Environmental and genetic effects on cadmium accumulation capacity and yield of maizeOriginal PaperVlado Kovačević, Imre Kádár, Luka Andrić, Zvonimir Zdunić, Dario Iljkić, Ivana Varga, Jurica JovićCzech J. Plant Breed., 2019, 55(2):70-75 | DOI: 10.17221/5/2018-CJGPB Maize (Zea mays) is an economic crop suitable for use in phytoremediation in low to moderately cadmium (Cd)-contaminated soils due to its ability to accumulate high concentration of Cd in parts of maize that are not used in human diet. The aim of this study was to test Cd content in nine female parents of the commercial maize hybrids (C1 = ♀2-48; C2 = ♀1767/99; C3 = ♀87-24; C4 = ♀135-88, C5 = ♀84-28; C6 = ♀84-44; C7 = ♀438-95; C8 = ♀30-8; C9 = ♀B-73) grown under field conditions in two soils (B1: eutric cambisol, B2: stagnosol) during three growing seasons (A1: 2006, A2: 2007, A3: 2008). The stationary trial was conducted in four replicates. The ear-leaves at flowering and grain at maturity were taken for chemical analysis. The average quantities of leaf-Cd were 0.081, 0.088 and 0.143 mg per kg of dry matter for A1, A2 and A3, 0.089 and 0.118, for B1 and B2, respectively. Grain-Cd was below the threshold (< 0.02 mg/kg). Five Cd-inefficient genotypes (C3, C5, C6, C7 and C9) had low leaf-Cd (average 0.049 mg/kg), while this content was about 6-times higher (average 0.299 mg/kg) in Cd-efficient genotype C4. The yield among the years ranged from 2.36 to 4.31 t/ha. Maize grown on B2 had about 26% lower yield than on B1. Five genotypes (C1, C2, C8 and C9) achieved yields less than 3.50 t/ha (mean 3.15 t/ha), while in two genotypes (C3 and C5) yields were above 4.00 t/ha (mean 4.14 t/ha). Very strong correlations (r) of leaf-Cd status among years (ranged from 0.52 to 0.77) confirmed high genetic effect on the capability of Cd accumulation in maize. However, correlations between Cd content and yield were low (ranged from -0.17 to 0.06). Cd-efficient C4 female parent could be used for development of maize hybrids suitable for phytoremediation, while Cd-inefficient female parents for hybrids could be suitable as forage maize crop contributing to the lower Cd input into food chain. |

Sci., 2018, 63(2):61-69 | DOI: 10.17221/56/2017-CJAS The objectives of this study were to estimate linkage disequilibrium (LD), describe and scan a haplotype block for the presence of genes that may affect milk production traits in Portuguese Holstein cattle. Totally 526 animals were genotyped using the Illumina BovineSNP50 BeadChip, which contained a total of 52 890 single nucleotide polymorphisms (SNPs). The final set of markers remaining after considering quality control standards consisted of 37 031 SNPs located on 29 autosomes. The LD parameters historical recombinations through allelic association (D') and squared correlation coefficient between locus alleles frequencies ( r2) were estimated and haplotype block analyses were performed using the Haploview software. The averages of D' and r2 values were 0.628 and 0.122, respectively. The LD value decreased with increasing physical distance. The D' and r2 values decreased respectively from 0.815 and 0.283 at the distance of 0-30 kb to 0.578 and 0.090 at the distance of 401-500 kb. The identified total number of blocks was 969 and consisted of 4259 SNPs that covered 159.06 Mb (6.24% of the total genome) on 29 autosomes. Several genes inside the haplotype blocks were detected; CSN1S2 gene in haplotype block 51 on BTA 6, IL6 and B4GALT1 genes in haplotype blocks 6 and 33 on BTA 8, IL1B and ID2 genes in haplotype blocks 19 and 29 on BTA 11, and DGAT1 gene in haplotype block 1 on BTA 14. The extension of LD using BovineSNP50 BeadChip did not exceed 500 kb and its parameters r2 and D' were less than 0.2 and 0.70, respectively, after 70-100 kb. Consequently, the 50K BeadChip would have a poor power in genome wide association studies at distances between adjacent markers lower than 70 kb. |
Production of low protein and gluten-free cookies for phenylketonuria (PKU) and/or celiac patientsOriginal PaperOzen Parlak, Ayse Neslihan DundarCzech J. Food Sci., 2021, 39(1):29-34 | DOI: 10.17221/145/2020-CJFS Abstract: The aim of this study was to analyse the production of gluten-free, low protein cookies, which can easily be consumed by celiac and phenylketonuria (PKU) patients. The formula was adopted to prepare the control cookies (without dried fruits) from maise starch, stabilisers, salt, sodium bicarbonate, protein-reduced milk powder, and sunflower oil. Different cookies were made by adding raisins (R), dried black currants (Cu), dried dates (D), dried apples and cinnamon (A) separately. In cookies, two different stabilisers were calculated according to starch weight [pectin (P): 0.15%, glycerol monostearate (GMS, G): 0.4%]. The effects of stabilisers and dried fruits on the physicochemical and sensory properties of cookies were investigated. The protein content was significantly higher (P < 0.05) in the cookies including R with pectin (PR), Cu with pectin (PCu), and A with pectin (PA). The amount of phenylalanine (Phe) was significantly higher (P < 0.05) in PR. The values of dietary fibre were the highest in the A cookies, while the lowest values appeared in the control (C). In the sensory evaluation (appearance, tasting properties, and affordability), it was found that PD (pectin with dries dates) was chosen as the best cookie (P < 0.05). |
Evaluation of genetic diversity of subdivided genealogical groups in Lithuanian Trakehner horse population using immunogenetic toolsOriginal PaperAlma Račkauskaité, Ruta ©veistiené, Violeta Razmaité, Virginija JatkauskienéCzech J. Anim. Sci., 2021, 66(6):200-210 | DOI: 10.17221/118/2020-CJAS The objective of this study was to evaluate the inter-breed genetic diversity of Lithuanian Trakehner horses. The current population in Lithuania numbers 1 039 Trakehner horses. The study included the immunogenetic data analysis of 316 blood samples. Pedigree was traced back to 7-11 generations. The horses were assigned to genealogical clusters originating from East Prussian Trakehner Stud, Thoroughbred and Arab sires. The first two clusters were subdivided into nine-horse groups according to the most important ancestors. The evaluation of genetic diversity was based on six blood groups and five protein polymorphism systems. The differences between genealogical groups were confirmed by principal component analysis and applied cluster analysis. According to the allelic frequency, the Trakehner population changed very slightly through a 30-year period, and only one rare (0.013) allele TfRR (frequent in other Trakehner populations), has been newly found. The analysis of genetic polymorphism systems based on a very low rate of alleles Qabc (0.093) and Ddkl (0.046), which are typical of other populations, indicated the exclusivity of the Trakehner population in Lithuania. The average expected heterozygosity by blood protein polymorphism and blood groups were 0.409 and 0.441, respectively. The genetic clustering diverged by observed heterozygosity of blood groups and by pedigree data in Pilger through Egoist, Bay Ronald through Dark Ronald and Dampfross through Hyperion subgroups. The determined distinctiveness of subdivided Trakehner horse groups suggests reconsidering the breeding strategies and conservation programme of Trakehner horses. Breeding and selection of subdivided sire lines could be among the appropriate solutions for the maintenance and extension of the genealogical structure. |

Z'Fix is an agent recommended as an addition to animal bedding to prolong its function and to lower ammonia emissions in stables. Concurrently, a positive effect on organic matter transformation in resulting manure is claimed. The experiment involved control, farmyard manure (FYM), and farmyard manure with Z'Fix (FYM_ZF) as variants. In-field sampling was conducted for cone index, water infiltration and implement a unit draft, where the latter two showed significant differences in favour of FYM_ZF. Also, concerning crop yields, FYM_ZF consistently attained the highest values, followed by FYM throughout all three seasons. Furthermore, remotely sensed data were analysed to describe crop status via normalised difference vegetation index where significant differences were found across all variants. Based on the study, FYM_ZF demonstrated positive effects both on soil properties and crop conditions. |

VicenteVet Med - Czech, 2018, 63(5):216-224 | DOI: 10.17221/87/2017-VETMED The aim of this study was to evaluate the applicability of B-mode, Doppler, acoustic radiation force impulse elastography and contrast-enhanced ultrasound exams in the differentiation of neoplastic and non-neoplastic benign mammary lesions in bitches. This research was conducted as a prospective secondary observational cohort (2014-2016) study, which included 36 mammary lesions, evaluated physically and by ultrasound (B-mode, Doppler, contrast-enhanced ultrasound and acoustic radiation force impulse) exams prior to mastectomy and histopathological classification as neoplastic or non-neoplastic lesions. All ultrasonographic parameters studied were compared between histopathological classifications using Fisher's or Student's tests and differences were considered significant when P < 0.05. Out of 36 benign mammary lesions evaluated, 25 were classified as neoplastic and 11 as non-neoplastic. The qualitative and quantitative parameters evaluated using the different ultrasound methods were not effective (P > 0.05) in differentiating between neoplastic and non-neoplastic mammary masses in bitches. Nevertheless, some B-mode variables, such as longitudinal length (P = 0.0292), width/length ratio (P = 0.0001) and width/height ratio (P = 0.0001) showed limited efficacy in the differentiation of mammary lesions types. In conclusion, ultrasonographic evaluation of benign canine mammary lesions did not allow differentiation between neoplastic and non-neoplastic tissues and only a few B-mode variables may aid in the prediction of specific tumour types. |

Eng., 2021, 67(2):84-91 | DOI: 10.17221/67/2020-RAE This research optimised the application of a hexane-methanol mixture as a binary solvent for the concurrent oil-resin extraction and separation from Calophyllum seeds on a pilot scale, in a direct stage. The optimum oil and resin yields were determined by optimising the extraction conditions using response surface methodology and a second order polynomial model. The extraction conditions affected the oil and resin yields, with the extraction time as the biggest influencing factor. Optimum oil (65%) and resin (16%) yields were predicted to be obtained at 5.2 h and 433 rpm. The model validation with these extraction conditions showed that the predicted results and actual oil (62%) and resin (15%) yields were in passable agreement. The oil was composed of 75.4% triglycerides with a density of 0.874 g.cm-3, a viscosity of 26.4 mPa.s-1, an acid value of 46.4 mg KOH.g-1, an iodine value of 98.0 g iodine.100 g-1, trace water and sediment contents, and zero ash content. The resin had a viscosity of 4 694.8 mPa.s-1, a total phenolic content of a 4.51% gallic acid equivalent, an antioxidant activity of an 8.82 mg ascorbic acid equivalent.g-1, and an acid value of 126.2 mg KOH.g-1. |
Sustainable alternatives to chemicals for weed control in the orchard - a ReviewReviewMd Jebu Mia, Francesca Massetani, Giorgio Murri, Davide NeriHort. Sci. (Prague), 2020, 47(1):1-12 | DOI: 10.17221/29/2019-HORTSCI This review is designed to address various alternative weed-control practices and their possibilities in the fruit orchard in terms of sustainability. Correct weed management and maintenance of adequate orchard biodiversity are crucial for sustainable orchard soil management. The key is to practice an alternative weed-management approach (single or integrated) rather than to use possibly harmful chemicals only. Integration of modern equipment with a shallow tillage system can provide effective weed control in tree rows, including optimised tree performance and soil biodiversity. Living mulch suppresses weeds and enhances orchard biodiversity, while selection of less competitive and less pest-attracting species is crucial. Plastic covers offer long-term weed control, but additional nutrient amendments are required to maintain the balanced fertility of the soil. Wood chip mulch is suggested where the materials are available on or near the farm, and where there is lower incidence of perennial weeds. High pressure water and robotic systems are still in their infancy for fruit orchards, and required more research to confirm their efficiency. |
Influence of various biofertilizers on root growth dynamics in sweet cherry (Prunus avium L.) cv. (Prague), 2021, 48(3):105-116 | DOI: 10.17221/119/2020-HORTSCI The experiment was established in the Pomological Orchard of The National Institute of Horticultural Research in Skierniewice in a system of randomized blocks. The aim of the experiment was to investigate the impact of innovative organic fertilizers: BioIlsa, BioFeed Ecomix, biostimulator Ausma and mycorrhizal inoculum Mykoflor on the fine roots growth characteristics of 'Vanda' sweet cherry trees in comparison with NPK mineral fertilization. The experiment involved five combinations, in three repetitions of three trees each, treated with tested preparations. The study assessed the influence of fertilization on the lifespan of the roots, the depth of their formation, their diameter and survivorship using minirhizotron camera. The highest numbers of roots were found in the treatment where the plants were fertilized with NPK and the lowest following the use of the biofertilizer BioFeed Ecomix. The longest lifespan was shown by the roots of the trees treated with BioFeed Ecomix - 347 days, and the shortest - by those fertilized with the Ausma - 225 days. The lifespan of the roots increased with their diameter. The roots that lived the longest had a diameter in the range from 0.9 to 1.0 mm - 568 days, and the shortest-living were the roots with a diameter smaller than 0.3 mm - 238 days. The roots that formed in late autumn and winter had the shortest median lifespan of 159 days, while the roots formed in the spring where characterized by the longest lifespan of 300 days. The lifespan of the roots formed close to the soil surface was the shortest - 225 days, while that of the roots formed at a depth of 10 to 20 cm was the longest - 326 days. Biological origin, organic nitrogen rich fertilizers positively influence on fine roots lifespan and longevity. Mineral fertilization increases number of new formed roots. |

Food Sci., 2021, 39(5):402-409 | DOI: 10.17221/261/2020-CJFS The present investigation was an attempt to compare the phytoceutic potential of ginger rhizome and ginger leaves of the Suravi variety. For this purpose, both rhizome and leaves were dried and used for the preparation of patties. After that, patties were assessed for colour tonality, texture, total phenolic content and hedonic response such as colour, taste, flavour, texture and overall acceptability. The results depicted that L* and b* values changed significantly during the storage interval; however, b* value was also affected by treatments whilst L* and a* values did not impart any momentous effect. For texture, the highest value was observed for patties with ginger rhizome powder (0.067 ± 0.0032 N) followed by patties with ginger leaf powder (0.060 ± 0.0029 N) and then control patties (0.057 ± 0.0026 N). For total phenolic content (TPC), maximum phenolic contents were observed as 84.80 ± 3.31 mg GAE 100 g-1 in treatment T2 followed by 75.68 ± 2.95 mg GAE 100 g-1 in T1 and 61.70 ± 2.41 mg GAE 100 g-1 in T0. For hedonic response, all the parameters changed significantly during the storage interval; however, flavour, taste and overall acceptability changed momentously with treatments. The findings of the current investigation demonstrated that ginger leaves have a higher antioxidant potential as compared to the ginger rhizome and control patties, and they should be incorporated into food products. |
Estimation of the prevalence of respiratory diseases in pigs in north-eastern Poland: Survey of pulmonary lesions in pigs at a slaughterhouseOriginal PaperP Przyborowska-Zhalniarovich, Y Zhalniarovich, K WasowiczVet Med - Czech, 2021, 66(6):242-247 | DOI: 10.17221/188/2020-VETMED A total of 29 520 animals, from 164 batches of pigs belonging to an identical number of herds, were involved in the study. The considered population of pigs were limited to the region of north-eastern Poland involving six voivodeships. From each herd, samples of blood were collected to evaluate the antibody titres to Mycoplasma hyopneumoniae, porcine reproductive and respiratory syndrome virus, Aujeszky's disease virus and swine influenza virus. At an abattoir, the lung lesions of each batch were scored and the enzootic pneumonia-like lesion average value was calculated. Lesions, indicative of enzootic pneumonia, were found in 57.8% of the lungs. For all lungs, the enzootic pneumonia-like lesion average value was 1.74, ranging from 0.42 to 3.56 among the 164 batches. In the examined pig population, 57.8% were considered suffering from swine respiratory disease, the majority of the affected pigs came from the Podlaskie (21.7%) and Greater Poland (17.25%) voivodeships. In the most affected voivodeships, 88.37% and 85.16% of the farms were considered as disease-susceptible for Greater Poland and Podlaskie, respectively. The findings indicate that, in pigs in north-eastern Poland, the major pathogens causing pneumonia-like lesions are Mycoplasma hyopneumoniae (68.9%) and porcine reproductive and respiratory syndrome virus (44%). |

(Prague), 2021, 48(1):30-37 | DOI: 10.17221/21/2020-HORTSCI Ornamental cabbages are cultivars of cabbages grown for decorative purposes due to the varied shape and colour of their internal leaves. The aim of the study was to determine the course of growth and to assess the decorative and nutritional value of two cultivars of ornamental cabbage ('Pigeon Red', 'Coral Prince') grown in a flowerbed along with annual plants: the French marigold (Tagetes patula nana L.) and flossflower (Ageratum houstonianum Mill.), using a pine bark mulch. The impact of the marigold and flossflower on the cabbage height and foliage changed with the plant growth. Cabbages growing without the proximity to the other species had the smallest diameter and formed the least leaves; and the plant's height was lower in September. The cabbage height, foliage and leaf colour were modified by the weather in the subsequent years of research. The proximity of the marigold or flossflower and mulching of the soil with bark did not affect the colour of the leaves. The nutritional value of the ornamental cabbage was high, and the content of the mineral components in the leaves depended largely on their colour. The coloured leaves were more abundant in nitrogen, phosphorus, potassium and magnesium, while the green leaves contained more calcium and sulfur, and slightly more dry matter. |
Spontaneous development of early successional vegetation improves Norway spruce forest soil after clear-cutting and renewal failure: a case study at a sandy-soil siteOriginal PaperOndřej ©pulák, Duąan KacálekJ. Sometimes, if the new crop is not established successfully, clearcut is left unreforested. This study focused on a site where early successional species such as silver birch (Bi) and rowan (Ro) were accompanied with Norway spruce (Sp) in 13-year-old stand from natural regeneration at 550 m of altitude at an acidic site with eastern aspect and 25% slope. We found five types of stand composition: treeless gaps, Ro-Bi, Ro-Bi-Sp, Bi-Sp and monospecific Sp. Besides these juvenile ones, adjacent 100-year-old spruce (Sp old) stand representing pre-harvesting conditions was studied. In addition to the performance of trees, organic layer (Hum), topsoil (Ah) and upper subsoil (B) horizons were sampled to study an expected shift of chemical properties after clear-cutting and secondary succession at the site of interest. Birch dominated the natural regeneration; rowan and spruce were present mostly in understorey. Old spruce was more acidic and nutrient-poorer compared to the juvenile treatments. The treeless treatment showed also slightly higher pH and comparable nutrients compared to the young mixtures. Young spruce was higher in nitrogen compared to Ro-Bi-Sp mixture. |

Sci., 2021, 67(10):464-476 | DOI: 10.17221/79/2021-JFS Extensive microbiological research was carried out in Tbilisi (capital of Georgia) and its surroundings to determine the causes of massive disease and dieback of urban coniferous plantations. The biological material was picked up in June-July 2020 from trees with various degrees of the disease in 42 different localities. 247 conifers (15 species) were examined microbiologically. 1 169 samples of microscopic fungi were isolated. Based on cultural-morphological and molecular-genetic (PCR) studies, 34 strains were identified to the species level, 17 to the genus level, and 1 strain to the family level. Ascomycota were represented by 15 families and 33 species, Basidiomycota by 1 family and 2 species and Zygomycota by 2 families and 2 species. Among the isolated strains, 9 species were clearly dominant and found in all studied coniferous species: Alternaria alternata (Fr.) Keissl, Sphaeropsis sapinea (Fr.) Dyko & B. Button, Epicoccum nigrum Link., Sordaria lappae Potebnia, Curvularia spp., Dothiorella spp, Nothophoma quercina (Sydow & P. Sydow) Q. Chen & L. Cai, Phoma odoratissimi Q. Chen, Didymella aliena (Fries) Q. Chen & L. Cai. It may be supposed that massive activation of pathogenic fungi is the result of weakening of plant immunity on the background of increasing abiotic stresses in Tbilisi over the years; which led to an imbalance between latent pathogens and host plants and eventually to the depressing consequences of trees dieback. |
New dimension of logistics innovations development in agricultural enterprises in SlovakiaOriginal PaperĄuboslav Szabo, Patrik Richnák, Klaudia GubováAgric. Econ. - Czech, 2021, 67(4):136-143 | DOI: 10.17221/444/2020-AGRICECON The research article's primary aim was to provide a comprehensive and systematic view of a new dimension of logistics in the form of logistics innovations in agricultural enterprises in Slovakia. The intention thereof was to define and then analyse selected logistics innovations in agricultural enterprises. The research object was formed by 95 agricultural enterprises in Slovakia. A questionnaire was used as the research instrument. MS Excel and IBM SPSS Statistics 20 were used to process the data in the questionnaire. The univariate descriptive statistics and multivariate descriptive statistics were used to interpret the data. The research results were tested and evaluated by means of Spearman's rank correlation coefficient. Medium-sized agricultural enterprises in Slovakia represented the largest segment (68%) of the study. The agricultural enterprises mostly understand the use of smart technologies as an innovation in logistics (mean = 4.43). The results of the research showed that the analysed agricultural enterprises intend to innovate mainly the production logistics (mean = 3.20). Out of the logistics processes, the agricultural enterprises want to innovate mostly the inventory management (mean = 4.55). Lean logistics is the most frequently implemented logistics innovation in agricultural enterprises (mean = 3.64). The hypothesis testing has proven that there is a statistically significant relationship between the utilisation of lean logistics and production logistics innovation. In addition, it was proven that there is a statistically significant relationship between the utilisation of radio frequency identification (RFID) and the logistics process innovation - inventory management. |

Food Sci., 2021, 39(3):197-207 | DOI: 10.17221/225/2020-CJFS The article presents the results of homogeneity assessments for selected food mixes based on computer image analysis. The study was conducted on müsli and condiment mixes standardly available on the consumer market. A total of 40 different ready-for-use products were analysed. Collected samples from three package of each product were placed in a special chamber and then photographed. Photographs were then subjected to computer image analysis to acquire information on the percentage content of individual components. Homogeneity assessment was based on the contents of a selected component called tracer and the coefficient of variation (CV). Lower CV values (3.02-27.31%) and thus better homogeneity was observed for condiment mixes as compared to müsli mixes (3.57-59.15%). Fourteen of twenty condiment mixes had adequate (acceptable at CV ≤ 10%) mixing degree. For the müsli, only six of the tested mixes had appropriate homogeneity. The presented results are a preliminary to developing an image-based methodology for determining the uniformity of granular dry food mixes. |

Hydrogen peroxide (H2O2) plays an essential role during stress response by acting as a signal molecule that activates multiple stress tolerance mechanisms. In this study, the effects of H2O2 on agronomic traits were studied in rice (Oryza sativa L.) cv. Khao Dawk Mali 105 (KDML 105) was subjected to drought stress. H2O2 was applied by either seed priming or foliar application method with a concentration of 1, 5, and 15 mmol/L. The results showed that both seed priming and foliar application with H2O2 improved some yield components. The tiller numbers, number of panicles, number of filled grains, filled grain weight, and harvest index were improved approximately 1.13, 1.04, 1.23, 1.21, and 1.1 times compared to the untreated plants. Foliar application, however, helps the plant by reducing yield loss as indicated by a 0.5-time reduction in the number of unfilled grain and lower unfilled grain weight. It was suggested that 5 mmol/L H2O2 was the most effective concentration to alleviate the effect of drought stress during the reproductive stage in rice. |
Phylogenetics of native conifer species in Vietnam based on two chloroplast gene regions rbcL and matKOriginal PaperMai Phuong Pham, Viet Ha Tran, Dinh Duy Vu, Quoc Khanh Nguyen, Syed Noor Muhammad ShahCzech J. Genet. Plant Breed., 2021, 57(2):58-66 | DOI: 10.17221/88/2020-CJGPB We used two chloroplast gene regions (matK and rbcL) as a tool for the identification of 33 local conifer species. All 136 sequences, 101 newly generated (14 species for gene matK; 16 species for gene rbcL) and 35 retrieved from the GenBank, were used in the analysis. The highest genetic distance (matK region) was recorded between the species in Cupressaceae with an average of 5% (0.1-8.5), Podocarpaceae with an average of 6% (0-8.5), Taxaceae with an average of 5% (0.2-0.5) and Pinaceae with an average of 20.4% (0.8-54.1). The rbcL region showed a low genetic distance between the species in Cupressaceae 2% (0-3.3), Podocarpaceae 3% (0.6-3.4), Taxaceae 1% (0-2.1) and Pinaceae 1.2% (0-5.82). The phylogenetic analyses using the Maximum likelihood (ML) and Bayesian inference (BI) bootstrap values obtained at the branching nodes of each species ranged from 62 to 100% (Maximum likelihood bootstrap - MLBS and Bayesian posterior probabilities - BPP) for the matK gene; from 66 to 100% (MLBS) and 60 to 100% (BPP) for the rbcL region. The rbcL region was not identified between the species of Taxaceae and Cephalotaxaceae. The matK gene region was very clear in the different species among the families (Cupressaceae, Podocarpaceae, and Cephalotaxaceae) and unsuitable for identifying closely related species in Amentotaxus (Taxaceae) and Pinus (Pinaceae). The gene (matK) is a useful tool as a barcode in the identification of conifer species of Cupressaceae, Podocarpaceae, and Cephalotaxaceae in Vietnam. |

Selenium salts in the form of sodium selenite and sodium selenate were applied in growth phases: end of tillering (BBCH 29) and flag leaf ligule and collar visible (BBCH 39). The effect of experimental years 2014/2015, 2015/2016 and 2016/2017 on the yield of wheat grain was not statistically significant within the observed variants. The achieved mean yields were in the range from 10.06 ± 0.81 to 11.07 ± 0.29 t/ha in 2014/2015, from 9.82 ± 0.54 to 10.32 ± 0.10 t/hain 2015/2016 and from 11.23 ± 0.76 to 11.64 ± 0.51 t/ha in 2016/2017. Selenate in comparison with selenite influenced the selenium accumulation in wheat grains more positively. However, a significant difference was recorded in variants with selenite application in the flag leaf growth phase in comparison with the end of tillering phase. The influence on the content of macroelements P, K, Ca and microelements Cu and Fe was observed in sodium selenite only; its application decreased the element content in comparison with the control variant. Statistically significantly higher values of fiber and fat were achieved after application of selenium in the flag leaf growth stage in comparison with the end of tillering. |
Effects of harvest stage and diurnal variability on yield and essential oil content in Mentha × piperita L.Original PaperMeryem Yeşil, Mehmet Muharrem ÖzcanPlant Soil Environ., 2021, 67(7):417-423 | DOI: 10.17221/114/2021-PSE This study aimed at investigating the effect of ontogenetic and diurnal variability on yield and quality criteria of Mentha piperita. The research was conducted in the Ordu province of Turkey for two years. The trial was established in a randomised complete block design with 3 replications according to factorial regulations. Four different harvesting periods (budding, first flowering, 50% flowering, 100% flowering) and three harvesting hours (9:00 a.m., 1:00 p.m., 5:00 p.m.) were evaluated. A significant difference was determined between the trial years in all characters examined: plant height 55.9-69.0 cm, fresh herb yield 3 236-16 438 kg/ha, drug-herb yield 1 748-2 862 kg/ha, fresh leaf yield 3 477-4 506 kg/ha, drug leaf yield 968-1 253 kg/ha, essential oil ratio 2.0-2.5% and essential oil yield that varied between 19-28 L/ha. The highest yields were obtained at the beginning of flowering in drug leaf yield and 100% flowering period for all other characteristics. Diurnal variability was statistically significant only in the rate of essential oil, and the highest rate occurred in the 9:00 a.m. harvest. |

The objective of this research was to design and develop an animal fodder chopping machine to be utilised by dairy farmers within their purchase range. The drawing of these machine parts was undertaken in AutoCAD software and the construction was performed in a local workshop. After development of this machine, performance tests were carried out on a farm. The chopping machine tests were carried out with commonly grown fodder (namely: straw, grass, and maize) in Bangladesh. The performance evaluation of the developed machine was carried out in terms of the chopping efficiency, machine productivity, and energy consumption. The economic analysis of the straw chopping machine was assessed by indicating the cost effectiveness to the poor farmers. Analysis of the data in regard to chopping efficiency and machine productivity varied from 93 to 96% and from 192 to 600 kg×h-1, respectively. The energy consumption during the chopping process ranged between 0.0025 and 0.01 kWh for the different types of fodder. The break-even point of the fodder chopping machine was 3 793 kg of cut straw and the payback period was within one year depending on the use. |

(Prague), 2020, 47(1):13-20 | DOI: 10.17221/39/2018-HORTSCI The cropping of six sweet cherry cultivars that originated in the Research and Breeding Institute of Pomology at Holovousy, and a standard one, 'Burlat', were evaluated on three rootstocks in the period of 2007-2017. Trees planted in a spacing of 1.5 m × 5.0 m were trained as tall spindle axes utilising their natural tendency to develop a central leader. On the standard rootstock, P-TU-2, 'Tim' was the most productive with a mean total harvest of 47.6 kg per tree. 'Sandra' yielded the most on the PHLC rootstock with 56.2 kg per tree and 'Helga' yielded the most on Gisela 5 with a mean total harvest of 55.9 kg per tree. The mean impact of the rootstock on the tree vigour, measured upon the trunk cross section area, ranged from 148.4 cm2 on the standard rootstock P-TU-2 to 114.1 cm2 on the PHLC and 125.2 cm2 on Gisela 5 . On the standard rootstock P-TU-2, the most vigorous one according to this criterion was 'Jacinta' (178.0 cm2) whereas 'Justyna' (109.7 cm2) was the least vigorous. On the PHLC, the most vigorous was 'Sandra' (147.2 cm2) and the least was 'Amid' (94.0 cm2). The other tree characteristics were mainly dependant on the cultivar and minimally, or not at all, influenced by the rootstock vigour. |

It is dictated by the organizational scheme that provides temporary stop multiplication of shoots for some months. To avoid subculturing to fresh media every 4 weeks, which is obligatory for gooseberry, they can be kept in conditions that protect them from ageing, by slowing down their metabolism. To develop a rational schedule of gooseberry micropropagation, two experiments were used to adopt a temperature and length of time for storage. The best results were obtained with storage conditions at 2 °C for two or four months for proliferating cultures. Under these conditions, the percentage of necrotic shoots was low (< 10%), and shoot proliferation in the subsequent passages was at a level similar to proliferation cultures incubated in the growth room and sub-cultured monthly. The rate of shoots > 1 cm was higher than in the control in the growth room. Storage at 4 °C increased the probability of necrotic shoots up to 80% and decreased the number of all shoots and shoots > 1 cm in subsequent passages. |
Distribution of minerals between orange juice and orange flesh in various cultivarsOriginal PaperEid Brima, Hatem MohamedCzech J. Different orange cultivars, such as Abousorah [Citrus sinensis (L.)], Aseear (Citrus aurantium), Afandi (Citrus reticulata Blanco), Helo (Citrus sinensis), and grapefruit (Citrus paradisi), were collected from local markets. Elemental analysis was carried out after microwave-assisted digestion using inductively coupled plasma mass spectrometry (ICP-MS) in 18 samples. Eleven elements (V, Cr, Mn, Co, Ni, Cu, Zn, As, Se, Cd, and Pb) were analysed. Their average concentrations ranged from 0.006 µg g-1 to 7.13 µg g-1 in both orange juice and flesh (wet weight). There was a high increase in the average concentrations of all elements in the juice compared to the flesh of up to 40-fold. |

Sci., 2021, 66(3):78-86 | DOI: 10.17221/231/2020-CJAS In the last decade, the pig sector in Slovakia has been changing continuously, especially in regard to the proportion of pigs coming from foreign breeding programs. These changes lead to changes in carcass characteristics and to a change in the distribution of carcasses within the classes of the Union scale for the classification of pig carcasses. Therefore, this study was undertaken to analyse the actual state of pig carcass classification in the Slovak Republic and to investigate the needs of updating the equations for the classification methods and possible steps to improve the quality of pork produced within the country. Results showed that an increased number of pig carcasses originated from Danish breeding program, and also from the Czech Republic and Poland. When compared to carcasses in 2009, these changes resulted in lower backfat thickness and higher muscle thickness of the longissimus dorsi muscle and thus higher lean meat content of carcasses. This results in a higher number of carcasses classified in the S and E classes. Slight differences between two instrumental classification methods were calculated. The study showed the need for updating the equations for classification methods, and also suggested possible further steps in order to improve the quality of pork. |

In this study, we analysed codon usage patterns for the acetolactate synthase (ALS) gene in eight monocot weeds and one model monocot. The base composition at the third codon position follows C3 > G3 > T3 > A3. The values of the effective number of codons (ENC or Nc) indicate low bias, and ENC or Nc vs. GC3 plot suggests that this low bias is due to mutational pressure. Low codon adaptation index and codon bias index values further supported the phenomenon of low bias. Additionally, the optimal codons, along with over- and under-represented codons, were identified. Gene design using optimal codons rather than overall abundant codons produce improved protein expression results. Our results can be used for further studies, including eliciting the mechanisms of herbicide resistance (occurring due to elevation of gene expression levels) and the development of new compounds, their efficiency and risk assessment for herbicide resistance evolution. |
Residual effect of superphosphate on the sulphur status of soil and plants in a long-term NPK fertilisation experiment on a Chernozem in HungaryOriginal PaperEvelin Kármen Juhász, Rita Kremper, Áron Béni, Andrea Balláné KovácsPlant Soil Environ., 2021, 67(11):625-632 | DOI: 10.17221/247/2021-PSE Recently, plant-available sulphur (S) in the soil is decreasing due to the limited use of S containing fertilisers and the reduction of atmospheric S deposition. The aim of this work was to evaluate the S status in a long-term fertilisation experiment on a Chernozem in Hungary, with control and 2 NPK rate treatments, considering that after 27 years of superphosphate (SP) use, SP was replaced by monoammonium phosphate in 2010. Plant and soil sampling were performed in 2017 at three different development stages of winter wheat. To assess the S status, the S balance was estimated (for 34 years), KCl soluble soil sulphate, S as well as nitrogen (N) concentration and some amino acids in wheat grain were measured. N/S ratios, S and N uptake of wheat were calculated. The residual effect of SP could be measured only in terms of KCl soluble SO42--S in soil. According to the wheat grain S concentration (0.08-0.10%) and N/S ratio (14.9-22.0), wheat was S deficient, despite the positive S balance in the fertilised plots. In this experiment, where S fertiliser was applied with 84 kg S/ha dose in 1983-2010, followed by a 7-year period without S fertilisation, S supply is necessary for achieving adequate wheat quality. |
Treatment of extremity fractures in 20 wild birds with a modified Meynard external fixator and clinical assessment of the resultsOriginal PaperK Gerbaga Ozsemir, K AltunatmazVet Med - Czech, 2021, 66(6):257-265 | DOI: 10.17221/5/2020-VETMED The aim of this study is to evaluate the outcome of the treatment on twenty wild birds presenting with fractures that were stabilised with modified Meynard external fixators. The study material consisted of a total of twenty birds of eight different species, six white storks (Ciconia ciconia), one eastern imperial eagle (Aquila heliaca), one European honey buzzard (Pernis apivorus), one long-legged buzzard (Buteo rufinus), three common buzzard (Buteo buteo), one northern goshawk (Accipiter gentilis), one Armenian gull (Larus armenicus), and six yellow-legged gulls (Larus michahellis), which were diagnosed with fractures in their extremity bones following clinical and radiological examinations. The lesions of the twenty cases evaluated in the study were diagnosed as humerus fractures in nine cases, ulna fractures in three cases, tibiotarsus fractures in seven cases and a femur fracture in one case. The external fixators were removed between weeks five and nine postoperatively. While healing was seen in twelve of the twenty evaluated cases, the complete functional recovery occurred in six of these and they were released into nature. It was concluded that modified Meynard external fixator, which was made applicable for fractures in birds, could be an alternative method for fracture treatments in wild birds. |

In this study, total phenolic content, antioxidant activity and volatile compounds of 13 different honey samples collected from various districts of Malatya province were investigated. As a result of this study, it was determined that the total phenolic content varied between 8.50 mg GAE 100 g-1 and 73.90 mg GAE 100 g-1 and it was observed that the honey samples were rich in aldehydes, aliphatic acid and esters, alcohols, hydrocarbons, carboxylic acid esters, ketones, terpenes, fatty acids and esters. In addition, the antibacterial effects of honey samples were determined against 18 different pathogenic bacteria using agar well diffusion (AWD) method, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). In AWD assay, it was recorded that inhibition zone diameters varied between 9 mm and 14 mm and honey samples were found to have a partial inhibitory effect against selected target pathogens. |

Sci., 2021, 66(10):412-419 | DOI: 10.17221/80/2021-CJAS Light during incubation can influence embryonic and postembryonic development of chickens, but the underlying mechanisms are poorly understood. Previous studies have demonstrated that red and blue lights during incubation had opposite effects on the development of embryonic melatonin biosynthesis; red light results in the highest and blue light in the lowest amplitude of the daily rhythm. Therefore, in this study, we investigated if exposure to monochromatic red (632 nm) and blue (463 nm) light during incubation can differently influence growth, selected biochemical (glucose, cholesterol, triacylglycerols) and endocrine (corticosterone and thyroid hormones) traits and behavioural parameters during postembryonic development in broiler chickens. For analysis, we used 10 and 11 hatchlings incubated in red and blue light, respectively and 10 birds per each group (six males and four females) in 3-weeks-old broilers. During the rapid growth phase (days 18, 20 and 21 of age), higher body weight was recorded in broilers incubated under red compared to blue light, whereas endocrine and metabolic traits did not differ between the treatments. The improved growth rate was related to behavioural traits, mainly because chickens incubated in red light exhibited more passive (resting, standing, preening, dust bathing) and less active behaviours (walking, foraging, fighting, wing-flapping) than the blue-light incubated birds. The time spent for eating and drinking and the results of the tonic immobility test did not differ between both groups. Our results suggest that red and blue monochromatic light during incubation can differently program the postembryonic development of broilers, with possible consequences for their growth and welfare. |
