Blood chemistry reference intervals of captive Asiatic black bears (Ursus thibetanus)Original Paper

Data on blood chemistry values can make fundamental contributions to our understanding of physiological changes. However, there is a lack of information regarding blood chemistry in Asiatic black bears (Ursus thibetanus). Thus, the objects of this study were to determine reference ranges for 29 blood chemistry variables, and to evaluate differences between age groups and between seasons. Blood samples (n = 138) were collected from 44 (20 males, 24 females; age range, 1-15 years) clinically healthy, captive Asiatic black bears (Ursus thibetanus) in the Republic of Korea. Young and adult bears showed significantly higher levels of creatinine and total cholesterol, and lower levels of blood urea nitrogen, blood urea nitrogen/creatinine ratio, lactate dehydrogenase and creatine kinase MB during hibernation compared to during non-hibernation. Adults also showed significantly higher levels of triglyceride, but lower levels of inorganic phosphorus, aspartate transaminase, alkaline phosphatase, high density lipoprotein cholesterol and creatine phosphokinase during hibernation than during non-hibernation. During hibernation, the urea nitrogen/creatinine ratio and levels of alkaline phosphatase, lactate dehydrogenase, and creatine phosphokinase in young bears were significantly higher than in adults, whereas creatinine levels were lower than in adults. During non-hibernation, the urea nitrogen/creatinine ratio and levels of calcium, alkaline phosphatase, lactate dehydrogenase, creatine phosphokinase and creatine kinase MB in young bears were significantly higher, whereas creatinine, total protein, albumin, gamma-glutamyl transferase and haemoglobin levels were lower than in adults. The results of this study provide reference values that will aid in understanding the physiology of Asiatic black bears and in assessing the health of these animals in captive environments.

Effects of selenium on selenoprotein synthesis and antioxidant parameters of bovine mammary epithelial cellsOriginal Paper

This study aimed to investigate the effects of selenium (Se) on the selenoproteins synthesis and antioxidant parameters of bovine mammary epithelial cells (BMECs). The experiment was conducted as a single factor completely randomized design to explore the effect of different levels of Se supplementation (0, 10, 20, 50, and 100 nmol/l) on selenoproteins synthesis and antioxidant parameters of BMECs, and to screen the appropriate dose of Se supplementation ensuring a better antioxidant function. Se supplementation increased cell proliferation, the activities of glutathione peroxidase (GPx) and superoxide dismutase, total antioxidant capacity and seleoprotein P (SelP) content, and decreased reactive oxygen species and malondialdehyde levels in a dose-dependent manner. Se supplementation of 50-100 nmol/l had a better effect. Se supplementation also increased thioredoxin reductase (TrxR) activity in a dose-dependent manner, and Se supplementation of 20-50 nmol/l had a better promoting effect. The dose-dependent response between Se supplementation and mRNA and protein expression of GPx1 and TrxR1, as well as SelP mRNA expression was also observed in this experiment. The mRNA and protein expression of GPx1 was up-regulated with the addition of 50-100 nmol/l Se, and the mRNA expression of TrxR1 and SelP was up-regulated with the addition of 20-100 nmol/l Se. Results indicated that Se supplementation of 50 nmol/l had a better promoting effect on the selenoproteins synthesis and antioxidant parameters of BMECs.

Environmental and genetic effects on cadmium accumulation capacity and yield of maizeOriginal Paper

Maize (Zea mays) is an economic crop suitable for use in phytoremediation in low to moderately cadmium (Cd)-contaminated soils due to its ability to accumulate high concentration of Cd in parts of maize that are not used in human diet. The aim of this study was to test Cd content in nine female parents of the commercial maize hybrids (C1 = ♀2-48; C2 = ♀1767/99; C3 = ♀87-24; C4 = ♀135-88, C5 = ♀84-28; C6 = ♀84-44; C7 = ♀438-95; C8 = ♀30-8; C9 = ♀B-73) grown under field conditions in two soils (B1: eutric cambisol, B2: stagnosol) during three growing seasons (A1: 2006, A2: 2007, A3: 2008). The stationary trial was conducted in four replicates. The ear-leaves at flowering and grain at maturity were taken for chemical analysis. The average quantities of leaf-Cd were 0.081, 0.088 and 0.143 mg per kg of dry matter for A1, A2 and A3, 0.089 and 0.118, for B1 and B2, respectively. Grain-Cd was below the threshold (< 0.02 mg/kg). Five Cd-inefficient genotypes (C3, C5, C6, C7 and C9) had low leaf-Cd (average 0.049 mg/kg), while this content was about 6-times higher (average 0.299 mg/kg) in Cd-efficient genotype C4. The yield among the years ranged from 2.36 to 4.31 t/ha. Maize grown on B2 had about 26% lower yield than on B1. Five genotypes (C1, C2, C8 and C9) achieved yields less than 3.50 t/ha (mean 3.15 t/ha), while in two genotypes (C3 and C5) yields were above 4.00 t/ha (mean 4.14 t/ha). Very strong correlations (r) of leaf-Cd status among years (ranged from 0.52 to 0.77) confirmed high genetic effect on the capability of Cd accumulation in maize. However, correlations between Cd content and yield were low (ranged from -0.17 to 0.06). Cd-efficient C4 female parent could be used for development of maize hybrids suitable for phytoremediation, while Cd-inefficient female parents for hybrids could be suitable as forage maize crop contributing to the lower Cd input into food chain.

Effects of gravel-sand mulch on the runoff, erosion, and nutrient losses in the Loess Plateau of north-western China under simulated rainfallOriginal Paper

Gravel mulching is a characteristic agricultural technique that has been used for hundreds of years in the north-western Loess Plateau of China. However, the effects of the gravel-sand mulch on the processes of the runoff, soil erosion, and nutrient losses are neither fully distinguished nor even known in many parts of the world. This study investigated how different gravel particle sizes in the mulch affected the runoff, erosion as well as the extent of the nutrient losses in the surface runoff. The laboratory experiments were conducted using a rainfall simulator with three gravel mulch treatments: (1) fine gravel mulch (FG); (2) medium gravel mulch (MG); (3) coarse gravel mulch (CG) and a control group, bare soil (BS). The results of these rainfall simulation experiments gave estimates on how the grain size influences the runoff and losses of the soil and its nutrients. Applying the gravel mulch significantly delayed the runoff's starting time when compared with the bare soil. Both the total runoff and soil loss increased with the grain size of the gravel mulch. Compared with the bare soil, the lowest surface runoff and soil loss was observed from the fine gravel treatment. These results clearly show that gravel mulch plays an important role in the runoff and sediment generation processes, and that it significantly reduces the surface runoff and soil loss. The losses of the total nitrogen (TN), total phosphorus (TP), and total organic carbon (TOC) from the bare soil were much higher than those under the gravel mulching. The fluctuations in these nutrient-loss processes were the most intense in the CG treatment, while the TC content, in initial runoff, was significantly higher in the FG than the other treatments. Our findings suggest gravel mulch is a useful water and soil conservation technique in the loess area of north-western China, and these results can inform one on the theoretical principles for properly utilising gravel-mulched fields.

Soil physical properties and crop status under cattle manure and Z'Fix in Haplic ChernozemOriginal Paper

A three-year experiment was conducted to investigate the effect of Z'Fix on soil physical properties and crop status. Z'Fix is an agent recommended as an addition to animal bedding to prolong its function and to lower ammonia emissions in stables. Concurrently, a positive effect on organic matter transformation in resulting manure is claimed. The experiment involved control, farmyard manure (FYM), and farmyard manure with Z'Fix (FYM_ZF) as variants. In-field sampling was conducted for cone index, water infiltration and implement a unit draft, where the latter two showed significant differences in favour of FYM_ZF. Also, concerning crop yields, FYM_ZF consistently attained the highest values, followed by FYM throughout all three seasons. Furthermore, remotely sensed data were analysed to describe crop status via normalised difference vegetation index where significant differences were found across all variants. Based on the study, FYM_ZF demonstrated positive effects both on soil properties and crop conditions.

Clinical and immunohistochemical findings of splenic mast cell tumour in a cat: A case reportCase Report

A 6-year-old, spayed, female, domestic shorthair cat presented with a 4-month history of chronic intermittent vomiting and anorexia. The haematologic results indicated moderate anaemia and a circulating mast cell population. The abdominal radiography revealed a markedly enlarged spleen. The cytological analysis of the spleen showed a uniform population of mast cells, and a diagnosis of systemic mastocytosis (splenic mast cell tumour with mastocytaemia) was made. This diagnosis was subsequently confirmed by the histopathological examination of the spleen. The immunohistochemistry for KIT showed KIT pattern II (focal cytoplasmic expression). A splenectomy and chemotherapy with vinblastine and prednisolone resulted in remission of the anaemia and other clinical signs. This case report highlights the importance of cytological evaluations of peripheral blood smears and/or aspirates of enlarged spleens for diagnosing splenic mast cell tumours and for quickly initiating the appropriate treatment.

Evaluation of genetic diversity of subdivided genealogical groups in Lithuanian Trakehner horse population using immunogenetic toolsOriginal Paper

The objective of this study was to evaluate the inter-breed genetic diversity of Lithuanian Trakehner horses. The current population in Lithuania numbers 1 039 Trakehner horses. The study included the immunogenetic data analysis of 316 blood samples. Pedigree was traced back to 7-11 generations. The horses were assigned to genealogical clusters originating from East Prussian Trakehner Stud, Thoroughbred and Arab sires. The first two clusters were subdivided into nine-horse groups according to the most important ancestors. The evaluation of genetic diversity was based on six blood groups and five protein polymorphism systems. The differences between genealogical groups were confirmed by principal component analysis and applied cluster analysis. According to the allelic frequency, the Trakehner population changed very slightly through a 30-year period, and only one rare (0.013) allele TfRR (frequent in other Trakehner populations), has been newly found. The analysis of genetic polymorphism systems based on a very low rate of alleles Qabc (0.093) and Ddkl (0.046), which are typical of other populations, indicated the exclusivity of the Trakehner population in Lithuania. The average expected heterozygosity by blood protein polymorphism and blood groups were 0.409 and 0.441, respectively. The genetic clustering diverged by observed heterozygosity of blood groups and by pedigree data in Pilger through Egoist, Bay Ronald through Dark Ronald and Dampfross through Hyperion subgroups. The determined distinctiveness of subdivided Trakehner horse groups suggests reconsidering the breeding strategies and conservation programme of Trakehner horses. Breeding and selection of subdivided sire lines could be among the appropriate solutions for the maintenance and extension of the genealogical structure.

Cystic compound odontoma in a dog: Diagnosis and managementCase Report

Tumour of the oral cavity is a rare condition in young animals. The most frequent are odontomas - locally invasive tumours with no metastatic potential. The article describes a case report of 4-month-old male puppy diagnosed with mandibular odontoma with cystic appearance. Authors present clinical features, histopathological examination of odontoma and the surgical management using deep curettage and hydroxyapatite granules deposition. Odontomas can form bone cyst-like structure padded with membrane forming denticles. A deep curettage is acceptable method of treatment but should be supplied with hydroxyapatite deposition if the cyst is present.

Insights into the development of electrokinetic remediation technology: A bibliometric analysisOriginal Paper

Electrokinetic remediation (EKR) is a powerful technique aimed at pollutant removal in soil, sludge, mine tailings, and so on. In the current work, we performed a bibliometric analysis of the research on EKR for the period of 1900-2018 on the basis of the core database of the Science Citation Index Expanded. In addition to a basic analysis of the research characteristics, keywords were analysed for four major participants: USA, China, Spain, and South Korea. The periods of 1990-2001, 2002-2008, and 2009-2018 were studied using the keyword analysis method to gain insights into the development of EKR and predict its future trends. The results revealed that the related research field in the USA was broad during the study periods. Meanwhile, China was interested in fluorine pollution and contamination in red soil. Spain paid close attention to pollution due to agricultural contamination. South Korea focused on radioactive element pollution. The number of papers published over a period of 28 years increased steadily and continued to rise after 2008. The combined techniques of EKR + phytoremediation and EKR + bioremediation were successively utilised by scholars over time, and the latter is expected to demonstrate vitality in the future.

Principal component analysis of conformation traits in Hungarian Simmental cowsOriginal Paper

The aim of the current research was to analyze the linear type traits of Hungarian Simmental dual-purpose cows scored in the first lactation using principal component analysis and cluster analysis. Data collected by the Association of Hungarian Simmental Breeders were studied during the work. The filtered database contained the results of 8 868 cows, born after 1997. From the evaluation of main conformation traits, the highest correlations (r = 0.35, P < 0.05) were found between mammary system and feet and legs traits. Within linear type traits, the highest correlation was observed between rump length and rump width (r = 0.81, P < 0.05). Using the principal component analysis, main conformation traits were combined into groups. There were three factors having 84.5 as total variance ratio after varimax rotation. Cluster analysis verified the results of the principal component analysis as most of the trait groups were similar. The strongest relationship was observed between feet and legs and mammary system (main conformation traits) and between rump length and rump width (linear type traits).

Effects of hydrogen peroxide application on agronomic traits of rice (Oryza sativa L.) under drought stressOriginal Paper

Drought stress is a major environmental factor limiting crop growth and productivity. Hydrogen peroxide (H2O2) plays an essential role during stress response by acting as a signal molecule that activates multiple stress tolerance mechanisms. In this study, the effects of H2O2 on agronomic traits were studied in rice (Oryza sativa L.) cv. Khao Dawk Mali 105 (KDML 105) was subjected to drought stress. H2O2 was applied by either seed priming or foliar application method with a concentration of 1, 5, and 15 mmol/L. The results showed that both seed priming and foliar application with H2O2 improved some yield components. The tiller numbers, number of panicles, number of filled grains, filled grain weight, and harvest index were improved approximately 1.13, 1.04, 1.23, 1.21, and 1.1 times compared to the untreated plants. Foliar application, however, helps the plant by reducing yield loss as indicated by a 0.5-time reduction in the number of unfilled grain and lower unfilled grain weight. It was suggested that 5 mmol/L H2O2 was the most effective concentration to alleviate the effect of drought stress during the reproductive stage in rice.

Pathogenic and biological characterisation of T-DNA insertional mutants of a Colletotrichum gloeosporioides casual organism of apple anthracnoseOriginal Paper

Anthracnose leaf spot caused by Colletotrichum gloeosporioides is an important disease of apples, resulting in serious damage to the fruit production. In this paper, the pathogenic and physiological characters of sixteen isolates and the wild isolate "Stj16" were studied. In the current study, we generated C. gloeosporioides strains expressing green fluorescence by introducing a GFP gene via an Agrobacterium tumefaciens-mediated transformation (ATMT). To confirm the subcellular localisation of the A2799 gene, an A2799gfp fusion expression mutant was constructed. After observation of the fusion expression, the A2799gfp fusion expression protein was located in the peroxisomes of the cell. The pathogenicity results showed that the mutants A4204, M44, A1919, A3638 and A1598 lost the pathogenic capability and virulence, however, the virulence of the mutants A1764, A439, A3885, G1183, A3144, A1649 and A2675 increased significantly to the apple fruits compared to the wild isolate "Stj16". The biological study indicated that a Rose Bengal Agar (RBA) medium decreased the mycelium growth, but it can increase the sporulation for most of the isolates. The mutant A4204 does not grow well at pH 4.0 and pH 8.0, and mutant M44 just has the optimum growth at pH 8.0, and a 12 h light and 12 h dark condition stimulates the sporulation for most of the tested mutants, but the A1764 mutant more sporulated at regular dark conditions. All the mutants and "Stj16" grew vigorously at 25 °C-30 °C, for "Stj16", it produced the highest number of conidia at 30 °C compared with the other temperatures. Based on the biological study, we found the best growing and sporulation conditions for all the tested isolates. The information generated in the present study will facilitate molecular research on this devastating fungus.

Comparison of in vitro (fish cell line) and in vivo (fish and crustacean) acute toxicity tests in aquatic toxicologyOriginal Paper

The use of in vitro (fish cell lines) is a cost-effective, very rapid, and informative tool for toxicological assessments. Using the neutral red (NR) assay, we compared the in vitro acute toxicity (20hEC50) of twenty-six chemical substances on a rainbow trout gonad cell line (RTG-2) with their in vivo acute toxicity to Barbados Millions Poecilia reticulata (48hLC50, OECD 203) and crustacean Daphnia magna (48hEC50, OECD 202). The 20hEC50 values obtained by the NR assay were higher in nearly all the cases when compared to the 48hLC50 in P. reticulata and the 48hEC50 in D. magna, indicating that the sensitivity of the RTG-2 cell line was lower compared to P. reticulata and D. magna. A high (r = 0.89) and significant (P < 0.001) correlation was recorded between the 20hEC50 values of the RTG-2 and the 48hEC50 values of D. magna. The correlation between the 20hEC50 values of the RTG-2 and the 48hLC50 values of P. reticulata was lower (r = 0.65; P < 0.001), but also significant. The authors recommend use of the NR assay on the RTG-2 cell lines as a screening protocol to evaluate the toxicity of xenobiotics in aquatic environments to narrow the spectrum of the concentrations for the fish toxicity test.

Content of nitrates and nitrites in unprocessed raw beefOriginal Paper

The aim of the study was to examine the contents of nitrates and nitrites in raw beef coming from cattle of the Polish Holstein-Friesian breed, black-and-white type, originating from different parts of Poland. The research material comprised semitendinosus and longissimus thoracis muscles obtained from 176 heads of cattle. The content of nitrites in the examined muscles ranged from 0.6 to 13.3 mg kg-1 in rump muscles and from 0.8 to 13.7 mg kg-1 in muscles of the back. The most samples were characterised by the presence of nitrites in the range from 1 to 5 mg kg-1, and the least samples from 11 to 14 mg kg-1. The contents of nitrates were significantly higher, ranging from 10.2 to 73.5 mg kg-1 in the semitendinosus muscle and from 10.4 to 74.3 mg kg-1 in the thoracis longissimus muscle. Such and higher level of contamination may be the cause of meat discolouration after heat treatment despite the absence of curing ingredients. It is necessary to continuously monitor the concentration of nitrates in raw materials, water and feed to ensure the complete safety of food of animal origin.

Growth performance, mortality and body and carcass characteristics of rabbit fatteners related to crossbreeding of Mecklenburger Schecke sires with dam line of HYLA rabbitsOriginal Paper

The aim of the study was to evaluate the effect of crossing Mecklenburger Schecke (MS) males with the maternal commercial line of HYLA rabbits on the growth performance, mortality and body and carcass components of fattened rabbits under intensive farming conditions. The control (C) group consisted of albino crossbred rabbits of the HYLA combination AB sires × CD dams. The tested (T) group (MS sires × HYLA CD dams) consisted of rabbit crosses between the MS sires and the same dam line as in the C group. Rabbits of both groups were raised and fattened under identical nutritional and management conditions. The crossing of MS males with the maternal line of HYLA rabbits led to the acceptable growth performance of rabbits in the T group. The higher slaughter weight of rabbits in the T group as compared to rabbits in the C group (P < 0.01) as associated with the higher weight of hot carcass and higher proportions of fore and intermediate parts of carcass (P < 0.01). On the contrary, HYLA rabbits had a higher proportion of the hind part of carcass (P < 0.001); yields of hind leg meat and musculus longissimus thoracis et lumborum were not influenced by the genotype. However, since higher mortality was found in rabbits of the T group during fattening, it is not possible to recommend the MS breed as a common sire line used under conditions of intensive farming at this moment. Regarding the rabbit sex, slaughtered females showed a higher proportion of liver and a lower dressing percentage associated with the worse carcass compactness compared to males.

Selenium effect on wheat grain yield and quality applied in different growth stagesOriginal Paper

Small field plot experiments were carried out at the testing station of the Central Control and Testing Institute in Agriculture in Veµký Meder (Slovakia) in the experimental years 2014/2015, 2015/2016 and 2016/2017. Selenium salts in the form of sodium selenite and sodium selenate were applied in growth phases: end of tillering (BBCH 29) and flag leaf ligule and collar visible (BBCH 39). The effect of experimental years 2014/2015, 2015/2016 and 2016/2017 on the yield of wheat grain was not statistically significant within the observed variants. The achieved mean yields were in the range from 10.06 ± 0.81 to 11.07 ± 0.29 t/ha in 2014/2015, from 9.82 ± 0.54 to 10.32 ± 0.10 t/hain 2015/2016 and from 11.23 ± 0.76 to 11.64 ± 0.51 t/ha in 2016/2017. Selenate in comparison with selenite influenced the selenium accumulation in wheat grains more positively. However, a significant difference was recorded in variants with selenite application in the flag leaf growth phase in comparison with the end of tillering phase. The influence on the content of macroelements P, K, Ca and microelements Cu and Fe was observed in sodium selenite only; its application decreased the element content in comparison with the control variant. Statistically significantly higher values of fiber and fat were achieved after application of selenium in the flag leaf growth stage in comparison with the end of tillering.

Effects of harvest stage and diurnal variability on yield and essential oil content in Mentha × piperita L.Original Paper

This study aimed at investigating the effect of ontogenetic and diurnal variability on yield and quality criteria of Mentha piperita. The research was conducted in the Ordu province of Turkey for two years. The trial was established in a randomised complete block design with 3 replications according to factorial regulations. Four different harvesting periods (budding, first flowering, 50% flowering, 100% flowering) and three harvesting hours (9:00 a.m., 1:00 p.m., 5:00 p.m.) were evaluated. A significant difference was determined between the trial years in all characters examined: plant height 55.9-69.0 cm, fresh herb yield 3 236-16 438 kg/ha, drug-herb yield 1 748-2 862 kg/ha, fresh leaf yield 3 477-4 506 kg/ha, drug leaf yield 968-1 253 kg/ha, essential oil ratio 2.0-2.5% and essential oil yield that varied between 19-28 L/ha. The highest yields were obtained at the beginning of flowering in drug leaf yield and 100% flowering period for all other characteristics. Diurnal variability was statistically significant only in the rate of essential oil, and the highest rate occurred in the 9:00 a.m. harvest.

Stimulation of ex vitro growth of Rhododendron hybrids 'Nova Zembla' and 'Alfred' by inoculation of roots with Serendipita indicaOriginal Paper

Experiments were carried out to study the consequences of inoculating Rhododendron 'Nova Zembla' and 'Alfred' microcuttings and young plants with Serendipita indica (formerly Piriformospora indica), an endophytic fungus. The inoculation at the in vitro rooting stage decreased the in vitro root and shoot quality, but after planting microplants in a greenhouse in a non-fertilised peat substrate, the plants grew 33% taller, with 10% more leaves and with more abundant roots in comparison with the non-inoculated microplants. The young plants rooted in vitro, acclimated in a greenhouse in a peat substrate and inoculated with S. indica at the time of the transplantation to the pots displayed, after 15 months, an increase in the shoot lengths by 35% and 13%, depending on the cultivar, in the leaf number by 47% and in the chlorophyll content by 31% when compared with the non-inoculated plants. The beneficial effect of S. indica on the young Rhododendron plants indicates the possibility of its practical use in the nursery production of this plant.

The effect of low-intensive coherent seed irradiation on germinant growth of Scots pine and sugar beetOriginal Paper

The success of forest and agricultural plant establishment program mainly depends on the quality of reproductive material. The study intends to offer engineers and farm owners a solution for small-size seed improvement before sowing. The effect of low-intensity coherent light on the seeds of various crops is theoretically and empirically hypothesized. The seedlots of Scots pine (Pinus sylvestris L.) and sugar beet (Beta vulgaris L.) of Russian diploid hybrid RMS-127 were germinated in a controlled environment. The germinants were produced from six seed fractions, previously irradiated with 1.274 W.m-2 at the 632.8 nm wavelength with 1, 2, 3, 5, 10, 15 min exposure to a standard laser system, plus untreated control. Pine germinants were measured on day 15, beetroot on day 10 after germination. An increase in exposure time reduced Scots pine germination energy and capacity, while for sugar beet the results were not conclusive. On the contrary, increasing the exposure time had a positive effect on both the height and biomass growth of both Scots pine and sugar beet germinants. The 10-min exposure time resulted in maximum values for sugar beet height and biomass and Scots pine height, while the 15-min exposure time produced maximum Scots pine biomass.

Phylogenetics of native conifer species in Vietnam based on two chloroplast gene regions rbcL and matKOriginal Paper

We used two chloroplast gene regions (matK and rbcL) as a tool for the identification of 33 local conifer species. All 136 sequences, 101 newly generated (14 species for gene matK; 16 species for gene rbcL) and 35 retrieved from the GenBank, were used in the analysis. The highest genetic distance (matK region) was recorded between the species in Cupressaceae with an average of 5% (0.1-8.5), Podocarpaceae with an average of 6% (0-8.5), Taxaceae with an average of 5% (0.2-0.5) and Pinaceae with an average of 20.4% (0.8-54.1). The rbcL region showed a low genetic distance between the species in Cupressaceae 2% (0-3.3), Podocarpaceae 3% (0.6-3.4), Taxaceae 1% (0-2.1) and Pinaceae 1.2% (0-5.82). The phylogenetic analyses using the Maximum likelihood (ML) and Bayesian inference (BI) bootstrap values obtained at the branching nodes of each species ranged from 62 to 100% (Maximum likelihood bootstrap - MLBS and Bayesian posterior probabilities - BPP) for the matK gene; from 66 to 100% (MLBS) and 60 to 100% (BPP) for the rbcL region. The rbcL region was not identified between the species of Taxaceae and Cephalotaxaceae. The matK gene region was very clear in the different species among the families (Cupressaceae, Podocarpaceae, and Cephalotaxaceae) and unsuitable for identifying closely related species in Amentotaxus (Taxaceae) and Pinus (Pinaceae). The gene (matK) is a useful tool as a barcode in the identification of conifer species of Cupressaceae, Podocarpaceae, and Cephalotaxaceae in Vietnam.

Identification of the optimal codons for acetolactate synthase from weeds: an in-silico studyOriginal Paper

Although various studies of codon usage bias have been reported in a broad spectrum of organisms, no studies to date have examined codon usage bias for herbicide target genes. In this study, we analysed codon usage patterns for the acetolactate synthase (ALS) gene in eight monocot weeds and one model monocot. The base composition at the third codon position follows C3 > G3 > T3 > A3. The values of the effective number of codons (ENC or Nc) indicate low bias, and ENC or Nc vs. GC3 plot suggests that this low bias is due to mutational pressure. Low codon adaptation index and codon bias index values further supported the phenomenon of low bias. Additionally, the optimal codons, along with over- and under-represented codons, were identified. Gene design using optimal codons rather than overall abundant codons produce improved protein expression results. Our results can be used for further studies, including eliciting the mechanisms of herbicide resistance (occurring due to elevation of gene expression levels) and the development of new compounds, their efficiency and risk assessment for herbicide resistance evolution.

Residual effect of superphosphate on the sulphur status of soil and plants in a long-term NPK fertilisation experiment on a Chernozem in HungaryOriginal Paper

Recently, plant-available sulphur (S) in the soil is decreasing due to the limited use of S containing fertilisers and the reduction of atmospheric S deposition. The aim of this work was to evaluate the S status in a long-term fertilisation experiment on a Chernozem in Hungary, with control and 2 NPK rate treatments, considering that after 27 years of superphosphate (SP) use, SP was replaced by monoammonium phosphate in 2010. Plant and soil sampling were performed in 2017 at three different development stages of winter wheat. To assess the S status, the S balance was estimated (for 34 years), KCl soluble soil sulphate, S as well as nitrogen (N) concentration and some amino acids in wheat grain were measured. N/S ratios, S and N uptake of wheat were calculated. The residual effect of SP could be measured only in terms of KCl soluble SO42--S in soil. According to the wheat grain S concentration (0.08-0.10%) and N/S ratio (14.9-22.0), wheat was S deficient, despite the positive S balance in the fertilised plots. In this experiment, where S fertiliser was applied with 84 kg S/ha dose in 1983-2010, followed by a 7-year period without S fertilisation, S supply is necessary for achieving adequate wheat quality.

Optimisation of concurrent Calophyllum oil-resin extraction and separationOriginal Paper

This research optimised the application of a hexane-methanol mixture as a binary solvent for the concurrent oil-resin extraction and separation from Calophyllum seeds on a pilot scale, in a direct stage. The optimum oil and resin yields were determined by optimising the extraction conditions using response surface methodology and a second order polynomial model. The extraction conditions affected the oil and resin yields, with the extraction time as the biggest influencing factor. Optimum oil (65%) and resin (16%) yields were predicted to be obtained at 5.2 h and 433 rpm. The model validation with these extraction conditions showed that the predicted results and actual oil (62%) and resin (15%) yields were in passable agreement. The oil was composed of 75.4% triglycerides with a density of 0.874 g.cm-3, a viscosity of 26.4 mPa.s-1, an acid value of 46.4 mg KOH.g-1, an iodine value of 98.0 g iodine.100 g-1, trace water and sediment contents, and zero ash content. The resin had a viscosity of 4 694.8 mPa.s-1, a total phenolic content of a 4.51% gallic acid equivalent, an antioxidant activity of an 8.82 mg ascorbic acid equivalent.g-1, and an acid value of 126.2 mg KOH.g-1.

Morphological variation of male A. arguta plants affects their flowering potential and pollen efficiencyOriginal Paper

Actinidia arguta (Siebold et Zucc.) Planch. ex Miq. is functionally a cryptic dioecious plant and successful pollination is crucial for achieving high quality fruit. The extent and bases of morphological variability in female genotypes have been well studied, but here we focus on the males. Mature plants of seven male A. arguta genotypes were studied in 2016 and 2017 during which a suite of phenological and morphological features was measured on ten randomly chosen one-year-old canes on each plant. This analysis was complemented by two other, specialized measures potentially related to pollination efficiency, namely pollen quality, and quantity. The time of bud break was similar each year across all genotypes and the pollen quality was uniformly very high (viability - 95.0-99.9% and germination - 89.0-97.3%). However, the ten genotypes exhibited high variability in both the number of flowers per inflorescence and pollen quantity, indicating marked differences in their flowering potential and efficiency as pollinizers. The male kiwiberry indicators developed in this study - flowering potential and pollen efficiency, appear to be effective tools for the comparative evaluation of male A. arguta genotypes.

Intracranial meningioma in a pet rat: MRI findingsCase Report

We herein report the magnetic resonance imaging (MRI) findings in a brain meningioma in a pet rat (Rattus norvegicus domestica). A 1.5-year-old pet rat was referred for depression, ataxia, tremors and bilateral nystagmus; a brain MRI was elected suspecting an intracranial neoplasia. The study was performed with a 0.25 T scanner. The MRI revealed a well-defined oval mass with a heterogeneous appearance, hyperintense in T1 and T2, and with extra-axial localisation and severe brain compression, the mass appeared strongly and diffusely contrast-enhancing. Euthanasia was elected, with the necropsy confirming the presence of a voluminous extra-axial mass. Histologically, a well-differentiated, benign meningotheliomatous meningioma was diagnosed, with the histochemical stains allowing one to exclude a granular cell tumour. To the best of our knowledge, this is the first report of MRI investigations on a pet rat intracranial meningioma.

Identification of lactic acid bacteria and yeasts from traditional sourdoughs and sourdough production by enrichmentOriginal Paper

The subject of this study was to investigate lactic acid bacteria (LAB) and yeasts microbiota of traditional sourdough (n = 36) and to indicate characteristics of enriched sourdough that is produced from combinations of isolates. A total of 60 LAB and 40 yeasts were identified from sourdough by matrix-assisted laser desorption ionisation-time-of-flight mass spectrometry (MALDI-TOF MS) analysis. The dominant LAB microflora was Lactobacillus brevis (43.33%), Pediococcus acidilactici (21.67%) and Lactobacillus plantarum (18.33%). The dominant yeasts microflora was Saccharomyces cerevisiae (27.5%), Pichia kudriavzevii (25.0%) and Kluyveromyces marxianus (12.5%). The sourdough prepared with the combination of L. brevis, Leuconostoc mesenteroides, P. acidilactici and S. cerevisiae, K. marxianus showed the best physicochemical and microbiological properties while that with L. plantarum, L. brevis and P. kudriavzevii, Wickerhamomyces anumalus was the poorest. LAB and yeasts in the sourdoughs were ranged from 6.58 log CFU g-1 to 9.12 log CFU g-1 and from 6.12 log CFU g-1 to 7.88 log CFU g-1, respectively. Various chemical parameters such as pH, total titratable acidity (TTA), ethanol, and sourdough volume were differ depending on the type of microbial species. TTA was more pronounced in the sourdoughs produced with homofermentative LAB. Yeasts and LAB were dominated during continuous enriching of sourdough, supporting an important role during fermentation.

Sewage sludge enhances tomato growth and improves fruit-yield quality by restoring soil fertilityOriginal Paper

Among the various disposal strategies for sewage sludge (SS), soil application is the most suitable. This study was conducted to evaluate soil amendment with SS (0, 10, 20, 30 and 40 g/kg) and its impact on soil fertility and tomato (Solanum lycopersicum L.) growth. The SS significantly improved the agromorphological attributes, the number of produced fruits, and the fruit biomass of tomato plants. The 30 g/kg application of SS led to the highest growth rate and fruit yield. Considering the fruits, the best safe enrichment of metal nutrients was recorded at 30 g/kg, with a significant increase in the micronutrient metals Mn, Zn, Ni, Cu, and Fe with 624, 193, 125, 70, and 32%, respectively, compared to the control. The SS amendment enhanced soil fertility, and heavy metals were within the permissible ranges for agricultural soils. Bioaccumulation factors (BFs) indicated that SS application induced the accumulation of most of the studied metals in the roots, and the BF values of Zn, Cu, Ni, and Pb were > 1. The current study concluded that recirculating SS nutrient components to agricultural soils could offer a valid solution for the sustainable management of this organic waste and enhance plant-crop productivity.

Pedogenesis problems on reclaimed coal mining sitesReview

Open-cast coal mining presents a big global issue because of the large areas the mines occupy, which get entirely changed. Their ecosystems lose most of their functions, and a huge amount of fertile soil gets utterly destroyed. Reclamation is a process of returning the functions of the soil after the excavation is finished, most commonly achieved by establishing vegetation, which can sometimes be very difficult. This happens due to the physical, chemical and biological changes that occur on these sites, which are described in this paper. Also, some directions for mitigating these problems are given. Once the vegetation is successfully introduced, natural cycles that were compromised by the mining are established once again, and the process of soil formation begins. Some trends and problems related to pedogenesis research on reclaimed mine sites are presented and discussed, along with presumptions of how the process of soil formation evolves on afforested clayey Technosols of central Europe. The potential future research which would confirm these presumptions is discussed, with the emphasis on the need of research performed on older reclamation sites, as well as sites with similar ecological conditions and different tree species cover.

Observed changes in the agroclimatic zones in the Czech Republic between 1961 and 2019Original Paper

The paper shows a large-scale shift in agroclimatic zones in the territory of the Czech Republic (CR) between 1961 and 2019. The method used for agroclimatic zoning took advantage of high-resolution (0.5 km × 0.5 km) daily climate data collected from 268 climatological and 787 rain-gauge stations. The climate information was combined with soil and terrain data at the same resolution. The set of seven agroclimatic indicators allowed us to estimate rates of changes in agroclimatic conditions over the 1961-2019 period, including changes in the air temperature regime, global radiation, drought, frost risks and snow cover occurrence. These indicators are relevant for all main crops and agroclimatic zoning and account for local soil and slope conditions. The study clearly highlights major shifts in the type and extent of agroclimatic zones between 1961-2000 and 2000-2019, which led to the occurrence of entirely new combinations of agroclimatic indicators.

Prenatal effects of red and blue light on physiological and behavioural parameters of broiler chickensOriginal Paper

Light during incubation can influence embryonic and postembryonic development of chickens, but the underlying mechanisms are poorly understood. Previous studies have demonstrated that red and blue lights during incubation had opposite effects on the development of embryonic melatonin biosynthesis; red light results in the highest and blue light in the lowest amplitude of the daily rhythm. Therefore, in this study, we investigated if exposure to monochromatic red (632 nm) and blue (463 nm) light during incubation can differently influence growth, selected biochemical (glucose, cholesterol, triacylglycerols) and endocrine (corticosterone and thyroid hormones) traits and behavioural parameters during postembryonic development in broiler chickens. For analysis, we used 10 and 11 hatchlings incubated in red and blue light, respectively and 10 birds per each group (six males and four females) in 3-weeks-old broilers. During the rapid growth phase (days 18, 20 and 21 of age), higher body weight was recorded in broilers incubated under red compared to blue light, whereas endocrine and metabolic traits did not differ between the treatments. The improved growth rate was related to behavioural traits, mainly because chickens incubated in red light exhibited more passive (resting, standing, preening, dust bathing) and less active behaviours (walking, foraging, fighting, wing-flapping) than the blue-light incubated birds. The time spent for eating and drinking and the results of the tonic immobility test did not differ between both groups. Our results suggest that red and blue monochromatic light during incubation can differently program the postembryonic development of broilers, with possible consequences for their growth and welfare.
