Effect of housing system and age of laying hens on eggshell quality, microbial contamination, and penetration of microorganisms into eggsOriginal Paper

Hens of the laying hybrid ISA Brown were used in the study with the objective to evaluate eggshell quality, microbial contamination of eggshells, and penetration of microorganisms into the egg content in different housing systems (enriched cage: 60 hens, 10 hens per cage, 750 cm2 per hen vs free range: 60 hens, 9 hens per m2) and at different hen ages (26 vs 51 weeks) during storage time (0, 2, 7, 14, and 21 days). A significant interaction between the housing system and age was observed in egg weight and most of eggshell quality measurements. However, microbial contamination and penetration were affected mostly by the housing system and storage time. The numbers of Escherichia coli (P < 0.001, 4.51 vs 2.75 log cfu/eggshell) and Enterococcus (P < 0.001, 2.56 vs 1.11 log cfu/eggshell), and the total number of microorganisms (P < 0.001, 5.04 vs. 3.65 log cfu/eggshell) were higher in free range eggs compared to enriched cage eggs, respectively. The counts of Escherichia coli (P < 0.001, 4.23 vs 2.91 log cfu/eggshell) and Enterococcus (P < 0.001, 2.31 vs 1.27 log cfu/eggshell) decreased with storage time. A positive correlation between the total number of pores and penetration of Escherichia coli in both housing systems was observed in the albumen. It can be concluded that the housing system and age of laying hens significantly affected eggshell quality. Microbial contamination presumably affects the penetration of microorganisms. The correlation between the number of pores and penetration is assumed to be affected by the microbial species.

Examination of high-resolution feed intake data of grower finisher pigs confronted with typical short-term disturbances in stable routineOriginal Paper

Modern pig feeding systems allow the collection of highly detailed feeding data for each animal. These data enable the examination of individual feeding behaviours to assess an animal's wellbeing. As such, four different treatments ‒ undisturbed control, starving (no feed for 24 h, restrictive feeding), feed change (changes in feed composition) and social stress (exchanging of animals between the pens and short-term reduction of accessible water) ‒ were designed to simulate typical short-term disturbances in a practical stable routine. Each treatment was conducted over 2 pens with 12 animals each. Zootechnical performance and feed intake behaviour measures were assessed for each animal. Treatments did not affect zootechnical performance. Results showed that short-term disturbances did not influence feed intake behaviours, such as daily feed intake, amount of intake per feeder visit, number of daily feeder visits and daily feeding action with highest feed intake. Animals developed individual feeding patterns that persisted through artificial short-term disturbances. However, data suggested that an individual animal's behavioural pattern was strongly influenced by the group (pen) due to group dynamics among animals.

Effects of dietary energy level and guanidino acetic acid supplementation on growth performance, carcass quality and intestinal architecture of broilersOriginal Paper

Energy, known as the most expensive nutrient in broiler feed, is what strongly adjusts and affects the growth of broilers. Creatine has a key role in cellular energy metabolism and could be synthesised from guanidinoacetic acid (GAA) in the liver; however, its de novo synthesis is not able to adequately fulfil the demand of energy metabolism, especially in fast-growing modern broilers. So the aim of the study was to evaluate the efficiency of commercial GAA in energy-reduced broiler diets on performance and intestinal development. Overall, 11 400 day-old Ross 308 chicks were randomly allocated to six dietary treatments with ten replicates in each. Dietary treatments were designed as a 3 × 2 factorial arrangement with three levels of dietary metabolisable energy (AMEn) recommended by Aviagen for Ross 308 broilers (12.55 MJ/kg, 12.97 MJ/kg and 13.38 MJ/kg for starter, grower and finisher, respectively), 0.209 MJ/kg and 0.418 MJ/kg reduced and two levels of GAA (0.00% and 0.06%). There was no significant GAA × AMEn interaction for all performance parameters, carcass traits and jejunal morphological parameters (except for the villus width). Reduction of dietary AMEn (0.209 MJ/kg and/or 0.418 MJ/kg) caused a significant depression in body weight (BW) gain (P < 0.001) and feed conversion ratio (FCR) (P < 0.001). However, a decreasing AMEn level increased villus height (P < 0.003) and villus surface area (P < 0.03), while crypt depth and villus width were similar. The GAA improved final BW and FCR by 1.77% and 1.66%, respectively (P < 0.001). Birds fed low energy diets supplemented with GAA showed a significant improvement in the performance so that BW and FCR were the same as in the control birds; however, no such positive effects were obtained in jejunal villus development. Hence, it might be concluded that 0.06% GAA supplementation improves BW and FCR and can save at least 0.209 MJ/kg dietary AMEn in broiler diets.

Negative impact of heat stress on reproduction in cows: Animal husbandry and biotechnological viewpoints: A reviewReview

This review summarises current knowledge of the relationship between heat stress (HS) and reproduction in cattle. It focuses on research from the viewpoint of genetics (breed), from the viewpoint of reproduction physiology (in vivo and in vitro) and from the animal husbandry practice. From the viewpoint of animal husbandry, it was evidenced that heat stress influences reproduction before, during and after conception. Most publications suggest the negative impact of heat stress on the reproductive physiology of cows reflected in ovarian and follicular activity, in oocyte and embryo development, as well as in other processes studied under in vivo or in vitro conditions. There are also a number of products that the cell creates in response to heat stress, which is used as indicators of the stress (e.g. heat shock proteins). A number of publications also focus on how to prevent heat stress on the farm (e.g. shade, water shower) or during in vitro procedures, including the supplementation of the culture media with antioxidants like melatonin. Research of heat stress is very important in cattle breeding for preventing and reducing its effects on the farm and also in the context of climate changes and global atmospheric warming.

In situ ruminal degradability and fermentation characteristics of novel mixtures of winter cereal and Italian ryegrass plus winter cereal silagesOriginal Paper

This study was conducted using three multiparous non-lactating rumen-cannulated Holstein-Friesian dairy cows, with the objective of evaluating the in situ ruminal degradability and fermentation characteristics of novel mixtures of winter cereal and Italian ryegrass (Lolium multiflorum Lam.) plus winter cereal silages (mixture A: triticale, oats, barley and wheat; mixture B: triticale, barley and wheat; mixture C: Italian ryegrass and oats; mixture D: Italian ryegrass, oats, triticale, barley and wheat). The rumen fermentation study was conducted replacing the ensiled mixtures (experimental diets) with vetch-triticale haylage in a total mixed ration (control diet). It was found that the effective protein degradability at 0.08 rumen outflow rates was 80.6% (mixture A), 66.2% (mixture B), 79.7% (mixture C) and 79.3% (mixture D). The effective neutral detergent fibre (NDF) and acid detergent fibre (ADF) effective degradability at 0.08 rumen outflow rates was 18.0% and 17.7% (mixture A), 19.7% and 20.5% (mixture B), 19.1% and 17.0% (mixture C), and 15.2% and 14.6% (mixture D), respectively. Different dietary treatments did not change (P > 0.05) the rumen fermentation characteristics as there was no difference (P > 0.05) between control and experimental diets, and the inclusion of 40-55% Italian ryegrass (mixture C and D) did not cause any difference. These results suggest that the mixture of winter cereals and Italian ryegrass plus winter cereal-based silages had good potentially degradable dry matter, effective dry matter and effective protein degradability at 0.01, 0.05 and 0.08 rumen outflow rates without affecting the rumen environment maintaining neutral pH. The ensiled mixtures had a moderate level of potentially degradable NDF and ADF fractions.

The presence of pathogens in milk of ewes in relation to the somatic cell count and subpopulations of leukocytesOriginal Paper

Mastitis is a major health problem of the udder in dairy sheep breeds. For diagnosis of subclinical mastitis, somatic cell count (SCC) is commonly used. The presence of pathogens in the udder causes the increase of leukocytes and thus SCC in milk. Therefore, the aim of this study was to evaluate the presence of pathogens in the milk of ewes and the possible relationship with SCC. The changes of leukocytes subpopulation in milk samples with high SCC were evaluated as well. The experiment was carried out on a dairy farm with the Lacaune breed. This study was conducted on 45 ewes (98 milk samples) without signs of clinical mastitis. Based on somatic cell count, samples were divided to five SCC groups: SCC1 < 200 000 cells/ml (45 milk samples); 200 000 ≤ SCC2 < 400 000 cells/ml (10 milk samples); 400 000 ≤ SCC3 < 600 000 cells/ml (six milk samples); 600 000 ≤ SCC4 < 1 000 000 cells/ml (six milk samples); SCC5 ≥ 1 000 000 cells/ml (31 milk samples). No pathogens were observed in the majority of milk samples (60.20%). Coagulase-negative staphylococci (CNS) were the most commonly isolated pathogens from the milk of ewes (86.11%). Staphylococcus epidermidis had the highest incidence from CNS (35.48%). In the SCC5 group, up to 79.31% of bacteriological samples were positive. The percentage of leukocytes significantly increased (P < 0.001) in the samples with higher SCC (≥ 200 × 103 cells/ml) in comparison to the group SCC1. Also, the percentage of polymorphonuclear cells (PMNs) was significantly higher with increasing SCC (P < 0.001). In conclusion, the presented results showed that the high SCC was caused by the presence of the pathogen in milk. Thus SCC < 200 000 cells/ml and leukocyte subpopulation, especially PMNs, could be considered as important tools in udder health programs applied in dairy ewes.

Efficacy of poly (lactic-co-glycolic acid) microparticles as a gonadotropin-releasing hormone analogue delivery system to stimulate ovulation of peled Coregonus peledOriginal Paper

The aim of the study was to determine the efficacy of poly (lactic-co-glycolic acid) microparticles as a carrier of gonadotropin-releasing hormone analogues (GnRHa) for induction of ovulation in peled Coregonus peled. Female peled were injected intraperitoneally with 1) saline solution only (control), 2) mammalian GnRHa at 25 µg/kg body weight, 3) GnRHa in 753-type microparticles at 50 µg/kg, or 4) GnRHa in 653-type microparticles at 50 µg/kg. Blood plasma samples were taken on days 0, 4, 8, and 12 post-injection. All hormone treatments induced synchronous ovulation and higher cumulative ovulation compared to controls. Hormone treatments did not affect relative fecundity or the percentage of eyed eggs. Testosterone level decreased toward the onset of ovulation. On day eight of the trial, the testosterone level was significantly lower in hormone-treated groups compared to the control group. The level of 17β-oestradiol showed a decreasing trend post-injection, with the lowest observed level on day eight. Our results demonstrate that ovulation can be induced in the peled by the sustained - release of GnRHa in poly (lactic-co-glycolic acid) microparticles, but the treatment does not improve reproductive performance.

Intake and digestibility in cattle grazing temperate grass associated with legume and/or energetic supplementationOriginal Paper

The intercrop between grasses and legumes is an alternative to maintain and increase animal production. The study was conducted to evaluate the effect of grass-legume mixtures with or without supplementations on rumen fermentation, nutrient intake, and microbial protein synthesis. Six Holstein steers fitted with ruminal cannula were kept in a double 3 × 3 Latin square design. The treatments were: 1) oat, annual ryegrass, and supplement (GS), 2) oat, annual ryegrass, and vetch (GL), 3) oat, annual ryegrass, vetch, and supplement (GLS). Supplementation of ground maize was given daily at 11 h at 1% of body weight. Total digestible nutrient intake was higher in steers fed GS and GLS. Animals on GL ingested higher concentrations of nitrogen (N) compared to animals on GS and GLS diets. Ruminal pH and ammonia concentration were higher in GL. Grass-legume mixtures and supplements showed higher concentrations of sugar, α-amino acids, and peptides. The ruminal fermentative parameters, ruminal pH, ammonia, and sugars ranged cubically across the day. Microbial protein synthesis was similar amongst the treatments. Animals exclusively consuming temperate grass produce higher ruminal pH and ammonia concentrations. Therefore, using temperate legumes in pasture systems can be included in the cattle diet in lieu of utilizing energy supplements.

Biological response of piglets challenged with Escherichia coli F4 (K88) when fed diets containing intestinal alkaline phosphataseOriginal Paper

The aim of the study was to investigate the effect of intestinal alkaline phosphatase (IAP) added to diets on growth performance, diarrhoea incidence (DI), blood metabolites, relative organ weight, and intestinal morphometry of weaned piglets challenged with enterotoxigenic Escherichia coli F4 (K88). A total of 64 crossbred entire male piglets (25-day-old and 7.16 ± 0.28 kg body weight) were allocated into four treatments: control diet (CD-), CD- + antimicrobial growth promoter (AGP), CD- + 15 mg IAP/kg of diet and CD- + 30 mg IAP/kg of diet, with eight replications. At 15 days, all piglets were orally challenged with 6 ml of a solution containing K88 (106 colony forming units/ml). Microencapsulated IAP in acid solution showed 14.43% solubility and pH values of 1.69, 1.72, 1.51, and 1.52 at the different times measured (0.5 h, 1.0 h, 17.0 h, and 24 h); differently, IAP in basic solution had 4.10% solubility and pH values increased (5.95, 6.10, 6.32 and 6.63) according to the different times, respectively. On days 25-35, piglets that received 30 mg IAP and CD- showed a better feed conversion ratio (P = 0.075) compared to those fed 15 mg IAP. Piglets that consumed 30 mg IAP or CD- had higher (P = 0.004) average daily gain on days 35-44. On days 35-44, the piglet average daily feed intake was lower (P = 0.033) with 15 mg IAP compared to AGP. In the entire period, piglets fed 15 mg IAP showed a reduction in average daily gain (P = 0.040) and average daily feed intake (P = 0.092). Piglets on 30 mg IAP showed an improvement (P ≤ 0.05) in DI in the pre-and post-challenge periods. The relative spleen weight of the piglet increased (P = 0.043) in response to 30 mg IAP. Overall, the addition of 30 mg IAP to diets improves the growth performance, attenuates DI, and promotes an increase in spleen relative weight to maintain the healthy state of piglets.

The role of quercetin in primary culture of ovine spermatogonial stem cellsOriginal Paper

The aim of the present study was to examine the effect of quercetin on the survival and primary culture of ovine spermatogonial stem cells (SSCs). The two-time enzymatic digestion process was employed to obtain SSCs from lamb testes. In the next step, the use of filtration and differential plating methods caused an increase in the number of SSCs in the cell suspension resulting from enzymatic and mechanical digestions. Mitomycin-C-treated Sertoli cells were used to prepare the feeder layer. The stem cells were then cultured on the Sertoli cell feeder layer. The identification of the colonies was done through alkaline phosphatase staining methods and specific gene expression of ram's SSCs (nanog and Plzf). The results of methylthiazolyldiphenyl-tetrazolium bromide assay on SSCs 72 h after culture with different treatments of quercetin demonstrated that the highest percentage of survival was for 5 μM and 10 μM concentrations, respectively; however, compared to the control, no significant difference was observed. In comparison with the control, the concentration equal to and greater than 20 μM quercetin caused a significant decrease in the survival of SSCs (P < 0.05). Seven days after culture, 40 μM quercetin caused a substantial reduction in the mean number of colonies, compared to the control (P < 0.05). The results demonstrated that compared to the control, 5 μM to 40 μM of quercetin significantly reduced Plzf gene expression. Furthermore, the concentration equal to and higher than 10 μM quercetin significantly decreased bcl-2 gene expression in the cells under study (P < 0.05). Based on the findings of the present study, the use of quercetin for the primary culture of ovine SSCs is not recommended. It is suggested that the function of this antioxidant should be investigated on the differentiation of SSCs.

Effect of density and mixed culture of largemouth bass (Micropterus salmoides) and pikeperch (Sander lucioperca) on growth, survival and feed conversion rate in intensive cultureOriginal Paper

In this study, two experiments were performed with the aim to optimize intensive aquaculture of largemouth bass (Micropterus salmoides). In the first 140-day experiment the effect of the initial fish density was assessed at three levels: low density (LD) 23 kg/m3, medium density (MD) 35 kg/m3 and high density (HD) 46 kg/m3. All three densities provided the same final Fulton's condition coefficient (FC = 1.24-1.28), specific growth rate (SGR = 0.22-0.24%/day) and survival rate (97-100%). No cannibalism was observed at all tested densities. Feed conversion ratio (FCR = 1.39 ± 0.21 g/g) was the lowest for LD and the highest (1.61 ± 0.08 g/g) for MD. The highest fish biomass (25.7 ± 2.7 kg/m3) was obtained at HD and this density was considered as the most effective density of all tested ones during the intensive culture of largemouth bass. The second 60-day experiment tested the effect of largemouth bass and pikeperch (Sander lucioperca) monoculture and biculture of both species on production efficiency. Higher size heterogeneity was obtained in both (mono- and bicultural) groups of pikeperch (308.91-314.56‰/day) compared to the groups of largemouth bass (279.26-284.05 ‰/day). The higher FC (1.09) was found in both types of culture in largemouth bass compared to both methods of culture in pikeperch (0.74-0.78). The lowest SGR was evident in both types of largemouth bass cultures (1.20-1.28%/day). In contrast, the highest SGR was achieved in the bicultural pikeperch (1.88%/day). Similar results like for SGR were also assessed for FCR, where the highest value of FCR was in both cultures of largemouth bass (1.44-1.48 g/g) compared to the lowest FCR in the bicultural of pikeperch (0.73 g/g). Largemouth bass in both tested types of culture had higher survival rates (99.95-99.99%) compared to pikeperch (98.61-98.63%).

Modelling the growth of rearing cattleOriginal Paper

Scientific growth literature often uses the models of Brody, Gompertz, Verhulst, and von Bertalanffy. The versatile five-parameter Bertalanffy-Pütter (BP) model generalizes them. Using the least-squares method, we fitted the BP model to mass-at-age data of 161 calves, cows, bulls, and oxen of cattle breeds that are common in Austria and Southern Germany. We used three measures to assess the goodness of fit: R-squared, normalized root-mean squared error, and the Akaike information criterion together with a correction for sample size. Although the BP model improved the fit of the linear growth model considerably in terms of R-squared, the better fit did not, in general, justify the use of its additional parameters, because most of the data had a non-sigmoidal character. In terms of the Akaike criterion, we could identify only a small core of data (15%) where sigmoidal models were indispensable.

Molecular cloning, functional characterization, tissue expression and polymorphism analysis of buffalo PRDX6 geneOriginal Paper

PRDX6 is a bifunctional protein involved in antioxidant regulation and phospholipid metabolism. Previous studies have shown that PRDX6 is involved in some biological pathways and networks related to lactation. The aim of this study was to explore the characteristics, function, tissue expression and variation of buffalo PRDX6 gene. We cloned and characterized the complete coding sequence (CDS) of buffalo PRDX6. The CDS of PRDX6 for swamp and river buffalo is the same, which consists of 675 nucleotides and encodes a protein of 224 amino acids. Buffalo PRDX6 contains one PRX_1cys functional domain (AA 7-222), which is probably related to the regulation of oxidative stress. Multi-tissue differential expression analysis showed that buffalo PRDX6 was highly expressed in the muscle, brain, lung and small intestine during non-lactation and lactation, and there were significant differences in expression in all the tissues except the small intestine between the two periods. It is worth noting that the mRNA abundance of buffalo PRDX6 in non-lactating mammary gland is higher than that in lactating mammary gland. Among the two single nucleotide polymorphisms (SNPs) identified in the CDS in this study, c.261C>T is shared by the two types of buffalo with different allelic frequencies, and c.426T>G is found only in river buffalo. The c.426T>G is non-synonymous, resulting in the amino acid substitution p.Asn142Lys. Only one nucleotide differential site is identified in PRDX6 gene between buffalo and other species of Bovidae. Phylogenetic analysis indicated that buffalo PRDX6 has a closer genetic relationship with that of the species in Bovidae. These results indicate that PRDX6 probably plays a crucial role in the mammary gland of buffalo. This study provides the foundation for further functional studies of PRDX6 in buffalo.

Comparison of early-life jejunal microbiota diversity in two pig breedsOriginal Paper

The pig intestinal microbiota perform multiple physiological functions during the lifespan of the host. Host genetics is considered a major factor shaping the intestinal microbiota. This study explored the jejunal microbial diversity and potential functional differences between Dahe (DH) and Diannan small-ear (DS) pigs, two important native Chinese breeds with different growth characteristics. Nine piglets of each breed (DH and DS) were fed sow milk until the age of 35 days. Jejunal chyme samples were collected for 16S rRNA sequencing. The birth weight, final body weight at day 35, and average daily gain tended to increase more in DH piglets than in DS piglets. The jejunal microbial Shannon index was also higher in DH piglets than in DS piglets (P < 0.05). At the genus level, the relative abundances of Clostridium XI, Clostridium sensu stricto, Turicibacter, Megasphaera, Veillonella, Mitsuokella, and Selenomonas in DH pigs were higher than those in DS individuals, whereas Streptophyta, Enterococcus, Lactococcus, and Weissella were lower in DH pigs than in DS pigs (P < 0.05). Furthermore, linear discriminant effect size analyses revealed 25 differential bacterial taxa between DH and DS piglets. Spearman's analysis found that Enterococcus was negatively correlated with final body weight (P = 0.025, r = -0.56) and average daily gain (P = 0.034, r = -0.53), while average daily gain was positively correlated with Clostridium XI (P = 0.01, r = 0.63) and Mitsuokella (P =0.007, r = 0.64). Kyoto Encyclopedia of Genes and Genomes annotation identified eight functional categories related to amino acids and energy metabolism in DH piglets, while three categories were related to lipid metabolism in DS piglets. Our findings suggest that DH pigs have higher microbial diversity, while DS pigs may have higher fat deposition ability.

Changes in milk ketone and fatty acid concentrations during early lactation in Holstein and Fleckvieh cowsOriginal Paper

The aim of this study was to compare the changes in milk composition postpartum, especially ketones and milk fatty acids (FAs), in Holstein and Fleckvieh cows kept under identical management conditions. Milk composite samples were collected from 66 cows during afternoon milking, at weekly intervals from one to eight weeks postpartum, and their components were determined by Fourier transform infrared spectroscopy. The Holstein cows had higher (P < 0.05) concentrations of long-chain FAs (ranging from 6% to 16% in different weeks), monounsaturated FAs (6% to 12%), and C18:1 (5% to 16%), as well as lower (P < 0.05) concentrations of saturated FAs (3% to 8%) and short-chain FAs (7% to 17%) in their milk than the Fleckvieh cows for almost the entire monitored period. These differences can be explained by pronounced lipomobilization, due to a negative energy balance, when mainly long-chain FAs from adipose tissue are incorporated into milk and significantly inhibit the de novo synthesis of FAs in the mammary gland. In conclusion, it can be assumed that breed-related metabolic changes during the first weeks of lactation have a large effect on the milk FA composition. This reflection of the metabolic load changes and lipomobilization in differing milk FA profiles would allow for the use of selected milk FAs to detect energy imbalances and their associated diseases in early lactation cows.

Fleckscore system of exterior evaluation as a more accurate indirect predictor of longevity in Slovak Simmental dairy cowsOriginal Paper

The goal of the work was to discover the influence of conformation traits evaluated by the Fleckscore system on the length of productive life of Slovak Simmental dairy cows. Evaluation of body conformation traits according to the Fleckscore system was performed on 3 452 cows. The relationship of individual traits to longevity was analysed using the Weibull proportional hazard model. The results confirmed that smaller and longer cows, which were well muscular and with a deeper body, had a lower risk of early culling than cows in the reference group. Hock angularity, pastern and hoof height scored with low or high marks were associated with an early culling of cows. In terms of the length of productive life, a slightly more elastic pattern is required. Cows with a deeper udder and a stronger and tighter fore udder attachment, which were also average in notching and height central ligament and centrally placed teats, reached a longer productive life.

Copy number variation (CNV) of the AMY gene in humans has been enthusiastically studied for its association with starch digestibility and obesity. The alpha-amylase (AMY) is a major starch digestive enzyme in mammals. This study aimed to determine the association between CNV of the porcine pancreatic amylase (AMY2B) gene and feed efficiency. Improvement of feed efficiency in growing pigs is of great economic interest. We assayed the copy number of AMY2B by using real-time quantitative PCR (qPCR) in a Large White pig population. We identified three genotypes for AMY2B CNVs, namely I/I (homozygotes of haplotype I; a chromosome with one copy of AMY2B), I/II (heterozygotes of haplotype I and II; a chromosome with two copies) and II/II (homozygotes of haplotype II). We tested the genotypes of the parental generation consisting of six males, 21 females and 265 offspring piglets to validate the AMY2B CNV genotyping. With very few mistyping exceptions, copy numbers of AMY2B were transmitted to piglets in segregation ratios following Mendelian inheritance. Finally, we performed an association analysis between the CNV of the AMY2B gene and feed efficiency traits in 207 uncastrated male pigs. The generalised linear model analysis showed the significant effects of AMY2B CNV genotype on average daily feed intake, total feed intake and feed conversion ratio during growth from 30 kg to 100 kg body weight. However, it was not associated with average daily gain, backfat thickness and loin eye muscle area. Individuals with the genotype I/I had about 76.6 ± 27.1 g lower average daily feed intake, 5.35 ± 1.90 kg lower total feed intake and 0.089 8 ± 0.026 5 lower feed conversion ratio than individuals with I/II and II/II genotypes. Thus, AMY2B CNV has the potential to be an effective genetic marker that could reduce feed costs for pig farming.

Effects of folic acid supplementation to basal diets of broilers on growth performance, slaughter performance, IGF2 gene expression and methylationOriginal Paper

Folic acid (FA) is an important water-soluble vitamin and plays an important role as a cofactor and coenzyme in animal growth and development, and regulation of gene expression and methylation. A total of 270 female broiler chickens (1-day-old) were randomly allotted to three dietary treatments supplemented with 0 mg/kg (control group), 5 mg/kg, and 10 mg/kg FA in basal diets for 42 days, respectively. Each treatment had six replicate cages with 15 birds per cage. Dietary supplementation of 5 mg/kg FA significantly enhanced average body weight and average daily gain of 21-day-old broilers (P < 0.05), but significantly reduced subcutaneous fat thickness and widths of an intermuscular fat band of 42-day-old broilers by dietary FA treatments (P < 0.05). Also, a diet with 10 mg/kg FA supplementation significantly increased the relative heart weight of 42-day-old chickens (P < 0.05). Furthermore, dietary FA supplementation significantly improved the serum insulin-like growth factor 2 (IGF2) concentrations (P < 0.01) and IGF2 mRNA expression in the abdominal fat (P < 0.05), but no statistical differences were found in the methylation of IGF2 promoter (P > 0.05). The present study demonstrated that dietary FA supplementation may have positive effects on chicken growth through increased IGF2 gene expression.

Prevention of erosion in mountain basins: A spatial-based tool to support payments for forest ecosystem servicesOriginal Paper

This paper presents a spatial-based decision support system (DSS) to assist public and private forest managers in the analysis of potential feasibility in payments for forest ecosystem services (PES) for the prevention of soil erosion. The model quantifies the maximum willingness to pay (WTP) of managers of a reservoir to prevent soil loss. The minimum willingness to accept (WTA) of forest owners for the activation of a private market is also computed. The comparison of WTP and WTA identifies the forest area where PES are ideally feasible with additional potential for compensation to enable the schemes. The DSS highlights forest idiosyncrasies as well as local socio-economic and geomorphological characteristics influencing PES suitability at a geographic level. The potential applications and future improvements of the model are also discussed.

Methodological approaches to the valuation of forest ecosystem services: An overview of recent international research trendsReview

Forests represent the most important source of ecosystem services (ES) on a global level both for the production of goods and for the provision of services and externalities, nevertheless scientific research in the economic field is lacking. Currently the number of documents relating to ES is 16 673, of which only 1 379 concern the forestry sector. The aim of this study is to provide an overview of scientific research trends in the field of economic evaluation of forest ecosystem services (FES). To this end, an on-line bibliographic survey was carried out on the main scientific search engines, which made it possible to quantify the works and at the same time to detect the main evaluation methods used for the different FES. This survey allowed to collect 93 articles meeting the search criteria: the most active continents were Europe and Asia, whereas most of the articles focused on the joint evaluation of provisioning, regulation and cultural services, even if a good number of them only concerned cultural services. The most widely used valuation methodologies were the contingent valuation among the stated preference techniques and the market price among direct observation criteria.

Exploring the variability in elastic properties of roots in Alpine tree speciesOriginal Paper

Quantifying the soil reinforcement provided by roots is essential for assessing the contribution of forests to reducing shallow landslide susceptibility. Many soil-root models were developed in the literature: from standard single root model to fibre bundle model. The input parameters of all models are the geometry of roots (diameter and length) and the biomechanical properties (maximum tensile force and elastic modulus). This study aims to investigate the elastic properties estimated by the stress-strain curves measured during tensile tests. A standard procedure detected two different moduli of elasticity: one due to the root tortuosity, and the other due to the woody fibres of roots. Based on a large dataset of tensile tests on different Alpine tree species, the relationships between elastic modulus and root diameter was estimated for each series. Further, the interspecific and intraspecific variability in such relationships was investigated by a statistical analysis. The results showed more intraspecific differences in the elastic modulus vs. root diameter relationships compared to the interspecific ones. This outcome could be an important criterion of discrimination to explain the variability of the elastic properties and to provide representative biomechanical properties for specific environmental conditions.

Soil aggregate stability index and particulate organic matter in response to differently afforested lands in the temperate regions of IranOriginal Paper

Aggregate Stability Index (ASI) and particulate organic matter (POM) are strongly influenced by land use and management. This work illustrates the effects of plantations on ASI and POM-C and POM-N in northern Iran. Three plantations of P. deltoides (PD), T. distichum (TD), A. subcordata (AS), and a fourth site ‒ adjacent abandoned lands (BL, as control) were selected. Soil samples were taken within 16 quadrats of each plantation and BL from the two depths of 0-15 cm and 15-30 cm during the summer. Soil C was significantly higher under TD (2.10%) than under BL (2.02%) > PD (1.61%) > AS (1.30%). Soil N was found in ranked order of AS (8.99%) > TD (7.82%) > PD (5.30%) > BL (3.68%) (P < 0.019). The significantly higher ASI was found under TD (57.49) in comparison with PD (53.10), BL (51.23), and AS (36.57). The POM-C was as follows: TD (0.209%) > PD (0.141%) > AS (0.139%) > BL (0.075%) (P = 0.020). The highest POM-N was found under TD (0.035), followed by AS (0.0284%), PD (0.0288%), and BL (0.007%). The results indicate the positive effect of afforestation on soil ASI and POM-C and POM-N, especially in the surface layers of soil.

The influence of growing medium composition on pine and birch seedling response during the period of simulated spring droughtOriginal Paper

Climate change increases the earliness and effect of spring dry spells. The efforts to reduce their negative effects on tree regeneration include improvement in physical properties of soil aimed at an increase in water retention and availability. Clinoptilolite from the group of zeolites belongs among such water-absorbent natural materials. The article aims to assess the effect of clinoptilolite admixture in a growing medium, along with different fertilizer concentration, on the growth of pine and birch seedlings and their development during simulated drought. A common (reference) growing medium was tested along with 4 treatments of the growing medium with clinoptilolite. The birch responded to the fertilizer concentration more intensively than to the clinoptilolite admixture and was more vulnerable to drought damage due to a closer dependence of physiological responses on a decrease in the growing medium moisture. The onset of pine withering was slowest in the reference growing medium. A positive effect of clinoptilolite addition on the growth of pine seedlings was confirmed but its unambiguously positive effect on overcoming of spring dry spells was not demonstrated. Clinoptilolite addition with higher fertilizer rate was found as fully unsuitable.

Identification and genetic diversity of Acer ibericum (Aceraceae) in South CaucasusOriginal Paper

Internal transcribed spacer (ITS) sequence was tested in Acer ibericum for its suitability as a barcoding as well as population genetic marker. The marker was reliably used to identify A. ibericum as well as to gain preliminary insights into the genetic diversity of Georgian populations. MegaBLAST searches of studied samples against the GenBank database revealed that all studied accessions could be considered Acer ibericum. Populations from Vashlovani National Park revealed the presence of unique ribotypes, and together with the population from Shavi Mountain, they were identified as the most genetically diverse ones. Based on presented data the populations from Eastern Georgia should be prioritized if conservation measures are planned.

Woodland key habitat contribution to preserve biological diversity in Lithuania: assessing the difference between 2005 and 2017Original Paper

In response to the degradation of forest ecosystems, their habitats and the loss of species, many formal conservation policies and voluntary forest conservation tools have been proposed and implemented. The woodland key habitat (WKH) is one such initiative that aims to protect biodiversity. This generally involves two key actions: (i) the creation of policy (conservation action) and (ii) the consequences of the policy or initiative in the field. However, the final step of measuring their success in the field is often missing. The aim of this study is to assess the contribution of the WKH initiative to conserve biodiversity in Lithuania. We compared the changes in spatial distribution, species assemblages and richness within the WKH network between 2005 and 2017. Results showed that the spatial distribution of WKHs decreased in number and by area after 12 years. However, species occurrence, abundance and richness of the WHK network generally increased. In conclusion, we found the WKH initiative has contributed to the conservation of forest habitats and biodiversity in Lithuania. However, the future of the WKH network is uncertain due to the current voluntary system, lack of support and funding.

In Turkey, almost half of which has semi-arid conditions, steppe areas have continuously been increasing. These areas need to be afforested to prevent desertification. For this purpose, this research presents specifically the Anatolian black pine (Pinus nigra J.F. Arnold subsp. pallasiana /Lamb./ Holmboe) as a suitable forest tree species for afforestation activities, due to wide distribution both in Turkey and the broader region. After a thorough investigation of past and recent literature, and onsite activities, the review focuses on the production of seeds and seedlings of the species, land preparation and planting technique, post-planting site maintenance, success in the afforestation areas and considerations on Anatolian black pine seed transfer regioning. In conclusion, local ecological conditions regarding the species should always be taken into consideration. Another important issue for the success of afforestation in semi-arid lands is that the preparation activities of the land for afforestation are carried out with appropriate techniques timely and painstakingly. Moreover, after the 2000s, Turkish forestry focused on afforestation in semi-arid and anthropogenic steppe areas. Anatolian black pine is the most widely used tree species in semi-arid lands and anthropogenic areas and plays the major role in successful afforestation.

Stand structure and growth of Robinia pseudoacacia 'Jászkiséri' - 'Jászkiséri' black locustShort Communication

Black locust (Robinia pseudoacacia L.) is one of the most widespread tree species introduced into Europe and also into Hungary. It covers 24% of the total Hungarian forest area, providing 25% of the annual timber output of the country. Due to the demands of consumers, new cultivars are to be produced by means of improvement techniques and are to be introduced into the practical forestry use. Mono- and multiclonal cultivars were developed, then variety comparison trials and cultivation tests were established. Based on preliminary yield tests the locust cultivar 'Jászkiséri' (Robinia pseudoacacia 'Jászkiséri') proved to be one of the best black locust cultivars. Consequently, a more precise investigation of the stand structure of this cultivar may also make a significant contribution to the improvement of the relevant cultivation technology. In this study, based on full inventories of 13 stands in 7 subcompartments, age of 5 to 35 years, relationships of mean tree volume to diameter (R2 = 0.9797) and basal area (R2 = 0.9781), furthermore the relationship between mean tree volume and diameter of the stands (R2 = 0.9993) were examined. Besides that, the comparison of 15-year-old 'Jászkiséri' and common black locust (a case study) were presented in this paper, where 'Jászkiséri' proved to be better: significant differences (P < 0.05) were found in diameter, mean tree volume and stem form.

The impact of agricultural land afforestation on soil water content in Central BohemiaOriginal Paper

In the Czech Republic, the afforestation of agricultural land has been supported by providing subsidies from the government and the European Union. Afforestation of less-productive agricultural land provides many benefits including carbon sequestration, soil erosion control, biodiversity, water retention, cooling, social benefits, decreasing noise and light pollution, increasing air quality, wind speed reduction, oxygen production, wood production and non-wood products. In some aspects, it is possible to produce wood of the same quality on former agricultural land compared to permanent forest land. In this study, we attempted to find out the course of temperatures and volumetric water content as well as some other physical soil properties (at depths of 20, 40 and 60 cm) 9 years after the afforestation of agricultural land (warm, mild dry region of the Czech Republic) with a mixture of broadleaved tree species (Quercus robur L., Quercus rubra L. and Acer platanoides L.) or monospecific Pinus sylvestris L. stand; the study was performed in the period from April to the beginning of November 2020. Concerning the studied physical soil properties, the value of bulk density was higher (and total porosity lower) at a depth of 20 cm in Pinus sylvestris L. compared with agricultural land or the mixture of broadleaves; the water stability of soil aggregates was higher after the afforestation with the mixture of broadleaves. The temperature was lower in the soil of afforested plots (at all studied depths) compared to the agriculturally used land. Differences in rainfall interception, transpiration, soil(and forest floor) properties and other factors could influence the obtained values of water content in the soil of the studied plots. The average volumetric water contents were the highest in the plots with Scots pine (depth of 20 cm) and broadleaves (depth of 40 cm), and on the control plot (depth of 60 cm). The volumetric water content at a soil depth of 20 cm was not significantly (P > 0.05) different when the plot with Scots pine and agriculturally used land were compared. In all other cases and depths, the differences between plots were significant (P < 0.05).

Norway maple (Acer platanoides L.) is a key species of broadleaved forests whose population genetics is poorly studied using modern genetic tools. We used ISSR analysis to explore genetic diversity and differentiation among 10 Russian populations on the eastern margin of the species range of distribution, and to compare the revealed patterns with the results of our population genetic studies of pedunculate oak (Quercus robur L.). In the first set comparatively high heterozygosity and allelic diversity were found (expected heterozygosity HE = 0.160 ± 0.033, number of alleles na = 1.440 ± 0.080, effective number of alleles ne = 1.271 ± 0.062) in comparison with strongly fragmented and geographically isolated small maple stands of the second set (HE = 0.083 ± 0.011, na = 1.281 ± 0.031, ne = 1.136 ± 0.019). A relatively high genetic differentiation among populations was detected (the proportion of the inter-population component of total genetic variation, GST = 0.558 ± 0.038). In the Cis-Urals, local groups of populations that are confined to the northern, middle and southern parts of the Urals were identified. On the contrary, the current significant fragmentation of the pedunculate oak distribution area in the same study area did not lead to any noticeable genetic differentiation among the majority of populations, the values of the population genetic diversity were very similar in different parts of the Southern Urals.

Comparison of Scots pine growth dynamics in Polissya and Steppe zone of UkraineOriginal Paper

The goal of the study was to compare the dynamic changes in individual biometric indices of Scots pine in different natural zones of Ukraine, Polissya and Steppe. Scots pine stands were mainly concentrated in the Polissya zone, and their area was 3.6 times larger than that of forests in the Steppe zone, and the total wood stock by 4.6 times. The regression equations for biometric indices of artificial modal stands were developed. A comparison of the average height, diameter, and wood stock for pine stands of site index Іа, I, and II growing in Polissya and Steppe was made. It was found that the average differences in the average height (from 1.9 to 2.6 m) are observed at the age of 15-20 years. For the average diameter, the difference increases with age and the growth of the site index. The difference in the average stock is constantly increasing with age. At the age of 20 years this difference is 20-30 m3, and at the age of 120 years from 100 to 120 m3 depending on the site index. The developed growth models can be used in planning and prioritizing appropriate forestry activities for pine stands growing in specific regional conditions.
