Phenotyping winter wheat for early ground coverOriginal PaperYuksel KayaCzech J. Genet. Plant Breed., 2022, 58(4):189-200 | DOI: 10.17221/91/2021-CJGPB The relationship between the early ground cover and the grain yield in winter wheat is not yet fully understood. In a winter wheat breeding programme, selection for early ground cover is traditionally made using visual scoring. Although visual scoring is preferred as a phenotypic screening tool by wheat breeders, its output may not be reliable, as it requires experience. A smartphone camera-based digital image technique can be recommended as a feasible, reliable, repeatable, affordable, and fast selection tool for early ground cover in wheat as an alternative to visual scoring. For this purpose, two wheat trials were conducted in the 2017-2018 and 2019-2020 seasons. In both seasons, 215 wheat genotypes in total, together with three checks from spring wheat, were tested under rain-fed conditions in the spring wheat zone in Turkey. All the tested wheat genotypes were grouped into spring, facultative, and winter growth habit using visual scoring. Simultaneously, photos were taken from each plot with a smartphone camera, and the early ground cover (%) was estimated using the smartphone camera-based digital image technique. The relationships between grain yield, visual scoring, and early ground cover could so be estimated. In both seasons, significant negative correlation between grain yield and visual scoring (r = -0.679** and r = -0.704**, respectively) and significant positive correlation between the grain yield and the early ground cover (r = 0.745** and r = 0.747**, respectively) were observed. The correlation between visual scoring and early ground cover were negative (r = -0.862** and r = -0.926**, respectively). The broad sense heritability estimates in both seasons were 0.51 and 0.85, respectively, for early ground cover, 0.91 and 0.94 for visual scoring, and 0.86 and 0.69 for grain yield. In this study, we revealed that testing winter wheat genotypes in the spring wheat zone rather than in the winter wheat zone could be a more effective way to unveil the positive relationship between the early ground cover and the grain yield. We have shown that the smartphone-based digital image technique is a useful selection tool for early ground cover in winter wheat. |
Dietary supplementation of Lactobacillus zeae regulated the gut microbiome in piglets infected with enterotoxigenic Escherichia coliOriginal PaperQian Zhang, Lijuan Zhang, Yang Lyu, Yutao Shi, Liangyun Zhu, Min Zhang, Yuyan Zhao, Di Zhao, Lei Wang, Dan Yi, Yongqing Hou, Tao WuCzech J. Sci., 2022, 67(1):27-38 | DOI: 10.17221/136/2021-CJAS This study was conducted to investigate the effect of Lactobacillus zeae LB2 on the gut microbiota in piglets infected with enterotoxigenic Escherichia coli (ETEC). Thirty-two healthy 7-day-old piglets were randomly divided into four treatment groups: control group (basal diet), LB2 group (supplemented with 1 × 108 CFU/pig/day L. zeae LB2), ETEC group (infected with 1 × 1010 CFU/pig/day ETEC) and ETEC+LB2 group (LB2 supplementation + ETEC infection). Intestinal contents were collected for DNA extraction and Illumina sequencing. Significant result was observed for alpha diversity in the four intestinal sections, and both ETEC infection and LB2 supplementation showed a higher Chao1 alpha diversity. At the phylum level, Firmicutes and Bacteroidetes were dominant in the healthy piglets, while Proteobacteria were dominant in the ETEC-infected piglets. At the genus level, ETEC infection decreased the abundance of Prevotella, Ruminococcaceae, Lactobacillus, Alloprevotella, Flavobacterium, and Sutterella and increased the abundance of Actinobacillus. The LB2 supplementation reduced the abundance of Ruminococcaceae, Actinobacillus, Porphyromonas, and Alloprevotella, and increased the abundance of Prevotella and Lactobacillus. Both ETEC infection and LB2 supplementation affected several functional pathways associated with cellular processes, environmental information processing, genetic information processing, diseases, metabolism, and organismal systems. In summary, ETEC infection induced dysbiosis of the gut microbiome in piglets, while L. zeae supplementation could positively regulate the gut microbiome during ETEC infection. Therefore, L. zeae LB2 may be an ideal probiotic for the prevention or treatment of ETEC infection. |
A comparative study on the effect of quantitative feed restriction in males and females of broiler chickens, rabbits and nutrias. I. Sci., 2022, 67(2):47-54 | DOI: 10.17221/185/2021-CJAS The aim of the study was to compare the effects of feed restriction on the growth performance and carcass composition of chicken, rabbit and nutria males and females. Feed restriction was applied at a rate of 70% ad libitum from 14 to 21 days of age in chickens, 70% ad libitum from 42 to 49 days of age in rabbits, and 70% ad libitum from 12 to 15 weeks of age in nutrias. Animals were fed ad libitum prior to and following restriction. Carcass composition was evaluated at common slaughter ages, i.e., 31 days in chickens, 70 days in rabbits, and eight months in nutrias. The results showed stronger effects of feed restriction, sex, and their interaction in broiler chickens than in rabbits and nutrias. In chickens, feed restriction increased the sex differences in final body weight, which were associated with interaction of feed restriction and sex (P = 0.009). Feed restriction as a fixed factor significantly affected growth and feed conversion in chickens and it was with no effect in rabbits and nutrias. The effect of feed restriction on carcass composition was negligible in all selected species. With respect to sex, significant differences were observed in chickens in final body weight, daily weight gain, dressing out percentage and breast percentage, in rabbits in loin percentage, and in nutrias in growth and perirenal fat percentage. Results of the study indicated that interactions of feed restriction and sex in growth might be associated with a short realimentation period. |
Influence of chemotherapy on development and production of virus free in vitro strawberry plantsOriginal PaperJiří Sedlák, František Paprštein, Jana SucháHort. Sci. (Prague), 2019, 46(2):53-56 | DOI: 10.17221/249/2017-HORTSCI The objective of the study was to determine effects of ribavirin on development and health status of in vitro grown strawberry cultivars 'Honeoye' and 'Elkat' infected with viruses Strawberry mild yellow-edge virus (SMYEV), Tomato ringspot virus (ToRSV) and Arabis mosaic virus (ArMV). Antiviral compound ribavirin was added in concentrations 20, 40, 80 and 160 mg/l to the same MS medium as for multiplication. Growth reduction was noted on medium with 160 mg/l ribavirin and to a lesser degree in the 40 and 80 mg/l treatments. At the end of chemotherapy, in vitro clones free of viruses detected previously in the initial plants were obtained for both selected cultivars across all ribavirin concentrations. The highest number of plants (94) with negative results of ELISA testing was noted on medium with the highest ribavirin concentration 160 mg/l and the lowest (73) on medium with the lowest concentration 20 mg/l of ribavirin. The treated plants look symptomless and appear morphologically equal to the untreated control plants. Results indicate that ribavirin treatment of in vitro plants is a suitable method for eliminating SMYEV, ToRSV and ArMV from strawberry. |

(Prague), 2022, 49(2):109-116 | DOI: 10.17221/102/2021-HORTSCI Plant growth regulators (PGRs) and proper fertilisers are used to increase quality of the Salak Pondoh, such as in the plant production, fruit number and size. This study aims to determine the type and dosage of PGRs and the best type of fertiliser to increase the production bunch and fruit of Salak Pondoh. The method used was a Factorial Completely Randomised Design (FCRD) with 36 treatments (excluding the control) and 3 replications. Three treatments were conducted: first, as a control, no PGR treatment + fertilisation (no fertilisation, manure, and NPK). Second, a 3 × 3 factorial scheme. Treatment of PGR GA3 (50, 100, 150 ppm) + fertilisation (no fertilisation, manure, and NPK). Third, a 3 × 3 × 3 factorial scheme was used. Treatment of PGR Gracilaria sp. extract (50, 100, 150 ppm) with different solvents (n-Hexane, methylene-chloride, ethanol) + fertilisation (no fertilisation, manure, and NPK). The parameters observed were the number length, area of the bunch, length and area of the fruit. The results showed the highest number of bunches was found in PGR GA3 50 ppm. While, the highest bunch area was found in PGR Gracilaria sp. ethanol 150 ppm. The PGRs only increased the formation of the bunches and did not increase the length and width of the bunches and the fruit size. Fertilisation affected the Salak production in the form of the number, length and width of the bunches, and the fruit size. In addition, the fertiliser increased the number of the bunches, bunch area, and length and area of the fruit. The interaction of the PGR and fertilisation had no significant effect on increasing the Salak Pondoh production. The best result in increasing the Salak Pondoh production was PGR Gracilaria sp. ethanol 100 ppm with manure fertilisation. |

Sci., 2022, 68(2):61-71 | DOI: 10.17221/128/2021-JFS Wild boar (Sus scrofa L.) is one of the most discussed game species, distributed across Europe, therefore the management of this species is considered important. This management should be based on data presented, population quality and preferences and craniometric dimensions show the development of the individual and its prosperity. A sample of 148 male and 153 female wild boar mandibles was studied to compare differences in craniometric measurements, especially to find out wild boar environmental demands and population trends. The width of the caput mandibulae and angle of the mandible showed significant difference between males and females. Measurements analysed with forest area size and other data also showed that larger craniometric dimensions were reached in hunting areas with at least 200 ha of forested area, which may be due to the wild boar's need for safety and vegetative cover in the first months of piglet development with respect to its home range. The development of young wild boar is dependent on rest and shelter in the first months of life. A forest cover of at least 200 ha appears to be sufficient in this respect. Information on habitat preferences and individual development can lead to improvements in wild boar management. |

Sci., 2022, 68(4):145-155 | DOI: 10.17221/16/2022-JFS Vegetation dynamics, soil properties, and the correlation between them following a wildfire are crucial to understanding the recovery of forest (natural or planted forest) ecosystems. We compared species composition and soil properties in two burned (Br) and unburned (UBr) sites of loblolly pine (Pinus taeda L.) stand in Northern Iran. We detected 39 plant species including 22 (56.4%) species that were common in both sites, 13 (33.3%) species specifically in the Br site, and 4 (10.3%) species specifically in the UBr site. Although species abundance was significantly higher in the UBr site, species richness was significantly higher in the Br site. Species composition was significantly different (F = 16.25, P-value = 0.001) between Br and UBr sites. Rarefaction-extrapolation revealed consistently and significantly higher species diversity in Br site compared to UBr site for all three Hill numbers. Only sand (t = 2.23, P-value = 0.030), pH (t = 2.44, P-value = 0.018) and electrical coductivity (t = 2.98, P-value = 0.004) were significantly higher (P-value ≤ 0.05) in the Br site due to the demobilization of base cations in burnt vegetation. In the Br site, the wildfire did not cause any marked changes in C and N stocks. |
Performance evaluation of spirodiclofen, spirotetramat and carboxymethyl cellulose-B for management of Cacopsylla pyriOriginal PaperĐura Nađ, Dušan MarinkovićPlant Protect. Sci., 2022, 58(1):70-75 | DOI: 10.17221/110/2021-PPS The pear psyllid [Cacopsylla pyri (L.), (Hemiptera: Psyllidae)] represents one of the most economically significant pests of pear in Serbia and worldwide. It causes direct damage throughout feeding on young stems and leaves, but also creates favourable conditions for the development of plant diseases. In this article susceptibility of C. pyri to spirodiclofen, spirotetramat, and their combinations with foliar fertilizer (carboxymethyl cellulose-boron, CMC-B) was investigated in order to assess their efficacy. Experiments were conducted in accordance with standard OEPP/EPPO methods, in field trials on two localities in the Republic of Serbia (Kula, Vrbas), during 2020/21. Results of the field trials indicate good efficacy for all variants in pear psyllid control in pear orchards. The high efficacy of spirodiclofen (87.4-95.4%), spirotetramat (82.5-91.8%), spirodiclofen + CMC-B (90.3-99.8%), spirotetramat + CMC-B (89.5-96.8%) was achieved at both localities seven days after treatment during 2020/21 field trials. Also, it can be concluded for examinations 14 days after treatment, where the efficacy of spirodiclofen (84.9-92.0%), spirotetramat (81.2-91.7%), spirodiclofen + CMC-B (88.9-97.5%), and spirotetramat + CMC-B (82.3-92.5%) was high at both of investigated localities. Based on the obtained results it can be concluded that the high efficacy of the researched insecticides is a good indicator of C. pyri susceptibility in pear orchards. |

However, a systematic analysis of LRR-RLKs has not been accomplished in the economically important cucumber. 189 LRR-RLK genes were identified in the cucumber genome and further divided into 22 subgroups based on the sequence similarities in this study. A total of 31 segmental duplication events and 15 tandem duplication events were present in the genome, indicating that the two duplications were the main driving forces for the expansion of the LRR-RLK family in the cucumber. The expression profile analysis revealed that most of the CsLRR-RLKs were upregulated during a downy mildew infection, and resistant cucumbers comprised more upregulated CsLRR-RLKs than the sensitive lines. Taken together, our results provided information on the LRR-RLK gene family in the cucumber and contributed valuable information for the further research of CsLRR-RLKs. |
First report of Seiridium cardinale on Cupressus sempervirens in SerbiaShort CommunicationIvan Milenković, Zlatan Radulović, Dragan KaradžićPlant Protect. Sci., 2022, 58(4):360-364 | DOI: 10.17221/54/2021-PPS While monitoring the health of different ornamental and shade trees in Serbia, symptoms indicative of cypress canker disease were observed in young Cupressus sempervirens trees in the Belgrade urban area. Symptoms included tree mortality (specimens were recorded with a change in needle colour, branch decline or longitudinal bark cankers on the stems with resin exudates) and the appearance of acervuli fruiting bodies on the bark and cones. Using light microscopy, cross sections of fruiting bodies on the cankered bark and cones were examined and numerous fusoid, six-cell conidia were recorded with four inner coloured cells and two hyaline cells at the ends. Based on the unique combination of the morphological features and the infected host, this pathogen was identified as Seiridium cardinale. This is the first report of S. cardinale on the common cypress in Serbia. Possible introduction pathways and the implications of the findings are discussed. |
Free amino acid regulation in fronds and roots of two Pteris cretica L. ferns under arsenic stressOriginal PaperVeronika Zemanová, Daniela Pavlíková, Milan PavlíkPlant Soil Environ., 2020, 66(10):483-492 | DOI: 10.17221/369/2020-PSE In the present study, free amino acid (AA) regulation in the arsenic (As) hyperaccumulating ferns was evaluated in a pot experiment to determine the relationship between As stress and the characteristic change in metabolism of AAs. The ferns Pteris cretica cv. Albo-lineata (Pc-Al) and cv. Parkerii (Pc-Pa) were exposed to As treatments at 0, 20, 100, and 250 mg As/kg for 90 days. Greater As content, as well as higher biomass production, were identified in Pc-Al compared with Pc-Pa. Ferns showed changes in the stress metabolism of free AA homeostasis. These results indicate a disturbance in nitrogen metabolism and depletion of pool assimilated carbon metabolism. In the fronds and roots, Pc-Pa accumulated higher amounts of free AAs than Pc-Al. The total free AA content, as well as the ratio of the main AA family pathway (glutamate family), were increased by the accumulation of toxic As in the ferns. Results suggest that Pc-Al tolerates higher As doses better due to changes in AA biosynthesis; however, at higher As doses, Pc-Pa upregulated AA biosynthesis due to As toxicity. The most abundant free AAs of ferns was glutamine, which was enhanced by As. Furthermore, the ratios of selected individual free AAs revealed a characteristic phenotype difference between ferns. |

Therefore, secondary B sources such as end-of-life cellulose fiber insulation (CFI) should be preferred for B fertiliser production over primary borates. In addition, crop B fertilisation is challenging because B is only weakly adsorbed in soils and prone to leaching in particular if the soil pH is below 7. The objectives of this study were to assess the effect of pyrolysed CFI (B-Biochar) on crop B uptake in the field and on B leaching in a lysimeter study. B-Biochar was pyrolysed at 600 °C and tested (1) in a field experiment with maize (Zea mays L.) and sunflower (Helianthus annuus L.), and (2) in a lab microlysimeter experiment to study B leaching under simulated rainfall. In the field experiment, B concentration in plant tissue increased by up to 100% with B-Biochar compared to the control (from 29.6 to 61.6 mg B/kg in young sunflower leaves) and was only slightly lower (-10% to -20%) than with water-soluble Na-tetraborate (Borax). This lower uptake was attributed to the slow-release properties of the B-Biochar. In the lysimeter experiment, 41% and 55% of added B through B-Biochar was leached below 16 cm depth when fertilised with 1 and 2 kg B/ha, respectively, but B concentration of the leachate remained below the 1 mg B/L threshold value for drinking water in the European Union. In conclusion, CFI has a strong potential as a secondary B source for fertiliser production, and pyrolysis appears to be a suitable process for that purpose. During the processing of CFI to fertiliser, more focus should be given to slow B release in the future in order to reduce losses by leaching. |
Decomposition of rice straw residues and the emission of CO2, CH4 under paddy rice and crop rotation in the Vietnamese Mekong Delta region - A microcosm studyOriginal PaperTran Van Dung, Tat Anh Thu, Vu Van Long, Chau Thi DaPlant Soil Environ., 2022, 68(1):29-35 | DOI: 10.17221/304/2021-PSE This study investigated the influence of soil undergoing different crop rotations on the CH4, CO2 emissions, and decomposition of rice straw. The studied soil undergoing crop rotation systems were rice-rice-rice (SR) and baby corn-rice-mungbean (SB). Two main microcosm set-ups: anaerobic (SR-AN, SB-AN) and aerobic (SR-AE, SB-AE) conditions. Litter bags containing rice stems were inserted into the soil and recollected at different time points for chemical analysing and the gas sampling was collected to measure the CO2 and CH4 emissions. The results indicated that the total carbon (TC) decreased around 30%, and the TC removal in anaerobic was significantly higher than in aerobic conditions. The residue cellulose content varied in a range from 68.2% to 78.6%, while the hemicellulose content varied from 57.4% to 69.3% at day 50 after incorporation. There were no significant differences in the total nitrogen removal, cellulose, hemicellulose, and lignin contents among the microcosm set-ups. CO2 emission increased in all the microcosm set-ups with the treatments without rice straw (CTSR, CTSB) in both aerobic and anaerobic conditions. CH4 release in the SR-AN treatments did not differ significantly compared with the SB-AN treatments. This study confirmed that the decomposition of rice straw residues is faster in the anaerobic paddy soil condition compared to the aerobic crop rotation condition. |
Regulation of nitrogen balance and yield on greenhouse eggplant under biochar addition in MollisolOriginal PaperYao Wang, Meng Zhou, Meng Hou, Yimin Chen, Yueyu Sui, Xiaoguang JiaoPlant Soil Environ., 2022, 68(1):36-48 | DOI: 10.17221/393/2021-PSE Maintaining nitrogen (N) balance and inhibiting N leaching loss in the soil-crop system is crucial to maintaining yield and reducing the environmental pollution. This study investigated the effects of soil NO3--N content and accumulation, eggplant yield, N leaching and balance response to biochar addition, including regular fertilisation and irrigation (W + F), biochar addition with regular fertilisation and irrigation (W + F + B), and biochar addition with 20% fertilisation and irrigation reduction (0.8W + 0.8F + B) treatments. Compared with W + F, W + F + B and 0.8W + 0.8F + B increased soil NO3--N content in 0-40 cm and soil NO3--N accumulation in 0-20 cm, and raised harvest index, N surplus and balance. Simultaneously, 0.8W + 0.8F + B compared to W + F enhanced N use efficiency and N partial factor productivity, conversely, it decreased N dry matter production efficiency, N surplus and balance. Stepwise regression analysis demonstrated that the effect of NO3--N leaching lasted in 60 cm under biochar addition in the first year, and lasted in 20 cm without biochar application in the next year. Altogether, biochar addition with 20% fertilisation and irrigation reduction is the most suitable management strategy to decrease N surplus and leaching, and maintain eggplant N uptake in a two-year cycle system on greenhouse vegetables in Mollisols. |

The effect of inoculation strain was evaluated under different soil conditions: with or without previously grown soybean and applied commercial inoculant. Molecular identification of inoculation strain and investigated rhizobial isolates, obtained from nodules representing inoculated treatments, was performed based on 16S rDNA and enterobacterial repetitive intergenic consensus (ERIC) sequencing. Inoculation strain showed a significant effect on the investigated parameters in both soils. Higher nodule occupancy (45% vs. 18%), nodule number (111% vs. 5%), nodule dry weight (49% vs. 9%), shoot length (15% vs. 7%), root length (31% vs. 13%), shoot dry weight (34% vs. 11%), shoot nitrogen content (27% vs. 2%), and nodule nitrogen content (9% vs. 5%) was detected in soil without previously grown soybean and applied commercial inoculant. Soil had a significant effect on the shoot, root and nodule nitrogen content, while interaction of experimental factors significantly altered dry weight and nitrogen content of shoots, roots and nodules, as well as number of nodules. Nodulation parameters were significantly related with shoot dry weight, shoot and nodule nitrogen content. Symbiotic performance of inoculation strains in the field could be improved through co-selection for their competitiveness and effectiveness. |
Effect of auxin foliar application on seed yield and fatty acids composition of two safflower genotypes under late-season droughtOriginal PaperSeyed Basir Mousavi, Saeed Sayfzadeh, Hamid Jabbari, Seyed Alireza Valadabadi, Esmaeil Hadidi MasoulehPlant Soil Environ., 2022, 68(2):82-88 | DOI: 10.17221/329/2021-PSE To assess the response of two safflower genotypes to auxin foliar application under late-season drought stress using a factorial split-plot experiment with the randomised complete block design, a two-year experiment (2016-2017 and 2017-2018) was conducted in Iran. The watering regime as specified in two levels including, regular irrigation and drought stress in main plots and two safflower genotypes and auxin foliar application in two levels including non-foliar application (control) and auxin foliar application at a concentration of 30 ppm were categorised in subplots. Drought stress at the seed filling stage reduced the safflower seed and oil yield. Moreover, a significant drought-induced decrease in linoleic acid, palmitic acid and behenic acid content, as well as an increase in oleic acids, was observed in two safflower genotypes. Among two safflower genotypes, the early maturing genotype less affected by drought and Goldasht had higher seed yield than the Padideh by 814 kg/ha, respectively. Auxin foliar application alleviates the adverse effects of drought, which led to an 18% increase in seed yield and components. However, the auxin application had no effect on the safflower seed oil content but palmitic acid content was affected by auxin foliar application. Generally, safflower along with auxin application can be recommended to develop safflower cultivation in semi-arid areas. |

Four N treatments in the field experiment were designed as UAN (urea ammonium nitrate solution was applied under traditional side-dressing method), urea (urea was applied under traditional side-dressing method), UWFI (UAN was applied underwater and fertiliser integration technology) and CK (no N applied). The results showed that the root length density, surface area density and volume of DH605 (mid-late hybrid) and DH518 (mid-early hybrid) under UWFI were higher than other treatments, especially in shallow layers. The root absorption area of each soil layer under N application treatments varied with the growth stage. The grain yield and the accumulations of N, P and K in the shoots showed the trend of UWFI > UAN > urea > CK. Compared with UAN and urea, the nitrogen agronomic efficiency of UWFI treatment increased by 40.5~78.6%, and the nitrogen partial factor productivity increased by 4.75~7.61%. Consequently, rational application of UAN would improve root traits, nutrient uptake and utilisation, and yield of summer maize. |

The investigated pre-treatment methods are hydrolysis of the material in NaOH (0.5% and 5%) and H2SO4 (0.5% and 5%) at temperatures of 90-100 °C for 2 h. This work aimed to compare the effects of these pre-treatment methods on the lignocellulosic composition of maize waste (maize stalks, leaves and cobs), rapeseed straw and wheat straw and the biogas yields from its subsequent anaerobic digestion. Pre-treatment by 0.5% NaOH increased the biogas production the most (by 159% for rapeseed straw, 240% for wheat straw and 59% for maize waste) also the degrees of solubilisation was higher, at the same time the proportions of total organic carbon (TOC) in the solid component appear to be sufficiently high, it could be assumed that there has been sufficient disturbance of the material structure. Alkaline methods have proven to be more suitable compared to acid pre-treatment methods, also the use of alkali with a lower concentration has shown to be more efficient, which is more advantageous for use in practice from an environmental and economic point of view. |

To investigate the range of biochar's effects on soil chemical properties (e.g., soil pH, electrical conductivity (EC), cation exchange capacity (CEC), soil organic carbon (SOC), soil total carbon (TC), and soil carbon-nitrogen ratio (C:N ratio)) in response to varied experimental conditions, a meta-analysis was conducted on previously published results. The results showed that the effect of biochar on soil chemical properties varied depending on management conditions, soil properties, biochar pyrolysis conditions, and biochar properties. The effect size (Hedges'd) of the biochar was greatest for SOC (0.50), the C:N ratio of soil (0.44), soil pH (0.39), TC (0.35), EC (0.21), and CEC (0.20). Among the various factors examined by aggregated boosted tree analysis, the effects of biochar on soil chemical properties were largely explained by the biochar application rate, initial soil pH, and soil sand content. In conclusion, our study suggests that improving soil chemical properties by adding biochar not only requires consideration of biochar application rates and chemical properties but also the local soil environmental factors, especially soil initial pH and sand content of the soil, should be considered. |

This study aimed to develop a model calibrated to predict the rhodinol content in citronella oil using near-infrared (NIR) spectroscopy. This research is the initial stage in developing a spectral smart sensor system that predicts the rhodinol content of citronella oil in the distillation and fractionating process. Citronella oil samples were scanned by NIRFlex liquid N-500 with a wavelength of 1 000-2 500 nm having an absorbance value (log 1/T). The accuracy of the prediction was achieved using the partial least square (PLS) model. Based on the NIR spectrum at a peak of around 1 620 nm, the rhodinol content in the citronella oil was estimated. The finest model to predict the rhodinol content was y = 0.9874x + 15.6439 with a standard error of the calibration set (SEC) = 2.78%, a standard error of the prediction set (SEP) = 2.88%, a ratio of the performance to the deviation (RPD) = 9.23, a coefficient of variation (CV) = 16.81%, and the correlation coefficient (r) = 0.99. The NIR and PLS models are possible to use for the initial stage in developing a spectral smart sensor system to determine the rhodinol content of citronella oils. |

A total of 99 clinically healthy, mixed-breed mesocephalic dogs were included in the study and divided into three different age groups according to the current human/pet analogy chart: 40 adults (23 males, 17 females, 1–8 years old, 3–70 kg), 21 seniors (14 males, 7 females, 6–11 years old, 7–42 kg), and 38 geriatrics (22 males, 16 females, 8–13 years old, 5–45 kg). All the dogs underwent an ophthalmic examination, including Schirmer tear test, tonometry, biomicroscopy, and ophthalmoscopy. Neither eye drops nor pharmacological sedatives were administered before the autorefractometry. The refractive states were assessed bilaterally using a hand-held Retinomax 3 (Righton) autorefractor. The results underwent statistical analysis using Statistica v12 software (ANOVA and t-test). A P-value < 0.05 was considered as significant. Emmetropia, defined as a refractive state > −0.5 D and < +0.5 D, was found in 36% of the adult, 43% of the senior, and 38% of the geriatric patients. Anisometropia was found in 1% of the adult, 9.5% of the senior and 5.5% of the geriatric dogs when the refractive power of the two eyes differed ≥ 1.0 myopia ≤ −0.5 D and hyperopia ≥ +0.5 D were found in 23% and 41% of the adult eye globes as well as 24% and 33% in the senior dogs and 15% and 47% in the geriatric dogs, respectively. The maximal values of the myopia in the adult and geriatric dogs were −2.5 D and −2.75 D, respectively. The maximal values of the hyperopia in the adult and geriatric dogs were 1.75 D and 2.5 D, respectively. No statistically significant correlation was found between the groups. Ametropia is a common refractive state for dogs of different ages. The most frequent refractive state in ametropic mixed-bed dogs in all age groups is hyperopia. |

Most teratomas are benign, but a histological diagnosis is important for clinicians. The objective of this article is to describe a benign ovarian teratoma in a dog, which was found on the street and was appearing like pregnant. A veterinary inspection by palpation documented an enlarged abdomen with a mass of tough matter located on the right side in the abdominal-pelvic part. An ultrasound examination presumed neoplastic mass in region of ovary. A bilateral ovariohysterectomy was performed and the subsequent histological evaluation revealed a benign ovarian teratoma with a histochemically and immunohistochemically verified nervous tissue. After one year, no distant metastases were found and the dog was recognised as being clinically healthy without problems. On the basis of the ultrasonography diagnostics and histopathological analyses, we have demonstrated the occurrence of a benign ovarian teratoma in a dog. |

Diagnosis is based on clinical evidence of a patellar luxation and stifle joint instability. However, diagnostic imaging is required to assess the number of skeletal deformities and signs of instability. Surgical options include both soft tissue and osseous techniques, although, in most cases, a combination of multiple procedures is necessary to correct the patellar luxation and restore the stifle joint stability. Complication rates are generally low, but can include reluxation and implant-associated complications. This article describes the patellar luxation and cranial cruciate ligament rupture signs in dogs, including the clinical presentation and diagnosis, and discusses current treatment options. |

This review provides a broad approach to the pathophysiological mechanisms of this syndrome, and the diagnostic and treatment methods to facilitate their approach in clinical routine and increase the survival rate of patients. |
Combination of immunosuppressive drugs and allogeneic stem cell treatment in a dog with suspected nephrotic syndromeCase ReportY Jin, Y Son, T Oh, S BaeVet Med - Czech, 2022, 67(9):492-496 | DOI: 10.17221/115/2021-VETMED The case study aims to describe the nephrotic syndrome (NS) in a castrated 3-year-old male Cocker Spaniel dog. The patient arrived at the hospital with a loss of appetite and weakness. Skin oedema with ascites was observed along with hypoalbuminaemia, hypoproteinaemia, hyperlipidaemia, hypercholesterolaemia, and proteinuria (urine protein to creatinine ratio = 22.4). Based on these findings, the patient was diagnosed with NS, although a renal biopsy was not conducted. Prednisolone (1 mg/kg, p.o. q12 h) and mycophenolate mofetil (10 mg/kg, p.o. q12 h) were prescribed as the immunosuppressive drugs, and previously cryopreserved allogeneic adipose tissue-derived mesenchymal stem cells (2 × 107 cells/kg) were injected intravenously. After several weeks of treatment, the patient recovered from NS. This is the first case report on immunosuppressive drugs and allogeneic mesenchymal stem cells being used to treat a dog with NS. |
Isolation and characterisation of Brucella melitensis by bacteriological and molecular methods from livestock in North CyprusOriginal PaperM Demirpence, AM Saytekin, B Sareyyupoglu, OM EsendalVet Med - Czech, 2022, 67(10):497-509 | DOI: 10.17221/150/2021-VETMED In this study, the isolation, biotyping and molecular characterisation of Brucella melitensis from cattle, sheep and goats in North Cyprus are reported on. A total of 319 raw milk samples obtained from seropositive dairy livestock (190 cattle, 74 sheep and 55 goats) and tissue samples including the liver, spleen and abomasal contents obtained from 32 aborted foetal samples (5 cattle, 18 sheep and 9 goats) were analysed for the presence and characterisation of the agent. B. melitensis was isolated and identified from 90 out of 319 (28.2%) milk and 19 out of 32 (59.4%) foetal samples by conventional bacteriological methods. Identification of all 109 isolates was confirmed by using real-time PCR with genus and species-specific primers. Following the preliminary identification, 27 selected isolates representing various counties and herds were further analysed by conventional methods. Twenty (74.1%) isolates were identified as B. melitensis biovar 1 and seven (25.9%) were identified as B. melitensis biovar 3. The Bruce-ladder multiplex PCR assay revealed that all the isolates were field strains. The results of the present study confirmed the presence of B. melitensis in livestock including the cattle population in North Cyprus. Even though the majority of the samples came from seropositive cattle, Brucella abortus was not isolated in the study. The results also revealed the potential public health risk of brucellosis in livestock emphasising the need of implementing strict control and eradication strategies against the disease in animal populations in order to protect human health. |
Successful management of feline CD4+ CD8+ T-cell mediastinal lymphoma with pericardial effusionCase ReportSJ Yu, HE Kim, HJ Han, JH KimVet Med - Czech, 2022, 67(10):544-551 | DOI: 10.17221/70/2021-VETMED A 2-year-old, castrated, male Russian blue cat presented with acute dyspnoea, cyanosis, and lethargy. A thoracic radiography revealed a large cranial mediastinal mass; the computed tomography revealed caudal lobe atelectasis of the right lung with pericardial and pleural effusions. The cytodiagnostic tests revealed high-grade CD4+ CD8+ T-cell mediastinal lymphoma as clinical stage Vb; l-asparaginase-cyclophosphamide-doxorubicin-vincristine-prednisolone (l-CHOP)-based chemotherapy was initiated, following which the mass shrunk rapidly; 1 week after the initiation of chemotherapy, the appetite-related and respiratory symptoms improved dramatically, and the pleural and pericardial effusion resolved. The patient remains in complete remission three years after the initiation of the l-CHOP chemotherapy. Therefore, the accurate diagnosis and instantaneous initiation of chemotherapy may resolve life-threatening pleural and pericardial effusions in cats with high-grade aberrant T-cell mediastinal lymphoma. |
Can biogenic amines cause ailments following the intake of edible mushroom meals?Original PaperEva Dadáková, Tamara Pelikánová, Pavla Wolfová, Pavel KalačCzech J. The causes of such complaints have not been explained yet. We, therefore, tested levels of biogenic amines (BAs). Fruit bodies of widely consumed wild-growing species, Imleria badia and Suillus variegatus were stewed, then preserved by freezing or canning and stored for up to 12 months. Contents of six amines were determined in the fresh matter, in each step of preservation and during storage. Histamine (HIM) and cadaverine (CAD) were not detected at all. Putrescine (PUT) occurred in fresh fruit bodies at levels of 700-1 500 mg kg-1 dry matter (DM), however, its contents considerably decreased, particularly during stewing. Undesirable phenylethylamine (PEA) and tyramine (TYM) occurred at lower levels. Stewing, the technological step necessary in both the tested preservation treatments, reduced the contents of all the amines alike as sterilisation, whereas following storage showed a limited effect. PUT seems to be the only amine that could participate in the reported ailments. |
The effects of arbuscular mycorrhizal fungi and deficit irrigation on the yield and sugar content of watermelons (Citrullus lanatus)Original PaperPeng-Ming Yang, Song-Tao HeHort. Sci. (Prague), 2022, 49(4):225-233 | DOI: 10.17221/108/2021-HORTSCI Many studies have demonstrated that arbuscular mycorrhizal fungi (AMF) and deficit irrigation (DI) have positive effects on the fruit yield or quality. This study aims to test whether the watermelon sugar content and yield can be improved by appropriate amounts of AMF and DI methods and to clarify the underlying physiological mechanism. Diploid and triploid watermelon cultivars and their pumpkin rootstock-grafted seedlings were treated with AMF, DI and DI + AMF in a randomised complete block design with five replications. The results showed that DI significantly reduced the relative water content (RWC), PN, alkaline α-galactosidase activity, but increased the insoluble acid invertase (IAI), sucrose synthase (SuSy) and sucrose phosphate synthase (SPS) activities compared with the well-watered (WW) treatment, which led to a decrease in the fruit yield and an increase in the fruit sugar content. Although the AMF improved the RWC, PN and alkaline α-galactosidase, IAI, SuSy and SPS activities in all the watermelon lines under both the DI and WW conditions, the improvement magnitude of these parameters was more pronounced in the pumpkin-root watermelon lines than the corresponding own-root watermelon lines, especially under the DI condition. The integrated application of AMF and DI increased the fruit yield to a level similar to the WW value in the pumpkin-root watermelon lines and sugar content to an optimal level in all the watermelon lines. |
Sodium selenite improves the vase life of Eustoma grandiflorum cut flowersOriginal PaperNinghai Lu, Limin Wu, Chaohui Zhang, Changjuan ShanHort. Sci. As a trace element, selenium (Se) has been proven to play an important role in extending the vase life of lily cut flowers. However, there is still no report on the effects of Se on the vase life of other cut flowers. In this study, we explored the role of inorganic selenium, named sodium selenite (Na2SeO3), in improving the vase life of Eustoma grandiflorum cut flowers. Compared with the control (distilled water) and the other Na2SeO3 concentrations, 4.0 mg/L Na2SeO3 significantly increased the superoxide dismutase, catalase, ascorbate peroxidase glutathione reductase, glutathione peroxidase and glutathione S-transferase activities, and decreased the malondialdehyde content and the electrolyte leakage. Meanwhile, compared with the control and the other of Na2SeO3 concentrations, 4.0 mg/L Na2SeO3 significantly increased the soluble sugar, proline and soluble protein contents, relative water content, average fresh weight change rate and average water balance value. The above results imply that Na2SeO3 showed an important role in strengthening the antioxidant capacity and maintaining the water balance. Besides, 4.0 mg/L Na2SeO3 significantly increased the flower diameter and the vase life. The above findings suggested that Na2SeO3 extended the vase life and the ornamental value of E. grandiflorum cut flowers by enhancing the antioxidant capacity and water balance, which provides new knowledge for the application of Na2SeO3 in improving the fresh keeping of E. grandiflorum cut flowers. |
