Effects of cholecystokinin-octapeptide and cerulein on small-intestinal motility in sheepK.W. RomańskiCzech J. Anim. Sci., 2010, 55(8):321-329 | DOI: 10.17221/1692-CJAS Cholecystokinin (CCK) affects the intestinal motility but in ruminants the question has not been entirely explored. The aim of this study was to examine the precise effects of CCK-octapeptide (CCK-OP)and its amphibian analogue, cerulein, on duodenal motor activity in unfasted rams in the course of chronic experiments. Five rams underwent the implantation of a strain gauge force transducer to the duodenal wall, and - additionally - the bipolar platinum electrodes to the duodenal bulb, distal duodenum, near the strain gauge force transducer, and proximal jejunum. During continuous motor recordings, 0.15M NaCl or CCK peptides were administrated intravenously. Injections of CCK-OP at doses of 20 (over 30 s), 200 (over 30 or 60 s), and 2 000 (over 30, 60, or 120 s) ng/kg of body weight and injections of cerulein at doses of 1, 10, or 100 ng/kg (given over the same periods) were each administered in the course of duodenal phase 1, 2a, or 2b of the migrating motor complex (MMC), i.e. 5 min after the onset of each phase. Injections of the smallest doses of CCK peptides exerted a slight and mostly insignificant effect on the duodenal areas under contraction (AUC). In the duodenum, the moderate doses of the hormones evoked short stimulatory effects followed by longer inhibitory biphasic effects on AUC. These effects were inversely related to the duration of the hormone injection. It is concluded that CCK evokes stimulatory and inhibitory (biphasic) physiological effects on duodenal motility in sheep. |
Microbiological quality of marketed fresh and frozen seafood caught off the Adriatic coast of CroatiaN. Topic Popovic, A. Benussi Skukan, P. Dzidara, R. Coz-Rakovac, I. Strunjak-Perovic, L. Brlek-GorskiVet Med - Czech, 2010, 55(5):233-241 | DOI: 10.17221/2997-VETMED Fresh and frozen seafood products (fish, shellfish, crustaceans, molluscs) in wide use in Croatia and typical of the Mediterranean diet, were examined for the presence of microbiological contamination through the winter and summer seasons. Total bacterial counts of aerobic mesophilic bacteria (AB), aerobic psychrophilic bacteria (AP), Salmonella spp., Enterobacteriaceae, Escherichia coli, Staphylococcus aureus, sulphite-reducing clostridia (SRC), Listeria monocytogenes, Vibrio cholerae and V. parahaemolyticus were measured. The microbiological quality of individual samples varied widely between animal species and also between winter/summer seasons regarding total counts of aerobic mesophilic and psychrophilic bacteria. The poorest quality was for (both summer and winter) fish samples, where 66.6 % of fresh and frozen fish were found unacceptable by Croatian standards. The overall prevalence of V. parahaemolyticus was 5%. Its recovery rate was higher in fresh/frozen shellfish in both seasons than in other specimens or other storage/season conditions. Fresh crustaceans sampled in winter demonstrated significantly higher aerobic mesophilic counts than frozen ones. Unacceptable Enterobacteriaceae levels were obtained in 40% of the fresh fish summer samples. The results of this survey constitute an indicator of bacteriological contamination of a variety of seafood. The findings could serve as a basis for future testing of seafood, and possibly as a template for developing a regional/Mediterranean testing scheme on the microbial contamination of seafood in order to establish data with comparative epidemiological and statistical values. |
Mycobacterium avium subsp. paratuberculosis in a mouflon herd without clinical symptoms monitored using IS900 real-time PCR: a case reportR. Pribylova, I. Slana, J. Lamka, V. Babak, K. Hruska, I. PavlikVet Med - Czech, 2010, 55(12):625-630 | DOI: 10.17221/2943-VETMED The aim of this study was to monitor over two years a farmed mouflon herd for the presence and persistence of Mycobacterium avium subsp. paratuberculosis (MAP) using an IS900 real-time PCR method. This study followed the previous monitoring of the herd using a cultivation method which showed only a minimal infection load among the animals. Although no mouflon showed clinical symptoms, 35.7% and 80% of ewes were IS900-positive in 2008 and 2009, respectively. In seven out of 21 adult ewes, the presence of the IS900 sequence was determined in 2008 as well as in 2009. Between the first and second sampling, twenty-three mouflon lambs born and kept with the ewes were examined. Almost one third of them (30.4%) were proven to have the MAP sequence in their faeces. Also, 75% environmental samples from the mouflon farm showed positivity. Infected animals without clinical symptoms which low sensitivity cultivation does not detect represent a source of infection for other animals. Therefore, real-time PCR has a crucial role in paratuberculosis control programs, especially in control of the disease by the culling of infected animals. |
Characteristics of coarse woody debris in successional stages of natural beech (Fagus orientalis) forests of Northern IranK. Sefidi, M. R. Marvie MohadjerJ. For. Sci., 2010, 56(1):7-17 | DOI: 10.17221/113/2008-JFS Coarse woody debris (CWD) is an important structural and functional component in forests in Northern Iran. In this study we determine the temporal patterns of CWD in Kheyroud Forests by examining the CWD volume in different decay classes and size classes along a chronosequence of secondary forest succession. The volume of CWD followed the general "U-shaped" temporal trend: the highest in the late successional forest (51.25 m3.ha-1), lowest in the middle successional forest (25.95 m3.ha-1) and intermediate in the early successional forest (37.05 m3.ha-1). The late successional forest had a larger amount of logs, snags and stumps than the other two forests. In contrast, the snag volume did not differ between the late and middle successional forest. CWD in decay classes III and V was greater in the late successional forest than that in the other two forests, while CWD in decay classes II and I did not differ among the three successional forests. CWD in class II and I was significantly higher in the early successional forest than that in the middle successional forest. In the early and middle successional forests, CWD in early decay class was dominated by Carpinus betulus L. followed by Fagus orientalis Lipsky. In the late successional forest, CWD in early decay class was dominated by Fagus orientalis while CWD in the late decay class was dominated by Carpinus betulus. While forest succession had a large influence on the amount of CWD in different decay classes, it had no effect on CWD distribution among the different size classes. Our results suggest that both anthropogenic and natural disturbances have had a long-term effect on the distribution of CWD among three forests. |
Effect of salinity stress on mycorrhizal association and growth response of peanut infected by Glomus mosseaeA.S. Al-KhalielPlant Soil Environ., 2010, 56(7):318-324 | DOI: 10.17221/204/2009-PSE Arbuscular mycorrhiza is a mutualistic association between fungi and higher plants, and play a critical role in nutrient cycling and stress tolerance. However, much less is known about the mycorrhiza-mediated enhancement in growth and salinity tolerance of the peanuts (Arachis hypogaea L.) growing in the arid and semi-arid areas. Therefore, mycorrhizal status of Glomus mosseae in diverse salinity levels on original substrate soil conditions was investigated. Different growth parameters, accumulation of proline content and salt stress tolerance were studied. These investigations indicated that the arbuscular mycorrhizal fungi could improve growth of peanuts under salinity through enhanced nutrient absorption and photosynthesis. Chlorophyll content and leaf water content were increased significantly under salinity stress by the inoculation with mycorrhizal fungi. Tolerance of the plants to salinity was increased and the mycorrhizal association was found highly effective in enhancing peanut growth and establishment in soils under salinity and deficient in phosphorus. |

The botanical compositions were determined at the sites listed in NATURA 2000 (Donovaly, Tajov, and Priechod). The respective site coordinates were determined using a Global Position System, and then put on the digital map of the Google server. A range of rare, endangered, and protected plant species were found at the monitored sites (e.g. Gymnadenia densiflora, Lilium bulbiferum, Lilium matragon, and Stipa joannis). |

SenVet Med - Czech, 2010, 55(9):463-471 | DOI: 10.17221/2982-VETMED Organic anthropogenic compounds such as polycyclic aromatic hydrocarbons, polychlorinated biphenyls, nitroaromatics, dioxins, various pesticides and natural compounds can enter the aquatic environment. These lipophilic compounds are readily taken up into the tissues of aquatic organisms where biotransformation via Phase I and Phase II metabolism can in part, determine the fate and toxicity of the xenobiotics. In this study on gilthead seabream (Sparus aurata), liver microsomal ethoxyresorufin O-deethylase (EROD), methoxyresorufin O-demethylase (MROD), penthyloxyresorufin O-depenthylase (PROD), benzyloxyresorufin O-debenzylase (BROD), aniline 4-hydroxylase (A4H), N-nitrosodimethylamine N-demethylase (NDMA-DE), aminopyrine N-demethylase (APND), caffeine N-demethylase (CN3D) and erythromycin N-demethylase (ERND) were determined. A4H, ERND, NDMA-ND, EROD and PROD activities increased in gilthead seabream (Sparus aurata) liver microsomes of different ages, while CN3D and APND activities decreased. MROD activities were barely detectable in mature fish whereas BROD activities were not detectable at all ages studied. Results were also recorded at the protein level by Western blotting using anti-CYP1A, CYP2E and CYP3A antibodies. In conclusion, elevated levels of gilthead seabream liver microsomal CYP450 enzyme activities might reveal possible exposure to various exogenous compounds, which might affect the desired responses to drugs, hormones and dietary supplements used during breeding. |
Relationship between carcass weight, skatole level and sensory assessment in fat of different boarsNenad Parunović, Milica Petrović, Vesna Matekalo-Sverak, Jasmina Parunović, Čedomir RadovićCzech J. Food Sci., 2010, 28(6):520-530 | DOI: 10.17221/243/2009-CJFS The purpose of this study was to investigate the relationship between the carcass weight and the level of skatole in boar back fat samples with descriptive sensory profiles (trained sensory panel) immediately after heating the fat samples (warm). A weak correlation was found between the carcass weight and skatole level in fat (P > 0.05). Between skatole levels in the fat of boars, whose carcass weight was below 70 kg, and of those with the carcass weight equal or above 70 kg, there was a statistically significant difference (P < 0.05). The average content of skatole in the fat tissue of the boars < 70 kg, (0.18 ± 0.09 mg/kg fat, respectively) was below the commonly used respective thresholds for tainted meat (0.20 mg/kg fat), 53% of the samples showed the values of ≤ 20 mg/kg, and 73% of the samples the values of ≤ 25 mg/kg. In the group ≥ 70 kg (0.40 ± 0.39 mg/kg fat, respectively), 80% of the samples revealed the values of ≥ 20 mg/kg, and 66% of the samples the values of ≥ 25 mg/kg. Our results show that a positive, compelling and statistically highly significant correlation exists between the skatole level and the sensory assessment of skatole intensity in fat. |

Landraces and obsolete cultivars of minor wheat species in the czech collection of wheat genetic resourcesZ. Stehno, J. Bradová, L. Dotlačil, P. KonvalinaCzech J. Genet. Plant Breed., 2010, 46(10):S100-S105 | DOI: 10.17221/2664-CJGPB The proportions of landraces in the Czech collection of wheat genetic resources significantly differentiates among wheat species, 4.2% in bread, 77.6% in emmer, and 80.0% in the einkorn wheat collections. A set of 10 selected emmer wheat landraces has been characterized by high molecular weight glutenin subunits (HMW-GSs). They were evaluated for 3 years in field trials, and described by grain quality parameters. Emmer wheat accessions differ considerably in the polymorphisms of HMW-GSs. Out of the total of 10 studied emmer wheat landraces, 5 accessions appeared to be homogeneous in the electrophoretic patterns of HMW-GSs; they were formed by a single glutenin line. Much higher crude protein content was detected in all of the emmer wheat accessions, in comparison with the control bread wheat cultivar. The proportion of this important component varied between 15.5% and 22.2%. On the other hand, SDS sedimentation, an important parameter of bread making quality, was very low (1.2-4.4 ml); and a similar situation has been recorded in the gluten index. Based on such results, the emmer wheat landraces can be considered potentially more suitable for other purposes than for the preparation of bread (e.g. for different grain mixtures, purée, etc.). |

Sci., 2010, 55(10):428-435 | DOI: 10.17221/1698-CJAS The aim of the study was to analyze the pattern of oviposition time in laying sequences in broiler breeder hens and to determine a relationship between egg position in the sequence and egg quality. The sequences were described using mean oviposition time (hour) within a sequence, mean lag of oviposition time between successive ovipositions, and mean and cumulative lag of oviposition for a sequence. Egg weight, percentage of egg components and shape index were determined for successive eggs in a sequence. The 2-, 3-, 4-, 5- to 6- and 7- to 9-egg sequences were considered. The light/dark regime was 16 h/8 h (05:00 a.m. to 09:00 p.m.). Hens laid the first egg in a sequence about 3.5 h after the beginning of the photoperiod. With increasing sequence length, the first egg was laid sooner after the beginning of the photoperiod and the intervals between successive ovipositions shortened. This suggests that when planning the frequency of egg collection in a flock of broiler breeder hens, one should account for changes in the egg sequence length during the production period. No significant relationship between egg position in the sequence and quality of egg components was observed. |
Tremorgenic mycotoxicosis caused by Paspalum paspaloides (Michx.) Scribner infected by Claviceps paspali: a case reportM.R. Moyano, A.M. Molina, A.J. Lora, J. Mendez, A. RuedaVet Med - Czech, 2010, 55(7):336-338 | DOI: 10.17221/2964-VETMED A natural poisoning case caused by ingestion of Paspalum paspaloides (Michx.) Scribner infected by Claviceps paspali in a bovine herd in Huelva (Spain) is described. This kind of intoxication, frequently described in South Africa and America is rare in Europe and this is the first description in Spain. Clinical signs, which affected 23 calves out of a total of 130 animals, consisted in a nervous syndrome characterized by tremor, hyperexcitability, incoordination, ataxia, depression and paralysis, signs that were aggravated when animals were subjected to certain types of exercises. The most relevant lesions were detected in the brain and consisted in microhaemorrhages diffused through the parenchyma of the brain, neuronal degeneration, satellitosis, neuronophagia, gliosis, and moderate neuropil degeneration in the peripheral zones of the brain. |
Visible foliar injury as a tool for the assessment of surface ozone impact on native vegetation: a case study from the Jizerské hory MtsL. Matoušková, R. Novotný, I. Hůnová, V. BuriánekJ. For. Sci., 2010, 56(4):177-182 | DOI: 10.17221/61/2009-JFS Surface ozone is considered to be a very phytotoxic gaseous air pollutant. Its negative impacts at both the cell and the organ level have been shown, mainly as a result of experiments. However, the demonstration of ozone negative impacts on native plants is not explicit. An assessment of ozone impact on vegetation and ecosystems using indicators based on ambient ozone concentrations is insufficient and assessment techniques based on internal ozone dose and on real plant damage are more appropriate. Such a possible technique is the mapping of ozone visible symptoms due to ozone influence. The ICP-Forest method concerning ozone visible symptoms and the assessment of ozone influence were applied for the purposes of this case study. The visible symptoms are characterized by a few typical signs. Nevertheless, the identification of visible symptoms in native conditions can be problematic and misleading conclusions could be drawn. Therefore it is necessary to complete the identification of visible symptoms with a validation in order to confirm ozone as the cause of plant injury. |

It is possible to achieve various aims in the field of fiscal and agricultural policies of state, consumption of cigarettes, protection of national market and in the other field by the regulation of excise duty imposed on cigarettes in the EU states. The question is, in frame of excise duty imposed on cigarettes in the EU member state, whether this tax should be imposed through specific rate, ad valorem rate, or by their combination. The choice between these options depends on the laid down aims of tax policy in each EU member states. |

It describes the diversification activities according to the Farm Structure Survey. In 2007, every third farm performed other gainful activities. The paper analyzes the public expenditures paid to the diversification in the pre-integration and post-integration period and reveals the covering of the beneficiaries in the past and new programming period. The conclusion presents the attitudes and opinions of the Diversification Measure beneficiaries connected with the evaluation of the implementing means and other aspects joined with the implementation of diversification activities within the sectoral programme documents. |
Field efficacy of Helicoverpa armigera nucleopolyhedro-virus isolates against H. armigera (Hübner) (Lepidoptera: Noctuidae) on cotton and chickpea in Tamil NaduSubramanian Jeyarani, Natarajan Sathiah, Palaniappan KaruppuchamyPlant Protect. Sci., 2010, 46(3):116-122 | DOI: 10.17221/30/2009-PPS Field efficacy of seven geographical isolates of Helicoverpa armigera nucleopolyhedrovirus (HaNPV) along with an insecticide control was evaluated against H. armigera on cotton and chickpea in the Coimbatore district of Tamil Nadu, India. Among the HaNPV isolates, CBE I (Coimbatore) and NEG (Negamum) applied at 3.0 × 1012 POB/ha to cotton and 1.5 × 1012 POB/ha to chickpea with an adjuvant, crude sugar, significantly reduced the H. armigera larval population and increased the yield. CBE I and NEG recorded the highest yield of 2038 kg/ha and 2033 kg/ha, which was on a par with endosulfan (2026.7 kg/ha) with cost/benefit ratios of 1:2.32, 1:2.48, and 1:1.12, respectively, on cotton. In chickpea grain yields of 980, 983, and 973.3 kg/ha and cost/benefit ratios of 1:1.36, 1:1.48 and 1:0.87, respectively, in CBE I, NEG and endosulfan treated plots were obtained. The isolate RAJ (Rajasthan) recorded the lowest yield comparable to that of the untreated control in both crops. |
The effect of time of cutting on yield and the quality of the hop hybrid varieties Harmonie, Rubín and AgnusK. Krofta, J. JežekPlant Soil Environ., 2010, 56(12):564-569 | DOI: 10.17221/187/2010-PSE The influence of mechanized cutting time on the yield and quality of hops was studied for three years (2007, 2008 and 2009) in three Czech hybrid varieties Harmonie, Rubín and Agnus. The cutting term classified as 'early' was carried out at the turn of March and April. Further cutting dates were made at intervals of 7-10 days and they were classified as 'middle' and 'late' ones. Yield and contents of alpha acids were evaluated during the machine harvest. The measured data revealed no statistical relationship between term of cutting, alpha acid content and yield in any of the tested cultivars. However, during the experimental period considerable inter-annual differences were observed, especially in the yield. Average yield in the range of 2.0-2.5 t/ha in the period 2007-2008 decreased to 1.2-1.9 t/ha in 2009 due to downy mildew attack. On the basis of the obtained data it can be recommended to cut the hybrid varieties Harmonie, Rubín and Agnus in the first ten days of April, taking into account other factors such as age and the location of hop garden and current weather conditions. |
Multiplex PCR assay to detect rust resistance genes Lr26 and Lr37 in wheatShort CommunicationTaťána SUMÍKOVÁ, Alena HANZALOVÁCzech J. Genet. Plant Breed., 2010, 46(2):85-89 | DOI: 10.17221/32/2010-CJGPB Multiplex PCR was developed and optimized for simultaneous detection of wheat leaf rust resistance genes Lr26 and Lr37. The presence of the genes was analyzed in 21 winter wheat cultivars registered in the Czech Republic. Gene Lr37 was detected in four tested cultivars (Bakfis, Biscay, Nicol, Mulan), gene Lr26 occurred only in one cultivar (Etela) and three cultivars (Clarus, Orlando and Rapsodia) were found to carry both these genes. Data obtained by PCR markers were compared with results of greenhouse and field tests. Seedling reactions of cultivars possessing Lr26 to seven different leaf rust isolates conformed to the results obtained by the marker analysis, however, there were found some discrepancies in the detections of Lr37, which could be detected in greenhouse seedling tests only with difficulties. |
Post-harvest content of free titratable acids in the grain of proso millet varieties (Panicum milliaceum L.), and changes during grain processing and storageV. Dvořáček, D. Janovská, L. Plant Breed., 2010, 46(10):S90-S95 | DOI: 10.17221/699-CJGPB A triennial evaluation of the content of titratable acids (TA) in grain was carried out on 12 varieties of proso millet (Panicum miliaceum L.). The model variety, Mironovskoe 94, harvested in 2004, was used for the monitoring of sensory changes in TA content, in relationship to different storage conditions and the application of different grain processing methods. The Czech state norm (CSN 56 0512-9 1995) Determination of Titratable Acids, with a slight modification, was used as the standard method. The TA contents in dry matter varied between 39 and 78 mmol/kg of grain dry matter, and significant differences between varieties and experimental years were found. The method of proso millet processing had a key role in the final sensitivity to rancidity. The storage conditions had a much lower influence on TA content than the processing technology. The clearly detected sensory changes found in scoured grains corresponded with TA contents within the range of 90 to106 mmol/kg. The monitoring of the TA content can be considered as a suitable tool for the prediction of rancidity processes in millet grains. |

Sci., 2010, 55(8):330-336 | DOI: 10.17221/317/2009-CJAS The effect of season and parity order on doe fertility and rabbit growth was evaluated. Two hundred and fifty Hyplus® does were divided into five groups according to kindling, each in a given season of the year (September, December, February, April and June). Another five groups were separated by parity order (from the first to the fifth kindling). After weaning, each group of rabbits was fed until 77 days of age. The season of the year significantly affected service number of pregnancy, litter size, litter weight at birth, litter weight at 21 days, litter size at weaning as well as live weight at 77 days. Parity order affected service number of pregnancy and litter weight at 21 days in a more limited way. Significant interactions of season and parity order were determined in service number of pregnancy, with the highest values occurring in December (2.2, P ≤ 0.010) and February (1.9, P ≤ 0.010) at the second kindling. Litter size was the largest in December and at the fifth kindling. Significant interactions (P ≤ 0.015) were also found in litter weight at 21 days, whereby litter weight had increased by the third kindling. However, the highest litter weight was reported after the fourth kindling in December. |

KimVet Med - Czech, 2010, 55(5):253-257 | DOI: 10.17221/2995-VETMED A ten-month old, male Black and Tan Coonhound dog was referred with ocular bleeding due to gunshot injury. His mental state was normal. A computed tomography revealed that the bullet was planted in the left cranium. It was presumed that the trajectory of the bullet penetrated from the right medial angle of the eye to the orbit, and changed its track to caudo-dorsal by penetrating the cranium, ending up at the left cranium. The bullet was removed by lateral rostrotentorial craniectomy. No complications were observed during a one-year follow-up except the blindness in the right eye. This is a rare case of gunshot-induced traumatic brain injury featuring a bullet which went through the orbit into the cranium. The damaged frontal lobe seemed to show no neurological signs at the time of first examination in this case. In conclusion, a less aggressive surgical approach is recommended to remove bullets when they are accessible. |

Food Sci., 2010, 28(1):1-8 | DOI: 10.17221/210/2008-CJFS The composition and selected physical and chemical parameters of 44 samples of fresh goat cheeses produced on a farm in the Czech Republic were determined. The following average values were obtained for the parameters analysed: pH 4.87 ± 0.14, titratable acidity (SH) 98.09 ± 4.93, dry matter 46.83 ± 1.57%, fat in dry matter 52.74 ± 5.24%, sodium chloride (NaCl) 2.08 ± 0.54%, and aw 0.979 ± 0.007. All samples showed excellent sensory characteristics and their compositions corresponded to those declared by the producer. Microbiological tests were used for the detection of Enterobacteriaceae spp., lactic acid bacteria, Escherichia coli, Enterococcus spp., Staphylococcus aureus, Bacillus cereus, Salmonella spp. and Listeria monocytogenes. Under the applicable regulations, the analysed fresh goat's cheeses were microbiologically safe and had the appropriate physical and chemical characteristics. |
Effect of zinc foliar application on grain yield of maize and its yielding componeJ. Potarzycki, W. GrzebiszPlant Soil Environ., 2009, 55(12):519-527 | DOI: 10.17221/95/2009-PSE Actual yields of maize harvested by farmers are at level much below attainable yield potential of currently cultivated varieties. Among many growth factors zinc was recognized as one of main limiting factors of maize crop growth and yielding. This hypothesis has been verified within a three-year field study, where zinc fertilizer was applied to maize plants at the 5th leaf stage. Maize crop responded significantly to zinc foliar application in two of three years of study. The optimal rate of zinc foliar spray for achieving significant grain yield response was in the range from 1.0 to 1.5 kg Zn/ha. Grain yield increase was circa 18% (mean of three years) as compared to the treatment fertilized only with NPK. Plants fertilized with 1.0 kg Zn/ha significantly increased both total N uptake and grain yield. Yield forming effect of zinc fertilizer revealed via improvement of yield structure elements. The number of kernels per plant showed the highest response (+17.8% as compared to the NPK plot) and simultaneously the highest dependence on N uptake (R2 = 0.79). For this particular zinc treatment, however, the length of cob can also be applied as a component of yield structure significantly shaping the final grain yield. |
Research work on Vitis vinifera ssp. sylvestris in SlovakiaD. Pospíšilová, R. Šimora, D. SekeraCzech J. Genet. Plant Breed., 2010, 46(10):S47-S49 | DOI: 10.17221/989-CJGPB By 2008, twelve localities had been investigated. At each location we assessed the degree of threats to the wild grapevine. Flowering plants were seldom found. The plants found were localized geographically, and 149 vines were described using the characteristics of shoot types and leaves, using the O.I.V. descriptor list. Morphological differences of the leaf shape, hair or bristle density are high. A few seeds which were found in dry black berries are much smaller in size than the seeds of Vitis vinifera ssp. sativa. From plants that were easily attainable, cuttings were taken with the objective to multiply the Vitis sylvestris and to establish an ex situ collection. In 2008, we repeated the procedure with 54 vines. The state of health of the vines in situ is very good. No fungal diseases were visible, and mites occurred only rarely. In the nursery, some plants were contaminated by mildew. |
Effects of long-term liquid commercial semen extender and storage time on the membrane quality of boar semenS. Frydrychová, J. Čeřovský, A. Lustyková, M. RozkotCzech J. Anim. Sci., 2010, 55(4):160-166 | DOI: 10.17221/62/2009-CJAS The objective of this study was to assess the sperm membrane integrity in extended boar semen during storage time using specific spectrum laboratory methods. Boar semen was diluted with the long-term liquid commercial extenders Androhep (A), Androstar (AS), Androstar plus (AS+), LD and M III and was stored up to 96 h. The sperm membrane integrity was evaluated by motility, viable spermatozoa, short hypoosmotic swelling test (sHOST) and by the activity of the enzyme aspartate aminotransferase (AST). Negative changes in the quality of sperm membrane in relation to storage time were observed after 48 h for sHOST, after 72 h for viable spermatozoa and after 72 h for motility. The percentage of viable spermatozoa was decreased by 0.27% each hour. A statistically significant difference between extenders A and LD was observed in sHOST after 72 h and 96 h storage (P < 0.05). The AST activity did not show any statistically significant differences in extenders and in storage time. In overall assessment Androhep was the best of the tested extenders, followed by AS, AS+, M III and LD in terms of motility, viable spermatozoa and sHOST. The correlations among laboratory methods were highly significant (P < 0.001). In conclusion, the results documented that the sperm membrane integrity was statistically significantly affected by extenders and storage time (P < 0.001). Boar semen quality was the best in extender A. sHOST is a very sensitive and relatively simple method for the assessment of sperm membrane integrity in diluted semen. |

The height, area and volume of the ventricles and brain were measured in 20 Yorkshire terriers (10 normal and 10 hydrocephalic dogs) using low-field MR imaging (at 0.2 Tesla). All measurements were averaged and the relative ventricle size was defined as a percentage (percent size of the ventricle/size of the brain). The difference between normal and hydrocephalic dogs was statistically significant for the average of each ventricle as well as for the percentage value. Five hydrocephalic symptoms were identified: circling, head tilting, seizures, ataxia, and strabismus. With respect to height, area and volume of the brain/ventricle, the difference between normal and hydrocephalic dogs was not significant. The ventricle/brain with height (1D) was related to the area (2D) and volume (3D). The correlations with area and volume were as good as the ventricle/brain height ratio in the case of hydrocephalic dogs. Therefore, one-, two- and three-dimensional quantitative methods may be complementary. We expect that the stage of hydrocephalic symptoms can be classified if statistical significance for ventricular size among symptoms is determined with the analysis of a large number of hydrocephalic cases. |
Effects of probiotic dietary supplementation on diarrhoea patterns, faecal microbiota and performance of early weaned calvesJ. Jatkauskas, V. VrotniakieneVet Med - Czech, 2010, 55(10):494-503 | DOI: 10.17221/2939-VETMED Twenty Lithuanian Black-and-White calves (10 bulls, 10 heifers) were used to evaluate the effects of the supplemental probiotic product, Enterococcus faecium M74 (2.4 g/day/calve), added to fresh milk and skimmed milk in a 56 day-study. The probiotic was administered by dietary supplementation to first group of calves and their respective pens (probiotic group), whereas the second group (control group) received no probiotic supplementation. The results of this trial indicate positive effects of the probiotic product Enterococcus faecium M74. The actual percentage of calves with diarrhoea was reduced from 50 % to 20% among the calves fed the pre-and probiotic diet. Probiotic supplementation reduced the faecal count of clostridia and enterococci. The calves fed Enterococcus faecium M74 weighed more at 20, 40 and 62 days of age by 4.9%, by 9.7% (P < 0.05) and by 9.4% (P < 0.01), respectively, than the control calves. The calves fed Enterococcus faecium M74 had increased daily weight gains compared with the calves not fed a probiotic product. The average weight gain and the daily weight gain of the probiotic-supplemented calves were by 7.8 kg (P < 0.01) and by 0.14 kg higher (P < 0.01) compared with the control calves. The calves given the Enterococcus faecium M74 also had forage and total DM intakes that were numerically higher than those fed the control diet, without any additive. During the 56 days experimental period, the average feed conversion rate was improved by 12.9% in the probiotic-treated group. |
Evaluation of effect of different sterilization methods on soil biomass phosphorus extracted with NaHCO3A.A.S. Sinegani, A. The temperature, time and method for killing soil organisms in soil biomass P determination are so important factors that affect the results. The aim of this study was to compare some methods of soil sterilization and soil microbial P (Pm) release in extraction with NaHCO3. Five samples of calcareous soils in three replicates were incubated in field capacity and 28°C. The texture of soils differed from loamy sand to sandy loam. They had 10-15% equivalent calcium carbonate, 1-3% organic carbon and 40-90 mg/kg available P content. After 1, 10, 30, and 50 days of incubation a portion of each soil was sterilized by five methods (chloroform fumigation, autoclaving at 121°C for 0.5 h, oven drying at 70°C for 48 h, oven drying at 180°C for 2 h, and ultrasonification). Phosphorus of the sterilized and control soils were extracted with 0.5M NaHCO3 (pH 8.5) and determined spectrophotometrically as blue molybdate-phosphate complexes under partial reduction with ascorbic acid. Results showed that the method of soil sterilization and soil type had significant effects on biomass P estimated in soils (P < 0.01). There were no significant differences between extracted biomass P at chloroform fumigation, autoclaving at 121°C for 0.5 h, and oven drying at 70°C for 48 h. However biomass P was significantly more extracted from soil by oven drying at 180°C for 2 h and by ultrasonification. Fixation of Pm in soil during the extraction period can reduce the precision of biomass P estimates. This effect was more obvious in biomass P determination by fumigation and oven drying at 70°C for 48 h. Biomass P determined by these methods and autoclaving had positive and significant correlations with soil sand contents, respectively due to long sterilization period and temperature. Ultrasonification released more P from these calcareous soils especially at the start of soil incubation. It released higher microbial P and reduced P sorption in soils; biomass P determined by this method compared to other methods had relatively higher correlations with fungi, glumales spore and P solubilizing bacteria numbers in soil. |

The first international documentation standards were developed by the FAO in the 1950s. In 1974, a specific body for plant genetic resources, the IBPGR (International Board for Plant Genetic Resources), was established within the FAO. Since the 1970s, the descriptor lists for main crop collections have been developed, including passport descriptor sets. For the most part, the passport descriptors were common to all crops, with only slight differences. In 1997, the first attempt to create a standard set of descriptors resulted in 24 passport descriptors developed by FAO and IPGRI (International Plant Genetic Resources Institute) working in cooperation. In 2001, the official Multi-Crop Passport Descriptor list (MCPD) was published, including a revised standard set of 28 descriptors. The European ex situ crop catalogue (EURISCO) enlarged the standard set of passport descriptors by adding 6 additional fields in 2002; and another 2 new descriptors were added in November of 2008. A global level of data compatibility is preferred; therefore projects aimed at world-wide utilization and standardization of systems, such as GRINGlobal, and Accession-Level Information System (ALIS), are supported. |

Sci., 2010, 55(10):436-444 | DOI: 10.17221/1701-CJAS The quantification of emissions of greenhouse gases from human activities is of prime importance for determining the importance of their effect on the environment. The aim of this study was to test a hypothesis that the interior concentration and emission of carbon dioxide in chicken housing is impacted by the age of animals and season of fattening period. Carbon dioxide (CO2) concentrations and emissions were assessed over six fattening periods in total. The major part of CO2 seemed to have its origin in bird respiration with assumed production of approx. 147 kg of CO2/h. CO2 emission was most affected by chickens towards the end of the grow-out period (P < 0.001) taking dominance over the process of natural gas burning by heaters. The mean CO2 emission from the chicken house ranged between 120 and 247 kg/h in the first quarter of periods and between 325 and 459 kg/h in the last ones. The heaters could be theoretically a possible source of approx. 39 kg each hour if they worked continuously. CO2 emissions were considerably more affected by ventilation rate (P < 0.001) than by CO2 concentration in the indoor air. |
