extensibility and swelling, the content of N-substances, the sedimentation test in the modification with SDS, the falling number and the content of starch, the cultivars Schwabenkorn and Rouquin are considered as the most suitable and of the highest quality. In general all the cultivars of Triticum spelta L. showed high contents of N-substances (x = 15.46%) and wet gluten contents (x = 37.12%). Their disadvantages are lower swelling values (x = 9.3 ml) and lower sedimentation values (x = 37.4 ml) which have a negative influence on the bread volume and the specific volume (under 310 ml.100 g-1). The predicted good baking quality of Schwabenkorn has been comfirmed in a baking experiment (direct method of evaluating the baking quality). There were good baking quality results for Bauländer Spelz as well. Unexpectedly bad results have been found with Rouquin, which showed the lowest water absorbing capacity of flour, the lowest bread volume, specific volume and baking extraction. The bread was just acceptable as far as taste is concerned. The baking from Holstenkorn was evaluated as excellent.

Causes of mountain meadow soil chemical degradation in long-term fertiliser experiment

The fertilising experiment was set up in 1968 on the mountain meadow (720 m a.s.l.) in Czarny Potok near Krynica (20°8' E, 49°4' N). The experiment was conducted on the acid Cambi soil and comprised objects fertilised with two nitrogen forms and two doses against the background of PK fertilisation, the untreated object, and plots with unilateral P and N fertilisation. The paper concerns 30 years of investigations (1968-1997) of the effect of different NPK fertilisation on the dynamic of yields and the meadow sward quality against a background of the same treatments. The dynamic of the botanical composition was presented as well as the dynamic of the grassland yield potential with systematic mineral fertilisation and liming. The application of nitrogen fertilisation with the rate of 90 N.ha-1 + PK under mountain conditions and systematic liming of the meadow enables to maintain or increase production over the long period, to decrease the production risk and to prevent degradation of the environment and natural resources.

Critical values of trace elements in soils from the viewpoint of the transfer pathway soil - plant

The development of soil limiting values of the protection of the quantity and mainly quality of the crop production tends from empiric values towards critical values, based on ecotoxicity. We present an attempt to derive transfer functions by the means of multiple regression analysis. The substitution of trace elements contents in crops in the prediction equations for fodder or food standards or phytotoxicity limits satisfies the present ecotoxicological demands. We preferred polyfactor relations to simple ones. The exceeding of reference values must be verified by the determination of the hazardous transfer in the field. Therefore the reference values are called testing values. They were derived especially for Cd, Pb, As, Cu, Zn, Ni and Mn. For some trace elements, only protective values can be set up (especially for Cr, Hg, but also for Tl, Be, V). They reflect minimum values that guarantee growing crops without any risks. Experimental data (pot trials) were compared with results obtained in field investigations. The resulting critical values were also compared with the values proposed in Germany.

Influence of different cultivation factors on the yield structure and on changes of soil properties

The monitoring was carried on in the years 1996-2000 in the polyfactorial P-A field trial in Lukavec, Pelhřimov district. The given locality is characterized by low fertility sandy-loamy cambisoils, by long-term average annual rainfall of 653 mm, by average annual temperature of 7°C and its altitude is 620 m. The field trial included different organic fertilizations; graduate N-doses, different soil tillage as well as different forecrops. It manifested in this group of field trials that the forecrop value of red clover and dung manured potatoes before winter wheat was practically the same in the given locality. The optimal N dose for potato yields was about 80 kg N.ha-1. In oats the yields after red clover with using the Horsch system in spring were significantly lower than after embedding of red clover in autumn. In cereals, graduated N doses increased the numbers of ears (panicles) per m2, but the mass of 1000 grains often decreased. In addition, N content in grain and straw increased with N doses, while pH annually decreased by 0.1 to 0.4.

Influence of different organic mineral fertilization on the yield structure and on changes of soil properties

The monitoring was carried on in the years 1996-2000 in the international IOSDV (Internationale Ökologische Stickstoff Dauer Versuche) field trial running since 1983 in Lukavec, Pelhřimov district. In the given locality, there are low fertility sandy-loamy cambisoils, long-term average annual rainfall 653 mm, average annual temperature 7°C, altitude 620 m. In the field trials there were introduced different organic fertilizations and graduate N-doses. According to the complex diffusion analysis, statistically significant for the yields of winter wheat after potatoes was the impact of N fertilization, years, and double interactions, with the exception of different organic fertilization. The N doses up to 120 kg.ha-1 after potatoes proved to be optimal from the viewpoint of winter wheat yields and quality. The situation was similar for winter barley. The results of diffusion analysis for individual years were analogical also in straw yields of both cereals. The highest N dose to potatoes (200 kg.ha-1) resulted in the highest tuber yields, but the starch content was significantly lower. Graduated N doses increased in cereals the numbers of ears per m2, but the mass of 1000 grains mostly decreased. In addition, N content in grain and straw increased with N doses, while pH annually decreased by 0.1 to 0.4. On plots without organic fertilization the N-balance was equilibrated with the annual application of 120 kg N.ha-1 with straw embedding after cereals at 90 kg N.ha-1 and on plots with stall dung to potatoes already at 70 kg N.ha-1. On plots without N fertilization, the balance was passive also in P and K on plots without organic fertilization. A balance surplus was obtained already with the annual application of 15 kg P.ha-1 and 83 kg K.ha-1 and organic fertilization. With the annual fertilization with 22 kg P.ha-1 and 83 kg K.ha-1 and stall dung (30 t.ha-1) once every three years the P and K content increased annually by 1 to 3 mg.kg-1 of soil. Maximum levels of crop growing profitability were obtained with those doses of fertilizers with which also an equilibrated balance of nutrients was obtained.

Comparison of important parameters of spring and winter barley cultivated in sugar beet production area of Czech Republic

Both spring and winter barley were sown after a forecrop of winter rape. Three registered model varieties of six row and two row winter barley reached in the period 1999-2001 significantly higher yield than three model varieties of spring barley. Two row spring and winter varieties produced significantly higher thousand grain weight (TGW) than six row winter barley. The resistance to diseases and lodging (correlation coefficient with resistance to powdery mildew represent 0.68**, rust 0.72*, lodging 0.61**) was the most important factor determining the yield. Resistance to rust has influenced sieving on 2.5 mm, correlation coefficient reached value 0.88**. No significant differences were determined in any malting quality parameter when varieties of two row winter barley variety (Tiffany), and spring barley varieties (Akcent and Tolar) were compared. Variety Tolar reached higher parameters for friability and a beta glucan content. Between years, significant differences were found in following parameters: protein content and apparent degree of attenuation. Due to reduction of area sown with spring barley and reduced yield potential, two row winter barley varieties with comparable malting parameters are recommended to be cultivated as the reserve crop for malting barley industry demands.

Yield evaluation of varieties from the world collection of birdsfoot trefoil (Lotus corniculatus L.)

In field trials in 1998-2000, ten varieties of the world collection of birdsfoot trefoil (Lotus corniculatus) were evaluated for herbage and hay yields. In all the years of testing yields from three cuttings and total annual production were evaluated. In 1999, seed yield was also recorded. As a control, alfalfa (Medicago sativa) variety was included in the experiment. The test varieties showed good productivity in the first and especially in the second year of testing, most of them exceeded alfalfa in herbage yield in individual cuttings and in total productions. As for hay yield, the differences were not so great. Local varieties showed very good productivity, predominantly in herbage yields. Of foreign varieties, the best herbage, hay and seed yielder was the Hungarian variety Puszta. There were no statistical differences in seed yield between the varieties. The yields were, however, relatively low.

Improvement of an elutriation method for estimation of weed seedbank in the soil

A model experiment was conducted to compare hand extraction (elutriation) under running water and elutriation using the device Analysette 3, the two methods for estimation of weed seedbank in the soil. Technical parameters have been assessed for efficient operating of the device. We studied the time required for elutriation of soil samples and the time for collecting, counting and identifying the separated seeds. No significant difference in the effect of the used elutriation method on results of qualitative and quantitative estimation of weed seedbank densities has been found at any of the three locations differing in soil texture (silty loam, loam and clay loam soil). The time necessary for elutriation of soil samples was highly significantly shorter if the device was used, by 35.5 to 42.9% depending on soil texture vs. hand elutriation. The shortest time was assessed for silty loam soil. By contrast, the time needed for selecting, counting and identifying seeds was shortest for clay loam soil. This time was 46 and 92% longer for loam and silty loam soil, respectively. These differences were statistically highly significant. Comparing the seedbank in the soil, a significantly lower number of weed seeds as well as species was found on silty loam soil vs. the two locations with heavier soils. Amaranthus retroflexus was a dominating species at all locations, and on silty loam soil also Chenopodium album. Of a total number of 32 weed species detected in our experiment, 28 were annual and only four perennial (Cirsium arvense, Elytrigia repens, Rumex obtusifolius and Sonchus arvensis).

Yield performance of two buckwheat genotypes grown as a full-season and stubble-crop

Traditional way of growing buckwheat (Fagopyrum esculentum L.) in Slovenia is stubble-crop production, but grown as a full-season crop it yields more. Genotypes that are adapted to stubble-crop system may not necessarily be the best adapted for the full-season crop. The objective of this study was to determine yield performance of two buckwheat genotypes under stubble-crop and full-season production system. The experiments (randomised block design) were conducted in Podravje region with two common determinant buckwheat genotypes (land race population and cultivar Darja) in 1997 and 1998. Buckwheat grown as a full-season crop had a greater leaf area index, more flower clusters, more developed seeds and 42% higher yield than the stubble-crop buckwheat. Although cultivar Darja had 10% less flower clusters than the land race population, the number of flowers and the number of developed grains were higher. The 35% higher grain yield of cultivar Darja was associated with larger leaf area index than land race population (4.0 vs. 2.3). Cultivar Darja outperformed the land race population under full-season crop production, whereas the yield difference between the two varieties was not significant under the stubble-crop production. These results suggest that the best yielding buckwheat genotypes should be determined separately for stubble-crop and/or full-season production system.

The content of Pb, Zn and Cd in hydroameliorated soil and drainage water and their uptake by plants

The goal of the investigations was to assess the average contents of heavy metals (Pb, Zn and Cd) in the surface soil layer (0-30 cm), in drainage water and their uptake by growing plants, in the experimental amelioration field for four different variants of drainpipe spacing (15, 20, 25 and 30 m) during the three-year period. Maize and winter wheat were grown in all variants throughout the trial period. In all variants of drainpipe spacing, heavy metals in soil, drainage water and plant material were within the limits. The highest average level of Pb was 13.5 mg/kg, Zn 88 mg/kg and Cd 0.7 mg/kg in soil. Drainage water concentration of heavy metals was below 50 µg/dm3 for Pb, below 2 µg/dm3 for Cd, while the highest average value of Zn amounted to 20 µg/dm3. In grain of the crops grown, the concentration of Pb was below 0.4 mg.kg-1 and that of Cd below 0.1 mg/kg.

Analysis of chromosome termini in potato varieties

Chromosomes of potato (Solanum tuberosum) are terminated by telomeres, which are formed by tandemly repeated [TTTAGGG]n oligonucleotide sequence. The total length of blocks of telomeric DNA has been known to vary largely among plant species and their varieties, and also among individual chromosome arms within a single nucleus. To check for such differences in potato varieties, which could be of a possible use in genotyping, we performed pulsed-field gel electrophoretic analysis of terminal restriction fragments in selected potato varieties. We found a typical range of telomere lengths of 20-60 kb in most analysed varieties. In one of analysed varieties (Monalisa), telomeres of wider span (up to 80 kb) have been observed. Most of restriction enzymes (PvuII, HaeIII, TaqI) produced a resulting smeared hybridisation pattern of telomeres. When using BglII, however, a doublet hybridisation band could be observed. This may reflect differences in composition of telomere-associated sequences at different chromosome ends.

Rapid hydroponic screening for molybdenum tolerance in rice through morphological and biochemical analysis

High yielding varieties of rice (Oryza sativa) cultivars were tested for their tolerance to different levels of molybdenum (Mo) (0.1µM - control, 0.2, 0.4, 0.8 and 1.6µM) in nutrient solution at pH 6.8. Seeds of rice were germinated and grown in presence of molybdenum under controlled environmental conditions. Standard growth parameters such as root length, shoot length, root/shoot dry biomass and root/shoot tolerance index were tested as markers of molybdenum toxicity. Measurements as early as 48 hours after the germination did not yield consistent results. However, root measurement on 3rd, 6th and 9th day after root emergence showed significant differences among cultivars of rice. Rice cultivars Annapurna, Kusuma, Deepa and Vaghari developed better root system while, Paridhan-1, Pusa-2-21 and Ratna showed poor growth of the roots in presence (0.8µM) of molybdenum. The root tolerance index (RTI) and the shoot tolerance index (STI) in Annapurna, Kusuma and Deepa in rice were high indicating their tolerance to molybdenum; Paridhan-1 and Ratna, however, showed low RTI and STI. Based on the growth parameters, twenty cultivars of rice were ranked in respect of their tolerance to molybdenum: Annapurrna > Deepa > Kusuma > Vaghari > Hamsa > Vikram > Bharati > Paridhan-2 > Aswathi > Subhadra > Sankar > Sakti > Nilgiri > Rudra > Hema > Pragati > Pusa-2-21 > Ratna > Paridhan-1, respectively. Molybdenum toxicity was correlated with increased peroxidase and catalase activity in different cultivars of rice. This method can be employed for quick screening of rice cultivars for molybdenum tolerance in breeding programmes.

The effect of Cd and Zn contents in plants on Fe binding into organic substances of spinach biomass

Partial characterization of iron binding compounds in spinach above ground biomass and differences between binding of Cd, Zn and Fe in spinach biomass were investigated on two soils treated by sewage sludge in model pot experiment. After application of sludge to both soils, Cd and Zn accumulation in spinach biomass was increased in sludge treatments. Total Fe content in biomass showed opposite trend with these treatments and was decreased 2.5-2.7 fold. Results of sequential analysis of spinach biomass showed lower portions of iron in all individual fractions of both sewage sludge treatments compared to control treatments. The largest differences in iron content between control and sludge treatments were determined in ethyl acetate fraction. While majorities of Cd and Zn were found in fractions methanol + H2O and H2O and methanol + H2O + HCl, iron content determined in these two fractions was only 7-8% in control treatments and 15-20% in sludge treatments of total iron. In fraction, Fe portion was 87.7-90.7% in treatments without sludge and 76.1-83.5% in sludge treatments in spinach biomass. Results showed tighter iron binding to plant cell membranes in contrast to cadmium and zinc.

The availability of DTPA extracted heavy metals during laboratory incubation of contaminated soils with glucose amendments

The laboratory incubation with glucose treatment was carried out in order to estimate the possible effects of increased microbial activity on heavy metal availability. The soils from vicinity of a lead smelter operating for more than 200 years were used for the experiment. The DTPA-extractable heavy metal contents increased after glucose addition and mostly reached the highest values the second day of the incubation. The comparative study, where the chloroform fumigation was used prior to the incubation in order to decrease the microbial activities, showed especially from second day of incubation significantly lower DTPA-extractable metal contents compared to non-fumigated treatments. The interactions among the maximum possible availability of DTPA-extractable heavy metal fractions and native soil microbial characteristics were studied in differently contaminated arable and grassland soils. Irrespective of different heavy metal contents in soils, significant correlations were found among the maximum percentage increase of DTPA-extractable Pb and Cd and the ratio Bc/TOC and metabolic quotient (qCO2) which may be a result of the important role of organic matter and microbial characteristics in soils on the heavy metal availability.

Gene typing of the colonisation factor K88 (F4) in enterotoxigenic Escherichia coli strains isolated from diarrhoeic pigletsOriginal Paper

More than 4 000 E. coli strains isolated from diarrhoeic piglets in 111 pig herds in the Czech Republic during the period 1995-2000 were examined for serogroup and virulence factors. Gene typing of the K88 marker by polymerase chain reaction (PCR) was used for the examination of 283 enterotoxigenic strains (ETEC) which agglutinated with antisera against K88 or adhered to intestinal brush borders. The K88 gene was detected in 237 strains; among them 232 strains possesed the K88 variant. Genotype K88ab was found in two strains of the serogroup O8 from one herd and the gene K88ad was detected in three strains of the serogroup O8 originating from another herd. The results show that the type K88ac is predominant in ETEC strains with colonisation factors K88 in pig herds in the Czech Republic.

Collagen binding by vaginal aggregative lactobacilliOriginal Paper

Ten autoaggregating vaginal Lactobacillus strains (five of these strains were selected among isolates from sows' vaginal swabs and the other five among isolates from cows' vaginal swabs) were investigated for their ability to bind type Icollagen (Cn-I). All 10 autoaggregating strains in the range of A570nm readings 0.118-1.806 bound to immobilised Cn-I (at concentration of 100 μg/ml) in wells of microtitre plates, however, Lactobacillus acidophilus SV31 was much more adherent than the rest of the tested strains. The influence of culture medium on Cn-I binding was confirmed only in 50% of the tested strains when agar-grown cells bound significantly more Cn-I than broth-grown cells. The specificity of the binding was confirmed since the Cn-I binding by lactobacilli was abolished after their preincubation with this protein. The effect of heparan sulphate and hyaluronic acid was tested on 5 vaginal strains displaying the best Cn-I binding in microtitre plates after their cultivationon MRS agar plates. Both selected inhibitors significantly (P < 0.001 or P < 0.01) reduced Cn-I binding by the majority of strains. The presence of the gene coding APF (aggregation-promoting factor) was detected in seven strains (all five sows' and two cows' Lactobacillus strains) by PCR.

Monoclonal antibodies to bovine coronavirus and their use in enzymoimmunoanalysis and immunochromatographyOriginal Paper

Two monoclonal antibodies (MAb) to the outer structural protein E2 (spike peplomeric protein) and two MAb to the inner capsid protein N of bovine coronavirus (BCV) were prepared and identified by Western blotting to be used for increasing the specificity and sensitivity of BCV detection. The MAb were checked by the haemagglutination inhibition test and immunoperoxidase tests and no cross reactivity with rotavirus was demonstrated by the immunoperoxidase test and ELISA. A mixture of all the four MAb at predetermined optimum concentrations was first used in sandwich ELISA and then, in combination with an anti‑coronavirus polyclonal antibody, for the development of a simple and rapid immunochromatographic test (ICT). The results of which can be read visually within 10 min. The inclusion of MAb into ELISA and ICT allows the detection of both intact and incomplete BCV virions. ELISA and ICT were used in the examination of a set of 74 faecal samples collected from calves suffering from diarrhoea. ELISA, used as the golden standard verified by electron microscopy, detected BCV in 15 samples (20.3%) and ICT in 16 samples. Three of the ICT‑positive samples were negative by ELISA. On the other hand, two of the 58 ICT‑negative samples were positive by ELISA. Sensitivity and specificity of ICT were 94.9% and 86.7%, respectively

Variations in contamination by mercury, cadmium and lead on swine farms in the district of Hodonin in 1994 to 1999Original Paper

Environmental samples (n = 254) were collected at regular intervals from 1994 to 1999 on three swine farms in the area of the district town Hodonín, Czech Republic. The samples of feed mixtures (n = 85), barn dust sediments (n = 44), and muscular (n = 42), hepatic (n = 41), and renal (n = 42) tissues of feeder pigs were analysed for the concentrations of mercury, cadmium and lead. The concentrations of mercury showed decreasing tendencies in animal tissue samples (1994-1996). Compared with the mean for the Czech Republic, the concentration of mercury was markedly higher in liver and kidney samples (0.064 mg/kg and 0.114 mg/kg, respectively) collected in 1998, the concentration of cadmium was moderately higher in feed, muscle, liver, and kidney samples collected in 1996-1998. Statutory limits were exceeded in two liver and two kidney samples and one feed sample only. The concentration of lead in muscle, kidney, and feed samples (1994-1996) and barn dust samples (1996-1998) showed also a decreasing tendency. However, the limit was exceeded in 16 muscle samples (0.11 to 0.23 mg/kg) collected in 1994-1999 and two kidney samples collected in 1996 (0.74 mg/kg) and 1999 (0.77 mg/kg). Heavy metals present in dust sediments apparently did not contribute significantly to their content in animal tissue. While the health risk resulting from the contents of mercury and cadmium can be classified as moderate, the contents of lead must be regarded as markedly more dangerous.

Numerous data point to the fact that Czech people played an important role in the cultural development of Croatia. Professor Josip Ubl was one of the many outstanding Czech veterinarians who greatly contributed to the Croatian veterinary medicine. Prof. Josip Ubl was born on the 4th April 1844 in Chudenice, in the Plzeň district. He descended from a respectable family, which highly influenced his schooling. He finished his primary and secondary education at his birthplace. He graduated from the k. u. k. Militär-Thierartznei-Institute in Vienna in 1867. Prof. Josip Ubl first worked as an assistant lecturer and as a professor of animal husbandry and veterinary medicine at the School of Farming and Farming Crafts in Doubravice near Loštice and Mohelnice in Moravia. Later on he was appointed a teacher of veterinary medicine, animal husbandry, anatomy and zoology at the Kraljevsko gospodarsko-šumarsko učilište i ratarnica (Royal School of Farming and Forestry) in Križevci in Croatia. He was an exceptionally prolific writer and wrote seven veterinary manuals. Besides being an outstanding teacher he was also engaged in social work and was awarded for his contribution to this field of work several times. As the author of the first veterinary works in Croatian language and the creator of the Croatian veterinary terminology he has gained a prominent place in the history of Croatian veterinary medicine.

Enumeration of bifidobacteria in animal intestina samples

TPY agar supplemented with mupirocin (100 mg/liter) and glacial acetic acid (1 ml/liter) was effective in the enumeration of bifidobacteria in animal (11 species) fecal or intestinal samples. On the other hand, less than 50% of isolates from pig feces were identified as Bifidobacterium sp. using this medium. Subsequently TPY agar modified by the addition of mupirocin (100 mg/liter), glacial acetic acid (1 ml/liter), and colistin (25 mg/liter) was developed for the enumeration of bifidobacteria in pig feces. The results suggest that the bifidobacteria selective media should be chosen in respect of the animal species origin of the samples tested.

Phagocytic and bactericidal activity of blood thrombocytes in carps (Cyprinus carpio)

The studies aimed at monitoring development of phagocytic and bactericidal activity in carps in the course of their ontogenetic development. The studies were performed using the techniques described by Mantur et al. (1986a, b), adapted to fish. Results were expressed in thrombocyte phagocytic index (Ipt), percentage of phagocyting thrombocytes (%tp), percentage of ingested bacteria (%bp) and in the index of intracellular killing by thrombocytes (Ibt). Number of thrombocytes was also examined using the technique of Dessi. Results of testing thrombocyte capacity to ingest the standard Staphylococcus aureus 209P strain showed that Ipt and %tp increased gradually in carps between the age of 3 and 17 months. In older carps, 19 to 29 months of age, as well as in spawners the growth in activity paralelled the ontogenetic development within Ipt values while %tp values remained at a similar level. Phagocytic activity of thrombocytes expressed in Ipt was reflected also by %bp values, which was particularly evident in fish aging 23 months to 5 years. In the case of Ibt, values of the index were not related in any way to stage of ontogeny or sex of the fish. Number of thrombocytes in carps aged 3 months to 5 years was increasing in parallel to their ontogenetic development.

Characterization of superoxide dismutase in the rumen bacteriumStreptococcus bovis

Superoxide dismutase (SOD) isoenzymes of the rumen bacterium Streptococcus bovis 4/1 were studied. Native PAGE showed a single band of Mn-SOD, unaffected by 10 mM cyanide or 5 mM hydrogen peroxide under both aerobic and anaerobic growth conditions. When the metals were removed from the growth medium by Chelex 100, the addition of manganese increased enzymatic activity, while addition of iron inhibited SOD activity. Changes in Mn-SOD and glutathione peroxidase (GSHPx) activities evoked by paraquat and increased values of TBARS indicated that these enzymes were not able to sufficiently prevent oxidative stress at given paraquat concentrations.

Incidence of bovine tuberculosis in cattle in seven Central European countries during the years 1990-1999

The post-eradication incidence of bovine tuberculosis in seven Central European Countries (Bosnia and Herzegovina, Croatia, Czech Republic, Hungary, Poland, Slovakia and Slovenia) was studied between 1990 and 1999. The majority of cattle to the age of 24 months were screened by tuberculin skin test on annual basis. Tuberculous lesions observed during meat inspection at abattoirs were further laboratory examined by direct microscopy, cultivation and histology for the presence of mycobacteria. Data describing the incidence of the disease in animals for the whole period were obtained from all countries except Bosnia and Herzegovina, where data were obtained after the year 1995. Between the years 1990 and 1999, bovine tuberculosis was diagnosed in a total of 1 084 cattle herds. Nine hundred and seventy five (89.9%) outbreaks of the disease were reported in small herds (10 cows) and 109 (10.1%) outbreaks in large cattle herds (>10 cows). The last outbreak of bovine tuberculosis in cattle was diagnosed in Slovakia, Slovenia, the Czech Republic, Croatia and Bosnia and Herzegovina in 1993, 1993, 1995, 1999 and 1999, respectively. Bovine tuberculosis was diagnosed under proper quarantine in Slovenia in 37 fattening bulls imported from two European countries before animals were introduced to the targeting farms.

Brachyspira hyodysenteriae: detection, identification and antibiotic susceptibility

101 samples of faeces, colonic mucosa and rectal swabs taken from 100 pigs (29 commercial herds) were cultivated on Trypticase Soy Agar with 5% of sheep blood, spectinomycin (200 mg/l), vancomycin (50 mg/l), rifampicin (12.5 mg/l) and colistin (12.5 mg/l). Plates were incubated in an anaerobic container at 37°C for 5-7 days. 25 samples (10 faeces, 15 scrapings of colonic mucosa) were examined by darkfield microscopy for the presence of spirochaetes. In 80 samples (21 faeces, 31 rectal swabs, 28 scrapings of colonic mucosa) from diarrhoeic pigs 44 isolates of spirochaetes were identified by PCR method as Brachyspira hyodysenteriae. In 21 samples (20 rectal swabs, 1 scraping of colonic mucosa) taken from apparently healthy pigs in herds with swine dysentery were isolated weakly haemolytic spirochaetes: B. intermedia in 5 samples and phenotypic group III brachyspirae in 4 samples. Minimal inhibitory concentration (MIC) of tylosin, tiamulin and valnemulin was determined by the agar dilution method. To tylosin 16 out of 17 strains of B. hyodysenteriae were resistant (MIC 64 and 256 µg/l), one strain was susceptible (MIC l µg/ml). To tiamulin 17 out of 19 strains were susceptible (MIC from 0.016 to 0.25 µg/ml), one strain was intermediately susceptible (MIC 2 µg/ml) and one resistant (MIC 32 µg/ml). To valnemulin 17 out of 19 strains were susceptible (MIC from 0.016 to 0.064 µg/ml), one strain was intermediately susceptible (MIC2µg/ml) and one was resistant (MIC 8 µg/ml). Valnemulin resistant strain was also resistant to tiamulin.

Gene typing of the colonisation factors F18 of Escherichia coli isolated from piglets suffering from post-weaning oedema disease

Production of verotoxin Stx2e and expression of F18 and K88 colonisation factors were investigated in 222 strains of Escherichia coli isolated from weaned piglets. Sixty-two, 30, and 11 of the 129 verotoxigenic strains were classified to the serogroups O139, O141, and O157, respectively. Other serogroups were identified only sporadically and sixteen strains were unclassifiable. No colonisation factors were detectable in 15 (24.2%) of the 62 verotoxigenic strains classified with the serogroup O139. The fedA gene shared by the colonisation factors F18 was detected by PCR in 47 of the O139 strains (all but one F18ab). Gene fedA, in which the amplification product was digested with endonuclease NgoMI (F18ac), was peculiar to the serogroup O141. No colonisation factors were detected in 19 (14.7%) strains. Genes encoding the colonisation factors F18ac and F18ab were demonstrated in 22 (23.7%) and 11 (11.8%), of the 93 nonverotoxigenic and mostly enterotoxigenic strains, respectively.

Folic acid and its biologically active derivates (folacin or folates) rank among the B-group vitamins due to their hydrophilic character. This acid occurs in nature either in the free form or it is bound to proteins or polysaccharides. Folic acid belongs to the group of vitamins which, in the past were not considered important because the relevance of the acid in biological processes of the living cell has been discovered by the scientific research only in recent years. Folates play an important role as co-enzymes in the synthesis of RNA, DNA, proteins and of some essential amino acids. Therefore they are inevitable for the haematopoiesis, growth and reproduction of cells. In this sense a widely oriented research named "Maternal Folate Status" is proceeding in the U.S.A., Australia and France.
